Complete biosynthesis of the clinical agent escin Ia.

Escin Ia is a barrigenol-type triterpenoid specific to Aesculus seeds and is widely utilized in clinical practice for the treatment of cerebral edema, swelling, and venous insufficiency. Here, we fully elucidated its biosynthetic pathway by identifying ten previously uncharacterized enzymes from Aesculus chinensis. AcCYP716BX1/2 catalyze the C-22α and C-28 hydroxylation of the β-amyrin scaffold, and AcCYP714E67 mediates the C-24 hydroxylation to produce protoaescigenin. Sequential glycosylation at the 2'-O and 4'-O positions is catalyzed by AcUGT94AK1/3/6 and AcUGT87AZ3, respectively, forming

Attenuation of Cardiac Autonomic Neuropathy by Escin in Diabetic Rats.

Cardiac autonomic neuropathy (CAN) is a least diagnosed complication of diabetes. Inflammation and oxidative stress play a crucial role in the pathophysiology of cardiomyopathy and neuropathy. Escin has anti-inflammatory activity and antioxidant activity. Hence, the present study was designed to evaluate the effect of escin in the management of CAN. Diabetes was induced in Sprague Dawley rats with streptozotocin (STZ). Diabetic animals were randomized in different groups after 6 weeks. Animals in the diabetic control group received no treatment, while animals in other groups received escin at

Escin inhibits lipopolysaccharide-induced inflammation in human periodontal ligament cells.

Periodontitis is a chronic inflammatory disease associated with gram-negative subgingival microflora infection. Accumulating experimental evidence suggests that escin exerts anti-inflammatory and anti-edematous effects. This study was designed to investigate the in vitro effects of escin on the inflammatory reaction of human periodontal ligament cells (hPDLs). hPDLs were stimulated with lipopolysaccharide (LPS). The cells were treated with various concentrations of escin. The viability of hPDLs was evaluated using the MTT method. The expression of Toll-like receptor 2 (TLR2) in hPDLs and the l

The Role of Escin as a Topical Agent for Lymphedema Treatment in a Rat Model.

Escin, a naturally derived material isolated from horse chestnut, is used as an anti-inflammatory and anti-edema agent. This study aimed to evaluate its effects on lymphedema in a rat tail model. We divided the rats into five groups. The treatment groups received topical application of escin gel at concentrations of 20%, 10%, 2%, and 0.5% for 4 weeks. The fifth group served as a control. We performed volumetric (water displacement) tests, H&E staining, and LYVE-1 immunohistochemical staining, followed by statistical evaluation. All treatment groups showed significant volumetric reductions comp

Protective effects of escin against indomethacin-induced gastric ulcer in mice.

Escin, a natural mixture of triterpenoid saponin isolated from the seed of the horse chestnut, is reported to have a potent antiulcer activity against ethanol-induced gastric mucosal lesions. This study investigated the possible mechanisms underlying the gastroprotective effect of escin against indomethacin-induced gastric ulcer in mice. Gastric ulceration was induced by a single intragastric administration of indomethacin (18 mg/kg). The mice underwent intragastric treatment with escin at doses of 0.45, 0.9 or 1.8 mg/kg. Gastric lesion was estimated morphometrically and histopathologically 6

β-escin activates ALDH and prevents cigarette smoke-induced cell death.

BACKGROUND: The tobacco use is one of the biggest public health threats worldwide. Cigarette smoke contains over 7000 chemicals among other aldehydes, regarded as priority toxicants. β-escin (a mixture of triterpenoid saponins extracted from the Aesculus hippocastanum. L) is a potent activator of aldehyde dehydrogenase (ALDH) - an enzyme catalyzing oxidation of aldehydes to non-toxic carboxylic acids. PURPOSE: The aim of this study was to evaluate the effect of β-escin on ALDH activity, ALDH isoforms mRNA expression and cytotoxicity in nasal epithelial cells exposed to cigarette smoke extract

Pilot postoperative ileus study of escin in cancer patients after colorectal surgery.

BACKGROUND: Postoperative ileus, a common complication in patients after abdominal surgery, brings no benefit to the recovery of postoperative patients, and treatment targeted at restoring gastrointestinal motility may shorten the hospital stay. Studies have shown that escin accelerates gastrointestinal transit in mice and improves gastrointestinal motility in patients after abdominal surgery. A pilot study of escin's effect on the recovery of gastrointestinal motility was conducted in colorectal cancer patients in anticipation of a multiple-center randomized controlled trial. METHODS: A tota

Beta-escin has potent anti-allergic efficacy and reduces allergic airway inflammation.

BACKGROUND: Type I hypersensitivity is characterized by the overreaction of the immune system against otherwise innocuous substances. It manifests as allergic rhinitis, allergic conjunctivitis, allergic asthma or atopic dermatitis if mast cells are activated in the respective organs. In case of systemic mast cell activation, life-threatening anaphylaxis may occur. Currently, type I hypersensitivities are treated either with glucocorticoids, anti-histamines, or mast cell stabilizers. Although these drugs exert a strong anti-allergic effect, their long-term use may be problematic due to their si

Intracellular calcium stores in beta-escin skinned rat and guinea-pig bladders.

Intracellular Ca2+ stores in rat and guinea-pig bladders and taenia caecum were studied in beta-escin skinned smooth muscle strips. 30 min of skinning with 40 microM and 80 microM beta-escin were the best parameters found to obtain good calcium response curves (10(-7)-10(-4) M) in rat and guinea pig, respectively. Calmodulin (1 microM) increased the calcium contractions significantly. pCa 6 was used to load intracellular stores and application of carbachol (50 microM) in all tissues then only contracted the tissues in the presence of guanosine-5'-triphosphate (GTP; 100 microM). Use of escin as a perforating agent on the IonWorks quattro automated electrophysiology platform.

The automated electrophysiology platform IonWorks has facilitated the medium-throughput study of ion channel biology and pharmacology. Electrical and chemical access to the cell is by perforated patch, afforded by amphotericin. Permeation of the amphotericin pore is limited to monovalent cations. We describe here the use of the saponin escin as an alternative perforating agent. With respect to the number and robustness of seals formed across a variety of cell and ion channel types, the performance of escin is equal to that of amphotericin. Escin also permits the permeation of larger molecules

[New findings on the efficacy and mode of action of the horse chestnut saponin escin].

In order to complete previous results of the efficacy of the horse chestnut saponin escin, its activity was tested on two further models of inflammation: the rat serous peritonitis provoked by i.p. injection of formalin solution, and the rat serous pleurisy provoked by intrapleural injection of Evans Blue/carrageenan. The results showed escin to be an anti-exudative substance with regard to its exudation inhibitory effect determined by the reduction of exudative fluid. As to peritonitis the diminution of protein permeation into the abdominal cavity was determined: with increasing doses escin t

[Treatment of upper limb lymphedema after mastectomy with escine and levo-thyroxine].

Lymphoedema of the upper limb is the most serious and most crippling complication of mastectomy. An account is given of the factors possibly responsible, and a classification of the different forms is proposed. When fibrosis has set in, treatment is followed by very disappointing results, being at best able to bring about a partial, but transitory reduction in limb volume and consistency. Reference is also made to results obtained in 70 cases with two substances, 1-thyroxine and escine (somatoline), applied by massotherapy in postural drainage, ionophoresis, and pressotherapy, according to the

Escin: inhibiting inflammation and promoting gastrointestinal transit to attenuate formation of postoperative adhesions.

Postoperative peritoneal adhesions are common, serious complications of general abdominal and gynecologic surgery that can lead to chronic abdominal pain, intestinal obstruction, and infertility. As yet, there are no ideal drugs that may be prescribed for patients to prevent adhesion formation effectively. In this study the effects of escin, a natural drug, on the various steps of adhesion formation were investigated. The effects of escin on increased vascular permeability induced by acetic acid in a mouse model of acute inflammation, granuloma formation in a subchronic inflammatory rat model,

Efficient gram-scale synthesis and preclinical evaluation of an escin-derived therapeutic for ovarian cancer.

Ovarian cancer is the most lethal gynecologic malignancy, characterized by a poor five-year survival rate for patients with advanced-stage disease. EV-S008, an escin derivative, exhibits potent anti-ovarian cancer activity; however, its preclinical development has been hindered by inefficient synthetic methodologies. In this study, we report a rational, seven-step synthetic route for EV-S008 that achieves a 40-fold increase in yield and produces material with 99.3% purity on a gram scale. EV-S008 demonstrated broad cytotoxicity across multiple human ovarian cancer cell lines, with IC50 values
