Novel Fibrillar and Non-Fibrillar Collagens Involved in Fibrotic Scar Formation after Myocardial Infarction.

Following myocardial infarction (MI), adverse remodeling depends on the proper formation of fibrotic scars, composed of type I and III collagen. Our objective was to pinpoint the participation of previously unreported collagens in post-infarction cardiac fibrosis. Gene (qRT-PCR) and protein (immunohistochemistry followed by morphometric analysis) expression of fibrillar (types II and XI) and non-fibrillar (types VIII and XII) collagens were determined in RNA-sequencing data from 92 mice undergoing myocardial ischemia; mice submitted to permanent (non-reperfused MI, n = 8) or transient (reperfu

Next-Generation Sequencing and Variant Analysis of Fibrillar (Pro)Collagens.

Genetic defects in the genes encoding fibrillar collagen types I, II, III, V, and XI result in a variety of heritable connective tissue disorders. Next-generation sequencing (NGS) applications have facilitated the screening of these disease genes, but the interpretation of the obtained variants can be challenging. We describe a protocol for both targeted gene panel and exome-wide library preparation, bioinformatic pipeline, and variant classification and interpretation of germline variants in types I, II, III, V, and XI collagen.

COLdb, a database linking genetic data to molecular function in fibrillar collagens.

Fibrillar collagens are ubiquitous proteins essential for the structural integrity of bones, skin, blood vessels, and other tissues. Mutations in collagen genes result in disorders including osteogenesis imperfecta, chondrodysplasias, and Ehlers-Danlos syndromes, but the molecular basis for the heterogeneity of clinical phenotypes is not well understood. A more complete understanding of the relationship between sequence and phenotype requires synthesis of multiple facets of collagen structure and function. To facilitate such an analysis, we developed COLdb, a freely available database integrat

Astrocytes in culture express fibrillar collagen.

The use of monoclonal antibodies has led to much progress in the characterization of extracellular matrix components of the CNS. F1C3 is a monoclonal antibody raised against the astrocytic cell line, Neu7. Analysis by immunoprecipitation and Western blots of the F1C3 antigen in Neu7 cell lysates and conditioned medium reveals a recognition of several protein bands around 140-230 kD. Internal peptide sequence data from these bands indicate that they are highly homologous to fibrillar collagens, and the F1C3 antigen is specifically digested by the collagenase I protease. Other glial cell lines s

Methods for Quantifying Fibrillar Collagen Alignment.

Recent evidence has implicated collagen, particularly fibrillar collagen, in a number of diseases ranging from osteogenesis imperfecta and asthma to breast and ovarian cancer. A key property of collagen that has been correlated with disease has been the alignment of collagen fibers. Collagen can be visualized using a variety of imaging techniques including second-harmonic generation (SHG) microscopy, polarized light microscopy, and staining with dyes or antibodies. However, there exists a great need to easily and robustly quantify images from these modalities for individual fibers in specified

Molecular insights into prolyl and lysyl hydroxylation of fibrillar collagens in health and disease.

Collagen is a macromolecule that has versatile roles in physiology, ranging from structural support to mediating cell signaling. Formation of mature collagen fibrils out of procollagen α-chains requires a variety of enzymes and chaperones in a complex process spanning both intracellular and extracellular post-translational modifications. These processes include modifications of amino acids, folding of procollagen α-chains into a triple-helical configuration and subsequent stabilization, facilitation of transportation out of the cell, cleavage of propeptides, aggregation, cross-link formation,

New Altered Non-Fibrillar Collagens in Human Dilated Cardiomyopathy: Role in the Remodeling Process.

BACKGROUND: In dilated cardiomyopathy (DCM), cardiac failure is accompanied by profound alterations of extracellular matrix associated with the progression of cardiac dilation and left ventricular (LV) dysfunction. Recently, we reported alterations of non-fibrillar collagen expression in ischemic cardiomyopathy linked to fibrosis and cardiac remodeling. We suspect that expression changes in genes coding for non-fibrillar collagens may have a potential role in DCM development. OBJECTIVES: This study sought to analyze changes in the expression profile of non-fibrillar collagen genes in patients

Influence of collagen source on fibrillar architecture and properties of vitrified collagen membranes.

Collagen vitrigel membranes are transparent biomaterials characterized by a densely organized, fibrillar nanostructure that show promise in the treatment of corneal injury and disease. In this study, the influence of different type I collagen sources and processing techniques, including acid-solubilized collagen from bovine dermis (Bov), pepsin-solubilized collagen from human fibroblast cell culture (HuCC), and ficin-solubilized collagen from recombinant human collagen expressed in tobacco leaves (rH), on the properties of the vitrigel membranes was evaluated. Reinforcement of Fibrillar Collagen Hydrogels with Bioorthogonal Covalent Crosslinks.

Bioorthogonal covalent crosslinking stabilizes collagen type I hydrogels, improving their structural integrity for tissue engineering applications with encapsulated living cells. The chemical modification required for crosslinking, however, interferes with the fibrillar nature of the collagen, leading instead to an amorphous network without fibers. We demonstrate an approach to perform bioconjugation chemistry on collagen with controlled localization such that the modified collagen retains its ability to self-assemble into a fibrillar network while also displaying functional groups for covalen

Steady-State and Pulse-Chase Analyses of Fibrillar Collagen.

Steady-state and pulse-chase collagen analyses are powerful approaches to investigate in vitro the structure of collagen and its kinetic of secretion, respectively. The electrophoretic analysis of purified 3H-proline-labeled collagen allows to determine the nature and post translational modifications of its α chains, whereas short-pulse labeling can be used to follow collagen secretion over time.

Cardiac interstitium in health and disease: the fibrillar collagen network.

Composed of type I and III collagens, the valve leaflets, chordae tendineae and collagen matrix of the myocardium form a structural continuum. Synthesized by cardiac fibroblasts, these fibrillar collagens support and tether myocytes to maintain their alignment, whereas their respective tensile strength and resilience resist the deformation, maintain the shape and thickness, prevent the rupture and contribute to the passive and active stiffness of the myocardium. An acquired or congenital defect in this collagen network can lead to abnormalities in myocardial architecture, mechanics or valve fu

Development and ageing of the articular cartilage of the rabbit knee joint: distribution of the fibrillar collagens.

The development of the articular cartilage of the rabbit knee joint from the 17-day fetus to the 2-year adult rabbit has been examined. At 17 days, the developing femur and tibia are separated by the interzone. Cavitation occurs around 25 days; the cells of the intermediate layer flatten and move onto those of the chondogenous layers to create the articular surfaces. After birth, growth of the cartilage is mainly the result of matrix production. Ossification of the epiphyses is complete by 6 weeks postpartum. Horizontal zones can be distinguished in the articular cartilage; the superficial cel

Insights on the evolution of prolyl 3-hydroxylation sites from comparative analysis of chicken and Xenopus fibrillar collagens.

Recessive mutations that prevent 3-hydroxyproline formation in type I collagen have been shown to cause forms of osteogenesis imperfecta. In mammals, all A-clade collagen chains with a GPP sequence at the A1 site (P986), except α1(III), have 3Hyp at residue P986. Available avian, amphibian and reptilian type III collagen sequences from the genomic database (Ensembl) all differ in sequence motif from mammals at the A1 site. This suggests a potential evolutionary distinction in prolyl 3-hydroxylation between mammals and earlier vertebrates. Using peptide mass spectrometry, we confirmed that this

Fibrillar Collagen Quantification With Curvelet Transform Based Computational Methods.

Quantification of fibrillar collagen organization has given new insight into the possible role of collagen topology in many diseases and has also identified candidate image-based bio-markers in breast cancer and pancreatic cancer. We have been developing collagen quantification tools based on the curvelet transform (CT) algorithm and have demonstrated this to be a powerful multiscale image representation method due to its unique features in collagen image denoising and fiber edge enhancement. In this paper, we present our CT-based collagen quantification software platform with a focus on new f

Molecular changes in fibrillar collagen in myxomatous mitral valve disease.

INTRODUCTION: Myxomatous mitral valve disease (MMVD) is the single most common acquired cardiac disease of dogs and is a disease of significant veterinary importance. It also bears close similarities to mitral valve prolapse in humans and therefore is a disease of emerging comparative interest. We have previously mapped the structure of collagen fibrils in valve leaflets using synchrotron X-rays and have demonstrated changes in collagen structure associated with the regions of disease. METHODS: Differential scanning calorimetry (DSC),biochemical assay of collagen content, high-performance liq

Type V and VI collagen for cohesion of dermal fibrillar structures.

Type V and VI collagen were capable to joint each other and with type I and IV collagen, as well as mucopolysaccharides. This capability suggested that these collagens function for cohesion of fibrillar tissue components of dermis. This study demonstrated the locality of these types of collagen in dermis. Fresh specimens of normal skin were fixed in 2% paraformaldehyde in phosphate-buffered saline, overnight. Besides, in order to loosen the twist of collagen fibril, some pieces of the skin specimens were treated by citrate buffer pH 3.0, prior to fixation. The specimens were embedded in Techno

Collagen-mediated biofilm formation and fibrillar organization in Staphylococcus epidermidis.

The symbiotic skin bacterium Staphylococcus epidermidis moisturizes human skin, contributes to protection against external pathogens, and supports innate immune stimulation. However, the interaction between collagen, a major skin protein, and S. epidermidis remains insufficiently explored. In this study, biofilm development was investigated using scanning electron microscopy with ionic liquid pretreatment, confocal laser scanning microscopy, biofilm quantification assays, and growth curve analysis. In the presence of collagen, cells adhered to the substrate, secreted microvesicles, formed web-

SNED1 fibrillar assembly in the extracellular matrix requires fibronectin and collagen I.

The extracellular matrix (ECM) is a meshwork of proteins that orchestrates a broad range of cellular phenotypes, including proliferation, adhesion, migration and differentiation. SNED1 is a newly characterized ECM glycoprotein that promotes cell adhesion and is essential for embryonic development. Its upregulation is also associated with breast cancer metastasis and poor prognosis for breast cancer patients. We have recently shown that SNED1 assembles into fibrillar structures, but the mechanisms guiding its incorporation into the ECM scaffold remained unknown. Here, combining biochemical assa
