Erwinia amylovora, the causative agent of fire blight, causes significant economic losses for farmers worldwide by inflicting severe damage to the production and quality of plants in the Rosaceae family. Historically, fire blight control has primarily relied on the application of copper compounds and antibiotics, such as streptomycin. However, the emergence of antibiotic-resistant strains and growing environmental concerns have highlighted the need for alternative control methods. Recently, there has been a growing interest in adopting bacteriophages (phages) as a biological control strategy.

This study investigated whether AcrR-mediated regulation of the acrAB efflux system is associated with tetracycline susceptibility in Erwinia amylovora. We compared tetracycline susceptibility, acrAB expression, intracellular tetracycline accumulation, intracellular pH, ATP-associated luminescence, envelope-associated phenotypes, virulence, and responses to exogenous acyl-homoserine lactones (AHLs) among the wild type (WT), an acrR mutant (ΔacrR), the same mutant after prior tetracycline exposure (ΔacrRTc), and an acrR-complemented mutant (ΔacrR(acrR)). Relative to WT, ΔacrR showed fourfold in

Erwinia amylovora causes fire blight in rosaceous plants. In nonhost Arabidopsis thaliana, E. amylovora triggers necrotic symptoms associated with transient bacterial multiplication, suggesting either that A. thaliana lacks a susceptibility factor or that it actively restricts E. amylovora growth. Inhibiting plant protein synthesis at the time of infection led to an increase in necrosis and bacterial multiplication and reduced callose deposition, indicating that A. thaliana requires active protein synthesis to restrict E. amylovora growth. Analysis of the callose synthase-deficient pmr4-1 muta

rsmB(Ecc) specifies a nontranslatable RNA regulator that controls exoprotein production and pathogenicity in soft rot-causing Erwinia carotovora subsp. carotovora. This effect of rsmB(Ecc) RNA is mediated mostly by neutralizing the function of RsmA(Ecc), an RNA-binding protein of E. carotovora subsp. carotovora, which acts as a global negative regulator. To determine the occurrence of functional homologs of rsmB(Ecc) in non-soft-rot-causing Erwinia species, we cloned the rsmB genes of E. amylovora (rsmB(Ea)) and E. herbicola pv. gypsophilae (rsmB(Ehg)). We show that rsmB(Ea) in E. amylovora po

The bacterium Erwinia amylovora is responsible for the destructive disease known as fire blight in pear trees. This highly detrimental condition poses a significant threat to the health and vitality of these trees. The existing strategies for managing fire blight disease involve the regular use of copper compounds and streptomycin, particularly during periods when environmental factors are conducive to the spread of the infection. Silver nanoparticles, also known as SNPs, are tiny specks of silver ranging in size from 10 to 100 nm. These particles are created through various chemical and biolo

Erwinia amylovora causes fire blight, a destructive disease of rosaceous plants very difficult to control. We demonstrated that copper, employed to control plant diseases, induces the "viable-but-nonculturable" (VBNC) state in E. amylovora. Moreover, it was previously reported that copper increases production of its main exopolysaccharide (EPS), amylovoran. In this work, the copper-complexing ability of amylovoran and levan, other major EPS of E. amylovora, was demonstrated. Following this, EPS-deficient mutants were used to determine the role of these EPSs in survival of this bacterium in AB

Understanding the phage resistance mechanisms in Erwinia amylovora is crucial for improving phage therapy applications targeting this plant pathogen, which is responsible for fire blight disease in apple and pear orchards. This study investigated E. amylovora phage resistance in the presence of peptidylarginine deiminase (PAD) inhibitors, which have been found in other studies to prevent the conversion of arginine to citrulline as a post-translational modification that may initiate outer membrane vesical (OMV) formation. Infection of the E. amylovora Ea6-4 strain by ϕEa21-4 phages, in the pres

Fire blight, caused by the Gram-negative bacterium Erwinia amylovora, is one of the most destructive diseases of pome-fruit crops worldwide. Despite decades of research, its management remains challenging because efficient and sustainable control options are limited, and many commercially important apple and pear cultivars remain highly susceptible. Conventional treatments for bacterial disease suppression are often hindered by inconsistent efficacy, phytotoxic effect, environmental concerns and regulatory restrictions, necessitating new sustainable alternative options. Virulent bacteriophages

Erwinia amylovora, the bacterial species responsible for fire blight, causes major economic losses in pome fruit crops worldwide. Chemical control is not always effective and poses a serious threat to the environment and human health. Social demands for eco-sustainable and safe control methods make it necessary to search for new biocontrol strategies such as those based on antagonists. A bacterial collection from different fire blight-free Mediterranean environments was tested for antagonistic activity against Spanish strains of E. amylovora. Antagonistic assays were carried out in vitro in cu

Erwinia amylovora, the causal agent of fire blight, poses a severe and ongoing threat to the global production of Rosaceous crops. We synthesized 54 garlic-derived disulfides and identified compound b as the most potent candidate (EC50 = 2.85 μg/mL), which significantly surpassed allicin and zhongshengmycin (ZSM). In planta assays on Cuiguan pear demonstrated that b provided exceptional curative and protective efficacy, effectively suppressing fire blight infections in flowers, leaves, shoots, and fruits. Mechanistically, b triggered lethal ROS accumulation by inhibiting catalase (CAT) and sup

BACKGROUND: The Gram-negative bacterium Erwinia amylovora induces fire blight disease in Rosaceae hosts, severely damaging pome fruit trees including pear, apple, and hawthorn cultivars. Traditional control methods, such as antibiotics and copper-based agents, are associated with plant damage and the emergence of resistant bacterial strains, and present significant challenges. This study aims to design and synthesize a novel eco-friendly bactericide.
RESULTS: In this study, we designed and synthesized 30 small-molecule compounds by combining 1,3,4-thiadiazole, a key moiety of zinc thiazole, w

Erwinia amylovora (EA) is a phytopathogenic bacterium, the causative agent of bacterial fire blight, a disease that affects Rosaceaes. In order to replace antibiotics and copper, the antimicrobial activity of three extracts of Moringa oleifera Lam., methanolic (MeOH-MOE), hydroalcoholic (HA-MOE) and hydroalcoholic with maltodextrins (HAMD-MOE), was tested on eleven strains of EA isolated from apple trees by the Emilia-Romagna Phytosanitary Department. MIC and MBC have been evaluated; biofilm formation, swarming motility and amylovoran production were performed with the crystalviolet, soft-agar

Fire blight, caused by the bacterium Erwinia amylovora, is a disease devastating the production of rosaceous crops, primarily apple and pear, with worldwide distribution. Fire blight begins in the spring when primary inoculum is produced as ooze, which consists of plant sap, E. amylovora, and exopolysaccharides. Ooze is believed to be transferred to healthy tissues by wind, rain, and insects. However, the mechanisms by which insects locate and transmit ooze are largely undocumented. The goals of this study were to investigate the biological factors affecting acquisition of E. amylovora from oo

Erwinia amylovora, the causal agent of fire blight, is an enterobacterial pathogen of Rosaceous plants including apple and pear. We have been studying the response of E. amylovora to oxidative stress because, during infection, the bacterium elicits an oxidative burst response in host plants. During the screening of a transposon mutant library for hydrogen peroxide sensitivity, we identified a mutant carrying an insertion in waaL, a gene involved in lipopolysaccharide biosynthesis, that was more sensitive to hydrogen peroxide than the parental wild-type strain. We also confirmed that a waaL mut

We assessed the effect of three antibiotics (streptomycin, oxytetracycline, and kasugamycin) on populations of the fire blight pathogen Erwinia amylovora on apple flower stigmas during three field seasons. Application timing relative to E. amylovora presence on flower stigmas had little impact on population dynamics and subsequent disease incidence. Although E. amylovora populations on water-treated flowers increased to 106-7 cfu flower-1 after 4 to 5 days during each experiment, the antibiotics streptomycin and kasugamycin caused statistically significant reductions in stigma populations by a

Iron is crucial for bacterial growth and virulence. Under iron-deficiency bacteria produce siderophores, iron chelators that facilitate the iron uptake into the cell via specific receptors. Erwinia amylovora, the causative agent of fire blight, produces hydroxamate-type desferrioxamine siderophores (DFO). The presented study reassesses the impact of DFO as a virulence factor of E. amylovora during its epiphytic phase on the apple flower. When inoculated in semisterile Golden Delicious flowers no difference in replication and induction of calyx necrosis could be observed between E. amylovora CF

With the rise of antibiotic-resistant pathogens, bacterial infections have become an increasingly significant threat. Bacteriophages have the potential to complement the current treatment options. To ensure that phage biocontrol is efficient and long-lasting, the risk of resistance development must be minimized, and phage resistance mechanisms should be thoroughly investigated. In this study, a Tn5 transposon mutagenesis library of the fire blight pathogen Erwinia amylovora was screened to identify multi-phage-resistant mutants. These mutants had transposon insertions in the genes Eamy_RS26425

Fire blight, caused by Erwinia amylovora, is one of the major bacterial disease of apple and pear, causing enormous economic losses worldwide. Several control measures against E. amylovora have been reported till date, however, none of them have proved to be effective significantly against the pathogen. In this study, mechanisms of the copper-based control agents (CBCAs): copper oxychloride (COCHL), copper oxide (COX), copper hydroxide (CHY), copper sulfate basic (CSB), and tribasic copper sulfate (TCS) and their disease severity reduction efficacy against E. amylovora were analyzed. Fire blight, caused by Erwinia amylovora, is a significant threat to fruit crops, with limited biocontrol methods. This study aimed to develop a nanosystem using mesoporous silica nanoparticles (MSNs) loaded with a phenolic plant extract (ZP) derived from Myrtus communis, Thymus vulgaris, and Curcuma longa, and coated with natural biopolymers Gum Tragacanth (GT) and sodium alginate (SA). The MSNs were synthesized and characterized by XRD, FTIR, and TEM, exhibiting a specific surface area of about 750 m2/g and an average pore diameter of 5 nm. ZP was effectively loaded into the MSNs with a load
