Versatile biotechnological applications of Euglena gracilis.

Euglena gracilis is a freshwater protist possessing secondary chloroplasts of green algal origin. Various physical factors (e.g. UV) and chemical compounds (e.g. antibiotics) cause the bleaching of E. gracilis cells-the loss of plastid genes leading to the permanent inability to photosynthesize. Bleaching can be prevented by antimutagens (i.e. lignin, vitamin C and selenium). Besides screening the mutagenic and antimutagenic activity of chemicals, E. gracilis is also a suitable model for studying the biological effects of many organic pollutants. Due to its capability of heavy metal sequestrat

Ethanol alleviates glucose stress from Euglena gracilis.

Euglena gracilis, a microalga, can be cultured under photoautotrophic, heterotrophic, or mixotrophic conditions. This species can produce proteins, vitamins, and lipids. Based on previous reports, paramylon, a storage polysaccharide of E. gracilis and a type of β-1,3-glucan, can inhibit the development of skin lesions. Glucose increases the proliferation of E. gracilis and the accumulation of paramylon. However, excess glucose decreases the proliferation of E. gracilis because of high osmotic pressure. In this study, we found that E. gracilis cells cultured with high concentrations of glucose

Peculiar proteome of dark-cultivated Euglena gracilis.

Euglena gracilis is a flagellate photosynthetic microalga that, thanks to its metabolic adaptability, can grow under both autotrophic and heterotrophic conditions. This adaptability makes euglena an interesting species for applied biotechnology. We focused on the proteome of E. gracilis cultivated in Cramer-Myers medium (supplemented with ethanol) in dark and light conditions. Cultures grown in the light showed a characteristic green coloration, while cultures incubated in the dark were bright yellow. When cultured in the dark, microalga showed reduced concentration of chlorophylls (a, b, and

Responses of the photosynthetic flagellate, Euglena gracilis, to hypergravity.

Motility and orientation has been studied in the unicellular photosynthetic flagellate, Euglena gracilis, using real time image analysis capable of tracking up to 200 cells simultaneously in the slow rotating centrifuge microscope (NIZEMI) which allows one to observe the cells' swimming behavior during centrifugation accelerations between 1 g and 5 g. At 1 g the cells show a weak negative gravitaxis, which increases significantly at higher accelerations up to about 3 g. Though most cells were capable of swimming even against an acceleration of 4.5 g, the degree of gravitaxis decreased and some

Trophic Condition Shapes UVC Responses in Euglena gracilis.

Short-wavelength ultraviolet radiation can impair biological systems by causing DNA damage, oxidative stress, and disruption of photosynthetic processes. Although ultraviolet C (UVC) at 254 nm is widely used as a controlled laboratory stressor, the extent to which trophic condition influences repeated UVC tolerance in phototrophic protists remains unclear. Here, we examined the response of Euglena gracilis grown under photoautotrophic or ethanol-supported mixotrophic conditions and exposed to daily UVC pulses for five days. Cell growth, photosynthetic pigments, intracellular oxidative stress m

Cd2+ transport and storage in the chloroplast of Euglena gracilis.

Euglena gracilis lacks a plant-like vacuole and, when grown in Cd2+-containing medium, 60% of the accumulated Cd2+ is located inside the chloroplast. Hence, the biochemical mechanisms involved in Cd2+ accumulation in chloroplast were examined. Percoll-purified chloroplasts showed a temperature-sensitive uptake of the free 109Cd2+ ion. Kinetics of the uptake initial rate was resolved in two components, one hyperbolic and saturable (Vmax 11 nmol 109Cd2+ min(-1) mg protein (-1), Km 13 microM) and the other, linear and non-saturable. 109Cd2+ uptake was not affected by metabolic inhibitors or illum

Nomen omen: Euglena gracilis possesses a rhodopsin-based photoreceptor.

The unicellular microalga Euglena gracilis has always been considered the ideal alga to investigate photoreceptive responses and systems, and it has been the subject of hundreds of articles. Moreover, because of its detectable photoreceptor, it has been given a key role in the evolution of photoreception, from single and simple cells to complex visual system of higher organisms. In this article, we report the Raman spectra recorded in vivo on photoreceptors of E. gracilis and Bos taurus retina. The almost perfect superimposability (correlation coefficient r = 0.955) of these spectra states tha

Chromium uptake, retention and reduction in photosynthetic Euglena gracilis.

Photosynthetic Euglena gracilis grown with different K(2)CrO(4) concentrations was analyzed for its ability to take up, retain and reduce Cr(VI). For comparison, cells were also exposed to CrCl(3). Cellular Cr(VI) uptake at pH 7.2 showed a hyperbolic saturation pattern with K (m) of 1.1 mM, V (m) of 16 nmol (h x 10(7) cells)(-1), and K (i sulfate) of 0.4 mM. Kinetic parameters for sulfate uptake were similar, K (m) = 0.83 mM, V (m) = 15.9 nmol (h x 10(7)cells)(-1) and K (i chromate) = 0.3 mM. The capacity to accumulate chromium depended on the ionic species, external concentration and pH of th

Phytochelatin-cadmium-sulfide high-molecular-mass complexes of Euglena gracilis.

High-molecular-mass PC complexes (PC-HMWCs) constituted by phytochelatins (PCs), cadmium and sulfide are synthesized by several organisms after exposure to cadmium. In this study, PC-HMWCs were isolated from photoheterotrophic Euglena gracilis and purified to homogeneity, resulting in compounds of molecular mass 50-380 kDa depending on the CdCl2 and sulfate concentrations in the culture medium. In contrast with plants and some yeasts, PC-HMWCs from E. gracilis mainly comprise (57-75%) monothiol molecules (Cys, gamma-glutamylcysteine, GSH) and, to a lesser extent (25-43%), PCs. Paramylon from Euglena gracilis Prevents Lipopolysaccharide-Induced Acute Liver Injury.

Acute liver injury (ALI) is a life-threatening syndrome with high mortality and lacks effective therapies. Rodents under LPS (lipopolysaccharide)/D-Gal (D-galactosamine) stress mimic ALI by presenting dramatically increased inflammation and cell death in the liver. Euglena gracilis, functioning like dietary fiber, is commonly used as a paramylon (Pa)-rich nutritional supplement that has various biological effects such as regulating immune system, anti-obesity, and anti-tumor. Here, we found that Pa or sonicated and alkalized paramylon (SA-Pa) alleviated the LPS/D-Gal-induced hepatic histopatho

Euglena gracilis as an eukaryotic test organism for detecting mutagens and antimutagens.

The unicellular flagellate Euglena gracilis was used in order to assess the inhibition of N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) and N-methyl-N-nitrosourea (MNU) mutagenicities, which induce white mutants due to the irreversible loss of chloroplasts. All tested compounds, including o-aminobenzoic acid and p-aminobenzoic acid, salicylic acid, acetylsalicylic acid, sodium salicylate and p-aminosalicylic acid, were not mutagenic per se and inhibited MNNG mutagenicity by at least 50%. The last two compounds inhibited by at least 50% also MNU mutagenicity.

Influence of tetracyclines or cetylpyridinium bromide on activity of fluoroquinolones in Euglena gracilis.

To verify the hypothesis that the fluoroquinolone inhibitors of DNA gyrase generate some oxidant species and subsequently free radicals, the effects of simultaneous action of fluoroquinolones (ofloxacin, fleroxacin) and tetracyclines (tetracycline, doxycycline, thiatetracycline) or cetylpyridinium bromide as possible antioxidants on the flagellate Euglena gracilis were examined. Chloroplasts due to their bacterial character were injured and subsequently eliminated by fluoroquinolone. The loss of chloroplasts was accompanied by bleaching of originally green euglena cells. The Glow Up: Optimizing Growth Conditions and Carotenoid Profiling in Euglena gracilis.

Until now, only five photoactive pigments have been identified in Euglena gracilis and there has been contradictory reporting of the major carotenoid. The impact of various environmental factors on the carotenoid content of E. gracilis has not been well studied. Using an improved HPLC UV-VIS method, combined with mass spectrometry, we have now identified 12 photoactive pigments from E. gracilis extracts and confirmed diadinoxanthin as the major carotenoid produced in E. gracilis. After testing cultivation media and major carbon sources (addition of glucose-2-20 g/L, ethanol-0.2%-2.0% w/v), we

Long-time behavior of swimming Euglena gracilis in a heterogenous light environment.

The cell motion of Euglena gracilis in homogeneous and heterogeneous light environments was analyzed. Homogeneous and heterogeneous environments were prepared, with only a red color or with a red circle surrounded by brighter white regions, respectively. In a heterogeneous environment, the cells move into the red circle. Swimming orbits at 1/25 s intervals for 120 s were analyzed. The speed distribution of the 1 s-averaged cell orbits in a homogeneous environment was different from that in a heterogeneous environment, where the faster swimming fraction was enhanced. The relationship between sp

Indications for acceleration-dependent changes of membrane potential in the flagellate Euglena gracilis.

The effects of the calcium sequester EGTA on gravitactic orientation and membrane potential changes in the unicellular flagellate Euglena gracilis were investigated during a recent parabolic-flight experiment aboard of an Airbus A300. In the course of a flight parabola, an acceleration profile is achieved which yields subsequently about 20 s of hypergravity (1.8 g(n)), about 20 s of microgravity, and another 20 s of hypergravity phases. The movement behavior of the cells was investigated with real-time, computer-based image analysis. Membrane potential changes were detected with a newly develo

Effects of iron-, manganese-, or magnesium-deficiency on the growth and morphology of Euglena gracilis.

Iron-, manganese-, or magnesium-deficiency has been induced in Euglena gracilis. Each arrests cell proliferation, decreases the intracellular content of the deficient metal, and increases that of several other metals. Light and electron microscopy of stationary phase cells reveal that Fe-deficient (-Fe) cells are similar in size and shape to control organisms. Magnesium-deficient (-Mg) cells, however, are larger, and approximately 14% are multilobed, containing 2 to 12 lobes of equal size emanating from a central region. Individual (-Mg) cells and each lobe of multilobed cells contain a single
