The fungal endophytes: does the rate of mycelium growth decrease with host age and host generations?

Vertical transmission of endophytic fungi is imperfect. Fungal endophytes do not always enter seeds of the host on which they live. One of the factors affecting the effectiveness of vertical transmission may be the fungal growth rate. We have hypothesized that the rate of mycelium growth of the endophytic fungus Epichloë poae decreases with host plant age and in successive years of the host and fungal generations. The hypothesis was tested in vitro, by using passaging of fungal mycelium to imitate vertical transmission in plants. Growth rate of the fungus decreased with host plant age and in s

Incidence, Identification, and Mycoparasitic Activity of Clonostachys epichloë, a Hyperparasite of the Fungal Endophyte Epichloë typhina.

Fungi of the genus Epichloë (Clavicipitaceae, Ascomycota) cause choke disease in many grass species. The disease manifests itself as fungal stromata that form around developing inflorescences, thereby suppressing their maturation. Economic losses in agricultural production due to choke have long been known in the U.S.A. and France, but attempts to control choke disease have not been successful. The interaction between Epichloë typhina (Pers.) Tul. & C. Tul. and its naturally occurring fungal hyperparasite, Clonostachys epichloë (Speg.) Schroers (sexual morph Bionectria epichloë) was investigat

Cross-species transcriptomics identifies core regulatory changes differentiating the asymptomatic asexual and virulent sexual life cycles of grass-symbiotic Epichloë fungi.

Fungi from the genus Epichloë form systemic endobiotic infections of cool season grasses, producing a range of host-protective natural products in return for access to nutrients. These infections are asymptomatic during vegetative host growth, with associations between asexual Epichloë spp. and their hosts considered mutualistic. However, the sexual cycle of Epichloë spp. involves virulent growth, characterized by the envelopment and sterilization of a developing host inflorescence by a dense sheath of mycelia known as a stroma. Microscopic analysis of stromata revealed a dramatic increase in

EgSPE, a secreted protein from Epichloë gansuensis, modulates symbiotic establishment and host drought tolerance.

BACKGROUND: Epichloë endophytes form beneficial symbioses with cool-season grasses, enhancing host tolerance to abiotic stresses such as drought while maintaining normal plant growth. However, the molecular mechanisms underpinning this symbiosis, particularly the role of fungal-secreted protein, remain largely unexplored. RESULTS: In this study, we identify EgSPE, a secreted protein from Epichloë gansuensis, as a key regulator of symbiotic establishment and host drought adaptation in drunken horse grass (Achnatherum inebrians). Transcriptome profiling during host colonization revealed EgSPE a

High-affinity iron uptake is required for optimal Epichloë festucae colonization of Lolium perenne and seed transmission.

Epichloë festucae uses a siderophore-mediated system to acquire iron, which is important to maintain endophyte-grass symbioses. Here we investigate the roles of the alternative iron acquisition system, reductive iron assimilation (RIA), via disruption of the fetC gene, which encodes a multicopper ferroxidase, either alone (i.e., ΔfetC) or in combination with disruption of the gene sidA, which encodes a siderophore biosynthesis enzyme (i.e., ΔfetC/ΔsidA). The phenotypic characteristics of these mutants were compared to ΔsidA and wild-type (WT) strains during growth under axenic culture conditio

The Epichloë festucae antifungal protein has activity against the plant pathogen Sclerotinia homoeocarpa, the causal agent of dollar spot disease.

Epichloë spp. are naturally occurring fungal endophytic symbionts of many cool-season grasses. Infection by the fungal endophytes often confers biotic and abiotic stress tolerance to their hosts. Endophyte-mediated disease resistance is well-established in the fine fescue grass Festuca rubra subsp. rubra (strong creeping red fescue) infected with E. festucae. Resistance to fungal pathogens is not an established effect of endophyte infection of other grass species, and may therefore be unique to the fine fescues. The underlying mechanism of the disease resistance is unknown. E. festucae produce

Unlocking the power of Pseudomonas: network-guided isolation enhances plant growth on Achnatherum inebrians.

The tripartite interplay among plants, bacteria, and fungal endophytes is crucial for maintaining host plant fitness. However, how Epichloë endophytes influence plant-associated bacterial communities in cool-season grasses, particularly in ecologically important yet understudied species, such as Achnatherum inebrians, remains unclear. Using phylogenetic molecular ecological network analysis, we determined that seed-borne (seed epiphytic) and phyllosphere bacterial communities of Epichloë-infected (EI) A. inebrians exhibited reduced network complexity compared to Epichloë-free (EF) plants. Epichloë Endophytes Potentially Facilitate Host Plant Recruitment of Rhizosphere Microbiota Carrying Beneficial Traits.

Plant-microbe symbiotic relationships drive ecosystem evolution. This study employed metabolomics and metagenomic technologies to investigate the effects of the aboveground-restricted endophytic fungus Epichloë guerinii in the host plant Melica transsilvanica on the rhizosphere microbial community structure and functional traits. Our results revealed that the presence of E. guerinii significantly increased the secretion of organic acids, amino acids, and sugar alcohols from the host root system. These exudates correlated strongly with abundant, plant growth-promoting rhizosphere microorganisms

Transcriptome analysis of Epichloë strains in tall fescue in response to drought stress.

Epichloë coenophiala, a systemic fungal symbiont (endophyte) of tall fescue (Lolium arundinaceum), has been documented to confer to this grass better persistence than plants lacking the endophyte, especially under stress conditions such as drought. The response, if any, of the endophyte to imposition of stress on the host plant has not been characterized previously. Therefore, we investigated effects on gene expression by E. coenophiala and a related endophyte when plant-endophyte symbiota were subjected to acute water-deficit stress. Plants harboring different endophyte strains were grown in

Fungal endophytes relieve the growth-defence trade-off of their plant hosts through the production of bioactive alkaloids.

A central paradigm in plant biology is that there is a trade-off between growth and defence. We propose that Epichloë fungal endophytes relieve this trade-off in plants by the fungal-derived production of antiherbivore alkaloids. Lolium perenne plants without (nil) and with Epichloë LpTG-3 sp. strain AR37 able (wild type (wt), ΔidtA) and unable (ΔidtM) to produce bioactive indole diterpene alkaloids were subjected to an exogenous application of gibberellin (GA) hormone followed by a challenge with Rhopalosiphum padi aphids. The GA-derived plant growth promotion increased the susceptibility to

The plant hormone salicylic acid interacts with the mechanism of anti-herbivory conferred by fungal endophytes in grasses.

The plant hormone salicylic acid (SA) is recognized as an effective defence against biotrophic pathogens, but its role as regulator of beneficial plant symbionts has received little attention. We studied the relationship between the SA hormone and leaf fungal endophytes on herbivore defences in symbiotic grasses. We hypothesize that the SA exposure suppresses the endophyte reducing the fungal-produced alkaloids. Because of the role that alkaloids play in anti-herbivore defences, any reduction in their production should make host plants more susceptible to herbivores. Lolium multiflorum plants

Dendrobium officinale Endophytes May Colonize the Intestinal Tract and Regulate Gut Microbiota in Mice.

Dendrobium officinale is a traditional Chinese medicine for treating gastrointestinal diseases by nourishing "Yin" and thickening the stomach lining. To study whether D. officinale endophytes can colonize the intestinal tract and regulate gut microbiota in mice, we used autoclave steam sterilizing and 60Co-γ radiation to eliminate D. officinale endophytes from its juice. Then, high-throughput ITS1-ITS2 rDNA and 16S rRNA gene amplicons were sequenced to analyze the microbial community of D. officinale endophytes and fecal samples of mice after administration of fresh D. officinale juice. Genetic Characterisation of the Bacterial Microbiota Associating With a Strain of Epichloë Fungal Endophyte of Perennial Ryegrass and the Interaction With Its Paenibacillus Members.

Plant-associated fungi can host unique bacterial microbiota to provide multiple benefits to their fungal hosts. Here it was characterised the bacterial microbiota associated with an Epichloë fungal endophyte (strain AR135) isolated from perennial ryegrass (Lolium perenne) via both 16S rRNA gene sequencing and direct microbial isolation and investigated the microbe-microbe interactions between these bacteria and the fungus. The bacterial microbiota of AR135 was dominated by members within the genus Paenibacillus, with 99% of abundance (on average); although bacteria within genera Delftia and Br

Impact of Low-Level Ergot Alkaloids and Endophyte Presence in Tall Fescue Grass on the Metabolome and Microbiome of Fall-Grazing Steers.

Fescue toxicosis (FT) is a mycotoxin-related disease caused by the ingestion of tall fescue, naturally infected with the ergot alkaloid (EA)-producing endophyte Epichloë coenophiala. Some grazing on endophyte-free (E-) or non-toxic (NT), commercial endophyte-infected pastures takes place in the US as well. Earlier, we found that grazing on toxic fescue with low levels of EAs during fall affects thermoregulation, behavior, and weight gain. Building on these findings, the current study aimed to investigate how the presence of low EA-producing E+ or NT endophytes can influence animal metabolome,

Toward Sustainable Management of Spodoptera frugiperda (Lepidoptera: Noctuidae): A Review on the Role of Endophytic Fungi in Crop Protection.

The fall armyworm (FAW), Spodoptera frugiperda, is a highly polyphagous pest that has rapidly expanded across Africa, Asia, and Oceania, threatening food security and agricultural productivity. Reliance on synthetic insecticides for FAW management is increasingly challenged by insecticide resistance, environmental contamination, and adverse effects on non-target organisms and human health. This review synthesizes current knowledge on the potential of endophytic fungi as a sustainable strategy for FAW management. Following a systematic literature search, 59 original research articles published

Influence of Plant Age and Endophyte Status on the Nematotoxicity of Festulolium loliaceum to Trichodorus primitivus and Quantification of Active Phytochemicals.

Festulolium hybrids are cool-season forage grasses that form symbiotic relationships with the fungus Epichloë uncinata, which produces loline alkaloids that protect the host from herbivores. This study evaluated the nematotoxicity of shoot and root extracts of Festulolium loliaceum against the stubby root nematode Trichodorus primitivus. Methanolic root and shoot extracts from plants aged 8, 12, 16, and 20 weeks were tested in vitro at five concentrations (312.5-5000 µg mL-1) over 24, 48, and 72 h. Interaction of ergovaline with serotonin receptor 5-HT2A in bovine ruminal and mesenteric vasculature.

Ergot alkaloids from endophyte-infected (Epichloë coenophiala) tall fescue (Lolium arundinaceum) induce vasoconstriction. Previous work has shown that serotonin receptor subtype, 5HT2A, is present in bovine ruminal (R) and mesenteric (M) vasculature, plays a role in vasoconstriction, and could be influenced by ergot alkaloids. To determine the influence of ergot alkaloids on 5HT2A, the vasoactivity of an agonist selective for 5HT2A, (4-bromo-3,6-dimethoxybenzocyclobuten-1-yl) methylamine HCl (TCB-2), was evaluated using bovine ruminal and mesenteric arteries and veins (RA, RV, MA, MV) that wer
