Endothelins and anti-endothelins.

Endothelins, endothelin-1 (ET1), endothelin-2 (ET2) and endothelin-3 (ET3), are the most potent vasoconstrictor peptides released by endothelial cells. ET production is stimulated by vasopressor hormones, platelet-derived factors, coagulation products and cytokines, whereas nitric oxide and prostacyclin reduce ET production. ET bind to ETA and ETB receptors and produce marked and sustained rise in blood pressure, intense vasoconstriction of coronary arteries and have positive inotropic and chronotropic effects on myocardium. Besides, they influence neuroendocrine, renal and smooth muscle funct

[Endothelins. I. Physiology].

At the beginning of 1988 in Japan in a unique manner a new peptide substance derived form the endothelium was described, so far the most powerful vasoconstrictor substance-endothelin. This started an information boom which soon led to evidence of other substances of this group, naturally related substances; the detailed structure of all endothelins was established, gradually these substances were synthetized, very sensitive radioassays for their estimation were developed. Experimental and clinical work on endothelins and their effects penetrate at present all spheres of medicine and intensive

Endothelins and kidney acidification.

Endothelins (ETs) were initially identified for their vasoactive properties, but they also influence kidney tubule transport including acidification. They contribute importantly to enhancing kidney acidification in response to an acid challenge to systemic acid-base balance and enhance tubule acidification in settings of chronically reduced glomerular filtration rate (GFR) as in chronic kidney disease. ETs also contribute pathophysiologically to the increased kidney acidification in some forms of chronic metabolic alkalosis. In addition, chronically increased ET activity in the kidney, as in c

Endothelins and the lung.

Since endothelins were discovered by Yanasigawa in 1988 it has been recognised that they may have an important role in lung pathophysiology. Despite their biological importance as vasoconstrictors the physiological role of endothelin has not yet been defined within the lungs. This review explores their role in acute and chronic disease. During acute inflammation and ischaemia-reperfusion injury cytokines may induce release of endothelin. This is important in the realm of acute lung injury and during surgical procedures such as cardiopulmonary operations including lung resections and transplant

Endothelins as cardiovascular peptides.

The normal functional state of the vasculature and the events leading to the development of significant arterial disease involve the interaction of important vasoactive substances, which play important modulating or initiating roles in the development of hypertension and arteriosclerosis. Three endothelins have now been identified, of which ET-1 is the best characterized. ET-1 is produced by epithelial, mesangial, neuronal and glial, and liver cells, and is the most potent vasoconstrictor yet found. Each endothelin is derived from a different gene on separate chromosomes, and each binds to at

Endothelins in inflammatory neurological diseases.

Endothelins were discovered more than thirty years ago as potent vasoactive compounds. Beyond their well-documented cardiovascular properties, however, the contributions of the endothelin pathway have been demonstrated in several neuroinflammatory processes and the peptides have been reported as clinically relevant biomarkers in neurodegenerative diseases. Several studies report that endothelin-1 significantly contributes to the progression of neuroinflammatory processes, particularly during infections in the central nervous system (CNS), and is associated with a loss of endothelial integrity

Endothelins in health and disease.

Endothelins are powerful vasoconstrictor peptides that also play numerous other roles. The endothelin (ET) family consists of three peptides produced by a variety of tissues. Endothelin-1 (ET-1) is the principal isoform produced by the endothelium in the human cardiovascular system, and it exerts its actions through binding to specific receptors, the so-called type A (ET(A)) and type B (ET(B)) receptors. ET-1 is primarily a locally acting paracrine substance that appears to contribute to the maintenance of basal vascular tone. It is also activated in several diseases, including congestive hear

Highlights on endothelins: a review.

The endothelins (ET) are a family of contractile peptides made up of 21 amino acids. They are synthesised from larger precursors and they are expressed in different tissues. ET-1 is synthesised in endothelial cells by means of a specific endothelin converting enzyme and it is assumed that most of it is secreted into the basolateral compartment. It acts in a paracrine manner on the ETA and ETB2 receptors located on the surface of the vascular smooth muscle to elicit an increase in intracellular calcium and vasoconstriction. The circulating ET-1 can also activate endothelial ETC and ETB1 recepto

Endothelins as local activators of adrenocortical cells.

Besides the classical corticotropic hormones, ACTH and angiotensin II, various regulatory peptides produced by the adrenal gland are thought to participate in the control of corticosteroid secretion. Here, we review the evidence that endothelins (ETs) synthesized within the adrenal cortex may act as autocrine and/or paracrine factors to regulate adrenocortical cell activity. The expression of ETs has been detected in normal, hyperplastic and neoplastic adrenocortical cells. The occurrence of ET receptors has been described in the different zones of the cortex. ETs stimulate the secretion of bo

Update on endothelins - biology and clinical implications.

In the decade since the initial discovery and characterization of endothelin, its biology as a powerful vasoconstrictor has been dramatically demonstrated. Studies have clarified the existence of endothelin isoforms, complex mechanisms of biosynthesis, interaction with specific receptors, and pathogenic implications. We are on the brink of using endothelin antagonism as a clinical treatment for disease processes where endothelin plays an important role, including congestive heart failure and hypertension. Novel observations have been made about the unexpectedly profound contribution endothelin

Human alveolar macrophages synthesize endothelins by thrombin.

Alveolar macrophages (AM) play pivotal roles in the defense mechanism and the regulation of inflammatory processes in the airways. Macrophages express receptors for thrombin on their surfaces, and thrombin induces the chemotaxis and the proliferation of macrophages. Thrombin acts on vascular endothelial cells to synthesize endothelin (ET)-1. AM have been known to express prepro ET-1 mRNA. Thus, we hypothesized that thrombin stimulates AM so as to synthesize ET. Surgically resected human lungs were irrigated by saline to remove intravascular blood, then saline was instilled into the bronchus, a

Molecular biology and biochemistry of the endothelins.

Endothelin-1 (ET-1), the first member of the newly discovered mammalian endothelin family of biologically active peptides, was originally identified as a 21 residue potent vasoconstrictor peptide in vascular endothelial cells. However, it has since been demonstrated to possess a wide variety of pharmacological activities in tissues both within and outside the cardiovascular system, and peptides with a striking similarity to ET-1 have been found to be the major toxic component of a snake venom. Moreover, recent studies have suggested that mammals including humans produce three distinct members

Endothelins and airways--a short review.

Endothelins (ETs) are a multifunctional large family of polypeptides. There are three well recognized members in this family (ET-1, ET-2, and ET-3) of which ET-1 appears to be the most important. They have been shown to play an important role in the pathogenesis of many life threatening diseases of humans and animals. They also perform a wide variety of physiological roles. The most important property of ETs is smooth muscle contraction, which allows them to play an important role in the pathogenesis of many vascular, gastrointestinal, urogenital and airway diseases. Another important feature

Endothelins as regulators of vascular tone in man.

1. The vascular pharmacology, physiological relevance and pathophysiological roles of the endothelium-derived vasoconstrictor peptide endothelin-1 have been unclear. These issues were investigated, in vivo in man, using infusion of drugs into the brachial artery or dorsal hand vein, with responses measured by forearm plethysmography and the hand vein displacement technique respectively. 2. Endothelin-1 is a potent and sustained constrictor of resistance and capacitance vessels in vivo in man, acting through both subtypes (ETA and ETB) of endothelin receptors. Endothelin-1 stimulates generation

Endothelins Inhibit Junctional Permeability in Cultured Mouse Astrocytes.

Endothelins, a family of potent vasoconstrictor peptides initially characterized in peripheral tissues, have also been reported to be synthesized in the brain. In this structure several cell types, including astrocytes, endothelial cells and certain neurons, are potential targets for these peptides. In astrocytes, endothelins induce changes in the concentration of several second messengers (calcium, diacylglycerol, arachidonic acid, cAMP) known to be involved in the regulation of gap junction channels. Using the scrape loading/dye transfer technique we have observed that two isoforms of endoth
