[Deviation of the life cycle of Dipetalonema viteae (Filarioidea)].

Dipetalonema viteae is a filarial that can evoluate among hosts zoologically broadly apart (Ixodides and Argasides), but always gathered from meriones burrows. Its evolution is, on the contrary, blocked among most of the other ticks, particularly among Ornithodoros erraticus morphologically very similar to the normal vector. Our work concerns the experimental deviation of the cycle of Dipetalonema viteae, with its possible adaptability in the bosom of an intermediate of fowl tropism and a permanent host, different from a gnawing.

Cryopreservation of infective larvae of Onchocerca volvulus (Filarioidea: Onchocercidae).

Infective larvae (L3) of Onchocerca volvulus were procured in Liberia, West Africa, in the natural black fly vector, Simulium yahense. A cryobiological technique was developed to preserve L3 of O. volvulus that were fully viable after thawing. Larvae were treated before cooling with 4 cryoprotective compounds. Three compounds, dimethyl sulfoxide (DMSO), glycerol, and ethylene glycol, were prepared with distilled water. The fourth compound was DMSO prepared in different concentrations with 0.25 M sucrose. The treatment with DMSO + 0.25 M sucrose cryoprotectant resulted in the highest survival o

Ultrastructural analysis of Wuchereria bancrofti (Nematoda: Filarioidea) body wall.

Bancroftian filariasis constitutes the principal mosquito-borne nematode infection of humans and the surface of adult of Wuchereria bancrofti seems to be especially important in the intricate interplay between host and parasite. The study of the parasite's surface structure might help to understand the localization and function of various organelles. W. bancrofti adult worms were recovered from untreated patients during hydrocele repair surgery and studied by transmission electron microscopy. The body wall of adult parasite is composed of cuticle, hypodermis and muscular layer. Cuticle is the

Scanning electron microscopy of adult Wuchereria bancrofti (Nematoda: Filarioidea).

Despite the great importance of Wuchereria bancrofti in causing human lymphatic filariasis, only conventional morphological studies have been completed with adult forms of this filaria. No ultrastructural studies have been carried out, mainly due to the difficulty in obtaining viable parasites from human tissues and the lack of a suitable experimental model or in vitro cultivation. With the recent success in using ultrasound to localize adult worms in living tissues and their surgical recovery from human lymphatic vessels, we show in the present paper the first ultrastructural observations of

Onchocerca sweetae (Nematoda: Filarioidea): notes on the intermediate host.

Microfilariae of Onchocerca sweetae are broadly distributed in the superficial layers of the dermis of the water buffalo (Bubalus bubalis). A total of 2855 insects representing 20 species were collected from O. sweetae-infected bait buffaloes. Only one species, Culicoides sp. "M", ingested microfilariae from buffalo skin. Larval development of O. sweetae was observed in the thorax of this species. Atotal of 829 insects, representing 7 species and including 749 parous Culicoides spp. were collected from light and Manitoba traps. Developing filarioid larvae were observed only in Culicoides sp. Polyamine uptake by a rodent filariid, Acanthocheilonema viteae (Nematoda: Filarioidea).

Helminth parasites lack the capacity to synthesize polyamines de novo. With the view to characterise alternative pools for the replenishment of polyamines, uptake of spermidine and spermine, were studied in Acanthocheilonema viteae, a parasite known to infect rodents. Motile worms recovered from the subcutaneous tissues of experimentally infected Mastomys natalensis were incubated in vitro with radiolabelled polyamines for the uptake assays. A time-dependent, temperature-sensitive, energy-requiring and saturable-uptake of the polyamines was observed. Male worms exhibited better uptake than the

Immunocytochemical studies on several developmental stages of Dipetalonema viteae (Filarioidea).

The binding of antibodies to infective larvae (L3), eggs, uterine contents and blood microfilariae was demonstrated by light microscopy employing the peroxidase-antiperoxidase (PAP) method. Antigen-antibody reactions were located on the shell of filarial eggs as well as on the cuticle and the interior of L3 and microfilariae. Using sera from Meriones unguiculatus, Mastomys natalensis and golden hamsters infected with D. viteae obtained at 7, 14, 21 and 28 weeks p.i. it was observed that the intensity of the immunostaining on several developmental stages of the filariae decreased with increasin

Brugia timori sp. n. (nematoda: filarioidea) from Flores Island, Indonesia.

Brugia timori sp. n. from experimentally-infected Mongolian jirds (Meriones unguiculatus) is described. The adult male differs from other Brugia species, except Brugia malayi, in having a spicular ratio of 3 : 1; it differs from B. malayi in having greater numbers of subventral adanal papillae (up to 5 on each side) that are loosely spaced and irregularly positioned about the cloaca, a greater diameter of the capitulum of the left spicule, greater lengths of the proximal- and midsections of the left spicule, and a greater length of the proximal section of the right spicule. The ultrastructure of adult Brugia malayi (Brug, 1927) (Nematoda: Filarioidea).

The ultrastruct of the adult subperiodic Brugia malayi (Brug, 1927) within pulmonary arteries of male jirds (Meriones unguiculatus) was studied by transmission electron microscopy. The cuticle consists of 10 sublayers (2 of which are prominently banded) and a typical outer unit membrane. Evidence is presented showing that the subcuticular region of the lateral chords comprises a functional complex of basal infoldings, multivesicular bodies, and associated mitochondria, which is probably engaged in the exchange of solutes across a permeable cuticle. Effects of albendazole on Litomosoides chagasfilhoi (Nematoda: Filarioidea) females in vivo.

Gerbils (Meriones unguiculatus) experimentally infected with Litomosoides chagasfilhoi were treated with a single oral dose of 40 or 80 mg of albendazole, respectively. Observation of the microfilaremia after the treatment showed that both single oral doses of albendazole decreased the microfilaremia in L. chagasfilhoi infection. The body wall was composed of a cuticle, a hypodermis, and a muscular layer, and treated nematodes showed no morphological alterations. The ultrastructural alterations produced by treatment with 40 mg of albendazole included a higher number of membrane invaginations i

Moulting and exsheathment of the infective larvae of Onchocerca lienalis (Filarioidea) in vitro.

Fourth-stage larvae (L4) of the cattle filarial parasite Onchocerca lienalis were produced from third stage forms (L3) and maintained in vitro using a variety of culture media and feeder cell layers. Up to 50% of the L3 larvae moulted to the fourth stage in the presence of two jird (Meriones unguiculatus) cell lines, a monkey kidney cell line and also with Vero cells. Moulting took place 2-5 days after of the initiation of the cultures: These L4 parasites could be maintained in a healthy condition for up to 88 days, although no significant increases in size of these larvae were observed during

Ultrastructure of the rectum of infective-stage Wuchereria bancrofti (Nematoda: Filarioidea).

The authors have examined the ultrastructure of the rectum of infective-stage Wuchereria bancrofti by transmission electron microscopy. Our observations show that the rectum is divided into anterior and posterior segments. The cells of the anterior rectum appear to be derived from the microfilarial R (rectal) cells described by other authors. In both stages, these cells show voluminous nuclei, abundant mitochondria, and small cytoplasmic processes which contain fibrillar components. Amorphous material associated with these processes appears throughout the larval rectum and may protrude from th

Zoonotic Dirofilaria repens (Nematoda: Filarioidea) in Aedes vexans mosquitoes, Czech Republic.

The surveillance of vectors for arthropod-borne pathogens is nowadays an important tool in surveillance programmes throughout Europe. Whereas many studies have been performed to screen arthropods for viruses or bacterial pathogens, only limited information is available concerning the geographical distribution and vector range of pathogenic filariae in Central Europe. To consider the prevalence of filarial parasites in mosquito vectors, we performed a molecular survey of mosquitoes for filarial DNA. Mosquito collection was conducted at six study sites in the South Moravian region (Czech Republi

Redescription of Parafilaria bovicola Tubangui, 1934 (Nematoda: Filarioidea) from Swedish cattle.

Parafilaria bovicola was introduced into Sweden and identified for the first time in Swedish cattle in 1978. Since then the parasite has become well established and has been responsible for substantial economic losses in Swedish beef production. Although studies have been undertaken on serodiagnosis, life cycle, vector and geographical distribution within Sweden this is the first full description of P. bovicola from Swedish cattle and the first study to use scanning electron microscopy. P. bulgarica Daskalov, 1944 is regarded as a synonym of P. bovicola Tubangui, 1934. Tooth-like thickenings d

Identification of a potential protective microfilarial antigen from Litomosoides carinii (Nematoda, Filarioidea).

The proteins of the filarial parasite Litomosoides carinii that are presented to the host in the course of a natural infection were assessed by surface and intrinsic labelling, microfilarial agglutination assays and ELISA. Surface labelling revealed several proteins present on all parasite stages which were recognised by infected animals. Only one major surface protein of 55 KD was found on microfilariae, while in all other stages (adult males, adult females and infective larvae isolated from pool animals) a range of proteins could be labelled. Immunisation with live microfilariae obtained fro

Characterization of aortic and brachiocephalic filariasis by Filarioidea sp (Nematoda:Spirurida:Filarioidea) in Mexican ramphastids.

The manuscript presented herein documents the findings of filaria nematodes in 5 keel-billed toucans, and one emerald toucanet, originated from 2 private aviaries in Mexico City during two years. The birds displayed ruffled feathers, depression, inability to perch, convulsions, and sudden death. Furthermore, thickened wall of the aortic and brachiocephalic arteries, with connective tissue proliferation and chondroid metaplasia were observed. Molecular characterization matched Filarioidea sp (Nematoda: Spirurida: Filarioidea). To the authors' knowledge, this is the first documented report of fi

Cytochemical characterization of the third-stage larva of Wuchereria bancrofti (Nematoda: Filarioidea).

In the present paper, we report the results we obtained using several cytochemical techniques to analyze the infective larva of Wuchereria bancrofti. An imidazole osmium tetroxide solution was used to visualize unsaturated fatty acids. A highly contrasted material forming a continuous structure was observed on the larval surface and over the epicuticle. A strong reaction was observed on the esophagus and also on the inner secreted material. Carbohydrates containing vic-glycol groups were not observed on the cuticle of the third-stage larva of W. bancrofti submitted to the Thiéry technique. Usi
