Evolution of avian encephalomyelitis virus during embryo-adaptation.

Wild-type avian encephalomyelitis virus (AEV) causes neurological signs in young chicks but no disease in pullets after oral or intracutaneous infection. However, if the virus gets embryo-adapted by serial passaging in chicken embryos, it will cause AE after intracutaneous infection in chickens of all ages. Recently, several cases of AE in layer pullets occurring shortly after intracutaneous vaccination were described. The present investigation was initiated to determine if vaccines that had inadvertently been embryo-adapted were responsible for these outbreaks. Virus isolation was done from t

EXPERIMENTAL STUDIES OF THE VIRUS OF INFECTIOUS AVIAN ENCEPHALOMYELITIS.

The results of investigations thus far carried out on experimental avian encephalomyelitis indicate that the virus of this newly described disease conforms to the group of definitely established viruses. It was essential to determine its taxonomy since the only prior record of its study (1) defines the infective agent as a virus because the usual cultural attempts failed to reveal a visible microorganism to be identified with it, and because the transmissible agent passed through Seitz and Berkefeld N filters. At the present time such determinants fail completely to satisfy the criteria for de

Avian host preference by vectors of eastern equine encephalomyelitis virus.

An important variable in the amplification and escape from the enzootic cycle of the arboviral encephalitides is the degree of contact between avian hosts and mosquito vectors. To analyze this interaction in detail, blood-fed mosquitoes that were confirmed vectors of eastern equine encephalomyelitis (EEE) virus were collected in 2002 from an enzootic site in central Alabama during the time this virus was actively transmitted. Avian-derived blood meals were identified to the species level of the host, and the proportion derived from each species was compared with the overall composition of the

Localization of viral protein in avian-encephalomyelitis-virus-infected hens.

Eighteen 2-year-old hens were infected orally with wild avian encephalomyelitis virus and examined using the immunofluorescent-antibody technique for 30 days. Viral antigen was detected in 10 organ tissues (liver, spleen, kidney, pancreas, and ovary) in 7 birds during the first half of the observation period and sporadically in alimentary tracts of 12 birds throughout this period, especially the lower parts of the intestines, but it was not detected in other organ tissues. Thirteen test-positive hens showed more localization of the antigen than baby chicks of other studies did.

Assessing the efficacy of a live vaccine against avian encephalomyelitis virus.

Avian encephalomyelitis virus (AEV) causes typical neurological symptoms in young chicks and a transient drop in egg production and hatchability in adult laying birds, resulting in huge economic losses in the poultry industry. An effective way to control and prevent this disease is vaccination of the flocks. Here, we assessed the efficacy of the live vaccine candidate strain GDt29 against avian encephalomyelitis virus. The GDt29 strain has low virulence, was confirmed safe, and showed no signs of pathogenicity. High titers of AEV-specific antibodies were detected in GDt29-vaccinated hens (S/P

Susceptibility of chickens to avian encephalomyelitis virus. III. Behavior of the virus in growing chicks.

A chick embryo-adapted strain of avian encephalomyelitis virus was inoculated subcutaneously and orally into 40-day-old (middle-aged) and 110-day-old (advanced-aged) chicks to examine the behavior of the virus in the chick body. In the middle-aged chicks, the virus appeared in the muscle at the site of inoculation, liver, spleen, pancreas, lumbar and cervical portions of the spinal cord, and brain 1 approximately 9 days after subcutaneous inoculation, and remained mostly in the central nervous system up to 17 days after the inoculation. The virus was found in large amounts in the muscle at the

A New Variant of Avian Encephalomyelitis Virus Associated with Neurologic Signs in Turkey Poults.

Avian encephalomyelitis (AE) is a disease caused by the avian encephalomyelitis virus (AEV) of the genus Tremovirus in the family Picornaviridae. Recently, cases of turkey poults showing neurological signs were submitted to the veterinary diagnostic laboratories at South Dakota State University and the University of Minnesota. The affected birds were showing nervous neurological signs such as tremors, inability to stand, torticollis, and wing drop. Clinical signs were observed by 3 weeks of age. Necropsy of birds revealed no significant gross lesions in the internal organs, including the brain

Histopathological diagnosis and genetic diversity of avian encephalomyelitis virus circulating in broiler farms in Egypt.

BACKGROUND: Avian encephalomyelitis (AE) is an imperative viral disease of avian species worldwide caused by the AE virus (AEV). AIM: This retrospective study aimed to genetically characterize field AEV strains circulating in the local governorates of Egypt and assess their genetic divergence in comparison with commercially available vaccines. To the best of our knowledge, this is the first described study of AEV-VP1 gene sequencing and phylogeny in broilers in Egypt. METHODS: A total of 150 tissue specimens from broilers of four Egyptian governorates were used for isolation and genetic char

Identification of virulence-related amino acid mutations of avian encephalomyelitis virus associated with vaccination methods.

Avian encephalomyelitis virus (AEV), a picornavirus, primarily infects the central nervous system of 1 to 2-week-old young chickens but not pullets. When wild-type AEV undergoes serial passaging in chicken embryos, it becomes to be embryo-adapted and can cause avian encephalomyelitis in chickens of all ages following intracutaneous infection through parenteral routes. This study was conducted to explore whether an outbreak of AEV in 95-day-old chickens was linked to inadvertent embryo adaptation of the AEV vaccine and its association with vaccination method. In this study, an AEV strain AEV/JS

Limited interdecadal variation in mosquito (Diptera: Culicidae) and avian host competence for Western equine encephalomyelitis virus (Togaviridae: Alphavirus).

Historically, western equine encephalomyelitis virus (WEEV) caused large equine and human epidemics in the Americas from Canada into Argentina. Despite recent enhanced surveillance for West Nile virus, there have been few reports of equine or human cases and little documented enzootic activity of WEEV. During the past three years, WEEV has been active again in California, but without human or equine cases. In the current study, we compared host and vector competence of representative WEEV isolates made during each decade over the past 60 years using white-crowned sparrows, house sparrows, and

Dynamic distribution and tissue tropism of avian encephalomyelitis virus isolate XY/Q-1410 in experimentally infected Korean quail.

Avian encephalomyelitis (AE) is an important infectious poultry disease worldwide that is caused by avian encephalomyelitis virus (AEV). However, to date, the dynamic distribution of AEV in quails has not been well described. Quantitative real-time polymerase chain reaction (qPCR) and immunohistochemistry (IHC) assays were used to investigate the dynamic distribution and tissue tropism of AEV in experimentally infected Korean quail. AEV was detected in the cerebrum, cerebellum, proventriculus, intestine, liver, pancreas, spleen, bursa, lung and kidney as early as 3 days post-infection (dpi). T

Viral encephalomyelitis of pigeons. IV. Growth of the virus in tissue cultures.

Growth and cytopathogenicity of pigeon herpes encephalomyelitis virus (PHEV) in avian and mammalian cell cultures were investigated. The virus was cytopathogenic to all avian primary cell cultures tested and produced large syncytia with intranuclear inclusions. Viral antigen was detected in the nuclei of infected cells 6 hr postinoculation. Infective virus, however, was obtained 8 hr post-inoculation. Maximum virus yields in avian cell cultures were reached 72 hr postinoculation. In mammalian cell lines tested, the virus proved to be cytopathogenic except in swine embryo kidney cell lines. The

The brain-specific upregulation of CARD11 in response to avian brain-neurotropic virus infection serves as a potential biomarker.

Avian neurotropic viruses are critical problems in poultry industry causing severe central nervous system (CNS) damage with neuroinvasive and neurovirulence properties. Biomarker of neurotropic viral intracranial invasion is of great application value for the diagnosis, but that of avian neurotropic viruses remains elusive. Previously, we found that chicken caspase recruitment domain family, member 11 (CARD11) was only upregulated in virulent Newcastle disease virus-infected chickens and in chicken primary neuronal cells. In this study, CARD11 was systemically expressed in chickens and pigeons

Progress in research on the molecular biological detection techniques of avian encephalomyelitis.

Avian encephalomyelitis (AE) is a highly infectious disease caused by the avian encephalomyelitis virus (AEV), which primarily affects the central nervous system of 1- to 4-week-old chicks and causes significant economic losses in the worldwide poultry industry. Despite heavy dependency on vaccine immunization, AEV has persisted on farms for extended periods, which increases its virulence and makes quick and accurate detection crucial to preventing and controlling the disease. Classical diagnostic methods have been unable to meet the current requirements for rapid diagnosis of AE cases. To add

Innovation and discovery: the application of nucleic acid-based technology to avian virus detection and characterization.

Polymerase chain reaction (PCR)-based approaches to the detection, differentiation and characterization of avian pathogens continue to be developed and refined. The PCRs, or reverse transcriptase-PCRs, may be general, designed to detect all or most variants of a pathogen, or to be serotype, genotype or pathotype specific. Progress is being made with respect to making nucleic acid approaches more suitable for use in diagnostic laboratories. Robotic workstations are now available for extraction of nucleic acid from many samples in a short time, for routine diagnosis. Following general PCR, the D

Detection of eastern equine encephalomyelitis viral antigen in avian blood by enzyme immunoassay: a laboratory study.

An enzyme immunoassay (EIA) was evaluated for its efficacy at detecting eastern equine encephalomyelitis (EEE) virus in avian blood and brain specimens. Preliminary analysis of blood from experimentally infected house sparrows and naturally infected whooping cranes showed that EEE antigen could be detected with the EIA. Polyclonal mouse antibodies were selected for antigen capture, and rabbit antibodies were selected for antigen detection. Overnight antigen incubation increased sensitivity. The lower limit of EEE antigen detection was 10(3.5) TCID50/ml for a stock of virus. Sensitivity was 10%
