Akabane and bovine ephemeral fever virus infections.

Akabane and bovine ephemeral fever viruses are exotic to the American continent. Both viruses are spread by insect vectors, and each causes disease of varying severity in food-producing animals. However, there are few other similarities between the agents and the diseases that they cause. They do not share the same insect vectors, the mammalian host range is different, and the clinical manifestations of virus infection vary markedly. Akabane virus is a cause of severe congenital defects, but adult animals show no signs of infection. In contrast, bovine ephemeral fever virus causes a febrile il

The occurrence of ephemeral fever in Kenya, 1968-1988.

Ephemeral fever occurs in Kenya in epidemics which have been associated with periods of unusually heavy and prolonged rainfall. The epidemics persist for 2-3 years and involve most of the country. They occur in the same years as epizootics of Rift Valley fever, which is a mosquito-borne Phlebovirus infection. Sporadic outbreaks of the disease have been associated with local heavy rains; though one outbreak occurred during a dry period. Culicoides spp. were generally present in large numbers at the site of the outbreaks. Seroconversions were detected in sentinel cattle, when there had been no a

Ataxia and paralysis associated with bovine ephemeral fever infection.

Three adult cattle that had been ataxic for 5 to 7 months and a bull that had been paralysed for 24 days following bovine ephemeral fever infection were studied clinically and pathologically. Severe bilaterally symmetrical Wallerian degeneration was demonstrated in the spinal cords of all 4 cattle; in 3 animals an area of primary damage was present in the first cervical segment of the cord and in the other animal it was at the level of the lumbar cord. Pressure on the cord resulting from trauma was considered to be the most likely aetiology because of the similarity of the lesion to other synd

The effect of bovine ephemeral fever virus on the bovine foetus.

Six pregnant heifers that were experimentally infected with bovine ephemeral fever virus produced normal calves. Foetuses of 8 heifers that were immune to bovine ephemeral fever were inoculated with bovine ephemeral fever virus. One was aborted after 36 days, but the cause of abortion could not be determined. One was sacrificed after 28 days. The foetus and its membranes were normal and neither virus nor antibody was demonstrated. The other 6 heifers produced normal calves. One calf, 501, inoculated after 160 days gestation, contained high levels of neutralising antibody in serum before ingest

IRF8 facilitates bovine ephemeral fever virus replication by downregulating IRF9.

Interferon regulatory factor 8 (IRF8), an essential member of the IRFs protein family, serves as a critical transcriptional regulator in cytokine signaling, gene transcription, and the differentiation and proliferation of immune cells. However, its function on the bovine ephemeral fever virus (BEFV) infection has not been described. In this study, we demonstrate that BEFV infection upregulates the expression of IRF8, and IRF8 promotes the replication of BEFV. Subsequent investigations reveal that IRF8 suppresses the type I IFN signaling pathway via the degradation of IRF9 in the context of BEF

Studies on the pathogenesis of bovine ephemeral fever. IV: A comparison with the inflammatory events in milk fever of cattle.

The study of ephemeral fever in cattle has defined a range of haematological and biochemical changes in blood which are characteristic of an inflammatory response. One of the clinical signs of ephemeral fever, a temporary paralysis reversible by treatment with calcium borogluconate, is similar to that in milk fever (parturient paresis), a disease of multiparous dairy cows. Three separate groups of cows were studied. Four multiparous cows were observed and sampled repeatedly during calving, three similar cows and one cow calving for the first time in a dairy herd were sampled daily before and a

The effect of anti-inflammatory agents on the clinical expression of bovine ephemeral fever.

The effect of two anti-inflammatory drugs on the development and persistence of clinical signs in cattle experimentally infected with bovine ephemeral fever (BEF) virus was investigated by their administration, either before or after the commencement of fever. A total of 16 cattle was given phenylbutazone sodium (PBZ). The drug prevented fever and other clinical signs in six cattle when given daily during the incubation period, and at 8-h intervals for 5 days when clinical disease might be expected. When treatment with PBZ was deferred until 2-4 h after the commencement of fever, the rectal te

Antigenic variation of bovine ephemeral fever viruses isolated in Iran, 2012-2013.

Bovine ephemeral fever virus (BEFV) is an economic arthropod-borne virus distributed in Africa, Asia, and Australia. Based on the sequence of the gene encoding the surface glycoprotein G, the viral antigenic determinant, BEFV has been phylogenetically classified into three clusters, including Australia, East Asia, and the Middle East. Here, we provide evidence for antigenic variations among the BEFV isolates in Iran during the period of 2012 to 2013 and also the exotic YHL strain, which are all classified into the East Asian cluster of the virus. For this propose, the entire length of the G ge

Molecular epizootiology of bovine ephemeral fever virus in Iran during 2015 to 2022.

Bovine ephemeral fever (BEF) is a debilitating disease of cattle and water buffaloes. Bovine ephemeral fever viruses (BEFVs) form four phylogenetic lineages including the Middle East, East of Asia, Australia and Africa, while the exotic viral strains have also been detected in different geographic areas. We characterized eight BEFVs from different regions of Iran during a period of seven years from 2015 to 2022. Sequencing the entire length of the G gene, the BEFVs were classified in the Middle Eastern lineage with the maximum of 99.73% and minimum of 97.30% nucleotide identity. The all Irania

Immunogenicity of a freeze-dried combined vaccine against Rift Valley fever and bovine ephemeral fever in cattle.

BACKGROUND: The target of vaccination is to encourage a strong, covering and long-lasting immune response against antigens. For achieving these objectives; effective adjuvant and new vaccine strategies are demanded to make the vaccine sufficiently immunogenic to instigate a powerful immune response. AIM: This study was completed for elaboration and evaluation of freeze-dried combined vaccine against both Rift Valley fever (RVF) and bovine ephemeral fever (BEF) viruses using different stabilizers. METHODS: Three formulae were prepared from such vaccine including: formula (1): stabilized with

Safety, immunogenicity and duration of immunity elicited by an inactivated bovine ephemeral fever vaccine.

Bovine ephemeral fever (BEF) is an economically important viral vector-borne cattle disease. Several live-attenuated, inactivated and recombinant vaccines have been tested, demonstrating varying efficacy. However, to the best of our knowledge, duration of immunity conferred by an inactivated vaccine has never been reported. In the last decade, Israel has faced an increasing number of BEF outbreaks. The need for an effective vaccine compatible with strains circulating in the Middle East region led to the development of a MONTANIDE™ ISA 206 VG (water-in-oil-in-water), inactivated vaccine based o

Resurgence of bovine ephemeral fever in mainland Japan in 2015 after a 23-year absence.

In September and October 2015, suspected cases of bovine ephemeral fever (BEF) were reported in the mainland region of Kagoshima Prefecture and on Tanegashima Island. The genome of the BEF virus (BEFV) was detected in the diseased cows and the cows that had recovered. The serum obtained from the affected cows contained high titers of BEFV-neutralizing antibody. In total, 18 affected cows were demonstrated to be infected with BEFV during the outbreak. Our findings showed evidence that BEF occurred in mainland Japan after a 23-year absence. Phylogenetic analysis based on the surface glycoprotein

Characterization of a recombinant Newcastle disease virus expressing the glycoprotein of bovine ephemeral fever virus.

Bovine ephemeral fever (BEF) is caused by the arthropod-borne bovine ephemeral fever virus (BEFV), which is a member of the family Rhabdoviridae and the genus Ephemerovirus. BEFV causes an acute febrile infection in cattle and water buffalo. In this study, a recombinant Newcastle disease virus (NDV) expressing the glycoprotein (G) of BEFV (rL-BEFV-G) was constructed, and its biological characteristics in vitro and in vivo, pathogenicity, and immune response in mice and cattle were evaluated. BEFV G enabled NDV to spread from cell to cell. rL-BEFV-G remained nonvirulent in poultry and mice comp

SSBP1 promotes bovine ephemeral fever virus replication by antagonizing antiviral immune responses via degrading MAVS.

Mitochondrial single-stranded DNA-binding protein 1 (SSBP1), a component of the mitochondrial DNA replisome, is involved in DNA replication, repair and maintenance of mitochondrial DNA. However, its function in the bovine ephemeral fever virus (BEFV) infection has not been described. Herein, we found that SSBP1 acted as an essential negative regulator of mitochondrial antiviral signaling protein (MAVS) to maintain immune homeostasis. Under BEFV infection, SSBP1 was upregulated in host cells, which boosted the susceptibility to virus. SSBP1 dramatically impaired BEFV-triggered antiviral immune

Low Match Between the Emerging 'Mayotte-like' Strain and the 919-Strain Bovine Ephemeral Fever Vaccine.

Background/Objectives: Bovine ephemeral fever virus (BEFV) causes an economically important arthropod-borne cattle disease. In 2023, a lineage I "Mayotte-like" (ML) strain emerged in Israel during an outbreak in vaccinated herds, raising concerns about its antigenic match with the live-attenuated vaccine based on Australian strain 919. We compared G-protein neutralizing sites and cross-neutralization among the vaccine strain, Israeli lineage IIIa strains from 2000 and 2021, and ML. Methods: G1-G3 neutralizing-site sequences were compared. Serum-neutralization assays tested all four viruses usi

Biopanning of polypeptides binding to bovine ephemeral fever virus G1 protein from phage display peptide library.

BACKGROUND: The bovine ephemeral fever virus (BEFV) glycoprotein neutralization site 1 (also referred as G1 protein), is a critical protein responsible for virus infectivity and eliciting immune-protection, however, binding peptides of BEFV G1 protein are still unclear. Thus, the aim of the present study was to screen specific polypeptides, which bind BEFV G1 protein with high-affinity and inhibit BEFV replication. METHODS: The purified BEFV G1 was coated and then reacted with the M13-based Ph.D.-7 phage random display library. The peptides for target binding were automated sequenced after fo

Viral neurotropism, peripheral neuropathy and other morphological abnormalities in bovine ephemeral fever virus-infected downer cattle.

OBJECTIVE: This study assessed the neurotropism of bovine ephemeral fever (BEF) virus (BEFV) and described histomorphological abnormalities of the brain, spinal cord and peripheral nerves that may causally contribute to paresis or paralysis in BEF. METHODS: Four paralysed and six asymptomatic but virus-infected cattle were monitored, and blood and serum samples screened by qRT-PCR, virus isolation and neutralisation tests. Fresh brain, spinal cord, peripheral nerve and other tissues were qRT-PCR-tested for viral RNA, while formalin-fixed specimens were processed routinely and immunohistochemi

The immunogenicity of the secretory GΔTM protein of bovine ephemeral fever virus stably expressed by mammalian cells.

Bovine ephemeral fever virus (BEFV) causes an acute febrile disease in cattle and water buffalo. The disease has an impact on dairy and beef production in tropical and subtropical countries. Vaccination is used for disease prevention and control. In this study, we developed a recombinant lentivirus to produce mammalian stable cells expressing histidine-tagged BEFV G protein with a deleted transmembrane domain (GΔTM) as a secretory protein. In addition, guinea pigs were immunised with the purified GΔTM protein and booster immunised at a 3-week interval. The mammalian stable cells were able to c
