Phase-variable outer membrane proteins in Escherichia coli.

Escherichia coli contains at least two phase-variable proteins in its outer membrane. One, termed antigen 43 (Ag43), is the product of the metastable flu gene located at min 43.6 on the E. coli chromosome and is responsible for colony form variation and for autoaggregation in liquid media. Ag43 is composed of two proteinaceous subunits, alpha 43 and beta 43 in 1:1 stoichiometry. alpha 43 (apparent M(r) 60,000) is surface expressed, extends beyond the O-side chains of smooth lipopolysaccharide and is bound to the cell surface through an interaction with beta 43 (apparent M(r) 53,000), itself an

Unique and overlapping pollutant stress proteins of Escherichia coli.

Exposure of growing batch cultures of Escherichia coli to nine different "model micropollutants" (benzene, cadmium chloride, chlorpyrivos, 2,4-dichloroaniline, dioctylphtalate, hexachlorobenzene, pentachlorophenol, trichloroethylene, and tetrapropylbenzosulfonate) led to the induction of 13 to 39 proteins, as analyzed by two-dimensional gel electrophoresis. Some of these proteins overlapped with heat shock and carbon starvation proteins, but at least 50% were unique to a given chemical. The stress protein induction showed a temporal pattern, indicating sequential gene expression. Chemical stre

Heterodimer formation between Escherichia coli Rep and UvrD proteins.

DNA helicases catalyze the essential process of unwinding duplex DNA to form the single-stranded DNA intermediates required for DNA metabolic processes including replication, recombination, and repair. Most cells, possibly all, encode multiple helicases that function selectively in different processes, although some helicases can complement each other in vivo. Thus, although Escherichia coli can survive mutations or deletions of either the uvrD gene (encoding Helicase II) or the rep gene (encoding Rep helicase) separately, deletion of both rep and uvrD genes is lethal (Washburn, B. K., and Kus

Cooperative Immune Suppression by Escherichia coli and Shigella Effector Proteins.

The enteric attaching and effacing (A/E) pathogens enterohemorrhagic Escherichia coli (EHEC) and enteropathogenic E. coli (EPEC) and the invasive pathogens enteroinvasive E. coli (EIEC) and Shigella encode type III secretion systems (T3SS) used to inject effector proteins into human host cells during infection. Among these are a group of effectors required for NF-κB-mediated host immune evasion. Recent studies have identified several effector proteins from A/E pathogens and EIEC/Shigella that are involved in suppression of NF-κB and have uncovered their cellular and molecular functions. A nove

Chemotaxis in Escherichia coli: associations of protein components.

Interactions between protein components of the chemotaxis mechanism in Escherichia coli were investigated by using the cleavable cross-linking reagent, dithiobis(succinimidyl propionate). Two methods were used to allow detection of chemotaxis-specific proteins in intact cells. The first method was to program their synthesis in the presence of [35S]methionine using lambda E. coli hybrid phages which carry the chemotaxis genes. The second method was to label endogenous methyl-accepting chemotaxis proteins (MCP's), with the methyl donor S-adenosyl-L-[methyl-3H]methionine, after permeabilizing the

Muconic Acid Production Using Gene-Level Fusion Proteins in Escherichia coli.

This study reports on the improving of muconic acid (MA) production by using metabolically engineered Escherichia coli. Three MA synthesis pathways separately were introduced into E. coli. After 72 h of cultivation, two of the three strains, i.e., one carrying the Pathway 1 (1.00 g/L) and the other carrying the Pathway 3 (1.34 g/L) produced MA. To increase MA production, the enzymes of the shikimate pathway (AroC and AroD) were overexpressed in these strains. Although the overexpression of AroC increased the MA production (1.59 g/L) by the Pathway 1, AroD overexpression decreased it by the Pat

Effects of chlorpromazine on the cell envelope proteins of Escherichia coli.

Chlorpromazine (CPZ), at a concentration of 60 micrograms/ml of medium completely inhibited the replication of Escherichia coli. At concentrations below this MIC, CPZ caused transient induction of filamentation, such that by the end of 5 h, all of the cells were filaments, and by the end of 24 h, only rod-shaped E. coli were present. The reversion to normal morphology in the presence of CPZ was not due to either the degradation of CPZ or the selection of CPZ-resistant mutants. The electrophoretic pattern of proteins extracted from isolated cell envelopes of CPZ-induced filaments as well as fro

Genomic island-encoded regulatory proteins in enterohemorrhagic Escherichia coli O157:H7.

Enterohemorrhagic Escherichia coli (EHEC) is an important zoonotic pathogen that is a major cause of foodborne diseases in most developed and developing countries and can cause uncomplicated diarrhoea, haemorrhagic colitis, and haemolytic uraemic syndrome. O islands (OIs), which are unique genomic islands in EHEC O157:H7, are composed of 177 isolated genomic features and harbour 26% of the total genes that are absent in the non-pathogenic E. coli K-12 genome. In the last twenty years, many OI-encoded proteins have been characterized, including proteins regulating virulence, motility, and acid

Enterotoxigenic Escherichia coli infection induces tight junction proteins expression in mice.

Enterotoxigenic Escherichia coli (ETEC) causes diarrhea in travelers, young children and piglets, but the precise pathogenesis of ETEC induced diarrhea is not fully known. Recent investigations have shown that tight junction (TJ) proteins and aquaporin 3 (AQP 3) are contributing factors in bacterial diarrhea. In this study, using immunoblotting and immunohistochemistry analyses, we found that ETEC increases the protein abundance of TJ proteins (occludin, claudin-1, zonula occludens-1) in mice. Enterotoxigenic Escherichia coli induced the expression of TJ proteins in mice through pathways by in

SPOR Proteins Are Required for Functionality of Class A Penicillin-Binding Proteins in Escherichia coli.

Sporulation-related repeat (SPOR) domains are present in many bacterial cell envelope proteins and are known to bind peptidoglycan. Escherichia coli contains four SPOR proteins, DamX, DedD, FtsN, and RlpA, of which FtsN is essential for septal peptidoglycan synthesis. DamX and DedD may also play a role in cell division, based on mild cell division defects observed in strains lacking these SPOR domain proteins. Here, we show by nuclear magnetic resonance (NMR) spectroscopy that the periplasmic part of DedD consists of a disordered region followed by a canonical SPOR domain with a structure simi

SsrA-Based Targeted Protein Degradation in Escherichia coli.

Targeted protein degradation is an emerging concept of drug discovery to selectively eliminate the pathogenic proteins by activating their degradation in cells. The proteolysis-targeting chimeras (PROTACs) are bifunctional small molecules that induce the degradation of a protein of interest (POI) by proteasome. Here we describe a method that utilizes an ssrA-based BacPROTAC (bacterial PROTACs) to target the drug-resistant proteins CTX-M-14, a class A β-lactamase commonly found in extended-spectrum beta-lactamase (ESBL) plasmids of antimicrobial resistance (AMR) bacteria. This strategy could re

Protein Expression Profiles of Antiseptic-Adapted Escherichia coli.

Repeated exposure to subinhibitory concentrations of antiseptics may lead to reduced susceptibility or resistance and potentially promote cross-resistance to antibiotics. However, the underlying molecular mechanisms remain incompletely understood. This study investigated whether antiseptic-adapted Escherichia coli strains exhibit altered protein expression profiles compared with wild-type (WT) E. coli. Protein expression was analysed in E. coli strains previously adapted over ten passages to subinhibitory concentrations of chlorhexidine (CHX), cetylpyridinium chloride (CPC), and benzalkonium c

Cardiolipin activation of dnaA protein, the initiation protein of replication in Escherichia coli.

ATP binding to dnaA protein is essential for its action in initiating the replication of plasmids that bear the unique origin of the Escherichia coli chromosome (oriC). ADP bound to that site renders dnaA protein inactive for replication. Diphosphatidylglycerol (cardiolipin), a diacidic membrane phospholipid, displaces the bound nucleotide, and in the presence of components that reconstitute replication, fully reactivates the inert ADP form of dnaA protein. The monacidic phosphatidylglycerol is one-tenth as active as cardiolipin, whereas the neutral phosphatidylethanolamine, the principal E. c

Recombinant multiepitope proteins expressed in Escherichia coli cells and their potential for immunodiagnosis.

Recombinant multiepitope proteins (RMPs) are a promising alternative for application in diagnostic tests and, given their wide application in the most diverse diseases, this review article aims to survey the use of these antigens for diagnosis, as well as discuss the main points surrounding these antigens. RMPs usually consisting of linear, immunodominant, and phylogenetically conserved epitopes, has been applied in the experimental diagnosis of various human and animal diseases, such as leishmaniasis, brucellosis, cysticercosis, Chagas disease, hepatitis, leptospirosis, leprosy, filariasis, s

Nitric oxide-induced bacteriostasis and modification of iron-sulphur proteins in Escherichia coli.

The nitric oxide (NO) cytotoxicity has been well documented in bacteria and mammalian cells. However, the underlying mechanism is still not fully understood. Here we report that transient NO exposure effectively inhibits cell growth of Escherichia coli in minimal medium under anaerobic growth conditions and that cell growth is restored when the NO-exposed cells are either supplemented with the branched-chain amino acids (BCAA) anaerobically or returned to aerobic growth conditions. The enzyme activity measurements show that dihydroxyacid dehydratase (IlvD), an iron-sulphur enzyme essential for

Effect of environmental conditions on proteins secreted by enterohemorrhagic Escherichia coli O26:H11.

Infections due to Shiga toxin-producing Escherichia coli (STEC) are responsible for severe diarrheal diseases in humans, and these bacteria have recently emerged as a leading cause of renal failure and encephalitis in children and the aged. In this study, we examined the environment-dependent production of proteins secreted from a strain of STEC O26:H11 by trichloroacetic acid precipitation, SDS-PAGE, Western blotting and N-terminal amino acid sequence analysis. Growth of bacteria in essential minimum medium (M9) led to the detection of secreted proteins of 104, 80,40, 37 and 25 kDa (P104, P80

Characterization of lipid-modified immunogenic proteins of Treponema pallidum expressed in Escherichia coli.

This study describes the sequence of the immunodominant Treponema pallidum surface protein TpD and its expression in Escherichia coli. The translated TpD DNA sequence revealed the presence of a putative site for lipid-modification downstream from the signal sequence of this membrane protein. Growth of TpD-expressing E. coli in the presence of radioactive palmitic acid revealed that TpD was lipid-modified. Three other, previously characterized cloned proteins of T. pallidum were also lipid-modified. The N-termini of two of three sequenced T. pallidum proteins contain a tetrapeptide sequence cha

Structure of GlgS from Escherichia coli suggests a role in protein-protein interactions.

BACKGROUND: The Escherichia coli protein GlgS is up-regulated in response to starvation stress and its overexpression was shown to stimulate glycogen synthesis. RESULTS: We solved the structure of GlgS from E. coli, a member of an enterobacterial protein family. The protein structure represents a bundle of three alpha-helices with a short hydrophobic helix sandwiched between two long amphipathic helices. CONCLUSION: GlgS shows structural homology to Huntingtin, elongation factor 3, protein phosphatase 2A, TOR1 motif domains and tetratricopeptide repeats, suggesting a possible role in protein
