Vaccines for enterotoxigenic Escherichia coli: current status.

PURPOSE OF REVIEW: Enterotoxigenic Escherichia coli (ETEC) remain major causes of infantile diarrhea in the developing world and of travelers' diarrhea in visitors to these areas from industrialized countries, although the relative importance of these pathogens in these settings may be decreasing. The pathogenesis of ETEC infections has been well understood for almost two decades, and there is good evidence for acquired immunity after infection. Nevertheless, currently there is no effective ETEC vaccine. This review evaluates the current status of ETEC vaccine development. RECENT FINDINGS: ET

Vaccines for preventing enterotoxigenic Escherichia coli (ETEC) diarrhoea.

BACKGROUND: Infection with enterotoxigenic Escherichia coli (ETEC) bacteria is a common cause of diarrhoea in adults and children in developing countries and is a major cause of 'travellers' diarrhoea' in people visiting or returning from endemic regions. A killed whole cell vaccine (Dukoral®), primarily designed and licensed to prevent cholera, has been recommended by some groups to prevent travellers' diarrhoea in people visiting endemic regions. This vaccine contains a recombinant B subunit of the cholera toxin that is antigenically similar to the heat labile toxin of ETEC. This review aims

Development of oral vaccines against enterotoxinogenic Escherichia coli diarrhoea.

Even though enterotoxin-producing Escherichia coli (ETEC) is the most important cause of diarrhoea in developing countries and among travellers, no vaccine for use in humans is yet available. New knowledge about virulence factors and protective antigens of ETEC, however, suggests that development of a useful vaccine may soon become possible. Such a vaccine should be given orally and ideally evoke both anticolonization and antitoxic immune responses in the gut. An oral cholera vaccine, containing a component (B subunit) which crossreacts immunologically with the major, heat-labile enterotoxin (

Development of oral vaccines against cholera and enterotoxinogenic Escherichia coli diarrhea.

An oral cholera vaccine consisting of the immunogenic but completely nontoxic B subunit of cholera toxin in combination with heat- and formalin-killed cholera vibrios has been developed. This vaccine, which was designed to evoke antitoxic as well as antibacterial intestinal immunity, has in extensive clinical trials including a large field trial been shown to confer, without any side-effects, protection against cholera lasting for at least 3 years. The vaccine also induced protection of shorter duration against diarrhea caused by enterotoxinogenic Escherichia coli (ETEC). Significant progress

Research progress on multivalent genetically engineered vaccines against enterotoxigenic Escherichia coli.

This article systematically elaborates on the etiological characteristics, epidemiology and current status of prevention and control of diarrhea caused by enterotoxigenic Escherichia coli (ETEC). ETEC is an important pathogenic bacterium responsible for diarrhea in young animals and its pathogenicity is closely associated with specific adhesins and enterotoxins. The article focuses on the serotype diversity of ETEC and its relationship with pathogenicity. It also analyzes the differences in susceptibility among various animal hosts and age groups. Furthermore, it outlines the epidemiological f

[Advances in new vaccines against human enterotoxigenic Escherichia coli--A review].

Enterotoxigenic Escherichia coli (ETEC) is the most common cause of diarrhea, which is a second leading cause of death for the children under five years old from all over the world. The key factors of ETEC contain both colonization factors (CFs) and enterotoxins including heat-labile enterotoxin (LT) and heat-stable enterotoxin (ST). CFs mediated the binding of bacteria to the host intestinal epithelial cells, whereas LT and ST stimulated the over-secretion of body fluids and electrolytes, resulting in the destruction of the host fluid balance and leading diarrhea. The vaccine against CFs and

Designing vaccines to neutralize effective toxin delivery by enterotoxigenic Escherichia coli.

Enterotoxigenic Escherichia coli (ETEC) are a leading cause of diarrheal illness in developing countries. Despite the discovery of these pathogens as a cause of cholera-like diarrhea over 40 years ago, and decades of vaccine development effort, there remains no broadly protective ETEC vaccine. The discovery of new virulence proteins and an improved appreciation of the complexity of the molecular events required for effective toxin delivery may provide additional avenues to pursue in development of an effective vaccine to prevent severe diarrhea caused by these important pathogens.

Vaccines against Infections Caused by Salmonella, Shigella, and Pathogenic Escherichia coli.

Infectious diseases represent one of the most common causes of death worldwide, with the enteropathogenic bacteria Salmonella and Shigella and pathogenic Escherichia coli being among the most detrimental. Currently, vaccination represents the preferred method of preventing such infections. For stimulating the adaptive immune response, immunizations are frequently based on formulations which include inactivated whole-cell vaccines, live attenuated vaccines, or subunit vaccines. These can be administered via a parenteral or mucosal route, the latter having the advantage that it most closely mimi

Formaldehyde effects on kanamycin resistance gene of inactivated recombinant Escherichia coli vaccines.

OBJECTIVES: Earlier studies have demonstrated the use of inactivated recombinant E. coli (bacterins), to protect against Clostridium spp. in vaccinated animals. These bacterins have a simpler, safer, and faster production process. However, these bacterins carry expression plasmids, containing antibiotic resistance gene, which could be assimilate accidentally by environmental microorganisms. Considering this, we aimed to impair this plasmids using formaldehyde at different concentrations. RESULTS: This compound inactivated the highest density of cells in 24 h. KanR cassette amplification was f

Genetically modified enterotoxigenic Escherichia coli vaccines induce mucosal immune responses without inflammation.

OBJECTIVE: Enterotoxigenic Escherichia coli (ETEC) is a major cause of acute diarrhoea in children in the developing world, in travellers and in the military. Safe, effective vaccines could reduce morbidity and mortality. As immunity to ETEC is strain specific, the ability to create vaccines in vitro which express multiple antigens would be desirable. It was hypothesised that three genetically attenuated ETEC strains, one with a genetic addition, would be immunogenic and safe, and they were evaluated in the first licensed UK release of genetically modified oral vaccines. METHODS: Phase 1 stud

Confronting challenges to enterotoxigenic Escherichia coli vaccine development.

The enterotoxigenic Escherichia coli (ETEC) are a diverse and genetically plastic pathologic variant (pathovar) of E. coli defined by their production of heat-labile (LT) and heat-stable (ST) enterotoxins. These pathogens, which came to recognition more than four decades ago in patients presenting with severe cholera-like diarrhea, are now known to cause hundreds of millions of cases of symptomatic infection annually. Children in low-middle income regions of the world lacking access to clean water and basic sanitation are disproportionately affected by ETEC. In addition to acute diarrheal morb

Intestinal Pathogenic Escherichia coli: Insights for Vaccine Development.

Diarrheal diseases are one of the major causes of mortality among children under five years old and intestinal pathogenic Escherichia coli (InPEC) plays a role as one of the large causative groups of these infections worldwide. InPECs contribute significantly to the burden of intestinal diseases, which are a critical issue in low- and middle-income countries (Asia, Africa and Latin America). Intestinal pathotypes such as enteropathogenic E. coli (EPEC) and enterotoxigenic E. coli (ETEC) are mainly endemic in developing countries, while ETEC strains are the major cause of diarrhea in travelers

Engineering Escherichia coli-Derived Nanoparticles for Vaccine Development.

The development of effective vaccines necessitates a delicate balance between maximizing immunogenicity and minimizing safety concerns. Subunit vaccines, while generally considered safe, often fail to elicit robust and durable immune responses. Nanotechnology presents a promising approach to address this dilemma, enabling subunit antigens to mimic critical aspects of native pathogens, such as nanoscale dimensions, geometry, and highly repetitive antigen display. Various expression systems, including Escherichia coli (E. coli), yeast, baculovirus/insect cells, and Chinese hamster ovary (CHO) ce

Evaluation of a combined rotavirus and enterotoxigenic Escherichia coli vaccine in cattle.

A vaccine of rotavirus and K99 antigen from enterotoxigenic Escherichia coli was emulsified in oil adjuvant and administered intramuscularly to pregnant cows. Calves born to and reared on vaccinated dams were protected against experimental rotavirus infection at five days old when compared with calves from unvaccinated control cows. Field trials of the vaccine were carried out in 40 commercial herds, in which half the cows in each herd were selected at random for vaccination and half were left unvaccinated. In 31 herds (2641 cows) there was no significant diarrhoea problem (less than 10 per ce

Clinical Studies of Escherichia coli O157:H7 Conjugate Vaccines in Adults and Young Children.

Pediatric immunization has been the most effective measure to prevent and reduce the burden of infectious diseases in children. The recent inclusion of pneumococcal and meningococcal polysaccharide conjugates in infant immunization further reinforces their importance. Currently there is no human vaccine against enterohemorrhagic Escherichia coli (EHEC) infections. This review focuses on the human EHEC vaccine that has been studied clinically, in particular, the polysaccharide conjugate against E. coli O157. The surface polysaccharide antigen, O-specific polysaccharide, was linked to rEPA, reco
