Magnesium and embryonic development.

Important for energy metabolism, neurotransmission, bone stability, and other cellular functions, Mg(2+) has well-established and undisputedly critical roles in adult tissues. Its contributions to early embryonic development are less clearly understood. For decades it has been known that gestational Mg(2+) deficiency in rodents produces teratogenic effects. More recent studies have linked deficiency in this vital cation to birth defects in humans, including spina bifida, a neural fold closure defect in humans that occurs at an average rate of 1 per 1000 pregnancies. The first suggestion that M

Embryonic development and pattern formation.

During embryogenesis, information encoded in the genome is translated into cell proliferation, morphogenesis, and early stages of differentiation. Embryonic pattern arises from the spatial and temporal regulation and coordination of these events. The vitamin A (retinol) derivative retinoic acid (RA) is essential for normal development. Mammalian embryos are protected against vitamin A deficiency by maternal retinoid homeostasis until stored retinoids fall to very low levels. Retinol binding protein, which is synthesized in the yolk sac placenta of rodent embryos and in the syncytiotrophoblast

The role of retinoic acid in embryonic and post-embryonic development.

Retinoic acid (RA) is the bioactive metabolite of vitamin A (retinol) which acts on cells to establish or change the pattern of gene activity. Retinol is converted to RA by the action of two types of enzyme, retinol dehydrogenases and retinal dehydrogenases. In the nucleus RA acts as a ligand to activate two families of transcription factors, the RA receptors (RAR) and the retinoid X receptors (RXR) which heterodimerize and bind to the upstream sequences of RA-responsive genes. Thus, in addition to the well-established experimental paradigm of depriving animals of vitamin A to determine the ro

[Embryonic mechanisms of human holoprosencephaly development].

12 human embryos (stages 14-18) with different form of holoprosencephaly were studied. Spatial organization of the brain was reproduced with graphic reconstructions. It is demonstrated that the cause of different forms of holoprosencephaly is disturbed or delayed movement of the neurulation wave coming through the hemispheres of the forebrain. The cause of the defect may be an unspecific teratogenic factor or some cytogenetic damage. Severity and type of holoprosencephaly are associated not with the etiological factor but with the time of the action on the neurulation wave. Alobar holoprosence

Role of cholesterol in embryonic development.

We showed previously that 3 distal inhibitors of cholesterol synthesis are highly teratogenic in rats. AY 9944 and BM 15766 inhibit 7-dehydrocholesterol reductase, which catalyzes the last step of cholesterol synthesis, and triparanol inhibits Delta(24)-dehydrocholesterol reductase, which catalyzes the last step in another pathway. These molecules cause holoprosencephalic brain anomalies. Under certain experimental conditions, other anomalies (of the limbs and male genitalia) are also observed. Assays performed by gas chromatography-mass spectrometry (GC-MS) show hypocholesterolemia and an acc

Embryonic MTHFR contributes to blastocyst development.

PURPOSE: Reduction in methylenetetrahydrofolate reductase (MTHFR) activity due to genetic variations in the MTHFR gene has been controversially implicated in subfertility in human in vitro fertilization. However, there is no direct gene-knockdown study of embryonic MTHFR to assess its involvement in mammalian preimplantation development. The purpose of this study is to investigate expression profiles and functional roles of MTHFR in bovine preimplantation development. METHODS: Reverse transcription-quantitative PCR (RT-qPCR) and analysis of publicly available RNA-seq data were performed to re

Extra-embryonic function of Rb is essential for embryonic development and viability.

The retinoblastoma (Rb) gene was the first tumour suppressor identified. Inactivation of Rb in mice results in unscheduled cell proliferation, apoptosis and widespread developmental defects, leading to embryonic death by day 14.5 (refs 2-4). However, the actual cause of the embryonic lethality has not been fully investigated. Here we show that loss of Rb leads to excessive proliferation of trophoblast cells and a severe disruption of the normal labyrinth architecture in the placenta. This is accompanied by a decrease in vascularization and a reduction in placental transport function. We used t

[Embryonic and post-embryonic development of the parietal and visceral peritoneum in white mice].

Light and electron microscopy were used in order to investigate histogenesis of the parietal and visceral peritoneum of white mice in embryonic and postembryonic periods of development. Four periods were distinguished, during which gradual differentiation of the primordium material into tissue structures (mesothelium and the connective tissue) of the peritoneum were observed. Asynchronous differentiation of the mesothelium as well as certain correlation in the degree of differentiation of mesothelial and mesenchymal cells took place at all stages of the embryonic and postembryonic development.

Chromosome segregation errors during early embryonic development.

BACKGROUND: Mitosis maintains a genome's genetic information in daughter cells by accurately segregating chromosomes. However, chromosome aberrations are common during early mammalian embryogenesis. Chromosomal abnormalities during the early stages of embryogenesis result in the formation of mosaic embryos, wherein cells with normal genomes coexist with cells exhibiting abnormal genomes. The precise frequency and etiology of such abnormalities remain unclear. It is postulated that these aberrations contribute to the etiology of a number of conditions, including infertility and congenital disea

Embryonic Development of the Bicuspid Aortic Valve.

Bicuspid aortic valve (BAV) is the most common congenital valvular heart defect with an overall frequency of 0.5%-1.2%. BAVs result from abnormal aortic cusp formation during valvulogenesis, whereby adjacent cusps fuse into a single large cusp resulting in two, instead of the normal three, aortic cusps. Individuals with BAV are at increased risk for ascending aortic disease, aortic stenosis and coarctation of the aorta. The frequent occurrence of BAV and its anatomically discrete but frequent co-existing diseases leads us to suspect a common cellular origin. Although autosomal-dominant transmi

Smoc2 modulates embryonic myelopoiesis during zebrafish development.

BACKGROUND: SMOC2 is a member of the BM-40 (SPARC) family of matricellular proteins, reported to influence signaling in the extracellular compartment. In mice, Smoc2 is expressed in many different tissues and was shown to enhance the response to angiogenic growth factors, mediate cell adhesion, keratinocyte migration, and metastasis. Additionally, SMOC2 is associated with vitiligo and craniofacial and dental defects. The function of Smoc2 during early zebrafish development has not been determined to date. RESULTS: In pregastrula zebrafish embryos, smoc2 is expressed ubiquitously. As developme

Development of endometriosis from embryonic duct remnants.

Among 18 cases of endometriosis involving ovaries and fallopian tubes, we identified three cases with foci of endometriosis adjacent to the embryonic duct remnants in the fallopian tube. The serial sections of the blocks of tissue containing areas of interest showed segments of embryonic duct remnants with changes suggestive of gradual transformation to endometrial glands. The early changes consisted of replacement of the muscular coat surrounding the epithelium of embryonic duct remnants by the endometrial stroma with positive immunoreactivities for estrogen receptor. Subsequently there were

Development of the embryonic trisomy 1 mouse.

Trisomic and normal control embryos 9.5 through 12 gestational days were examined externally and on serial section with standard light microscopy techniques. Trisomic embryos showed significant retardation in length, weight, optic angle, and somite numbers at most times (p less than .05), and were 1/2 day behind normal animals in overall development. The eye showed the most defects, and these appeared at every period. They included aphakia, large intraretinal space, and dysmorphia of optic cup layers. Brain defects were found at all times. The most common were narrow brain vesicles and folding

Early Embryonic Development in Agriculturally Important Species.

The fertilization of oocytes ovulated by pigs, sheep, cows, and horses is not considered a limiting factor in successful establishment of pregnancy. Pig, sheep, and cow embryos undergo cleavage to the blastocyst stage, hatch from the zona pellucida, and undergo central-type implantation. Hatched blastocysts of pigs, sheep, and cows transition from tubular to long filamentous forms to establish surface area for exchange of nutrients and gases with the uterus. The equine blastocyst, surrounded by external membranes, does not elongate but migrates throughout the uterine lumen before attaching to

[New models to study human embryonic development].

Since 2021, assisted reproductive technologies (ART) are available to infertile couples, but also to single women and female couples. The process of in vitro fertilization (IVF) has allowed to cross the threshold of 5 million births worldwide, between 1978 and 2013. However, the failure rate per each IVF cycle is estimated to be around 75%. Therefore, there is a need to better understand human embryonic development in order to improve the success rate of IVF. Study models have evolved significantly in recent years: development of embryo culture, sequencing of the transcriptome of individualize

Embryonic ethanol exposure on zebrafish early development.

INTRODUCTION: Embryonic exposure to ethanol leads to a condition of physical, behavioral, and cognitive deficiencies named fetal alcohol spectrum disorders (FASD). The most severe variations are in fetal alcohol syndrome (FAS), which is easier to diagnose and not studied in animal models. On the other side, the pFAS (partial fetal alcohol syndrome) includes cases of alcohol-related congenital disabilities and neurodevelopmental disorder with an inconclusive diagnosis. In recent years, the zebrafish has become a valuable model to study FASD and its variations. METHODS: This study characterizes
