[Substance P in the central mechanisms of the escape reaction].

The role of substance P in the central mechanisms of escape reaction elicited by electrical stimulation of the ventromedial hypothalamus was investigated in chronic experiments on rabbits. Intravenous injection of substance P (30 micrograms/kg) led to a short-term (less than 10 min) increase in the threshold of stimulation of the ventromedial hypothalamus and to more stable (up to 1.5 h) disorders of the hippocampal-hypothalamic relations. After substance P injection the inhibitory effects of the dorsal hippocampus but not the facilitating influences of the midbrain reticular formation on exci

A Conformational Escape Reaction of HIV-1 against an Allosteric Integrase Inhibitor.

HIV-1 often acquires drug-resistant mutations in spite of the benefits of antiretroviral therapy (ART). HIV-1 integrase (IN) is essential for the concerted integration of HIV-1 DNA into the host genome. IN further contributes to HIV-1 RNA binding, which is required for HIV-1 maturation. Non-catalytic-site integrase inhibitors (NCINIs) have been developed as allosteric IN inhibitors, which perform anti-HIV-1 activity by a multimodal mode of action such as inhibition of the IN-lens epithelium-derived growth factor (LEDGF)/p75 interaction in the early stage and disruption of functional IN multime

Mucus escape reaction that involves the mandible: a case report and diagnostic imaging considerations.

We present a case of mucus escape reaction in which we encountered difficulty in interpreting the images acquired by plain radiography, computed tomography, and magnetic resonance imaging. The 48-year-old male Japanese patient was referred for evaluation of a gradual swelling subjacent to the inferior border of the left mandible. At the early imaging examinations, magnetic resonance imaging provided information crucial to resolving the issue of whether the lesion consisted of a central malignant disease process or a malignant disease in the submandibular space or both of these two separate dis

Varied influence of damage to the motor cortex on precision avoidance and escape reactions in dogs.

The influence of local extirpation of the representation of the forelimb in the motor area of the cortex on the performance of precision elevations of this limb was investigated. Even imprecise avoidance reactions to an acoustic signal did not recover spontaneously. Irregular motoric reactions which were insufficient in amplitude could easily be restored in the course of retraining, but precise movements (both rapid fused, as well as relatively slow stepwise movements) did not recover for at least a half year after the operation. By contrast with this, precise rapid reactions in response to el

Regulatory Programs of B-cell Activation and Germinal Center Reaction Allow B-ALL Escape from CD19 CAR T-cell Therapy.

Chimeric antigen receptor (CAR) T-cell therapy has led to tremendous successes in the treatment of B-cell malignancies. However, a large fraction of treated patients relapse, often with disease expressing reduced levels of the target antigen. Here, we report that exposing CD19+ B-cell acute lymphoblastic leukemia (B-ALL) cells to CD19 CAR T cells reduced CD19 expression within hours. Initially, CD19 CAR T cells caused clustering of CD19 at the T cell-leukemia cell interface followed by CD19 internalization and decreased CD19 surface expression on the B-ALL cells. CD19 expression was then repre

Deep brain stimulation of the dorsal raphe inhibits avoidance and escape reactions and activates forebrain regions related to the modulation of anxiety/panic.

One of the main neurochemical systems associated with anxiety/panic is the serotonergic system originating from the dorsal raphe nucleus (DR). Previous evidence suggests that the DR is composed of distinct subpopulations of neurons, both morphologically and functionally distinct. It seems that mainly the dorsal region of the DR (DRD) regulates anxiety-related reactions, while lateral wings DR (lwDR) serotonin (5-HT) neurons inhibit panic-related responses. In this study we used the technique of deep brain stimulation (DBS) to investigate the role played by the DRD and lwDR in defense. Male Wis

Post-contact immobility and half-lives that save lives.

A wide variety of animals become completely immobile after initial contact with a potential predator. This behaviour is considered to be a last-ditch escape strategy. Here, we test the hypothesis that such immobility should have an extremely unpredictable duration. We find that it spans more than three orders of magnitude in antlion larvae. We also analyse the second period of immobility that follows the first bout of immobility, and consider the distributions of both first and second immobility periods within the context of the intermittence that characterizes the movement of most organisms.

Kinematics of rotation in place during defense turning in the crayfish Procambarus clarkii.

The kinematic patterns of defense turning behavior in freely behaving specimens of the crayfish Procambarus clarkii were investigated with the aid of a video-analysis system. Movements of the body and all pereiopods, except the chelipeds, were analyzed. Because this behavior approximates to a rotation in place, this analysis extends previous studies on straight and curve walking in crustaceans. Specimens of P. clarkii responded to a tactile stimulus on a walking leg by turning accurately to face the source of the stimulation. Angular velocity profiles of the movement of the animal's carapace s

Neural Circuitry that Evokes Escape Behavior upon Activation of Nociceptive Sensory Neurons in Drosophila Larvae.

Noxious stimuli trigger a stereotyped escape response in animals. In Drosophila larvae, class IV dendrite arborization (C4 da) sensory neurons in the peripheral nervous system are responsible for perception of multiple nociceptive modalities, including noxious heat and harsh mechanical stimulation, through distinct receptors [1-9]. Silencing or ablation of C4 da neurons largely eliminates larval responses to noxious stimuli [10-12], whereas optogenetic activation of C4 da neurons is sufficient to provoke corkscrew-like rolling behavior similar to what is observed when larvae receive noxious st

Rolling away: a novel context-dependent escape behaviour discovered in ants.

For animals facing dangers, the best option to optimize costs and benefits of defence sometimes may be avoidance. Here we report the discovery of a peculiar strategy adopted by Myrmecina graminicola, a cryptic ant living in forest floor. Experiments showed that when disturbed these ants respond with immobility. However, upon perceiving disturbance but under specific inclinations of the substrate, they shift to an active escaping strategy: rolling away. This is a context-dependent behaviour adopted only in appropriate circumstances. During rolling, the ants assume a ball-like shape using antenn

Dissociation of interference with the speed and accuracy of escape produced by inescapable shock.

Benzodiazepines and naltrexone administered before inescapable shock block behavioral consequences of the inescapable shock such as poor shuttle box escape, reduced activity in reaction to shock, reduced social interaction, and so on. Anxiogenic beta-carboline derivatives such as FG-7142 can produce these effects by themselves. In the present study, neither diazepam nor naltrexone had any effect on the interference with Y-maze choice escape accuracy produced by inescapable shock even though they both eliminated the reduction in Y-maze escape response speed produced by inescapable shock. Visual Threat Assessment and Reticulospinal Encoding of Calibrated Responses in Larval Zebrafish.

All visual animals must decide whether approaching objects are a threat. Our current understanding of this process has identified a proximity-based mechanism where an evasive maneuver is triggered when a looming stimulus passes a subtended visual angle threshold. However, some escape strategies are more costly than others, and so it would be beneficial to additionally encode the level of threat conveyed by the predator's approach rate to select the most appropriate response. Here, using naturalistic rates of looming visual stimuli while simultaneously monitoring escape behavior and the recruit

Reef fish escape responses selectively match predator attack speeds.

Animals must continually balance foraging with the risk of predation. In complex natural environments, this means quickly distinguishing between threats and harmless situations. We investigated how site-associated coral reef fishes escape in response to visual cues mimicking predator attacks, using controlled underwater presentations of looming stimuli at varying speeds. We measured escape responses across species and social contexts, and compared them to predator attack speeds observed in the same habitat. Escape probability increased sharply with the speed of the looming stimulus, with no re

Innate visual attraction before, during and after escape from adverse substrates in carpenter ants.

Many animals exhibit an innate attraction to dark areas or objects, driving orientation behaviours such as beacon aiming. In ants, some species do not appear to display beacon aiming. Here, we show that in one such species, Camponotus japonicus, the behaviour is triggered when crossing liquid-covered surfaces, regardless of locomotor pattern and the presence of water in the liquid. Once initiated, beacon aiming persisted even after the ants transitioned from water to dry substrates, as evidenced by their reorientation towards a displaced beacon. Beacon aiming could be observed before the ants

Synergistic effects of musking and autohaemorrhaging on the duration of death feigning in dice snakes (Natrix tessellata).

Predation exerts a significant selection pressure on prey, shaping a multitude of traits that serve as antipredator defences. In turn, natural selection could favour combinations of antipredator defences with synergistic effects that enhance prey survival. An especially interesting antipredator defence is death feigning (DF), present in a wide variety of taxa and usually characterized by the prey lying motionless often along with defaecation, musking and autohaemorrhaging (AH). All these aspects of the DF display should work in conjunction with one another, intensifying the overall effect of t

Neural circuits underlying habituation of visually evoked escape behaviors in larval zebrafish.

Larval zebrafish that are exposed repeatedly to dark looming stimuli will quickly habituate to these aversive signals and cease to respond with their stereotypical escape swims. A dark looming stimulus can be separated into two independent components: one that is characterized by an overall spatial expansion, where overall luminance is maintained at the same level, and a second, that represents an overall dimming within the whole visual field in the absence of any motion energy. Using specific stimulation patterns that isolate these independent components, we first extracted the behavioral alg

Not so fast: giant interneurons control precise movements of antennal scales during escape behavior of crayfish.

High-speed video recordings of escape responses in freely behaving crayfish revealed precisely coordinated movements of conspicuous head appendages, the antennal scales, during tail-flips that are produced by giant interneurons. For tail-flips that are generated by the medial giants (MG) in response to frontal attacks, the scales started to extend immediately after stimulation and extension was completed before the animal began to propel backwards. For tail-flips that are elicited by caudal stimuli and controlled by the lateral giants (LG), scale extensions began with significant delay after t
