Single-Cell Photoacoustic Microrheology.

Rheological properties, such as elasticity and viscosity, are fundamental biomechanical parameters that are related to the function and pathological status of cells and tissues. In this paper, an innovative photoacoustic microrheology (PAMR), which utilized the time and phase characteristics of photoacoustic (PA) response, was proposed to extract elastic modulus and viscosity. The feasibility and accuracy of the method were validated by tissue-mimicking agar-gelatin phantoms with various viscoelasticity values. PAMR realized single cell elasticity and viscosity mappings on the adipocyte and my

Quantitative viscoelastic assessment of copolymer-in-oil tissue-mimicking phantoms using plane-wave and ARFI-induced shear wave elastography.

Tissue-mimicking phantoms play a crucial role in evaluating new imaging modalities, simulating clinical scenarios, and ensuring the quality and reliability of imaging systems. Styrene-ethylene/butylene-styrene (SEBS) is an extremely stable and versatile material that has been recently used in ultrasound, elastography, and photoacoustic imaging across diverse applications. Despite its extensive use, a systematic viscoelastic characterization of different SEBS-based formulations reported in the literature, particularly using shear wave elastography (SWE), remains limited. To address this gap, th

Cross-plane shear wave elastography for viscoelasticity imaging.

Objective.Shear wave elastography (SWE) is widely used for elasticity imaging, but conventional implementations remain confined to two-dimensional (2D) measurement planes and lack sensitivity to viscosity, an emerging biomarker linking shear wave dissipation and disease progression. The objective of this work is to extend SWE for viscoelasticity imaging beyond the measurement plane through methodological and acquisition-based advances.Approach.We introduce a cross-plane acquisition strategy in which multiple measurement planes are recorded per acoustic radiation force push to capture dissipati

[New Diagnostic Imaging Technique - Shear Wave Elastography].

Shear wave elastography (SWE) is a new non-invasive diagnostic imaging technique, that maps the elastic properties of tissues. Nowadays this modality develops increasingly in medicine across its disciplines and opens a new era of high-quality ultrasound examination because it increases the specificity and thus improves diagnostic assurance. This method is similar to manual palpation, shows elastic properties of biological tissues and provides a kind of reconstruction of the internal structure of soft tissues based on measurement of the response of tissue compression. Various biological tissues

A noninvasive surface wave technique for measuring finger's skin stiffness.

The purpose of this work was to develop a compact surface wave elastography (CSWE) device for measuring finger's skin stiffness. The motivation was to develop a noninvasive technique for assessing limited cutaneous systemic sclerosis (lcSSc) in accordance with new ACR/EULAR clarification criteria. Currently, the Modified Rodnan Skin Score (MRSS) is widely used for assessing systemic sclerosis but is challenging for assessing patients with lcSSc. The novelty of CSWE is to develop a noninvasive technique to measure the elastic properties of skin of fingers. In the CSWE device, a local harmonic v

Factors influencing the Shear Wave Elastography evaluation of the patellar tendon.

AIM: There is no clear standardization for tendon Shear Wave Elastography (SWE), and data regarding the factors that influence the correctness of evaluation are scarce. We aimed to determine the intra- and interobserver agreement in patellar tendon SWE, and to establish the influence of various factors on elasticity values. MATERIALS AND METHODS: We recruited 37 healthy volunteers; SWE of the patellar tendon was performed by two examiners. The following factors were analyzed: probe frequency, degree of joint flexion, size of region of interest (ROI), distance of the color box from the probe f

Volumetric imaging of Young's modulus and nonlinear elasticity on the micro-scale.

Compression optical coherence elastography (OCE) provides micro-scale imaging of tissue and biomaterial mechanical properties in three dimensions (3-D), showing promise in a range of biomedical applications. Despite substantial progress, most 3-D OCE techniques do not account for the nonlinear elastic response of tissue, resulting in deformation-dependent variations in estimated Young's modulus within an image that reduce reliability and confound comparison both within and between samples. In this work, we overcome these limitations by explicitly accounting for sample deformation and present t

Establishing an Elastography calibration standard: Validation of a shear wave TOF device for measuring Elasticity and Viscosity in tissue-mimicking phantoms using rheometry.

This study designed a novel shear wave Time of Flight (TOF) device to measure frequency-dependent shear wave velocity in tissue-mimicking materials, from which viscoelastic parameters were estimated through Kelvin-Voigt fractional derivative modeling to establish a reliable calibration standard. Tissue-mimicking phantoms were fabricated using 10 wt% polyvinyl alcohol (PVA) and 2 wt% α-alumina powder, with mechanical properties modulated through freeze-thaw cycling. Bimorph transducers operating in the 40-180 Hz range induced and captured shear waves. A single-cycle sine wave excitation ensures

Physics-Informed Neural Networks for Tissue Elasticity Reconstruction in Magnetic Resonance Elastography.

Magnetic resonance elastography (MRE) is a medical imaging modality that non-invasively quantifies tissue stiffness (elasticity) and is commonly used for diagnosing liver fibrosis. Constructing an elasticity map of tissue requires solving an inverse problem involving a partial differential equation (PDE). Current numerical techniques to solve the inverse problem are noise-sensitive and require explicit specification of physical relationships. In this work, we apply physics-informed neural networks to solve the inverse problem of tissue elasticity reconstruction. Our method does not rely on num

A Theoretical Approach in Applying High-Frequency Acoustic and Elasticity Microscopy to Assess Cells and Tissues.

Medical ultrasound is a diagnostic imaging modality used for visualizing internal organs; the frequencies typically used are 2-10 MHz. Scanning acoustic microscopy (SAM) is a form of ultrasound where frequencies typically exceed 50 MHz. Increasing the acoustic frequency increases the specimen's spatial resolution but reduces the imaging depth. The advantages of using SAM over conventional light and electron microscopy include the ability to image cells and tissues without any preparation that could kill or alter them, providing a more accurate representation of the specimen. After scanning the

Soft elastic hydrogel couplants for ultrasonography.

Couplants play significant roles in ultrasonography. To ensure imaging quality, it is critical to maintain conformal contact of the couplant with both the skin surface and the ultrasound probe in clinical applications. In addition, either the probe or the couplant should not deform the skin surface significantly, which will result in an overestimated modulus of the tissue for elastography imaging. However, existing liquid gel couplants cannot bear external compressive force, while existing solid gel couplants cannot maintain a conformal contact with skin surface. Especially, the nonconformal c

Comparison of three cut-offs to diagnose clinically significant portal hypertension by liver stiffness in chronic viral liver diseases: a meta-analysis.

BACKGROUND: Transient elastography-based liver stiffness value (TE-LSV) has been investigated for assessing clinically significant portal hypertension (CSPH). The aetiology of CSPH is an important factor determining TE-LSV. There is insufficient evidence for selecting cut-off values. AIMS: This study performed a meta-analysis to compare the three most widely used cut-off values (around 13.6 kPa, 18 kPa and 22kPa) of TE-LSV for the diagnosis of CSPH in patients with chronic viral liver disease. METHODS: The PubMed, Ovid, Web of Science and Cochrane Library databases were searched. Diagnostic

Nonlinear fourth-order elastic characterization of the cornea using torsional wave elastography.

Measuring the mechanical nonlinear properties of the cornea remains challenging due to the lack of consensus in the methodology and in the models that effectively predict its behaviour. This study proposed developing a procedure to reconstruct nonlinear fourth-order elastic properties of the cornea based on a mathematical model derived from the theory of Hamilton et al. and using the torsional wave elastography (TWE) technique. In order to validate its diagnostic capability of simulated pathological conditions, two different groups were studied, non-treated cornea samples (n=7), and ammonium h

[New trends in ultrasound diagnostics of carotid stenoses].

The B-mode image quality of vessels has markedly increased with the advent of new transducer technologies. Morphological changes of the vessel wall, ulcerations, dissections or wall lesions benefit from non-Doppler dependent flow imaging methods. Two-dimensional imaging of the elasticity of the vessel wall may in future benefit from high speed imaging techniques. The biggest improvement in vessel imaging is probably the prudent use of contrast-enhanced ultrasound by which neovascularization can be visualized. Plaque vascularization is looked upon as an important risk factor in the evaluation o

Assessment of the uncertainty of shear wave speed measurements in ultrasound elastography.

Accurate measurement of tissue mechanical properties is crucial for diagnosing and characterising various pathological conditions. Among these properties, shear wave speed (SWS) is an indicator of tissue stiffness and has been widely studied across multiple imaging modalities. Despite its advantages, the quantification of SWS is subject to various sources of uncertainty, which can impact its clinical and research applications. The uncertainty of SWS measurements based on acoustic radiation force impulse (ARFI) technology implemented using a research ultrasound system is investigated in this st

Quantitative breast elastography from B-mode images.

PURPOSE: Elastography images provide information about the mechanical properties of soft tissue in a noninvasive way and can be useful to identify abnormalities and ascertain differential diagnoses of suspicious prior findings obtained through mammography ultrasound. In this work we investigate, from a physics point of view, the feasibility of quantifying the Young's modulus of breast tissue from the autocorrelation of a diffuse acoustic field computed from a sequence of B-mode images acquired through conventional ultrasound scanners. METHODS: Inspired in the seismological approach of retriev

In vivo non-contact measurement of human iris elasticity by optical coherence elastography.

Quantifying the mechanical properties of the iris can offer valuable insights into the pathophysiology of primary angle closure glaucoma. However, current techniques for iris elastography remain ex vivo with limited clinical applications. This article describes a proposition for a non-contact and non-invasive air-puff optical coherence elastography (OCE) system that can evaluate iris elasticity in vivo. Ten eyes recruited from seven subjects underwent OCE imaging acquisition under three different illumination conditions. The Young's modulus of each eye was detected and shown to be inversely pr
