Anchoring structure of the calvarial periosteum revealed by focused ion beam/scanning electron microscope tomography.

An important consideration in regeneration therapy is the fact that the tissue surrounding an organ supports its function. Understanding the structure of the periosteum can contribute to more effective bone regeneration therapy. As a cellular source, the periosteum also assists bone growth and fracture healing; this further necessitates its direct contact with the bone. However, its anchoring strength appears to be inexplicably stronger than expected. In this study, we used focused ion beam/scanning electron microscope tomography to investigate ultrathin serial sections as well as the three di

Three-dimensional ultrastructural analysis of cells in the periodontal ligament using focused ion beam/scanning electron microscope tomography.

The accurate comprehension of normal tissue provides essential data to analyse abnormalities such as disease and regenerative processes. In addition, understanding the proper structure of the target tissue and its microenvironment may facilitate successful novel treatment strategies. Many studies have examined the nature and structure of periodontal ligaments (PDLs); however, the three-dimensional (3D) structure of cells in normal PDLs remains poorly understood. In this study, we used focused ion beam/scanning electron microscope tomography to investigate the whole 3D ultrastructure of PDL cel

Superiority of focused ion beam-scanning electron microscope tomography of cardiomyocytes over standard 2D analyses highlighted by unmasking mitochondrial heterogeneity.

BACKGROUND: Cardioprotection by preventing or repairing mitochondrial damage is an unmet therapeutic need. To understand the role of cardiomyocyte mitochondria in physiopathology, the reliable characterization of the mitochondrial morphology and compartment is pivotal. Previous studies mostly relied on two-dimensional (2D) routine transmission electron microscopy (TEM), thereby neglecting the real three-dimensional (3D) mitochondrial organization. This study aimed to determine whether classical 2D TEM analysis of the cardiomyocyte ultrastructure is sufficient to comprehensively describe the mi

Comparison of lymphoblast mitochondria from normal subjects and patients with Barth syndrome using electron microscopic tomography.

Barth syndrome (BTHS) is a mitochondrial disorder that is caused by mutations in the tafazzin gene, which affects phospholipid composition. To determine whether this defect leads to alterations in the internal three-dimensional organization of mitochondrial membranes, we applied electron microscopic tomography to lymphoblast mitochondria from BTHS patients and controls. Tomograms were formed from 50 and 150 nm sections of chemically fixed lymphoblasts and the data were used to manually segment volumes of relevant structural details. Normal lymphoblast mitochondria contained well-aligned, lamel

Ultrastructural characterization of secretory canals in Peucedanum praeruptorum roots using microscopic sectioning, transmission electron microscopy, and computed tomography.

Plant secondary metabolites are mainly synthesized and stored in secretory tissues. Secretory canal development has been mainly characterized in Apiaceae. The secretory canals of Peucedanum praeruptorum contain pharmacologically active coumarins, but their organ-specific distribution and developmental dynamics remain poorly understood. This study integrated light microscopy (LM), transmission electron microscopy (TEM), X-ray microcomputed tomography (µ-CT), and high-performance liquid chromatography (HPLC) to investigate canal development, distribution, ultrastructure, 3D architecture, and cou

The synaptic organization of the human temporal lobe neocortex by high-resolution transmission, focused ion beam scanning, and electron microscopic tomography.

Fine-scale transmission electron microscopy (TEM), focused ion beam scanning EM (FIB), and EM tomography have opened a new window on the synaptic organization of the normal, developing and pathologically altered brain in experimental animals. Progress in the human brain has been slower, due to technical challenges and the problem of tissue availability from donors that underwent epilepsy or tumor surgery. The present manuscript is in part an overview of the geometry of synaptic boutons in surgical biopsy samples taken from human temporal lobe neocortex ('hTLN'). Here, the number, size, and sha

Zika virus replication in glioblastoma cells: electron microscopic tomography shows 3D arrangement of endoplasmic reticulum, replication organelles, and viral ribonucleoproteins.

Structural changes of two patient-derived glioblastoma cell lines after Zika virus infection were investigated using scanning transmission electron tomography on high-pressure-frozen, freeze-substituted samples. In Zika-virus-infected cells, Golgi structures were barely visible under an electron microscope, and viral factories appeared. The cytosol outside of the viral factories resembled the cytosol of uninfected cells. The viral factories contained largely deranged endoplasmic reticulum (ER), filled with many so-called replication organelles consisting of a luminal vesicle surrounded by the

Biological aspects of the nasal turbinates of the Anatolian shepherd dog captured from Egypt: Using computed tomography, histological, and scanning electron microscopic observations.

The current study was designed to describe the nasal turbinates of 15 heads of Anatolian shepherd dogs using the histology and scanning electron microscope. The caudal part of the nasal cavity is almost occupied by the ethmoidal concha that is related to the high dog's smelling. Keratinized stratified squamous epithelial lining of the rostral part of dorsal and ventral concha were interdigitated with the underlying lamina propria, with numerous sebaceous and sweat glands. The pseudostratified squamous epithelium lining of the middle part of the dorsal and ventral conchae had simple seromucous

Cryo-iCLEM: Cryo correlative light and electron microscopy with immersion objectives.

Correlative light and electron microscopy (CLEM) is a powerful tool for investigating cellular structure and function at the molecular level. However, while electron microscopy is often performed to great advantage at cryogenic temperatures, this is not always the case for light microscopy. One key challenge is the lack of cryo-compatible immersion objectives. In recent years, multiple cryoimmersion light microscopy (cryo-iLM) approaches have been described, but these techniques have never been used in correlative approaches. Here we present a novel workflow for correlative cryoimmersion light

Cryo-Electron Tomography of Toxoplasma gondii Indicates That the Conoid Fiber May Be Derived from Microtubules.

Toxoplasma gondii (T. gondii) is the causative agent of toxoplasmosis and can infect numerous warm-blooded animals. An improved understanding of the fine structure of this parasite can help elucidate its replication mechanism. Previous studies have resolved the ultrastructure of the cytoskeleton using purified samples, which eliminates their cellular context. Here the application of cryo-electron tomography to visualize T. gondii tachyzoites in their native state is reported. The fine structure and cellular distribution of the cytoskeleton are resolved and analyzed at nanometer resolution. Add

Controlling electron beam-induced structure modifications and cation exchange in cadmium sulfide-copper sulfide heterostructured nanorods.

The atomic structure and interfaces of CdS/Cu2S heterostructured nanorods are investigated with the aberration-corrected TEAM 0.5 electron microscope operated at 80 kV and 300 kV applying in-line holography and complementary techniques. Cu2S exhibits a low-chalcocite structure in pristine CdS/Cu2S nanorods. Under electron beam irradiation the Cu2S phase transforms into a high-chalcocite phase while the CdS phase maintains its wurtzite structure. Time-resolved experiments reveal that Cu(+)-Cd(2+) cation exchange at the CdS/Cu2S interfaces is stimulated by the electron beam and proceeds within a

Applications of emerging transmission electron microscopy technology in PCD research and diagnosis.

Primary Ciliary Dyskinesia (PCD) is a heterogeneous genetic condition characterized by dysfunction of motile cilia. Patients suffer from chronic infection and inflammation of the upper and lower respiratory tract. Diagnosis of PCD is confirmed by identification of a hallmark defect of ciliary ultrastructure or by identification of biallelic pathogenic mutations in a known PCD gene. Since the first description of PCD in 1976, assessment of ciliary ultrastructure by transmission electron microscopy (TEM) has been central to diagnosis and research. Electron tomography is a technique whereby a ser

Cryo-electron tomography reveals the binding and release states of the major adhesion complex from Mycoplasma genitalium.

The nap particle is an immunogenic surface adhesion complex from Mycoplasma genitalium. It is essential for motility and responsible for binding sialylated oligosaccharides on the surface of the host cell. The nap particle is composed of two P140-P110 heterodimers, the structure of which was recently solved. However, the interpretation of the mechanism by which the mycoplasma cells orchestrate adhesion remained challenging. Here, we provide cryo-electron tomography structures at ~11 Å resolution, which allow for the distinction between the bound and released state of the nap particle, displayi

Diffraction contrast in cryo-scanning transmission electron tomography reveals the boundary of hemozoin crystals in situ.

Malaria is a potentially fatal infectious disease caused by the obligate intracellular parasite Plasmodium falciparum. The parasite infects human red blood cells (RBC) and derives nutrition by catabolism of hemoglobin. As amino acids are assimilated from the protein component, the toxic heme is released. Molecular heme is detoxified by rapid sequestration to physiologically insoluble hemozoin crystals within the parasite's digestive vacuole (DV). Common antimalarial drugs interfere with this crystallization process, leaving the parasites vulnerable to the by-product of their own metabolism. A

Changes of arachnoid granulations after subarachnoid hemorrhage in cynomolgus monkeys.

This research explores ultrastructural changes of arachnoid granulations associated with hydrocephalus after subarachnoid hemorrhage in cynomolgus monkeys. It provides a theoretical basis for further study of the etiology and prevention of hydrocephalus. Female cynomolgus monkeys about one-year-old were selected. The position range of arachnoid granulations in superior sagittal sinus and transverse sinus was determined in a randomly selected control monkey. The morphology of normal arachnoid granulations in cynomolgus monkeys was observed under a transmission electron microscope. A primate mod

Ultrastructure and Nanoporosity of Human Bone Shown with Correlative On-Axis Electron and Spectroscopic Tomographies.

Mineralized collagen fibrils are the building block units of bone at the nanoscale. While it is known that collagen fibrils are mineralized both inside their gap zones (intra-fibrillar mineralization) and on their outer surfaces (extra-fibrillar mineralization), a clear visualization of this architecture in three dimensions (3D), combining structural and compositional information over large volumes, but without compromising the resolution, remains challenging. In this study, we demonstrate the use of on-axis Z-contrast electron tomography (ET) with correlative energy-dispersive X-ray spectrosc

FPGA-based scanner and SerialEM server for 4D-STEM electron tomography.

Four-dimensional scanning transmission electron microscopy (4D-STEM) enables the acquisition of diffraction patterns at every probe position in a dense array. For imaging applications this approach offers significant benefits in terms of spatial resolution and contrast enhancement. In this work, we present an update on our open SavvyScan platform, consisting of a synchronous scan generator integrated by software with SerialEM to enable the automation of complex experimental protocols such as tomography. The proposed hardware functions as an interface between SerialEM, the scan controls of the

Protocol for high-throughput electron tomography exemplified on centrioles in primary human plasma cells.

Electron microscopy is the gold standard to characterize cellular ultrastructure. However, production of significant morphometrical data is highly limited by acquisition time. Here, we describe a semi-automated high-throughput strategy using single-axis serial section electron tomography to investigate and analyze centriole ultrastructure in bone-marrow-derived, primary human CD138pos plasma cells. The protocol comprises steps for electron microscopy sample preparation, semi-automated transmission electron microscopy screening, and screening evaluation for cells of interest. Thereafter, we det

Cryo-tomography reveals rigid-body motion and organization of apicomplexan invasion machinery.

The apical complex is a specialized collection of cytoskeletal and secretory machinery in apicomplexan parasites, which include the pathogens that cause malaria and toxoplasmosis. Its structure and mechanism of motion are poorly understood. We used cryo-FIB-milling and cryo-electron tomography to visualize the 3D-structure of the apical complex in its protruded and retracted states. Averages of conoid-fibers revealed their polarity and unusual nine-protofilament arrangement with associated proteins connecting and likely stabilizing the fibers. Neither the structure of the conoid-fibers nor the
