The Role of Fatty Acid Binding Protein 3 in Cardiovascular Diseases.

Fatty acid binding proteins (FABPs) are proteins found in the cytosol that contribute to disorders related to the cardiovascular system, including atherosclerosis and metabolic syndrome. Functionally, FABPs serve as intracellular lipid chaperones, interacting with hydrophobic ligands and mediating their transportation to sites of lipid metabolism. To date, nine unique members of the FABP family (FABP 1-9) have been identified and classified according to the tissue in which they are most highly expressed. In the literature, FABP3 has been shown to be a promising clinical biomarker for coronary

Fatty acid binding protein 3 is associated with peripheral arterial disease.

BACKGROUND: Peripheral arterial disease (PAD) affects more than 150 million people worldwide and is associated with high rates of lower extremity amputation, myocardial infarction, stroke and death. Fatty acid binding protein 3 (FABP3) is released into circulation in patients with skeletal muscle injury. In this pilot study, we investigated a possible association between PAD and blood levels of FABP3. METHODS: Blood samples were collected from patients with clinical symptoms and diagnostic findings indicative of PAD (PAD group; ankle-brachial index [ABI] <0.9; n = 75) and in those without cli

The emerging role of fatty acid-binding protein 3 (FABP3) in cancer.

Fatty acid-binding proteins (FABPs) are intracellular transporter proteins that regulate lipid transport, signalling, and homeostasis. In this review, we assess whether upregulation of FABP3 is oncogenic. Cancers where overexpression of FABP3 was shown to be oncogenic included breast, brain, nonsmall cell lung cancer (NSCLC), gastric, melanomas, and leiomyosarcoma. FABP3 knockdown in brain, oesophageal, and melanoma cell lines reduced proliferation, migration, invasion, and colony formation in vitro, and attenuated glioma tumour burden in vivo. In tumours and the tumour immune microenvironment

Association between fatty acid binding protein 3 gene variants and essential hypertension in humans.

BACKGROUND: We have earlier identified a quantitative trait locus (QTL) on rat chromosome 5 that appears to be primarily under the control of the sympathetic nervous system. Because sympathetic overactivity is related to both hypertension and insulin resistance, FABP3 is a candidate gene for the link between this QTL and blood pressure regulation. In this study, therefore, we explored the role of FABP3 genetic variations in essential hypertension (EH) in humans. METHODS: We evaluated two single-nucleotide polymorphisms (SNPs) (rs2279885 and rs2271072) in 758 patients with EH and 726 controls.

Plasma heart-type fatty acid-binding protein, omega-3 fatty acids and adverse cardiac events in the general population.

BACKGROUND: Heart-type fatty acid-binding protein (H-FABP), a principal intracellular transporter of long-chain fatty acids (FAs), has been investigated as an early diagnostic biomarker for acute myocardial infarction (MI) and acute coronary syndrome. However, evidence regarding its associations with long-term risk of cardiac events and potential interaction with omega-3 FAs remains scarce. Therefore, we aimed to investigate these associations in the general population. METHODS: This study included 42 392 participants from the UK Biobank who were free of coronary heart disease (CHD) and heart

Comparison of Fatty Acid Binding Protein 3 and Ankle Brachial Index for Predicting Peripheral Artery Disease Outcomes.

Background: Peripheral artery disease (PAD) impacts more than 200 million individuals globally. Despite its prevalence, management remains suboptimal, partly due to the lack of reliable blood-based biomarkers. The ankle-brachial index (ABI), the current gold-standard test for PAD, is limited by inter-operator variability, misinterpretation, and reduced accuracy in patients with diabetes. Fatty acid binding protein 3 (FABP3) has emerged as a potential biomarker for PAD; however, its prognostic performance relative to ABI remains unclear. This study compared FABP3 and ABI for predicting PAD outc

The optineurin fatty acid-binding protein 3 axis: A novel regulator of autophagy and apoptosis in myocardial infarction.

BACKGROUND: Autophagy and apoptosis co-regulate the fate of cardiomyocytes after myocardial infarction (MI). OPTN is a well-characterized autophagy inducer, while FABP3 promotes apoptosis. However, the functional interaction between the two in MI remained unclear. METHOD: A rat MI model was established by left anterior descending coronary artery ligation, with electrocardiography for model validation and echocardiography for cardiac function assessment. Molecular expression was analyzed via immunofluorescence. AC16 cardiomyocytes were subjected to oxygen-glucose deprivation (OGD), with plasmi

Fatty acid-binding protein 3 contributes to ischemic heart injury by regulating cardiac myocyte apoptosis and MAPK pathways.

Fatty acid-binding protein 3 (FABP3), a low-molecular-weight protein, participates in lipid transportation, storage, signaling transduction, oxidation, and transcription regulation. Here, we investigated the expression and function of FABP3 in ischemic heart diseases and explored the mechanisms by which FABP3 affected remodeling after myocardial infarction (MI). We showed that ischemic or hypoxic conditions upregulated FABP3 expression in vivo and in vitro. Notably, overexpression of FABP3 induced more myocyte apoptosis in the infarction and border areas and aggravated cardiac dysfunction, wit

Inhibition of MPTP-induced α-synuclein oligomerization by fatty acid-binding protein 3 ligand in MPTP-treated mice.

Accumulation and aggregation of α-synuclein (αSyn) triggers dopaminergic (DAergic) neuronal loss in Parkinson's disease (PD). This pathological event is partly facilitated by the presence of long-chain polyunsaturated fatty acids (LC-PUFAs), including arachidonic acid. The intracellular transport and metabolism of LC-PUFAs are mediated by fatty acid-binding proteins (FABPs). We previously reported that heart-type FABP (FABP3) interacts with αSyn, thereby promoting αSyn oligomerization in DAergic neurons in the substantia nigra pars compacta (SNpc) following 1-methyl-1,2,3,6-tetrahydropyridine

[Increased circulating level of fatty acid-binding protein 3 (FABP3): a potential biomarker of muscle damage in inflammatory myopathies patients].

Inflammatory myopathies (IM) are autoimmune diseases characterized by chronic inflammation of skeletal muscle and muscle weakness. Despite immunomodulatory treatments, a high proportion of IM patients who reach low disease activity still suffer sustained disability. This condition, termed "damage", is frequent, and correlates with handicap and mortality. Our study aimed to investigate the association between various myokines imbalance, assessed at the time of inclusion, and the "total damages" in extension and severity, assessed using IMACS (International Myositis Assessment and Clinical Studi
