On-capillary fluorescent labeling of saccharides for capillary electrophoresis.

The feasibility of on-capillary derivatization of saccharides by aromatic amine-based fluorescent labeling agents was tested. To avoid the problematic evolution of gaseous hydrogen cyanide, the Schiff base reduction by sodium cyanoborohydride, as the second step of the standard reductive amination protocol, was omitted. Glucose was used as a model analyte and 7-amino-1,3-naphthalenedisulfonic acid as the labeling agent. Our experiments showed that the direct reaction of the saccharide with the labeling agent in 2.5-M acetic acid yields a labeled product that is sufficiently stable to be separa

Micro-injector for capillary electrophoresis.

A novel micro-injector for capillary electrophoresis for the handling of samples with volumes down to as little as 300 nL was designed and built in our laboratory for analyses in which the available volume is a limitation. The sample is placed into a small cavity located directly in front of the separation capillary, and the injection is then carried out automatically by controlled pressurization of the chamber with compressed air. The system also allows automated flushing of the injection chamber as well as of the capillary. In a trial with a capillary electrophoresis system with contactless

Myoglobinuria detection by capillary electrophoresis.

Capillary electrophoresis was used in this study to separate urinary myoglobin from hemoglobin based on its electrophoretic mobility. Urine was applied directly without any treatment. The separation was accomplished in less than 7 min. Myoglobin extracted from human muscle tissues was separated, in a borate buffer 150 mM, pH 8.7 containing 0.5% polyethyleneglycol at 6 kV, into two peaks (MI and MII) which were also resolved far from hemoglobin. Upon standing at room temperature, MII converted into MI. Horse myoglobin eluted close to MI. The addition of polyethyleneglycol to the buffer enhanced

High-Throughput Multi-Capillary Electrophoresis.

The potential of multi-capillary electrophoresis (CE) for high-throughput analysis of legacy CE applications was investigated by optimisation of the separation of six pharmaceutical compounds on a single-capillary instrument, followed by transference of the method to an eight-capillary BioPhase 8800 system from SCIEX. Multi-CE had superior performance, allowing construction of calibration curves in a single run, and a five-fold reduction of run time per sample, improved linearity, accuracy and limits of detection. Method transfer from the single-capillary system required minor instrumental par

Comparing capillary coatings for protein separation by capillary electrophoresis.

For intact protein separation by capillary electrophoresis, the application of a proper capillary coating is crucial to achieve high separation efficiencies. There are a large number of different capillary coatings and coating protocols, making it crucial to compare the coating performances in identical experimental conditions for intact protein separations. Here, the plate height vs. migration velocity was systematically plotted by doing electrophoretic experiments at different electric voltages, on five model proteins. For the first time, this study allows comparing the performances of seven

Analysis of agrochemicals by capillary electrophoresis.

An increasing amount of articles using capillary electrophoresis as an investigation tool for pesticides and environmental pollutants were found over the last few years in analytical chemistry oriented journals. This review covers a wide literature range of the 1990s and concentrates on the analysis of organic agrochemicals (herbicides, fungicides, insecticides, acaricides, etc.) with capillary electrophoresis (capillary zone electrophoresis, micellar electrokinetic chromatography with CE-UV-visible or laser-induced fluorescence detection) as well as with the on-coming hyphenated techniques li

Alcohols and wide-bore capillaries in nonaqueous capillary electrophoresis.

The feasibility of using C1-C5 alcohols as electrolyte solutions in nonaqueous capillary zone electrophoresis was investigated. The separation of basic narcotic analgesics and acidic diuretics was modified by changing the alcohol in an electrolyte solution containing alcohol-acetonitrile-acetic acid (50:49:1, v/v) and 20 mM ammonium acetate while other experimental conditions were kept constant. The alcohols studied were methanol, ethanol, 1-propanol, 2-propanol, 1-butanol, 2-butanol, and 1-pentanol. The results indicate that even longer-chain alcohols can be used in nonaqueous capillary zone

Microfluidic Breadboard Approach to Capillary Electrophoresis.

A breadboard approach for electrophoretic separations with contactless conductivity detection is presented. This is based on miniature off-the-shelf components such as syringe pumps, valves, and pressure controllers which could be set up in a very compact overall arrangement. It has a high flexibility for different tasks at hand, and the common operations of hydrodynamic injection and capillary flushing are automated. For demonstration of the versatility of the proposition, several very diverse configurations and modes of electrophoresis were successfully implemented, namely, standard capillar

Capillary zone electrophoresis for haemoglobinopathy diagnosis.

AIMS: Capillary zone electrophoresis (CE) at alkaline pH is increasingly used in haemoglobinopathy diagnosis. We report our evaluation of automated CE, using the Capillarys 2 Flex Piercing instrument, as a routine diagnostic method for the detection of variant haemoglobins and the diagnosis of β thalassaemia. METHODS: A Capillarys 2 Flex Piercing instrument with Phoresis software was evaluated in our laboratory over a 6-week period, comparisons being made with high performance liquid chromatography (HPLC) and, for haemoglobin A(2) quantification, with microcolumn chromatography. RESULTS: The

[Progress of highly reproducible capillary electrophoresis].

Following rapid developments in capillary electrophoresis (CE), this technology has become an established analytical technique owing to its microscale characteristics, high speed, high efficiency, and versatility. However, the challenges of poor peak stability and/or reproducibility have consistently hindered its wider applications. CE has long been used as a measurement tool for plotting signal intensities versus the migration time; however, the migration time is not an independent variable in CE, but is affected by many direct and indirect parameters, including capillary (length, diameter, a

Diagnosing AICA-ribosiduria by capillary electrophoresis.

AICA-ribosiduria is a recently discovered inherited metabolic disease caused by a defect in final steps of purine de novo biosynthesis-5-amino-4-imidazolecarboxamide ribotide (AICAR)-transformylase/inosinemonophosphate (IMP)-cyclohydrolase (ATIC). A rapid and selective capillary electrophoretic method for screening of patients with AICA-ribosiduria is described. The method is based on direct ultraviolet detection of 5-amino-4-imidazolecarboxamide (AICA) and 5-amino-4-imidazolecarboxamide riboside (AICAr) in untreated urine. Background electrolyte consists of 100mM malonic acid adjusted with ga

Nonaqueous capillary electrophoresis in pharmaceutical analysis.

Nonaqueous media have shown to be useful for CE separations. It is possible to obtain high selectivities of closely related compounds without using any additive in the electrophoresis medium. The physicochemical parameters of some selected solvents and their influence on separation are discussed. Separations of small compounds with equal or almost equal charge-to-mass ratios have been performed in both aqueous- and nonaqueous-based media and compared. Examples of the use of nonaqueous media for drug impurity testing are given, and precautions for obtaining quantitative and reproducible analysi

Potential of polyE-323 coated capillaries for capillary electrophoresis of lipids.

In this note the feasibility of a polyamine-based capillary coating, polyE-323, for capillary electrophoresis (CE) of lipids is explored. PolyE-323 has previously been demonstrated to be suitable to suppress analyte-wall interaction of proteins in CE. However, the full applicability range of polyE-323 has not been exploited yet and it might be useful in the analysis of hydrophobic analytes, such as lipids. In this study, the stability of polyE-323 when using highly organic background electrolytes (BGEs), which are needed to solubilize the lipid analytes, was studied. For this, we used three di

Separation of tetracyclines by high-performance capillary electrophoresis and capillary electrochromatography.

Separations of various tetracycline mixtures by high-performance capillary electrophoresis (HPCE) and a new form of electrochromatography (CEC) are compared. The new CEC method involves etching the inner wall of the capillary surface with an appropriate reagent (ammonium dihydrogen fluoride) in order to produce a significant increase in surface area. The etched surface is then modified by a silation/hydrosilation reaction sequence to first produce a hydride intermediate which is then further reacted to attach a C18 moiety. The bare and hydride capillaries are tested under HPCE conditions while

Analysis of anxiolytic drugs by capillary electrophoresis with bare and coated capillaries.

The analysis of anxiolytic drugs, including barbiturates and benzodiazepines, by CE with bare and coated capillaries was investigated. Two types of CE modes, capillary zone electrophoresis (CZE) and micellar electrokinetic chromatography (MEKC) were employed for the separation of the solutes. Charged, structurally similar compounds exemplified by hypnotic drugs such as barbiturates were separated by both CZE and MEKC. Optimal separation conditions were established by varying the pH, the concentration of surfactant, the strength of the electric field, and the addition of the organic solvent. CZ

An in-house made autosampler for capillary electrophoresis.

BACKGROUND: Researchers are developing customized capillary electrophoresis (CE) instruments due to their affordability, and versatility. CE's simplicity, compared to chromatography-based methods, allows for low-cost designs using commercially available components. Injection methods, however, necessitate particular attention because of the high reproducibility required for quantitative determinations. Traditional manual hydrodynamic and electrokinetic injections involve one or more actions by the user thus increasing the number of potential sources of error. Automation efforts, such as using s

Capillary electrophoresis of diuretics and probenecid in methanol.

Actual mobilities and dissociation constants of six diuretics (benzthiazide, bumetanide, ethacrynic acid, furosemide, hydrochlorothiazide and chlorothiazide) and probenecid were investigated in methanol by capillary zone electrophoresis (CZE). The actual mobilities were derived from the dependence of the effective mobilities of the analytes on the pH(*) of the methanolic background electrolyte (BGE) solution. The measurement of effective mobilities was carried out mainly by a pressure-mediated capillary electrophoresis (CE) method. For comparison, parallel measurements were carried for two of

Optimized methods for capillary electrophoresis of tetracyclines.

Optimized methods for the analysis of some tetracyclines by capillary electrophoresis are described. Different buffer systems were employed for the separation of tetracycline, oxytetracycline and demeclocycline from their respective major impurities, including the 2-acetyl-2-decarboxamido derivatives. The influence of buffer pH and buffer concentration was systematically investigated. Non-ionic surfactant Triton X-100 and methyl-beta-cyclodextrin were used to obtain improved selectivity in the case of oxytetracycline and demeclocycline. The results are compared with those of previously establi

Separation of neutral carbohydrates by capillary electrophoresis.

The basic strategies for analysis of neutral carbohydrates by capillary electrophoresis are summarized. Neutral carbohydrates are dissociated in strong alkali to give anions, hence they can be separated directly by zone electrophoresis based on the difference between their dissociation constants. However, neutral carbohydrates are not electrically charged under normal conditions. Therefore, they should be converted to ions prior to or during analysis. Precapillary introduction of a basic or an acidic group to a neutral carbohydrate gives the derivative positive (in acidic media) or negative (i

Evaluation of Urine Proteins by Capillary Electrophoresis.

Capillary electrophoresis (CE) is a laboratory method usually used to separate proteins in body fluids such as serum, cerebrospinal fluid, or urine. Separation of proteins in urine can have clinical applications for evaluating samples from healthy dogs and dogs with proteinuria in a qualitative way, which would not be possible with gel electrophoresis. Other advantages of CE over gel electrophoresis in serum include the reduced separation time (2 min vs. 20 min in a gel), reduction of waste harmful to humans and the environment, and ability to obtain a curve without the need for additional sta
