Soil microbial communities are pivotal to plant health and nutrient acquisition. It is becoming increasingly clear that many interactions, both among and between microbes and plants, are governed by small bioactive molecules or "secondary metabolites" that can aid in communication, competition, and nutrient uptake. Yet, secondary metabolite biogeography - who makes what, where, and why-is in its infancy. Further, secondary metabolite biosynthesis genes are often silent or weakly expressed under standard laboratory conditions, making it incredibly difficult to study these small molecules. To be

Secondary plant metabolites are chemical compounds produced by plants through a variety of metabolic pathways. It is known that these compounds, among others, monoterpenes, are characterized by holding valuable pro-health activities when consumed or applied. Taking into account the development of civilizational diseases, eating foods enriched with compounds such as the monoterpenes is highly recommended. Good sources of such health-promoting food items include common fruits and vegetables, seed sources and plant parts used to enhance flavour such as spices. It is known that monoterpene compoun

BACKGROUND: Leucine aminopeptidase (LAP) is a promising biomarker for the detection and diagnosis of liver diseases encompassing liver cancer and liver injury. Existing assays for LAP activity have limitations, as most of them rely on substrates that are cumbersome to synthesize and fail to amplify the detection signal.
RESULTS: Initially, L-pAP was synthesized to serve as a substrate for LAP through the condensation of p-aminophenol (pAP) with leucine. LAP triggered the hydrolyzation of L-pAP into pAP, which then reduced colourless Fe3+-ferrozine to purple Fe2+-ferrozine. Upon addition of tr

Although a number of reducing systems can release iron from ferritin, there is debate as to whether the process additionally requires a chelator. We have studied ferritin iron release by microsomes, paraquat and NADPH, by dialuric acid and by hypoxanthine and xanthine oxidase, using ferrozine to complex the released iron. In each case, Fe2+ (ferrozine) formation was detectable when the ferrozine was added at the beginning of the 10 min reaction period, but not at the end. However, with catalase present, up to 0.7 times as much Fe2+ could be measured with ferrozine added at the end. Further Fe2

A sensitive FerroZine assay is used to measure the membrane-bound ferric-chelate reductase activity in the Arabidopsis thaliana roots. In Arabidopsis, FRO2 (FERRIC CHELATE REDUCTASE 2) encodes the Fe(III) chelate reductase and its expression is induced by iron deficiency. As FRO2 reduces Fe(III) to soluble Fe(II), the resulting Fe(II) forms a purple-colored complex with the dye FerroZine. The concentration of the Fe(II)-FerroZine is directly proportional to the absorbance at 562 nm.

The routine method for LDH (Lactate dehydrogenase) activity determination is to monitor the increase of NADH concentration at 340 nm. There are some inconvenience in taking measurements in the near-UV region, especially in the case of serum samples analysis. In this work, two modifications of the routine LDH activity assay based on the use of reducing properties of NADH have been compared. Both methods involved the reduction of compounds that can be easily determined by well-known methods, ferric ion (with ferrozine) and nitrotetrazolium blue (NBT). A fully-mechanized Multicommutated Flow Anal

Blood iron levels play a vital role in oxygen metabolism and energy generation whilst transporter protein, transferrin, binds and delivers iron to the transferrin receptor of endosomal compartments of cells. Consequently, the iron-binding capacity of transferrin is an important indicator for many diseases, and its measurements are used in the diagnosis and treatment of anaemias. Various assays, including Total Iron Binding Capacity (TIBC), Unsaturated Iron-Binding Capacity (UIBC) and Transferrin Saturation (TS), were developed to assess the iron-binding capacity of transferrin. Clinically, UIB

Lactate, a crucial metabolic indicator, plays a vital role in diagnostics and treatment monitoring. Current lactate analysis often relies on enzymatic methods, employed in both clinical analyzers and point-of-care testing (POCT) devices. While these enzymatic assays offer selectivity and sensitivity, they suffer from limitations including higher costs, shorter lifetime, and reduced stability, particularly impacting POCT applications. This work introduces a novel POCT platform for lactate determination, utilizing a non-enzymatic approach. This analytical system is based on the photoreduction of

Fe(II) has been extensively studied due to its importance as a reductant in biogeochemical processes and contaminant attenuation. Previous studies have shown that ligands can alter aqueous Fe(II) redox reactivity but their data interpretation is constrained by the use of probe compounds. Here, we employed mediated electrochemical oxidation (MEO) as an approach to directly quantify the extent of Fe(II) oxidation in the absence and presence of three model organic ligands (citrate, nitrilotriacetic acid, and ferrozine) across a range of potentials (EH) and pH, thereby manipulating oxidation over

Vitamin C (ascorbic acid) has been known to prevent scurvy for many years. Recent research has shown its importance in lipid and iron metabolism. Vitamin C may also have some effect on the immune system. There is not as yet conclusive evidence that ascorbic acid may cure or prevent colds or cancer. The vitamin has few side effects even when ingested in large quantities. Several methods for analysis of ascorbic acid have been developed. These include titration and fluorometric methods, a ferrozine technique automated for centrifugal analyzers, a high performance liquid chromatography method, an

Protein distribution profiles may be used to characterize both physiological and pathophysiological cellular changes, but rigorous biochemical assays for measuring such movements are lacking. This paper reports on a protein redistribution assay that combines reversible metal chelate-based total protein detection with a four-fraction subcellular detergent fractionation procedure. TNF-alpha stimulated cultured human omental microvessel endothelial cells are fractionated into cytosol, membrane/organelle, nuclear (envelope and associated), and cytoskeletal/DNA compartments. Protein fractions are s

Limitations of the colorimetric assay involving tetramethylmurexide (TMM) to determine the extent of complex formation between metal ions and phenolic compounds have been studied. Older literature reports using this method to determine bound Fe(II). Our study shows the TMM assay is inadequate when determining the Fe(II) chelation activity of phenolic preparations rich in tannin constituents on account of the high absorbance values derived by control samples (i.e., those that do not contain the TMM reagent). Phenolic test samples comprising the TMM reagent, iron ions, and tannins could not yiel

We have investigated the mechanism of reduction of organically complexed iron(III) in the presence of superoxide, the one-electron reduced form of dioxygen that is produced in natural waters by thermal, photochemical, and biological pathways. Experimental results show that reduction of organically complexed iron(III) by superoxide may occur by either (or, in some instances, both) reaction of superoxide with inorganic iron(III) after its dissociation from the complex (dissociative reduction) or by direct reaction of superoxide with the complex (non-dissociative reduction). In the presence of lo

The mobility and toxicity of chromium (Cr) in soil and water systems are largely controlled by its oxidation state and interactions with solid phases. Relative to abiotic minerals, biogenic iron (Fe) (oxyhydr)oxides (BIOS) may enhance Cr(vi) adsorption and reduction due to their poorly ordered structures, large surface areas, and incorporation of cell derived organic matter. To determine the extent and mechanisms of the reaction between Cr(vi) and BIOS, sorption isotherm and kinetic studies were conducted using two-line ferrihydrite, BIOS, and BIOS amended with 0.135 M ferrozine (an Fe(ii) che

Previous studies documented the abnormal association of heme and heme proteins with the sickle RBC membrane. We have now examined RBC ghosts and inside-out membranes (IOM) for the presence of nonheme iron as detected by its formation of a colored complex with ferrozine. Sickle ghosts have 33.8 +/- 18.2 nmol nonheme iron/mg membrane protein, and sickle IOM have 4.3 +/- 3.0 nmol/mg. In contrast, normal RBC ghosts and IOM have no detectable nonheme iron. The combination of heme and nonheme iron in sickle IOM averages nine times the amount of membrane-associated iron in normal IOM. Kinetics of the

BACKGROUNDS: Joint iron overload in hemochromatosis induces M1 polarization in synovial macrophages, releasing pro-inflammatory factors and leading to osteoarthritis development. However, the mechanism by which iron overload regulates M1 polarization remains unclear. This study aims to elucidate the mechanism by which synovial iron overload promotes macrophage M1 polarization.
METHODS: In vitro, RAW264.7 macrophages were treated with iron and divided into five groups based on the concentration of the iron chelator, desferrioxamine (DFO): Ctrl, Fe, DFO1, DFO2, and DFO3. In vivo, rats were cate

Double radioactive label transport assays with iron, chromium, and gallium chelates were used to investigate the mechanism of iron uptake by Ustilago sphaerogena. In iron-deficient cells, ferrichrome A iron was taken up without appreciable uptake of the ligand. Iron-sufficient cells partially accumulated the ligand with the metal. The chromium- and gallium-containing analogs of ferrichrome A were transported as intact chelates. Ferrichrome A iron uptake was inhibited by dipyridyl. The data suggest that the intact ferrichrome A chelate binds to a specific receptor, the iron is then separated fr

Procaspase-3-activating compound 1 (PAC-1) induces procaspase-3 activation via zinc chelation. However, whether PAC-1 employs other mechanisms remains unknown. Here we systematically screened for potent PAC-1 targets using 29 enhanced green fluorescent protein-labeled reporter cell lines and identified hypoxia-inducible factor 1α (HIF1α) and RAD51 pathways as PAC-1 targets. These results were verified in HepG2 cells and two other cancer cell lines. Mechanistically, PAC-1 specifically blocked HIF1α hydroxylation and upregulated HIF1α target genes. In addition, DNA damage, G1/S cell cycle arrest

Mortality associated with mucormycosis remains high despite current antifungals. Iron-starvation strategies have been shown to have promising activity against Mucorales. We hypothesized that iron starvation enhances apoptosis in Rhizopus oryzae. Apoptosis was characterized in R. oryzae transformed with RNAi plasmid targeting FTR1 expression (iron permease mutant) or empty plasmid grown in iron rich (0.125% FeCl3) and iron depleted media (YNB+1mM ferrozine and 1 mM ascorbic acid). Increased apoptosis was observed with dihydrorhodamine-123 and rhodamine-123 staining in the iron starved mutant FT

Transition metal ions are a challenge to study in physiology because of problems associated with solubility, oxidation, binding, and attaining appropriate free activities in solution. This review discusses these problems and potential ways of accommodating them. Special attention is given to iron and zinc ions, but many of the concepts can be applied for studying other transition metals. Selection of reagents appropriate for metal work (including water, salts, noncomplexing pH buffers) is briefly discussed. Calculation of the solubility product (K(sp)) for common iron and zinc precipitates is
