Survival without photosynthesis: physiological characterization of long-lived albino beech (Fagus sylvatica).

Albinism is typically lethal in autotrophic plants due to the absence of photosynthetic pigments and functioning chloroplasts. Yet, rare exceptions occur where achlorophyllous individuals persist in natural ecosystems. We investigated the physiological, anatomical and isotopic characteristics of naturally occurring albino European beech (Fagus sylvatica L.) trees in the Moravian Karst, Czechia. One albino individual, estimated to be ~30 years old, represents an unprecedented case of long-term survival without photosynthesis in a woody angiosperm. Using a multi-parameter approach-including stab

Foliar Water Uptake Capacity in Sun and Shade Leaves of Fagus sylvatica L.

Foliar water uptake (FWU) is emerging as a key strategy for coping with drought, prompting more and deeper investigation into its role. This study examines the FWU dynamics by a series of time-dependent submersion experiments in sun and shade beech (Fagus sylvatica L.) leaves, a species known for its drought sensitivity. The results showed distinct FWU capacities, linked to their varying leaf water potentials, indicating that this factor is the driver for foliar absorption. Both sun and shade leaves experienced a proportional increase in leaf water content subsequent to FWU, with the same incr

Inter-Individual Budburst Variation in Fagus sylvatica Is Driven by Warming Rate.

The onset of the growing season in temperate forests is relevant for forest ecology and biogeochemistry and is known to occur earlier with climate change. Variation in tree phenology among individual trees of the same stand and species, however, is not well understood. Yet, natural selection acts on this inter-individual variation, which consequently affects the adaptive potential to ongoing environmental changes. Budburst dates of 146 mature individuals of Fagus sylvatica, the dominant natural forest tree of central Europe, were recorded over 12 years in one forest stand of 1 ha in the Müritz

Evolutionary Imprints on the Foliar Elementome: Biogeochemical Niche Differentiation Among Fagus Species.

Although phylogenetically related plant species often differ in competitive ability, the underlying causes remain poorly understood. The present study characterizes the biogeochemical niches (BN) of Fagus species to clarify interspecific variation in elemental acquisition, life history strategies, and competitive abilities. Quantifying the biogeochemical niche (BN) provides a mechanistic framework to address the shortfall in our understanding of how evolutionary history shapes trait-environment relationships and governs interspecific competition. We examined Fagus grandifolia Ehrh. from North

Legacy Effects in Buds and Leaves of European Beech Saplings (Fagus sylvatica) after Severe Drought.

Against the background of climate change, we studied the effects of a severe summer drought on buds of European beech (Fagus sylvatica L.) saplings and on leaves formed during the subsequent spring in trees attributed to different drought-damage classes. For the first time, we combined assessments of the vitality (assessed through histochemical staining), mass and stable carbon isotope ratios (δ13C) of buds from drought-stressed woody plants with morphological and physiological variables of leaves that have emerged from the same plants and crown parts. The number, individual mass and vitality

Short photoperiod reduces the temperature sensitivity of leaf-out in saplings of Fagus sylvatica but not in horse chestnut.

Leaf phenology is one of the most reliable bioindicators of ongoing global warming in temperate and boreal zones because it is highly sensitive to temperature variation. A large number of studies have reported advanced spring leaf-out due to global warming, yet the temperature sensitivity of leaf-out has significantly decreased in temperate deciduous tree species over the past three decades. One of the possible mechanisms is that photoperiod is limiting further advance to protect the leaves against potential damaging frosts. However, the "photoperiod limitation" hypothesis remains poorly inves

Bonding performance and surface characterization of cold-bonded acetylated beech (Fagus sylvatica L.) laminated veneer lumber.

Acetylation of wood with acetic anhydride reduces the wood-moisture interaction, improves the dimensional stability and resistance against biodegradation. However, the adhesive bonding is affected by the modification, which is crucial to manufacture engineered wood products, such as laminated veneer lumber (LVL). In this study we report the bonding of 8-layered acetylated beech (Fagus sylvatica L.) LVL boards to 2-layered LVL beams. The beams were glued together at room temperature adding three common load-bearing construction adhesives: melamine-urea-formaldehyde (MUF), phenol-resorcinol-form

The influence of O3, NO2 and SO2 on growth of Picea abies and Fagus sylvatica in the Carpathian Mountains.

At 17 long-term pollution monitoring sites throughout the Carpathian Mountains, tree growth patterns and variation in growth rate were examined to determine relationship of tree growth to specific pollutants. Canopy dominant Picea abies and Fagus sylvatica were selected at each site. Basal area increment (BAI) values were calculated from raw ring widths and used as an estimate of tree growth. Across all sites, BAI chronologies were highly variable, therefore local conditions and forest structure accounted for considerable variation. Several significant relationships, however, implicated a role

Effects of ambient ozone on seedlings of Fagus sylvatica L. and Picea abies (L.) Karst.

The effect of unfiltered versus filtered air was tested on tree seedlings at different sites with prevailing ozone pollution, at different altitudes. Growth and biomass data from different experiments were evaluated in relation to ozone dose above 40 nl l-1 during the exposure time. In beech (Fagtis sylvatica L.), biomass was significantly correlated with ozone dose whereas shoot length was not. There was no change in shoot/root ratio but there were more significant reductions in fine root biomass than in shoot biomass. The effect of ozone was cumulative within the duration of the experiments,

Genome-Wide Association Study in European Beech (Fagus sylvatica L.) for Drought Stress Related Traits.

Genome-wide association studies (GWAS) aim to identify loci associated with different traits, which could be evaluated in subsequent studies and later be prioritized in individual and seed stand selection. In trees, traits are measured at maturity or are only available after a long time, and juvenile-mature correlations are often unknown. Therefore, it is a promising approach to study natural populations directly. Data were collected from approximately 100 adult beech trees per stand in five locations in the Postăvarul Massif, South-Eastern Romanian Carpathians (Carpații de Curbură) along an a

Endophytic Bacterial Exopolysaccharide and Ethylene Hyperactivate Flavonoid Synthesis in Leaves of Fagus sylvatica L. with False Heartwood.

The presence of false heartwood in the stems of Fagus sylvatica L. significantly reduces timber quality; to date, its physiological triggers remain understudied. Research on beech stands in the forests of the Bukovinian Pre-Carpathians (Ukraine) was aimed at identifying functional links between leaf structure, synthesis of polyphenols, microbial elicitors, and false heartwood formation. Using morphometric analysis, epifluorescence microscopy, and spectrophotometry, trees with false heartwood (FH morphotype) were compared with those without it, the healthy control (HC morphotype). Reproduction-related variation in carbon allocation to woody tissues in Fagus crenata using a natural 13C approach.

The contribution of new photo-assimilates and stored carbon (C) to plant growth remains poorly understood, especially during reproduction. In order to elucidate how mast seeding affects C allocation to both reproductive and vegetative tissues, we measured biomass increase in each tissue, branch starch concentration and stable C isotope composition (δ13C) in bulk leaves, current-year shoots, 3-year branches and tree rings in fruiting and non-fruiting trees for 2 years, as well as in fruits. We isolated the effect of reproduction on C allocation to vegetative growth by comparing 13C enrichment i

Genetic transformation and CRISPR/Cas12a-mediated gene editing of European beech (Fagus sylvatica L.) employing a transient protoplast system.

Fagus sylvatica L. (European beech) is a dominant hardwood forest tree species across Central Europe, supporting diverse ecosystem services and forming the basis of a significant market for high-value timber. However, climate change increasingly threatens beech vitality and productivity, making molecular insights into its stress resilience and functional validation of underlying genes urgently needed. Here, we report a protocol for protoplast isolation from seedling leaves and demonstrate transient genetic transformation and CRISPR/Cas-mediated genome editing in F. sylvatica. PEG-mediated tran

Community Composition of Microbial Eukaryotes Transported by Stemflow from Fagus grandifolia Ehrh. (American Beech) Trees in Northeastern Ohio (USA).

Stemflow, the concentrated fraction of rainfall that drains down tree trunks, can translocate canopy biota to the forest floor, but its eukaryotic composition remains uncharacterized via eDNA methods. We collected stemflow from 18 Fagus grandifolia (American beech) trees during ten storms in northeastern Ohio (USA) and analyzed 18S rRNA eDNA to resolve transported microbial-eukaryote communities. Over 12 million reads (83 samples) revealed 920 zero-radius OTUs spanning fungi, algae, protists, and metazoans. Community composition differed significantly among storm events (PERMANOVA F = 3.6, r2

Heading for a fall: The fate of old wind-thrown beech trees (Fagus sylvatica) is detectable in their growth pattern.

Common beech (Fagus sylvatica) is one of the most important deciduous tree species in European forests. However, climate-change-induced drought may threaten its dominant position. The Sonian Forest close to Brussels (Belgium) is home to some of the largest beech trees in the world. This UNESCO world heritage site is famous for its high density of very large beech trees as a result of its climatic suitability, fertile soil conditions, and past management. Here we utilized tree-ring data from increment cores to investigate the growth of these old and monumental beech trees, evaluating their grow

Ultrastructure in leaves of Fagus silvatica and Carpinus betulus individuals tolerant and susceptible to SO2 and heavy metal pollutants.

The ultrastructure characteristics of leaves during the development of cell organelles, in the "greening process" of spring, have been studied in pollutant-tolerant and susceptible individuals of beech (Fagus silvatica) and hornbeam (Carpinus betulus) in an area polluted with SO2 (the main pollutant may reach 0.06-2.72 mg/m3), heavy metals, and other gases discharged by non-iron metal processing factories. Leaves from tolerant beech and hornbeam individuals did not undergo ultrastructural changes compared to control leaves in an unpolluted area. The susceptible and very susceptible individuals

Gene expression profiling in wounded and systemic leaves of Fagus sylvatica reveals up-regulation of ethylene and jasmonic acid signalling.

Wounding is a crucial threat to plants because of the physical damage caused and the possible entry of pathogens. Little is known about the wound reaction in forest trees. Therefore, leaves of young beech trees were wounded and the transcriptional response of wounded leaves and leaves directly above and below was analysed. A total of 123 genes exhibited significant regulation. The magnitude of regulation was slightly weaker in the downward leaves but the regulation pattern resembles that of the local and upward reactions. Thus, the signal was transduced in both vertical directions. Genes exhib

Down-regulation of defense genes and resource allocation into infected roots as factors for compatibility between Fagus sylvatica and Phytophthora citricola.

Phytophthora citricola is a wide spread and highly aggressive pathogen of Fagus sylvatica. The hemibiotrophic oomycete infects the roots and establishes a compatible interaction with F. sylvatica. To investigate the transcriptional changes associated with P. citricola infection, 68 custom oligo-microarray measurements were conducted. Hierarchical as well as non-hierarchical clustering was carried out to analyze the expression profiles. Experimental setup includes a time scale covering the biotrophic and necrotrophic stages of interaction as well as comparative analyses of the local and systemi

Different Wood Anatomical and Growth Responses in European Beech (Fagus sylvatica L.) at Three Forest Sites in Slovenia.

European beech (Fagus sylvatica L.) adapts to local growing conditions to enhance its performance. In response to variations in climatic conditions, beech trees adjust leaf phenology, cambial phenology, and wood formation patterns, which result in different tree-ring widths (TRWs) and wood anatomy. Chronologies of tree ring width and vessel features [i.e., mean vessel area (MVA), vessel density (VD), and relative conductive area (RCTA)] were produced for the 1960-2016 period for three sites that differ in climatic regimes and spring leaf phenology (two early- and one late-flushing populations)

Changes in the fine root proteome of Fagus sylvatica L. trees associated with P-deficiency and amelioration of P-deficiency.

UNLABELLED: Phosphorus is often the least available macronutrient in soil. Lack in phosphorus has detrimental effect on growth and biomass production of European Fagus sylvatica L., a major trees species in temperate forests. In contrast to leaf tissues, few studies have examined changes in the root system and no study has ever investigated the proteomic changes affected in beech roots by a lack in available phosphate (P). Here, we studied roots of young Fagus sylvatica L. trees in their native soils from two forests sites with contrasting availability of P: one P rich and P poor soil. To unde
