Characterization and antimicrobial resistance of Staphylococcus hyicus from swine exudative epidermitis in South Korea.

BACKGROUND: Staphylococcus hyicus causes porcine exudative epidermitis, predominantly affecting suckling and weaned piglets. This bacterium produces various exfoliative toxins (ExhA, ExhB, ExhC, ExhD, SHETA, and SHETB), which are responsible for the clinical manifestations of exudative epidermitis. However, treatment failure is common due to frequent antimicrobial resistance in porcine strains. Therefore, this study aimed to identify the genes encoding exfoliative toxins and assess the antimicrobial resistance profiles of S. hyicus. A total of 17 S. hyicus isolates were collected from piglets

Cloning of swine desmoglein 1 and its direct proteolysis by Staphylococcus hyicus exfoliative toxins isolated from pigs with exudative epidermitis.

Exudative epidermitis (EE) is an acute, often fatal skin disease of piglets caused by Staphylococcus hyicus. Clinical and histopathological manifestations of EE are similar to those of staphylococcal scalded skin syndrome (SSSS), a human blistering skin disease, in which exfoliative toxins produced by Staphylococcus aureus digest the extracellular domains of desmoglein (Dsg) 1 and cause loss of epidermal cell-cell adhesion. The aims of this study were to isolate and characterize cDNA for full length of swine Dsg1, and to determine whether the extracellular domains of swine Dsg1 produced by bac

Ulcerative glossitis and stomatitis associated with exudative epidermitis in suckling swine.

Thirty piglets 1 to 4 weeks old from five herds had epidermal, foot, conjunctival and renal lesions typical of exudative epidermitis. Ten piglets had a large central ulceration of the dorsum of the tongue. Three piglets had multiple erosions of the hard palate and one had mucoid degeneration of the urinary bladder epithelium and a thick viscous material in the renal pelvis, ureters and urinary bladder.

Complete genome sequences of 22 Staphylococcus spp. bacteriophages isolated from a single swine farm environment in Quebec, Canada.

Twenty-two bacteriophages infecting Staphylococcus spp. were isolated from water and manure collected on a Quebec swine farm affected by exudative epidermitis. Phylogenetic analysis of their genomes showed that 20 of them clustered into two groups, Twort- and ShyM-like. No CRISPR-Cas systems, antimicrobial resistance determinants, or virulence genes were identified.

A review of porcine circovirus 2-associated syndromes and diseases.

Clinical expression of porcine circovirus 2 (PCV2) infection in swine may result in several distinct syndromes and diseases including post-weaning multisystemic wasting syndrome (PMWS), porcine dermatitis and nephropathy syndrome (PDNS), reproductive failure, porcine respiratory disease complex, granulomatous enteritis, necrotizing lymphadenitis, and possibly exudative epidermitis. Association of PCV2 with congenital tremor in piglets is still controversial. The extent of the involvement of PCV2 in swine disease other than PMWS is currently poorly understood. This review concentrates on PCV-2-

Exudative Epidermitis in Combination with Staphylococcal Pyoderma in Suckling Piglets.

A case of generalized exudative epidermitis (EE) is described, which occurred in a very small piglet producing farm in Austria. The antimicrobial treatment prescribed by the herd veterinarian did not improve the clinical problem. Therefore, the University Clinic for Swine intervened in the case. Lab investigations were initiated in which Staphylococcus hyicus (SH) and Staphylococcus aureus (SA), both methicillin-resistant and susceptible strains, could be isolated from the skin of affected piglets. Poor hygiene and management practices were identified as predisposing factors on site. Adaptatio

Insights into the Virulence and Antimicrobial Resistance of Staphylococcus hyicus Isolates from Spanish Swine Farms.

Staphylococcus hyicus is a significant pathogen in swine, primarily causing exudative epidermitis. Addressing S. hyicus infections requires both the characterization of virulence and antimicrobial resistance (AMR) in farm-recovered isolates. This study aimed to characterize the virulence, AMR, and biofilm formation of S. hyicus isolates from Spanish swine farms. A total of 49 isolates were analyzed, originating from animals with cutaneous, reproductive, and systemic clinical signs. Half of the isolates (49.0%) were positive for at least one virulence factor (VF) gene, with SHETA being the most

Diversity of tet(L)-bearing plasmids found in Eastern Canadian Staphylococcus hyicus isolates from swine.

Staphylococcus hyicus is the etiological agent of exudative epidermitis (EE) in suckling and weaned piglets, which is an endemic disease of concern for animal health and welfare on swine farms. Staphylococcus hyicus is not widely studied, and there is no recent scientific literature on Canadian strains. In a preliminary study, we presented an Eastern Canadian S. hyicus strain isolated from an EE case and bearing a plasmidic tet(L) gene conferring resistance to tetracyclines, one of the most commonly used antibiotics in Canadian swine farms. The current study characterized the tet(L)-bearing pl

Systematic characterization of four novel Twort-group bacteriophages infecting Staphylococcus hyicus.

Exudative epidermitis (EE) in piglets, caused by the bacterium Staphylococcus hyicus, remains a major concern for the swine industry, particularly due to antimicrobial resistance. Bacteriophages represent a promising alternative treatment. This study aimed to isolate and characterize phages infecting S. hyicus from EE-affected swine farm environments in Quebec. Four novel phages (BQE1, BQE4, BQE7, BQL4) were isolated from water and manure samples. All produced clear plaques (<1 mm) and reached titers of 108-1010 PFU/mL. Electron microscopy revealed myovirus morphology. Their genomes (115.4-134

Staphylococcal exfoliative toxins: "molecular scissors" of bacteria that attack the cutaneous defense barrier in mammals.

Bullous impetigo and its generalized form, staphylococcal scalded-skin syndrome (SSSS), are highly contagious, blistering skin diseases caused by Staphylococcus aureus infection. Virulent strains of the bacteria produce exfoliative toxins (ETs) that cause the loss of keratinocyte cell-cell adhesion in the superficial epidermis. Recent studies have indicated that the three isoforms of ETs, i.e., ETA, ETB, and ETD, are glutamate-specific serine proteases that specifically and efficiently cleave a single peptide bond in the extracellular region of human and mouse desmoglein 1 (Dsg1), a desmosomal

Design and Pharmacodynamics of Recombinant Fungus Defensin NZL with Improved Activity against Staphylococcus hyicus In Vitro and In Vivo.

Staphylococcus hyicus is recognized as a leading pathogen of exudative epidermitis in modern swine industry. Antimicrobial peptides are attractive candidates for development as potential therapeutics to combat the serious threats of the resistance of S. hyicus. In this study, a series of derivatives were designed based on the NZ2114 template with the aim of obtaining peptides with more potent antimicrobial activity through changing net positive charge or hydrophobicity. Among them, a variant designated as NZL was highly expressed in Pichia pastoris (P. pastoris) with total secreted protein of

Staphylococcus pseudintermedius exfoliative toxin EXI selectively digests canine desmoglein 1 and causes subcorneal clefts in canine epidermis.

Staphylococcal exfoliative toxins are known to digest desmoglein (Dsg) 1, a desmosomal cell-cell adhesion molecule, thus causing intraepidermal splitting in human bullous impetigo, staphylococcal scalded skin syndrome and swine exudative epidermitis. Recently, a novel exfoliative toxin gene (exi), whose sequence shares significant homology with previously identified exfoliative toxins, was isolated from Staphylococcus pseudintermedius. Little is known about the pathogenic involvement of this toxin in canine pustular diseases such as impetigo. The aim of this study was to determine whether EXI,

Three Staphylococcus Bacteriophages Isolated from Swine Farm Environment in Quebec, Canada, Infecting S. chromogenes.

Exudative epidermitis (EE), caused by Staphylococcus hyicus, represents an issue for swine production, particularly due to antimicrobial resistance. In this project, we isolated bacteriophages using S. hyicus as host and studied them as a potential alternative to antibiotic treatment in Quebec, Canada. Three phages, STAE-4, STAF-3, and STAM-1, were isolated from swine farm samples using a single S. hyicus strain (SC366) as the host. Transmission electron microscopy revealed that all three phages exhibited a siphovirus-like morphology, and RAPD-PCR profiling indicated that the phages were genet

Antibiotic-resistance and plasmids in Staphylococcus hyicus isolated from pigs with exudative epidermitis and from healthy pigs.

A total of 100 S. hyicus strains isolated from healthy piglets and piglets with exudative epidermitis originating from 100 different herds was examined for drug-resistance and prevalence of plasmids. Resistance to macrolide/linosamide antibiotics could be related to plasmids in 55 (93%) of the 59 resistant strains: A plasmid of 2.4 kb mediating resistance to macrolides and lincosamides was observed in 25 strains, and a plasmid of 11.5 kb mediating resistance to both macrolides/lincosamides and tetracycline was observed in 30 strains. A plasmid with a molecular weight of 4.5 kb was shown by cur

Staphylococcal epidermolysins.

PURPOSE OF REVIEW: Staphylococcal epidermolysins are the major causative toxins of bullous impetigo and staphylococcal scalded skin syndrome. This disease is characterized by the splitting of the epidermis between two cell layers resulting in exfoliation. It predominantly affects newborn babies and exposes them to secondary infections. This leads to the risk of epidemics, especially in nurseries. With only an experimental model which consists of skin injections in newborn mice and the recent determination of three-dimensional structures, the essential function of these toxins remained controve

Lesions of porcine necrotic ear syndrome.

Porcine necrotic ear syndrome is a disease of swine characterized by large erosive lesions at the margin of the pinna(e). The gross and microscopic characteristics of the lesions were studied in 38 pigs selected from eight affected swine herds. The progression of the lesions was examined in a herd of 174 weaned pigs in a total confinement nursery. The lesions began as a superficial vesicular dermatitis associated with superficial auricular trauma and progressed to become exudative and encrusted. Localized lesions slowly healed or sporadically progressed to deep necrotic ulcers. The early lesio

[Increased piglet losses upon exudative epidermitis - a case report].

A massive outbreak of exudative epidermitis (EE) occurred on a Western German piglet producing farm with 350 productive sows. Gilts are produced on site. In one group of piglets, more than 50% of suckling and nursery piglets were clinically affected; furthermore, gilts as well as sows showed localized blackish-squamous skin lesions in the neck area. Generalized infection in suckling and nursery piglets resulted in mortality rates of up to 10% per weaning group. Swabs of moist, affected areas of skin taken on the farm in addition to swab and organ samples collected during necropsy were examined

A highly pathogenic strain of Staphylococcus sciuri caused fatal exudative epidermitis in piglets.

Staphylococcus sciuri are important human pathogens responsible for endocarditis, peritonitis, septic shock, urinary tract infection, pelvic inflammatory disease and wound infections. However, little information is known regarding the pathogenicity of S. sciuri to animals. From the pericardial fluid of a diseased piglet with exudative epidermitis (EE), we isolated a strain of Staphylococcus in pure culture. Surprisingly, this isolate was a member of S. sciuri rather than S. hyicus as identified by its biochemical traits and also by analysis of 23S ribosomal DNA using Internal Transcribed Space
