Detecting salient visual objects and orienting toward them are commonplace tasks for animals, yet the underlying neural circuit mechanisms remain poorly understood. The fruit fly is an ideal model for a comprehensive analysis of feature detection mechanisms given its complete synaptic wiring diagrams, robust behavioral assays, and cell-type-specific gene expression datasets. We previously showed that columnar visual neurons T3 are required for saccadic orientation toward landscape features during flight. Here, we examine how signals downstream of T3 are processed in the central brain. We ident

Sleep supports memory formation by neurophysiological mechanisms that are yet to be fully uncovered. We investigated the contribution of the direct coupling of neurons via electrical synapses (gap junctions). The administration of mefloquine (250 mg p.o. vs. placebo), an antimalarial, which blocks electrical synapses, to healthy young men before nocturnal sleep impaired the retention of word pairs learned before drug administration and disrupted the coupling of sleep spindles to EEG slow oscillations. In control experiments, in which participants received mefloquine before a consolidation inte

The mossy fiber-granule cell-parallel fiber system conveys proprioceptive and corollary discharge information to principal cells in cerebellum-like systems. In the dorsal cochlear nucleus (DCN), Golgi cells inhibit granule cells and thus regulate information transfer along the mossy fiber-granule cell-parallel fiber pathway. Whereas excitatory synaptic inputs to Golgi cells are well understood, inhibitory and electrical synaptic inputs to Golgi cells have not been examined. Using paired recordings in a mouse brain slice preparation, we find that Golgi cells of the cochlear nucleus reliably for

A mixed chemical/electrical synapse can generate variable output when the strength of each synaptic component is modulated. At mixed synapses of the lobster pyloric network, the chemical component is inhibitory. Without neuromodulation, the chemical component is weak or absent and the electrical component often dominates. Dopamine reverses the sign of these mixed synaptic interactions by a reduction in the strength of electrical coupling and an enhancement of chemical inhibition, including activation of silent chemical synapses. Sign reversal at mixed synapses by neuromodulators may contribute

Electrical coupling mediated by gap junctions is a defining feature of almost every neuron in the retina, as it is throughout the central nervous system. By allowing direct current flow between neurons, electrical coupling helps establish functional circuits, allows signals to be shared, and improves the signal-to-noise ratio, thereby regulating neuronal activity at the dendritic, single-neuron, and network levels. Remarkably, electrical coupling is not static or constitutive. It shows a high degree of plasticity, changing in strength on time scales ranging from milliseconds to days, thereby p

Electrical synapses play an important role in signaling between neurons and the synaptic connections between many neurons possess both electrical and chemical components. Although modulation of electrical synapses is frequently observed, the cellular processes that mediate such changes have not been studied as thoroughly as plasticity in chemical synapses. In the leech (Hirudo sp), the competitive AMPA receptor antagonist CNQX inhibited transmission at the rectifying electrical synapse of a mixed glutamatergic/electrical synaptic connection. This CNQX-mediated inhibition of the electrical syna

Slices of rat carotid bodies, or cultured glomus cells, were used to study intercellular coupling. This phenomenon occurs because gap junctions allow passage of currents and dyes from one cell to another. There is a two-way resistive coupling between glomus cells (GC/GC coupling), which is accompanied by activity of intercellular channels. Coupling between glomus cells and nerve endings is more complex. Coupling is mostly resistive from cell to nerve (GC/NE) but it is mostly capacitive in the opposite direction (NE/GC). Thus, slow electric events originating in the glomus cells can be transfer

The post-embryonic development of the rectifying Giant Fibre-Motor Giant (GF-MoG) synapse and the Giant Fibre-Segmental Giant (GF-SG) synapse has been investigated using electron-microscopy. In adults, the MoG and SG neurons make contact with the GFs by sending narrow 'finger-like' processes through the glial and connective tissue sheath surrounding each GF. The junctional region is characterized by closely apposed membranes (approximately 4 nm separation) traversed by regularly spaced connections, and large (60-80 nm) spherical vesicles in the presynaptic fibre. In newly hatched crayfish junc

Opioid receptors were shown to modulate a variety of cellular processes in the vertebrate central nervous system, including synaptic transmission. While the effects of opioid receptors on chemically mediated transmission have been extensively investigated, little is known of their actions on gap junction-mediated electrical synapses. Here we report that pharmacological activation of mu-opioid receptors led to a long-term enhancement of electrical (and glutamatergic) transmission at identifiable mixed synapses on the goldfish Mauthner cells. The effect also required activation of both dopamine

UNLABELLED: Most electrical synapses in the mammalian central nervous system are composed of Connexin 36 (Cx36). Electrical synapses are functionally plastic, changing their degree of coupling based on the activity of the cell or connected cells, or based on activation of neurotransmitter or neurohormone receptors. Plasticity can reach an extreme in which electrical synapses become functionally silent, which is a normal operational condition for some circuits. Cx36 coupling is regulated by phosphorylation, which opens the channels. In retinal circuits, Cx36 is often maintained in a poorly phos

The influences of the snake venom taipoxin on the cholinergic synapses of the electric organ of Torpedo mamorata were investigated. 1. In ultrathin sections presynaptic membrane indentations are noted under the influence of taipoxin/NaCl. 2. The presynaptic membranes often show small interruptions. 3. In taipoxin/horseradish peroxidase incubated organ pieces typical omega-shaped indentations are found, but on the postsynaptic side. 4. The vesicle density decreased under the influence of the taipoxin in contrast untreated cholinergic synapses. 5. In freeze etched preparations no membrane indent

Although hyperactivity is associated with a wide variety of neurodevelopmental disorders, the early embryonic origins of locomotion have hindered investigation of pathogenesis of these debilitating behaviors. The earliest motor output in vertebrate animals is generated by clusters of early-born motor neurons (MNs) that occupy distinct regions of the spinal cord, innervating stereotyped muscle groups. Gap junction electrical synapses drive early spontaneous behavior in zebrafish, prior to the emergence of chemical neurotransmitter networks. We use a genetic model of hyperactivity to gain critic

Electrical and chemical synapses usually reinforce one another, but the pyloric late-to-lateral pyloric (PL-to-LP) neuronal connections in lobster stomatogastric ganglia create an inverted U-shaped transfer function between the two neurons: regardless of whether the PL membrane voltage swings positive or negative, the postsynaptic LP voltage will go negative. When the presynaptic cell voltage goes negative, the effect on the LP voltage is due to electrical coupling. During positive presynaptic voltages, the strong contribution of graded chemical inhibition from the PL to the LP neuron override

Gap junction (GJ) channels have been recognized as an important mechanism for synchronizing neuronal networks. Herein, we investigated the participation of GJ channels in the pilocarpine-induced status epilepticus (SE) by analyzing electrophysiological activity following the blockade of connexins (Cx)-mediated communication. In addition, we examined the regulation of gene expression, protein levels, phosphorylation profile and distribution of neuronal Cx36, Cx45 and glial Cx43 in the rat hippocampus during the acute and latent periods. Electrophysiological recordings revealed that the GJ block

In the present study, we directly demonstrated electrical coupling between Bergmann glial cells (BG) and Purkinje neurones (PN) in acutely isolated cerebellar slices, prepared from 15 to 30 days old Sprague-Dawley rats. Electrical coupling between these two cells was identified by dual whole-cell voltage clamp, which allowed direct recording of junctional current. Whole-cell recordings from PN-PN, PN-BG and BG-BG pairs were made using Nomarski optics and infrared visualisation, which allowed precise morphological identification of cells. Junctional currents were recorded by applying hyper/and

Retinal AII amacrine cells are extensively coupled together by electrical synapses. Changes to the strength of these synapses affect how signals are routed through rod and cone retinal pathways during scotopic and photopic vision. Plasticity at these electrical synapses has not, to date, been characterized using electrophysiological approaches. We investigated the effects of NMDA receptor (NMDAR) activation on electrical coupling between AII cells using dual whole-cell patch-clamp electrophysiology in mouse retinal slices. NMDAR activation substantially decreased junctional conductance between

Initiating and regulating vertebrate reproduction requires pulsatile release of gonadotropin-releasing hormone (GnRH1) from the hypothalamus. Coordinated GnRH1 release, not simply elevated absolute levels, effects the release of pituitary gonadotropins that drive steroid production in the gonads. However, the mechanisms underlying synchronization of GnRH1 neurons are unknown. Control of synchronicity by gap junctions between GnRH1 neurons has been proposed but not previously found. We recorded simultaneously from pairs of transgenically labeled GnRH1 neurons in adult male Astatotilapia burtoni

Rectifying electrical synapses are commonplace, but surprisingly little is known about how rectification alters the dynamics of neuronal networks. In this study, we use computational models to investigate how rectifying electrical synapses change the behavior of a small neuronal network that exhibits complex rhythmic output patterns. We begin with an electrically coupled circuit of three oscillatory neurons with different starting frequencies, and subsequently add two additional neurons and inhibitory chemical synapses. The five-cell model represents a pattern-generating neuronal network with

Electrical signalling across distinct populations of brain cells underpins cognitive and emotional function. However, approaches that selectively regulate electrical signalling between two cellular components of a mammalian neural circuit remain sparse. Here we engineered an electrical synapse composed of two connexin proteins1 found in Morone americana (white perch fish)-connexin 34.7 and connexin 35-to accomplish mammalian circuit modulation. By exploiting protein mutagenesis, devising a new in vitro system for assaying connexin hemichannel docking, and performing computational modelling of

1. Two pairs of neurons in the pyloric network of the spiny lobster, Panulirus interruptus, communicate through mixed graded chemical and rectifying electrical synapses. The anterior burster (AB) chemically inhibits and is electrically coupled to the ventricular dilator (VD); the lateral pyloric (LP) and pyloric (PY) neurons show reciprocal chemical inhibition and electrical coupling. We examined the effects of dopamine (DA), serotonin (5HT) and octopamine (Oct) on these mixed synapses to determine the plasticity possible with opposing modes of synaptic interaction. 2. Dopamine increased net i
