Faecalibacterium prausnitzii is one of the most important butyrate-producing bacteria in the human gut. Previous studies have suggested the presence of several phylogenetic groups, with differences at the species level, in the species, and a taxonomic re-evaluation is thus essential for further understanding of ecology of the important human symbiont. Here we examine the phenotypic, physiological, chemotaxonomic and phylogenomic characteristics of six F. prausnitzii strains (BCRC 81047T=ATCC 27768T, A2-165T=JCM 31915T, APC918/95b=JCM 39207, APC942/30-2=JCM 39208, APC924/119=JCM 39209 and APC92

Faecalibacterium is one of the most abundant anaerobes in the human colon. At the genus level, this bacterium shows a strong positive association with human health. Expanding collections of isolates and metagenome-assembled genomes have revealed its species diversity, yet species-level functions remain so far underexplored. Here, we describe a novel species, Faecalibacterium harmsenii. In addition, we reclassify another isolate as a member of the recently reported Faecalibacterium langellae species. Despite close genomic relatedness, these isolates exhibit distinct physiological and biochemica

Faecalibacterium is an essential probiotic in the human gut; changes in its abundance are associated with various disease states in many studies. However, the causal nature of such associations remains obscure. Therefore, we aimed to thoroughly investigate the causal relationships between Faecalibacterium and its related diseases. A two-sample bi-directional Mendelian randomisation analysis was conducted using publicly available genome-wide association studies summary statistics for Faecalibacterium and its related diseases. We found that Faecalibacterium was negatively correlated with the ris

Faecalibacterium is among the most abundant bacterial genera in the healthy human colon, comprising approximately 10-15% of the total gut microbiota. Species within this genus ferment complex carbohydrates, including pectin, to produce butyrate, a short-chain fatty acid with anti-inflammatory and anti-carcinogenic properties. Butyrate is the primary energy source for colonocytes and in Faecalibacterium is synthesized via the butyryl-CoA:acetate CoA transferase pathway. Reduced levels of Faecalibacterium are often associated with increased abundance of Escherichia coli and may be linked to earl

In recent years, the incidence of metabolic disorders has exponentially increased worldwide mainly due to the misfunctioning of chemical reactions. Gestational diabetes mellitus (GDM) occurred during pregnancy when placenta prevent absorption of sufficient insulin in mother's body. Although this disorder usually disappears after delivery, it could raise the probability of developing complications in the future. GDM is associated with adverse effects on women and their children, such as obesity, cardiovascular disease, and type 2 diabetes. On the other hand, administration of probiotics as a li

There is an increasing interest in Faecalibacterium prausnitzii, one of the most abundant bacterial species found in the gut, given its potentially important role in promoting gut health. Although some studies have phenotypically characterized strains of this species, it remains a challenge to determine which factors have a key role in maintaining the abundance of this bacterium in the gut. Besides, phylogenetic analysis has shown that at least two different F. prausnitzii phylogroups can be found within this species and their distribution is different between healthy subjects and patients wit

Reports that bacteria within the Firmicutes phylum, especially the species Faecalibacterium prausnitzii, are less abundant in Crohn's disease (CD) patients and supernatants from cultures of this bacterium are anti-inflammatory prompted the investigation of the possible correlations between the abundance of F. prausnitzii and the response to treatment in patients with gut diseases and healthy controls. In a randomized, double-blind trial, faeces were collected from healthy volunteers, and from patients with active CD, ulcerative colitis (UC) and irritable bowel syndrome before and after treatme

As a paradigmatic next-generation probiotic, Faecalibacterium species play a major role in butyrate generation, intestinal barrier integrity, and immune homeostasis. However, its clinical translation is severely hindered by profound physiological fragility-namely, extreme oxygen sensitivity and inherent metabolic bottlenecks, such as its strict dependence on exogenous acetate for butyrate synthesis-which collectively impede in vitro cultivation, in vivo colonisation, and scalable production. To circumvent these constraints, leveraging the gut microbiota's "cross-feeding" network has emerged as

Faecalibacterium prausnitzii, a major butyrate-producing gut commensal with anti-inflammatory activity, is extremely oxygen-sensitive, limiting its use as a probiotic. Dietary prebiotics may enhance its growth and resilience, thereby influencing host immune responses. This study examined how distinct classes of prebiotics including oligosaccharides (fructooligosaccharides, arabinoxylan), nondigestible polysaccharides (inulin, pectin, resistant starch, golden kiwi fiber), and the vitamin riboflavin affect the growth kinetics, bile tolerance, and immunomodulatory properties of F. prausnitzii. Faecalibacterium has recently garnered attention for its potential health implications. To better understand its role, we developed and assessed real-time PCR assays for detecting and quantifying various Faecalibacterium species in human stool samples from both healthy individuals and Crohn's disease patients, either in flare or remission. The assays targeted the Microbial Anti-inflammatory Molecule (MAM) genes, which encode MAM proteins. These assays demonstrated 100% species-specificity using strains from six Faecalibacterium species: Faecalibacterium prausnitzii, Faecalibacterium taiwanense

Faecalibacterium prausnitzii (Fp), a potential candidate for a next-generation, is widely distributed in the feces and intestinal tract of mammals. Accumulating evidence indicates that Fp and its metabolites exert anti-inflammatory, immunomodulatory, and maintain intestinal barrier integrity. In this review, we summarize the biological characteristics of Fp and its nutritional cross feeding interactions with other microbial species. We further synthesize current evidence on the local roles of Fp in intestinal diseases and systemic effects on distant organs via the gut-organ axis. Furthermore,

BACKGROUND: Gut microbiota play a protective role against pneumonia in mice, probably by producing the immunomodulatory short-chain fatty acid butyrate. Yet, butyrate has limited potential for clinical use due to its challenging handling in practice. We performed mouse experiments and translational analyses to determine whether butyrate-producing gut commensals, Faecalibacterium prausnitzii and Anaerobutyricum soehngenii, could provide protection against bacterial pneumonia and serve as next-generation probiotics.
METHODS: We pre-treated C57BL/6J mice with butyrate, F. prausnitzii (recently r

OBJECTIVE: Although evidence indicates that modulating gut microbiota may mitigate psychological stress-induced obesity, the specific gut microbes and their mechanistic pathways remain unclear.
DESIGN: This study first examined the relationships between gut microbes, particularly Faecalibacterium prausnitzii (F.prausnitzii), psychological stress, and indicators of adiposity in adolescents, utilizing both cross-sectional (n = 124) and prospective cohort (n = 51) data. Second, fecal microbiota transplantation (FMT) was performed to investigate whether psychological stress induces obesity throug

Insomnia is associated with gut microbial dysbiosis, but the specific microbial metabolites mediating gut-brain communication remain elusive. Here, we integrate metagenomic sequencing from 171 individuals (primary insomnia, post-COVID insomnia, and controls) with functional pathway analysis and preclinical validation. We identify Faecalibacterium prausnitzii depletion and reduced L-arginine biosynthesis as consistent features in both insomnia subtypes, accompanied by elevated cortisol levels. Genomic and in vitro analyses confirm that F. prausnitzii is a key microbial contributor to L-arginine

BACKGROUND: Microbial anti-inflammatory molecule (MAM) is a key effector of the next-generation probiotic Faecalibacterium duncaniae A2-165, a species whose depletion in the gut microbiota is strongly linked to inflammatory bowel disease (IBD) and other conditions. Despite its importance, the direct anti-inflammatory effects of purified MAM have never been evaluated in vitro or in intestinal inflammation models. Prior studies have relied on bacterial supernatants, synthetic peptides, or DNA delivery systems, each with inherent limitations.
RESULTS: In this study, we produced and purified reco

The gut microbiota (GM) is a complex microbial ecosystem, and its alteration contributes to the development of several diseases including multiple sclerosis (MS). Progressive research concerning neurodegenerative diseases and GM reveals that GM dysbiosis has been linked to fostering the development and progression of MS. Among existing bacteria, F.prausnitzii and A.muciniphila are key species implicated in this disease. Thus, the present study systematically reviewed and synthesized the research on the involvement of F. prausnitzii and A. muciniphila in MS patients versus healthy individuals.

Ovarian cancer (OC) is a highly lethal gynecologic malignancy characterized by limited availability of treatment options and frequent recurrence. The gut microbiota has emerged as a key regulator of tumor progression; however, the anticancer potential of individual probiotic species remains insufficiently characterized and warrants further investigation. Ferroptosis is a regulated iron-dependent cell death with therapeutic potential in cancer. In this study, we initially observed that the traditional herbal pair, Scutellaria barbata D. This study investigated the effect of dietary oleanolic acid (OA) addition on broiler intestinal morphology, intestinal antioxidant capacity, changes in cecum microbiota, and the relationship between microbiota and antioxidant capacity. A total of 288 Arbor Acres broilers at 1-day-old broilers were reared, and they were randomly divided into four groups. The control and experimental groups were fed the basal diet and the basal diet supplemented with 50, 100, and 150 mg/kg OA for a total of 42 d respectively. The results showed that OA does not affect the performance of broiler chickens. The OA

BACKGROUND AND AIMS: Chronic fatigue is common in patients with inflammatory bowel disease (IBD). The gut microbiota, specifically, microbial diversity and butyrate-producing bacteria have been linked to the fatigue pathogenesis. High-dose oral thiamine reduces fatigue, potentially through gut microbiota modification. In this study, we investigated how the gut microbiota influences the efficacy of high-dose thiamine in alleviating chronic fatigue in quiescent IBD (qIBD).
METHODS: We analyzed the microbiota and short-chain fatty acids concentrations in fecal samples from patients with qIBD, wi
