Quantitative analysis of chlorpromazine by electron spin resonance (ESR) spectroscopy.

A simple and sensitive method is described for quantitative analysis of chlorpromazine in blood, serum, urine and tissue homogenate. The chlorpromazine cation radical produced by adding perchloric acid and 2,3-dichloro-5,6-dicyano-p-benzoquinone to the sample can be detected by the ESR method at room temperature. The sensitivity limit is 10 ng, that is, 20 microliters of the solution containing 0.5 microgram chlorpromazine/ml. The time needed for the measurement is within 10 min. The chlorpromazine radical thus produced is very stable; for example, 95% of the radical was observed after 24 h. T

The design of spin probes for electron magnetic resonance spectroscopy and imaging.

The problems that have to be addressed in designing suitable spin probes are manifold. In this article it will be shown that some the properties the probes need to possess are (i) a simple EMR spectrum (in some types of imaging or oximetry a single line is desirable), (ii) sharp lines with no unresolved splittings, to provide maximum sensitivity to detection, (iii) chemical, metabolic and thermal stability, (iv) easy possible synthesis to permit substitution with isotopes, prosthetic groups or groups to control the partition coefficients in lipid/water systems, and (v) electronic anisotropy, i

Electron spin resonance in scanning probe microscopy: From single-spin spectroscopy to atomically engineered quantum operations.

Electron spin resonance (ESR) is one of the most powerful spectroscopic methods for probing systems with unpaired electronic spins. Conventional ESR has usually relied on the detection of large spin ensembles, thereby averaging over local environments and suppressing microscopic information that is central to surface science, molecular magnetism, and quantum nanoscience. Scanning probe microscopy (SPM) changes this situation. By combining magnetic resonance with the spatial resolution and manipulation capabilities of scanning tunnelling microscopy (STM) and atomic force microscopy (AFM), ESR c

Electron spin resonance spectroscopy, exercise, and oxidative stress: an ascorbic acid intervention study.

Oxygen free radicals are highly reactive species that are produced in increased quantities during strenuous exercise and can damage critical biological targets such as membrane phospholipids. The present study examined the effect of acute ascorbic acid supplementation on exercise-induced free radical production in healthy subjects. Results demonstrate increases in the intensity of the alpha-phenyl-tert-butylnitrone adduct (0.05 +/- 0.02 preexercise vs. 0.19 +/- 0.03 postexercise, P = 0.002, arbitrary units) together with increased lipid hydroperoxides (1.14 +/- 0.06 micromol/l preexercise vs.

[Use of electron paramagnetic resonance spectroscopy spin labels in studying the properties of biological membranes].

Electron paramagnetic resonance (EPR) spectroscopy of spin labels has become a widely spread method in biochemistry, molecular biology, and medicine. After a brief view on the structure and functions of biological membranes, basic principles of EPR and particularly of the spin labeling technique are presented. Applications of this technique are demonstrated on the study of various properties of biological membranes, such as phase transition of lipids, properties of different membrane parts, permeability of membranes, inner volumes of liposomes and cells, membrane potentials, lateral diffusion,

Location of the cross-β structure in prion fibrils: A search by seeding and electron spin resonance spectroscopy.

Prion diseases are transmissible fatal neurodegenerative disorders spreading between humans and other mammals. The pathogenic agent, prion, is a protease-resistant, β-sheet-rich protein aggregate, converted from a membrane protein called PrPC . PrPSc is the misfolded form of PrPC and undergoes self-propagation to form the infectious amyloids. Since the key hallmark of prion disease is amyloid formation, identifying and studying which segments are involved in the amyloid core can provide molecular details about prion diseases. It has been known that the prion protein could also form non-infecti

Exploring Structure, Dynamics, and Topology of Nitroxide Spin-Labeled Proteins Using Continuous-Wave Electron Paramagnetic Resonance Spectroscopy.

Structural and dynamical characterization of proteins is of central importance in understanding the mechanisms underlying their biological functions. Site-directed spin labeling (SDSL) combined with continuous-wave electron paramagnetic resonance (CW EPR) spectroscopy has shown the capability of providing this information with site-specific resolution under physiological conditions for proteins of any degree of complexity, including those associated with membranes. This chapter introduces methods commonly employed for SDSL and describes selected CW EPR-based methods that can be applied to (1)

Use of spin traps to detect superoxide production in living cells by electron paramagnetic resonance (EPR) spectroscopy.

Detection of superoxide produced by living cells has been an on-going challenge in biology for over forty years. Various methods have been proposed to address this issue, among which spin trapping with cyclic nitrones coupled to EPR spectroscopy, the gold standard for detection of radicals. This technique is based on the nucleophilic addition of superoxide to a diamagnetic cyclic nitrone, referred to as the spin trap, and the formation of a spin adduct, i.e. a persistent radical with a characteristic EPR spectrum. The first application of spin trapping to living cells dates back 1979. Since th

Measurement of radical-scavenging ability in hepatic metallothionein of rat using in vivo electron spin resonance spectroscopy.

In this study, the ability of metallothionein (MT) to scavenge free radicals was determined by in vivo electron spin resonance (ESR) spectroscopy using a carbamoyl-PROXYL, nitroxyl radical, as a spin probe. Production of metallothionein was induced in the liver of rats with ZnSO(4) (0.2 mol/kg, ip) and the intensity of the carbamoyl-PROXYL ESR signal was measured at the upper abdominal level which is a position of the liver. After the injection of carbamoyl-PROXYL, the peak of ESR signal gradually decreased and showed a linear decay curve. The rate of decay of carbamoyl-PROXYL, the spin cleara

Distribution profiles of nitroxide spin probes in human skin--a combined study using spatially resolved electron spin resonance spectroscopy and mass spectrometry.

Electron spin resonance spectroscopy and mass spectrometry are two analytical methods that are very rarely used in combination. In this paper, we will show that the methods complement one another in the example of the distribution of stable nitroxide radicals in human skin, including the spatial resolution of these distribution processes. There are many ESR investigations dealing with this subject, but unfortunately, they are all limited to the detection of paramagnetic species. The combination with MS allows the successful examination of the distribution profile of the main biotransformation

Studies on electron spin resonance spectroscopy of biradical molecules containing (14)N-O and (15)N-O moieties.

The biradicals with (14)N-Oxide and (15)N-Oxide at the both ends of a molecule are synthesized for the molecular ruler of protein structure, and a potential device for quantum computing. We also establish a general synthetic method for reliable biradical formation. ESR spectra are recorded for the biradicals containing (15)N-Oxide and (14)N-Oxide with various interdistance separations. We find that two types of biradicals yielded different ESR spectra depending upon the distance between the (15)N-O and (14)N-O moieties in a molecule. This is due to electron spin dipole-dipole interaction occur

Hydroxyl radical formation in chondrocytes and cartilage as detected by electron paramagnetic resonance spectroscopy using spin trapping reagents.

Chondrocytes have been shown to produce superoxide and hydrogen peroxide, suggesting possible formation of hydroxyl radical in these cells. In this study, we used electron spin resonance/spin trapping technique to detect hydroxyl radicals in chondrocytes. We found that hydroxyl radicals could be detected as alpha-hydroxyethyl spin trapped adduct of 4-pyridyl 1-oxide N-tert-butylnitrone (4-POBN) in chondrocytes stimulated with phorbol 12-myristate 13-acetate in the presence of ferrous ion. The formation of hydroxyl radical appears to be mediated by the transition metal-catalyzed Haber-Weiss rea

Effects of aspirin, indomethacin, and sodium salicylate on human erythrocyte membranes as detected with electron spin resonance spectroscopy.

Electron spin resonance spectroscopy of probed samples was used to determine the structural changes in human erythrocyte membranes prior to and at intervals following ingestion of either 10 grains acetylsalicylic acid, 10 grains sodium salicylate, or 50 mg indomethacin by both male and female subjects. Analysis of erythrocytes from female subjects indicated a time-dependent disordering of the membrane over the eight hour period following aspirin ingestion while the cells of male subjects showed a slight membrane ordering over the same time period. Erythrocytes drawn from females at the beginni

Effect of anaesthesia-induced alterations in haemodynamics on in vivo kinetics of nitroxyl probes in electron spin resonance spectroscopy.

Although the advent of in vivo electron spin resonance (ESR) spectroscopy has allowed analysis of the redox status of living animals, whether the haemodynamic condition affects the signal decay rate remains unknown. Three kinds of haemodynamic conditions were generated by changing the anaesthetic dosage in mice. Haemodynamics was analysed (n=6 each) and in vivo ESR was performed to measure the signal decay rates of three nitroxyl spin probes (carbamoyl-, carboxy- and methoxycarbonyl-PROXYL) at the chest and head regions (n=6 for each condition and probe). Haemodynamic analysis revealed negativ

Detection of weak metal ion binding to cutinase-like enzyme using native mass spectrometry and electron spin resonance spectroscopy.

Weak metal ion binding to enzymes is often necessary for enzyme function; however, its detection remains difficult. The catalytic efficiency of Cut190, a cutinase-like enzyme, is higher with the weak binder Ca2+ than with the strong binder Mn2+. Upon the introduction of a disulfide bond into Cut190, the enzyme was stabilized and effectively degraded polyethylene terephthalate. Notably, Ca2+ binding to the stabilized Cut190 mutant remained weak and was necessary for its function. Here, we demonstrate that native mass spectrometry and electron spin resonance spectroscopy are effective methods fo

Increasing nitroxide lifetime in cells to enable in-cell protein structure and dynamics measurements by electron spin resonance spectroscopy.

There is increasing evidence that the stability, structure, dynamics, and function of many proteins differ in cells versus in vitro. The determination of protein structure and dynamics within the native cellular environment may lead to better understanding of protein behavior. Electron spin resonance (ESR) has emerged as a technique that can report on protein structure and dynamics within cells. Nitroxide based spin labels are capable of reporting on protein dynamics, structure, and backbone flexibility but are limited due to nitroxide reduction occurring in cells. In order to overcome this li

Characterization of lipid oxidation process of beef during repeated freeze-thaw by electron spin resonance technology and Raman spectroscopy.

In this study, electron spin resonance (ESR) and Raman spectroscopy were applied to characterize lipid oxidation of beef during repeated freeze-thaw (RFT). Besides the conventional indexes including peroxide values (PV), thiobarbituric acid-reactive substances (TBARS) and acid values (AV) were evaluated, the radical and molecular structure changes were also measured by ESR and Raman spectroscopy. The results showed that PV, TBARS and AV were increased (P<0.05) after RFT. This suggested that lipid oxidation was occurred during RFT. With the increase of radical signal intensity, lower oxidation

Combining Drosophila melanogaster somatic-mutation-recombination and electron-spin-resonance-spectroscopy data to interpret epidemiologic observations on chromium carcinogenicity.

Lung cancers are significantly increased among workers exposed to chromate (Cr6+, Cr3+), chromium pigments (Cr6+) and chromium plating (Cr6+). Chromium lung burdens and cancer risk increase proportionately with duration of employment at long latencies. However, this epidemiologic information alone is insufficient in determining whether Cr6+ or Cr3+ are equally important in causing cancer. We have attempted to combine epidemiologic data with data from the Drosophila melanogaster somatic-mutation-recombination-test and from the in vitro electron-spin-resonance spectroscopy study to demonstrate t
