Analytical applications of immobilized enzymes.

Several clinical laboratory methods using enzymes as reagents now may utilize immobilized enzymes. These are enzymes attached to solid surfaces by adsorption, covalent binding, cross linking or similar means. Immobilized enzymes are widely used presently in convenient tests for urine glucose and galactose or blood glucose and urea, and in serum glucose or urea determinations by automated methods. Advantages include enhanced stability, enzyme conservation, reuse and economy. Limitations in clinical analysis include the requirement for proper handling of both the immobilized enzyme and the speci

Immobilized Enzymes for ABO-Independent Blood Cell Transfusions.

The transfusion of blood products requires ABO compatibility due to A and B antigens on blood cells and their corresponding antibodies in plasma. The supply of red cell concentrates (RCCs) and platelet concentrates (PCs) is challenged by rising demand and declining numbers of donors. To counteract the supply limitations, a method is developed to generate universal RCCs and PCs. For this, the A antigen is enzymatically removed by two enzymes from Flavonifractor plautii working in concert and the B antigen is removed by galactosidases from Pedobacter panaciterrae (PpaGal_WT or the variant PpaGal

Effect of the microenvironment on the kinetic properties of immobilized enzymes.

A new immobilization method was developed in order to perform a systematic study of the influence of the microenvironment on the properties of immobilized enzymes. The enzyme, alcohol dehydrogenase, was first activated, then polypeptide arms of known composition were quantitatively grafted and finally the enzyme was covalently immobilized by co-polymerization of the activated ends of the peptide arms with acrylamide monomers. In this way, the polypeptide linker arms fully determine the properties of the microcavity of the gel in which the enzyme is immobilized by multipoint covalent linkages.

An immobilized enzyme reactor for the detoxification of bilirubin.

An immobilized enzyme reactor has been developed for the degradation of bilirubin as a potential treatment for neonatal jaundice. It utilizes the enzyme bilirubin oxidase from Myrothecium verrucaria, which in the presence of molecular oxygen converts bilirubin to biliverdin and other products that are much less toxic than bilirubin. Bilirubin oxidase was covalently attached to agarose beads using cyano transfer activation. Forty percent of the specific activity of bilirubin oxidase was retained after immmobilization, and preparations with 20 units of enzymatic activity per gram of drained wet

Calmodulin-mediated reversible immobilization of enzymes.

This work demonstrates the use of the protein calmodulin, CaM, as an affinity tag for the reversible immobilization of enzymes on surfaces. Our strategy takes advantage of the of the reversible, calcium-mediated binding of CaM to its ligand phenothiazine and of the ability to produce fusion proteins between CaM and a variety of enzymes to reversibly immobilize enzymes in an oriented fashion to different surfaces. Specifically, we employed two different enzymes, organophosphorus hydrolase (OPH) and beta-lactamase and two different solid supports, a silica surface and cellulose membrane modified

Development of novel enzyme immobilization methods employing formaldehyde or triethoxysilylbutyraldehyde to fabricate immobilized enzyme microreactors for peptide mapping.

The digestion of proteins with proteolytic enzymes has expedited the analysis of peptide mapping. Here, we compared the digestion efficiency of soluble chymotrypsin (CT) with two immobilized CT preparations using bovine serum albumin (BSA) as the substrate. An efficient method of immobilizing chymotrypsin using formaldehyde (FA) was optimized and the conditions were applied to assess a novel immobilization reagent, triethoxysilylbutaraldehyde (TESB). Efforts to determine the best enzyme-to-substrate (E : S) ratios during digestion of denatured BSA with single-use FA-CT enzyme particles were pe

Improvement of catalysis performance of pepsin and lipase enzymes by double enzyme immobilization method.

Ferrocene-coupled chitosan (CS-Fc) support polymer was synthesized for the immobilization of double enzymes and single enzymes. The immobilization study was carried out after a detailed characterization study. The binding of pepsin and lipase enzymes to the polymer was supported by the shift of peaks in the FTIR spectra of free enzymes. The particle sizes of chitosan (CS) and CS-Fc were evaluated using zeta potential measurement. The optimum conditions of the enzymes in the double enzyme system did not affect each other and each showed their activity. In the presence of double enzyme immobiliz

Metal-Organic Framework-Immobilized Mycotoxin-Degrading Enzymes: Interfaces, Host Design, and Food/Feed Applications.

Mycotoxin contamination remains a persistent threat to food and feed safety owing to the chemical stability of many mycotoxins, frequent co-occurrence, and matrix-dependent risks. Enzymatic detoxification enables structure-targeted transformation of toxicity-determining motifs, such as epoxide rings, reactive double bonds, amide linkages, and lactone structures. However, free mycotoxin-degrading enzymes are often constrained by poor operational stability, difficult recovery, and limited adaptability to complex matrices. Metal-organic frameworks (MOFs) provide programmable microenvironments for

Evaluation of the immobilized enzymes function in soil remediation following polycyclic aromatic hydrocarbon contamination.

The bioremediation of polycyclic aromatic hydrocarbon (PAHs) from soil utilizing microorganisms, enzymes, microbial consortiums, strains, etc. has attracted a lot of interest due to the environmentally friendly, and cost-effective features. Enzymes can efficiently break down PAHs in soil by hydroxylating the benzene ring, breaking the C-C bond, and catalyze the hydroxylation of a variety of benzene ring compounds via single-electron transfer oxidation. However, the practical application is limited by its instability and ease to loss function under harsh environmental conditions such as pH, tem

A study on the functional subunits of phospholipases A2 by enzyme immobilization.

Pancreatic and venom phospholipases A2 have complex and distinct oligomerization behaviour. Pancreatic enzymes are monomeric in solution, but their quaternary structure at interfaces is unknown. On the other hand, certain crotalid venom phospholipases A2 are dimeric in solution, and different reports have proposed either the monomer or the dimer as the catalytically functional subunit. In this study, enzyme immobilization was used as a tool for determining the functional subunits of these enzymes. The dimeric Crotalus atrox phospholipase A2 was covalently attached to agarose beads, via either

Electrostatically Mediated In Situ Polymerization for Enzyme Immobilization and Activation.

Enzyme immobilization in nanoparticles is of interest for boosting their catalytic applications, yet rational approaches to designs achieving both high enzyme loading and activation remain a challenge. Herein, we report an electrostatically mediated in situ polymerization strategy that simultaneously realizes enzyme immobilization and activation. This was achieved by copolymerizing cationic monomers with a cross-linker in the presence of the enzyme lipase (anionic) as the template, which produces enzyme-loaded nanogels. The effects of different control factors such as pH, lipase dosage, and cr

Magnetic Polymer Nanoparticles Immobilized with Enzymes for Stabilizing Pickering Emulsions: A Recyclable Interfacial Catalysis System.

The Pickering interfacial catalysis (PIC) system based on enzyme-immobilized carriers offers an efficient platform for the enzymatic reactions. In this study, we developed enzyme-immobilized magnetic polymer nanoparticles (MPNs) capable of stabilizing Pickering emulsions while enabling the rapid and stimuli-responsive recycling of catalytic products. Nano-Fe3O4 was prepared via coprecipitation, and MPNs with an average diameter of 252 nm were synthesized through hybrid emulsion polymerization of a mixture of styrene and methyl methacrylate containing a magnetic fluid. Enzyme-Immobilized Oxoammonium Nanogels: A Biocompatible and Injectable Platform for Enhanced Enzyme Stability and Reusability.

We report redox-responsive, oxoammonium-functionalized nanogels for mild, injectable, noncovalent enzyme immobilization. Amphiphilic PEG-b-poly(PMA-co-GMA) prepared by Reversible Addition-Fragmentation Chain Transfer (RAFT) polymerization was oxidized to nitroxide/oxoammonium states, forming nanogels that electrostatically complex with anionic proteins. Increasing oxoammonium content (30-70%) boosted encapsulation (85-98%) and loading (26-47%). Lipase and paraoxonase-1 (PON1) retained or exceeded native activity; the highest-charge formulation delivered 2.5-5-fold higher specific activity and

Investigations of reaction kinetics for immobilized enzymes--identification of parameters in the presence of diffusion limitation.

A method is proposed for identification of kinetic parameters when diffusion of substrates is limiting in reactions catalyzed by immobilized enzymes. This method overcomes conventional sequential procedures, which assume immobilization does not affect the conformation of the enzyme and, thus, consider intrinsic and inherent kinetics to be the same. The coupled equations describing intraparticle mass transport are solved simultaneously using numerical methods and are used for direct estimation of kinetic parameters by fitting modeling results to time-course measurements in a stirred tank reacto
