Entosis is cell cannibalism utilized by tumor cells to engulf live neighboring cells for pro- or anti-tumorigenic purposes. It is unknown whether this extraordinary cellular event can be pathogenic in other diseases such as microcephaly, a condition characterized by a smaller than normal brain at birth. We find that mice mutant for the human microcephaly-causing gene Pals1, which exhibit diminished cortices due to massive cell death, also exhibit nuclei enveloped by plasma membranes inside of dividing cells. These cell-in-cell (CIC) structures represent a dynamic process accompanied by lengthe

Entosis, a form of cell-in-cell (CIC) structure formation where one cell invades and becomes internalized by another, has long been observed in various malignancies and is considered a critical cellular process in cancer progression, with potential impacts on tumor heterogeneity, survival, and metastasis. Syndecan-1 (SDC1), a transmembrane proteoglycan involved in cell adhesion and communication, plays a pivotal role in these processes, yet its specific function in entosis within pancreatic ductal adenocarcinoma (PDAC) remains underexplored. This study aimed to investigate whether SDC1 facilit

Entosis is a non-apoptotic cell death process implicated in various important biological processes, such as tumorigenesis. Entotic death is preceded with the formation of cell-in-cell structures that are well known to be controlled by two spatially separated core elements: adherens junction and actomyosin. However, the molecular mechanism underlying their coordination remains a longstanding open question. In this study, by profiling isogenic breast cancer cells, ARHGAP36 was identified as a potent inducer of entotic cell-in-cell formation, consistent with multiple lines of tumor-suppressive ev

Entosis, a form of cell cannibalism, is a newly discovered pathogenic mechanism leading to the development of small brains, termed microcephaly, in which P53 activation was found to play a major role. Microcephaly with entosis, found in Pals1 mutant mice, displays P53 activation that promotes entosis and apoptotic cell death. This previously unappreciated pathogenic mechanism represents a novel cellular dynamic in dividing cortical progenitors which is responsible for cell loss. To date, various recent models of microcephaly have bolstered the importance of P53 activation in cell death leading

Homotypic entosis is a phenomenon in which one cancer cell invades a neighboring cancer cell and is closed entirely within its entotic vacuole. The fate of entosis can lead to inner cell death or survival. Recent evidence draws attention to entosis as a novel prognostic marker in breast cancer. Nevertheless, little is known about the quantity and quality of the process of entosis in human cancer specimens. Here, for the first time, we analyze the frequency of entotic figures in a case of NOS (Non-Other Specified) breast cancer with regard to location: the primary tumor, regional lymph node, an

BACKGROUND: Entosis is an actomyosin-dependent cell-in-cell process that may culminate in lysosome-mediated entotic cell death and is implicated in tumor evolution. Its clinical relevance in lung adenocarcinoma (LUAD) remains unclear.
METHODS: To construct an Entosis-related Prognostic Prediction Index (EPPI), this study integrated TCGA-LUAD transcriptomic, clinical, and somatic mutation data. Three independent GEO cohorts provided external validation. The initial candidate gene set covered components driving cell-in-cell formation and the intracellular degradation of internalized cells. Lysophosphatidic acid (LPA) species accumulate in the ascites of ovarian high-grade serous cancer (HGSC) and are associated with short relapse-free survival. LPA is known to support metastatic spread of cancer cells by activating a multitude of signaling pathways via G-protein-coupled receptors of the LPAR family. Systematic unbiased analyses of the LPA-regulated signal transduction network in ovarian cancer cells have, however, not been reported to date. Methods: LPA-induced signaling pathways were identified by phosphoproteomics of both patient-derived and OVCAR8 cells, RNA sequencing, measu

Cell-in-cell (CIC) structures are among the most intriguing cellular phenomena occasionally observed in human cancer specimens. Once regarded as incidental findings, accumulating evidence has linked specific CIC subtypes, particularly entosis, to tumor progression and patient prognosis. Despite growing interest, systematic investigation of entosis remains limited due to the labor-intensive nature of manual identification and analysis. Computational approaches are, therefore, needed to enable scalable and reproducible entosis detection. In this study, we developed and evaluated five morphology-

BACKGROUND: Osteomyelitis (OM) is a debilitating infectious disease characterized by inflammation of the bone and bone marrow. Emerging evidence suggests that multiple forms of programmed cell death (PCD) contribute to its pathogenesis. However, the specific roles and interactions of these PCD types in OM remain largely undefined.
METHODS: Microarray-based transcriptome datasets related to OM were retrieved from the Gene Expression Omnibus (GEO) database. Thirteen PCD modalities were defined from the literature and specialized databases, including classical forms (e.g., apoptosis, autophagy)

Resistant cancer phenotype is a key obstacle in the successful therapy of prostate cancer. The primary aim of our study was to explore resistance mechanisms in the advanced type of prostate cancer cells (PC-3) and to clarify the role of autophagy in these processes. We performed time-lapse experiment (48 hours) with ROS generating plumbagin by using multimodal holographic microscope. Furthermore, we also performed the flow-cytometric analysis and the qRT-PCR gene expression analysis at 12 selected time points. TEM and confocal microscopy were used to verify the results. We found out that autop

BACKGROUND: The implementation of proton beam irradiation (PBI) for breast cancer (BC) treatment is rapidly advancing due to its enhanced target coverage and reduced toxicities to organs at risk. However, the effects of PBI can vary depending on the cell type. This study aimed to explore the effects of PBI on two BC cell lines, MCF7 and MDA-MB-231.
METHODS: The relative biological effectiveness (RBE) of PBI was assessed using a clonogenic assay. DNA double-strand break (DSB) repair, epithelial-mesenchymal transition (EMT), and filamentous actin (F-actin) were evaluated using immunofluorescenc

BACKGROUND: Prostate cancer (PCa) exhibits highly heterogeneous clinical outcomes, underscoring the need for biomarkers that more closely reflect tumor biology. While programmed cell death (PCD) is fundamental to tumor suppression, the prognostic impact of individual PCD pathways remains poorly characterized in PCa.
METHODS: We constructed 14 PCD prognostic models in a training cohort (GSE116918, n = 248), integrated the top three (apoptosis, entosis, necroptosis) into an AEN score, and validated in two independent cohorts in PCa. Tumor microenvironment (TME), drug sensitivity, single‑cell tr

Cell death is a critical biological phenomenon that can be categorized into accidental cell death (ACD) and programmed cell death (PCD), each exhibiting distinct signaling, mechanistic and morphological characteristics. This paper provides a comprehensive overview of seven types of ACD, including coagulative, liquefactive, caseous, fat, fibrinoid, gangrenous and secondary necrosis, discussing their pathological implications in conditions such as ischemia and inflammation. Additionally, we review eighteen forms of PCD, encompassing autophagy, apoptosis, necroptosis, pyroptosis, paraptosis, ferr

Glomerular podocytes are characterized by terminally differentiated epithelial cells with limited proliferating ability; thus, podocyte loss could not be fully compensated by podocyte regeneration. A large body of clinical studies collectively demonstrated that podocyte loss correlated with glomerular diseases progression. Both podocyte death and podocyte detachment lead to podocyte loss; however, which one is the main cause remains controversial. Up to date, multiple mechanisms are involved in podocyte death, including programmed apoptotic cell death (apoptosis and anoikis), programmed nonapo

Diabetes mellitus is a common chronic metabolic disease, with its prevalence escalating annually. Diabetic cardiomyopathy is a leading cause of mortality among diabetic patients, characterized by intricate metabolic disturbances and myocardial cell demise. Various forms of cellular death pathways including apoptosis, pyroptosis, autophagic cell death, necroptosis, ferroptosis, and entosis have been identified in diabetic cardiomyopathy. Inhibiting myocardial cell death pathways has shown promise in mitigating diabetic cardiomyopathy progression. However, there are still gaps in understanding t

Regulated cell death (RCD) is a fundamental biological process essential for tissue homeostasis, and the elimination of damaged or malignant cells. In cancer, dysregulation of RCD is closely linked to tumor initiation, progression, therapeutic resistance, and remodeling of the tumor microenvironment (TME). In this review, we classify RCD in cancer into three broad groups. Classical cell death types include apoptosis, autophagy, necroptosis, and pyroptosis, which have well-established roles in controlling cell fate. Metal-dependent pathways, represented by ferroptosis, and cuproptosis, highligh

Implantation failure is a major cause of infertility, but its mechanisms remain unclear due to the lack of techniques for constructing organized endometrial structures and recapitulating the implantation process in vitro. Endometrial organoids have recently been developed, but they consist of only epithelial cells, and their apical surface faces inward, preventing blastocyst attachment. We developed an apical-out mouse endometrial organoid incorporating epithelial and stromal cells, and examined its ability to recapitulate implantation with mouse blastocysts. Mouse uteri were digested with col

Intervertebral disc degeneration (IVDD) represents a severe chronic condition characterized by diverse programmed cell death (PCD) mechanisms serving as critical pathological features. The identification of key genes associated with cellular demise in IVDD is crucial for enhancing diagnostic and prognostic strategies. We extracted microarray-based transcriptomic multi-datasets from the GEO database, comprising 34 normal specimens (grade I/II) and 38 IVDD cases (grade III/IV). Nineteen PCD-associated genes encompassing multiple death modalities (including apoptosis, pyroptosis, ferroptosis, aut
