Drosophila melanogaster is a model system for examining the mechanisms of action of neuropeptides. DPKQDFMRFamide was previously shown to induce contractions in Drosophila body wall muscle fibres in a Ca(2+)-dependent manner. The present study examined the possible involvement of a G-protein-coupled receptor and second messengers in mediating this myotropic effect after removal of the central nervous system. DPKQDFMRFamide-induced contractions were reduced by 70% and 90%, respectively, in larvae with reduced expression of the Drosophila Fmrf receptor (FR) either ubiquitously or specifically in

Vagus nerve comprises two distinct kinds of nerves, nodose and jugular ganglionic nerves. We tested pharmacological difference between two vagal nerves in the responsiveness to FMRFamide. The response probability to FMRFamide was significantly higher in nodose than jugular nerves in intracellular calcium measurement. Nodose nerves also depolarized membrane potential to FMRFamide more than jugular nerves did, in patch-clamp recording, although the probability of action potential discharge was same in both nerves. The inward current induced by FMRFamide was characterized as mixed cations. These

The second FMRFamide-gated Na(+) channel (HtFaNaC), from Helisoma trivolvis, has been cloned. HtFaNaC has some different pharmacological properties to HaFaNaC, from Helix aspersa, which has enabled a rational approach to be made to start to identify the FMRFamide recognition site. Several chimeras were made by switching sections between the channels. The differences in sensitivity to FMRFamide, and amiloride, were assessed after expression in Xenopus oocytes. The data suggest that a recognition site for FMRFamide, and the potentiating action of amiloride, resides in a sequence of about 120 ami

The distribution of FMRFamide-like immunoreactivity was determined in the brain and the pituitary of the goldfish, Carassius auratus. Immunoreactive perikarya were observed in the olfactory nerve, nucleus entopeduncularis, nucleus anterioris periventricularis, nucleus posterioris periventricularis, lateral part of the nucleus lateralis tuberis pars posterioris, midbrain tegmentum, and medulla oblongata. Immunoreactive fibers were widely distributed in the brain, in particular in the ventral telencephalon and the hypothalamus. A few immunoreactive nerve fibers were observed in the pituitary. Th

The possible role of centrally administered tetrapeptide FMRFamide (Phe-Met-Arg-Phe-NH2) on gastric acid secretion in pylorus ligated rats was investigated. Intracerebroventricularly administered FMRFamide stimulated the gastric acid secretion in a dose-dependent manner. This stimulatory effect was abolished by vagotomy and atropine pretreatment. The presence of FMRFamide in rat brain and the ability of FMRFamide to stimulate gastric acid secretion suggest that FMRFamide plays a physiological role in brain modulation of gastric acid secretion.

Neurons immunoreactive for the molluscan cardioactive peptide FMRFamide (FMRFi) in the brain of Carassius were demonstrated immunohistochemically with the peroxidase-antiperoxidase technique (PAP) and three antisera against this tetrapeptide. FMRFi perikarya were consistently found in the ganglion of the nervus terminalis, in some scattered neurons of the nucleus entopeduncularis, in the nucleus praeopticus pars magno- and parvocellularis, the nucleus ventromedialis, the nucleus posterior periventricularis, nucleus recessus lateralis and posterior. Some weak FMRFi neurons occurred in the rostr

The molluscan neuropeptide FMRFamide (Phe-Met-Arg-Phe-NH2) inhibits the beat of isolated ventricles of the freshwater clam Lampsilis claibornensis. The decline in frequency and diastolic tone precedes the dose- and time-dependent increase in cAMP also produced by the peptide. Phosphodiesterase inhibitors (theophylline, 3-isobutyl-1-methylxanthine) and cAMP analogs also inhibit Lampsilis hearts, but only at high doses (10(-5)-10(-3)M). Again, the inhibition by theophylline is accompanied by a rise in cAMP. Nevertheless, the delayed increase in cAMP levels following FMRFamide treatment may sugge

The development of the nervus terminalis system in the lungfish, Neoceratodus forsteri, was investigated by using FMRFamide as a marker. FMRFamide immunoreactivity appears first within the brain, in the dorsal hypothalamus at a stage around hatching. At a slightly later stage, immunoreactivity appears in the olfactory mucosa. These immunoreactive cells move outside the olfactory organ to form the ganglion of the nervus terminalis. Immunoreactive processes emerge from the ganglion of the nervus terminalis in two directions, one which joins the olfactory nerve to travel to the brain and the othe

The distribution of FMRFamide-like-immunoreactive peptides was investigated in the brain and pituitary of the elasmobranch fish Scyliorhinus canicula using the indirect immunofluorescence technique. FMRFamide-immunoreactive cells and fibers were mainly observed in the telencephalon and the diencephalon, while other brain structures were almost unstained. In the telencephalon, FMRFamide-like-containing neurons were seen in the caudal part of the area periventricularis pallialis, in the posterior area of the nucleus septi medialis and in the nucleus septi caudoventralis. In the diencephalon, num

FMRFamide-immunoreactive neurons were determined immunocytochemically by means of the PAP-technique for unlabeled antibodies. FMRFamide-containing perikarya were found in the frontoventral telencephalon, in the valvula and corpus cerebelli, and in the rostral and caudal medulla oblongata. FMRFamide immunoreactive cells were localized in the anterio lobe of the pituitary. FMRFamide fibres occurred in the bulbus olfactorius, in the anterior, medial and lateral parts of the telencephalon, in the commissura anterior, beneath the nucleus praeopticus, beneath the nucleus dorsomedialis, ventrolis and

Purification of small peptide components in the venoms of the solitary sphecid wasps, Sphex argentatus argentatus and Isodontia harmandi, led to the isolation of several major peptides. Analysis of MS/MS spectra by MALDI-TOF/TOF revealed the sequence of a new peptide Sa112 (EDVDHVFLRF-NH2), which is structurally very similar to leucomyosupressin (pQDVDHVFLRF-NH2) and SchistoFLRFamide (PDVDHVFLRF-NH2), the FMRFamide-like peptides from cockroach and locust, respectively. Indeed, this new peptide, like SchistoFLRFamide, inhibited the frequency and amplitude of spontaneous contractions of the locu

The extrabulbar olfactory system (EBOS) is a collection of nerve fibers which originate from primary olfactory receptor-like neurons and penetrate into the brain bypassing the olfactory bulbs. Our description is based upon the application of two neuronal tracers (biocytin, carbocyanine DiI) in the olfactory sac, at the cut end of the olfactory nerve and in the telencephalon of the developing clawed frog. The extrabulbar olfactory system was observed already at stage 45, which is the first developmental stage compatible with our techniques; at this stage, the extrabulbar olfactory system fibers

The present study employed an in situ hybridisation technique to detect the expression of a number of FMRFamide-like peptide encoding (flp) genes, previously identified from Globodera pallida, in whole-mount preparations of the J(2) stage of this worm. gpflp-1, encoding the FMRFamide-related peptide (FaRP) KSAYMRFamide, was expressed in neurones associated with the circumpharyngeal nerve ring and specifically in a number of cell bodies in the lumbar ganglia of the perianal nerve ring. The lumbar ganglia and pre-anal ganglia along with the BDU neurones and a number of cells in the retrovesicula

BACKGROUND: Trematoda Opisthorchis felineus Rivolta, 1884 is the causative agent of dangerous parasite disease-opisthorchiasis, widespread in the Russian Federation. The details of the neuroanatomical localization of the serotoninergic and FMRFamidergic neurotransmitter elements as well as their functional roles remain not studied enough in both adult and larval forms of O. felineus. The studies in this area are important in term of the development of a new pharmacological strategy of the struggle with the causative agent of opisthorchiasis affecting the neuronal signal substances and the func

A complementary DNA (cDNA) library was prepared from poly(A)(+) RNA isolated from the central ganglia of Helix aspersa from which two classes of FaRP-encoding cDNA clones were identified by hybridization with the Aplysia FMRF-1 clone and oligonucleotides based on known Helix peptides. One type of cDNA (exemplified by HF-1) encodes only the tetrapeptides (FMRFamide and FLRFamide) and is very similar to the tetrapeptide-encoding precursors of other molluscan species. The other type of cDNA (represented by HF-4) encodes no tetrapeptides, but only N-terminally extended peptides, including all of t

FMRFamide-like peptides (FLPs) are a diverse group of neuropeptides that are expressed abundantly in nematodes. They exert potent physiological effects on locomotory, feeding and reproductive musculature and also act as neuromodulators. However, little is known about the specific expression patterns and functions of individual peptides. The current study employed rapid amplification of cDNA ends-polymerase chain reaction (RACE-PCR) to characterize flp genes from infective juveniles of the root knot nematodes, Meloidogyne incognita and Meloidogyne minor. The peptides identified from these trans

Application of FMRFamide (Phe-Met-Arg-Phe-NH2) induced a slow depolarization in neurons of the Aplysia abdominal ganglion. In voltage-clamped cells, FMRFamide induced a slow inward current that increased when the membrane was depolarized beyond -85 mV, showing a negative slope conductance. However, this inward current never reversed to outward current when hyperpolarized beyond the equilibrium potential for K+. The FMRFamide-induced response was markedly augmented in Ca(2+)-free media, but depressed in Na(+)-free media. It was unaffected by a change in external potassium. Intracellular injecti

The molluscan neuropeptide, Phe-Met-Arg-Phe-NH2 (FMRFamide), the mammalian opioid peptide met-enkephalin, and their common analogues, met-enkephalin-Arg6-Phe7 (YGGFMRF) and Tyr-Gly-Gly-Phe-Met-Arg-Phe-amide (YGGFMRFamide), were injected into the lateral ventricle of the rat; the cardiovascular effects were studied. FMRFamide caused a rapid, transient elevation in blood pressure accompanied by a great increase in pulse pressure. These effects were followed by secondary increases in blood and pulse pressures. Met-enkephalin produced an initial reduction in blood pressure which was followed by a

The terminal nerve is a ganglionated cranial nerve with peripheral processes that enter the nasal cavity and centrally directed processes that enter the forebrain. Members of all classes of gnathostomes have been found to possess a terminal nerve, some components of which demonstrate immunoreactivity to the peptides Phe-Met-Arg-Phe-NH2 (FMRFamide) and gonadotropin-releasing hormone (GnRH). To explore the possibility that lampreys possess a terminal nerve, we examined the distribution of these peptides in the silver lamprey, Ichthyomyzon unicuspis, by using antisera to FMRFamide and to four for

The presence and localization of the serotoninergic and FMRFamidergic structures in the nervous system of the trematode Opisthioglyphe ranae, the marsh frog intestinal parasite, was studied using immunocytochemistry. The serotonin-immunoreactive nerve cells and fibers were revealed in the head ganglia, circular commissure, longitudinal nerve cords and their connective commissures, as well as around the oral and ventral suckers, oesophagus and genital pore. FMRF-specific immunoreactivity was observed in the head ganglia, longitudinal nerve cords and terminal parts of the reproductive system. Th
