Hormone levels and response during de-etiolation in pea.

The objective of this study was to increase our understanding of the hormonal regulation of de-etiolation by investigating endogenous hormone levels and response in etiolated pea ( Pisum sativum L.) seedlings after exposure to continuous white light. Recent reports suggest that de-etiolation may result from the down-regulation of an enzyme in the brassinosteroid (BR) biosynthesis pathway in pea. A subsequent review highlighted the need for direct measurements of BR levels to support this hypothesis. We have shown that endogenous castasterone and 6-deoxocastasterone levels are not greatly reduc

G protein signaling in UV protection: methods for understanding the signals in young etiolated seedlings.

A seed is competent to respond to light soon after imbibition. A new developmental program begins in or on the ground where the young seedling may be exposed to heat, cold, drought, flooding (anoxia), salts, varying levels of visible light, and the topic of this paper, ultraviolet radiation. Herein what is described is a method for growing and maintaining seedlings, then methods of UV irradiation in order to measure discrete effects of UV wavelengths in signal transduction, very early in seedling development. The physiological response to an abiotic signal is partly dependent on the developmen

A CqbZIP55-CqPIF3 regulatory module associated with light-responsive flavonoid biosynthesis during quinoa seedling de-etiolation.

Quinoa (Chenopodium quinoa) is an emerging leafy vegetable and microgreen crop rich in health-promoting flavonoids, yet the regulatory mechanisms linking light perception to early metabolic adaptation remain unclear. Here, we integrated phenotypic, transcriptomic, metabolomic, and molecular analyses to investigate early de-etiolation responses in quinoa seedlings. Short-term light exposure rapidly promoted seedling establishment and induced transcriptional programs associated with photosynthesis, carbon metabolism, hormone signaling, and flavonoid biosynthetic gene expression, whereas metaboli

On the role of calcium in indole-3-acetic acid movement and graviresponse in etiolated pea epicotyls.

To determine whether Ca2+ plays a special role in the early graviresponse of shoots, as has been reported for roots, we treated etiolated pea epicotyls with substances known to antagonize Ca2+ (La3+), to remove Ca2+ from the wall (spermidine, EGTA), to inhibit calmodulin mediated reactions (chlorpromazine), or to inhibit IAA transport (TIBA). We studied the effect of these substances on IAA and Ca2+ uptake into 7 mm long subapical 3rd internode etiolated pea epicotyl sections and pea leaf protoplasts, on pea epicotyl growth, and graviresponse and on lateral IAA redistribution during gravistimu

The DnaJ-like zinc finger domain protein ORANGE localizes to the nucleus in etiolated cotyledons of Arabidopsis thaliana.

Vitamin A deficiency (VAD) is a worldwide health problem. Overexpression of the DnaJ-like zinc finger domain protein ORANGE (OR) is a novel strategy for the biofortification of pro-vitamin A carotenoids in different staple crops to alleviate VAD. In plants, OR triggers the differentiation from non-pigmented plastids into carotenoid-accumulating plastids. There are different reports on the subcellular localization of this protein in either chloroplasts or the nucleus, both of which were supported by confocal observation and protein-protein interaction results. In this work, we studied the subce

Brassinosteroids rescue the deficiency of CYP90, a cytochrome P450, controlling cell elongation and de-etiolation in Arabidopsis.

The cpd mutation localized by T-DNA tagging on Arabidopsis chromosome 5-14.3 inhibits cell elongation controlled by the ecdysone-like brassinosteroid hormone brassinolide. The cpd mutant displays de-etiolation and derepression of light-induced genes in the dark, as well as dwarfism, male sterility, and activation of stress-regulated genes in the light. The CPD gene encodes a cytochrome P450 (CYP90) sharing homologous domains with steroid hydroxylases. The phenotype of the cpd mutant is restored to wild type both by feeding with C23-hydroxylated brassinolide precursors and by ectopic overexpres

Natural Etiolation in Spearmint (Mentha spicata L. var. crispa) Shoots Emerging From Rhizomes-Plastids, Glandular Hairs, and Phytochemical Profile.

Spearmint (Mentha spicata L. var. crispa) is a widely cultivated aromatic herb valued both as a culinary ingredient and as a source of bioactive compounds, such as essential oils and phenolics, which contribute to its traditional uses and potential medicinal applications. In this work, we provided the first evidence of natural etiolation with etioplast and prolamellar body formation in leaves and stems of spearmint shoots emerging in the soil from underground rhizomes. Soil-grown and hydroponically dark-forced shoots exhibited comparable etiolation symptoms, plastid ultrastructure, and chlorop

The miR396-GRFs Module Mediates the Prevention of Photo-oxidative Damage by Brassinosteroids during Seedling De-Etiolation in Arabidopsis.

The switch from dark- to light-mediated development is critical for the survival and growth of seedlings, but the underlying regulatory mechanisms are incomplete. Here, we show that the steroids phytohormone brassinosteroids play crucial roles during this developmental transition by regulating chlorophyll biosynthesis to promote greening of etiolated seedlings upon light exposure. Etiolated seedlings of the brassinosteroids-deficient det2-1 (de-etiolated2) mutant accumulated excess protochlorophyllide, resulting in photo-oxidative damage upon exposure to light. Conversely, the gain-of-function

Glutamine synthetases of green and etiolated leaves ofSinapis alba : Evidence of the identity of the respective enzyme proteins.

Studies on the glutamine synthetases (GS, EC 6.3.1.2) of green (GS2) and etiolated leaves (GSet) ofSinapis alba L. (cv. Steinacher) revealed striking similarities between the respective enzyme proteins. The enzymes showed corresponding chromatographic properties, both on dimethylaminoethyl-Sephacel and on hydroxylapatite columns. The purified GS proteins were also identical with regard to the molecular weight of their subunits. Isoelectrofocusing of pure GSet yielded two distinct polypeptide bands in the pH 5.6 region of the gels. This pattern corresponded to the two strong bands of GS2. Two c

Nitric oxide stimulates seed germination and de-etiolation, and inhibits hypocotyl elongation, three light-inducible responses in plants.

Seed germination, greening of etiolated plants and inhibition of hypocotyl elongation are stimulated by light, which is sensed by various types of photoreceptor. Nitric oxide (NO) has proven to be a bioactive molecule, especially in mammalian cells and, most recently, in plants. Like some phytochrome-dependent processes, many NO-mediated ones are accomplished through increases in cGMP levels. Given these similarities, we proposed that NO could take part in light-mediated events in plants. Here we show that NO promotes seed germination and de-etiolation, and inhibits hypocotyl and internode elo

Comparative transcriptome analyses provide novel insights into etiolated shoot development of walnut (Juglans regia L.).

In general, genes promoting IAA, CTK GA and ethylene biosynthesis were upregulated, while genes participating in ABA, chlorophyll and starch biosynthesis pathways performed opposite tendency during etiolation. Etiolation as a method for rejuvenation plays an important role in the vegetative propagation of woody plants. However, the molecular mechanism of etiolated shoot development remains unclear. In this study, we investigated changes at different etiolation stages of Juglans regia. The histology and transcriptome of J. regia were analysed using etiolated stems, which were treated in darknes

Establishment of an efficient tissue culture system for Paeonia ostii by combining vernalization and etiolation pretreatment with optimized culture conditions.

BACKGROUND: Paeonia ostii, an economically important oil-producing peony cultivar, faces challenges in large-scale cultivation due to low propagation rates and long cultivation cycles. This study aimed to optimize tissue culture protocols for P. ostii 'Fengdan No. 3' by evaluating vernalization and etiolation pretreatments on single-node and leaf explants. RESULTS: Vernalization and etiolation treatments significantly enhanced in vitro regeneration of P. ostii, resulting in improved organogenic responses and reduced browning. Optimal sterilization and culture conditions were established for b

phyA dominates in transduction of red-light signals to rapidly responding genes at the initiation of Arabidopsis seedling de-etiolation.

Contrary to expectations based on the visible phenotypic behavior of seedlings undergoing de-etiolation in response to continuous red light (Rc), previous gene expression profiling showed that one or more of the five-membered phytochrome (phy) family of Arabidopsis, other than phyB, is predominantly responsible for transducing the Rc signals to light-responsive genes. To begin to identify which phys are involved, and to define potential primary targets of phy signaling, we have examined the genome-wide expression profiles of genes responding to Rc within 1 h (early response genes) of initial e

Restoration of mature etiolated cucumber hypocotyl cell wall susceptibility to expansin by pretreatment with fungal pectinases and EGTA in vitro.

Mature plant cell walls lose their ability to expand and become unresponsive to expansin. This phenomenon is believed to be due to cross-linking of hemicellulose, pectin, or phenolic groups in the wall. By screening various hydrolytic enzymes, we found that pretreatment of nongrowing, heat-inactivated, basal cucumber (Cucumis sativus) hypocotyls with pectin lyase (Pel1) from Aspergillus japonicus could restore reconstituted exogenous expansin-induced extension in mature cell walls in vitro. Recombinant pectate lyase A (PelA) and polygalacturonase (PG) from Aspergillus spp. exhibited similar ca

Mutation of Mg-chelatase subunit I causes distinct phenotypes of chlorophyll accumulation and seedling etiolation in watermelon (Citrullus lanatus) and melon (Cucumis melo).

Chlorophyll biosynthesis is essential for plant growth and development, as it directly modulates photosynthetic capacity. Here, we identified yellow mutants with chlorophyll deficiency and abnormal thylakoid stacking-disturbed in watermelon (lethal) and melon (non-lethal), and mapped the traits to SNPs (H359R in watermelon, T193A in melon) in ChlI, which encodes Mg-chelatase subunit I. Suppression of ClChlI in watermelon and CmChlI in melon caused reduced chlorophyll accumulation and leaf yellowing. Moreover, the mutated ChlI exhibited altered autologous protein structures, and weakened intera

Integrated Metabolome and Transcriptome Analyses Reveal Etiolation-Induced Metabolic Changes Leading to High Amino Acid Contents in a Light-Sensitive Japanese Albino Tea Cultivar.

Plant albinism causes the etiolation of leaves because of factors such as deficiency of chloroplasts or chlorophylls. In general, albino tea leaves accumulate higher free amino acid (FAA) contents than do conventional green tea leaves. To explore the metabolic changes of etiolated leaves (EL) in the light-sensitive Japanese albino tea cultivar "Koganemidori," we performed integrated metabolome and transcriptome analyses by comparing EL with green leaves induced by bud-sport mutation (BM) or shading treatments (S-EL). Comparative omics analyses indicated that etiolation-induced molecular respon

Regulation of protochlorophyllide synthesis genes in darkness by class I TCP transcription factors TCP14 and TCP15 prevents photodamage after illumination in Arabidopsis.

De-etiolation, which takes place when seedlings germinated in the dark are first exposed to light, involves profound morphological and physiological changes, among them the synthesis of chlorophylls from the highly reactive precursor protochlorophyllide (Pchlide). As excess Pchlide accumulation can cause cellular damage upon illumination, the entire chlorophyll synthesis pathway is subjected to tight transcriptional control. Here, we report that the activity of the class I TCP transcription factors (TCPs) TCP14 and TCP15 is required to repress genes involved in Pchlide synthesis in the dark, t

Time-Course Transcriptome Study Reveals Mode of bZIP Transcription Factors on Light Exposure in Arabidopsis.

The etiolation process, which occurs after germination, is terminated once light is perceived and then de-etiolation commences. During the de-etiolation period, monochromatic lights (blue, red and far-red) induce differences in gene expression profiles and plant behavior through their respective photoreceptors. ELONGATED HYPOCOTYL 5 (HY5), a bZIP-type transcription factor (TF), regulates gene expression in the de-etiolation process, and other bZIP TFs are also involved in this regulation. However, transcriptomic changes that occur in etiolated seedlings upon monochromatic light irradiation and
