Bioactivity of flavonoids.

Flavonoids are benzo-gamma-pyrone derivatives of plant origin. They possess wide spectrum of biological activity. From the therapeutical point of view the most important are their antioxidant properties. These are the result of high propensity to electron transfer, ferrous ions chelating activity and direct scavenging of reactive oxygen species. Flavonoids inhibit enormous number of enzymes. From the pharmacological point of view inhibition of cyclooxygenase and lipoxygenases as well as scavenging of superoxide anions seem to be essential. Flavonoids are antiinflammatory agents as the result o

Flavonoids as acetylcholinesterase inhibitors.

Flavonoids are new promising potential natural compounds for treating Alzheimer's disease (AD). Actually most promising drugs for symptomatic treatment of AD are acetylcholinesterase inhibitors (AChEI). Flavonoids with AChE inhibitory activity and due to their well known antioxidant activity could be new multipotent drugs for AD treatment. This work focuses on natural and synthetic flavonoids inhibitors of the enzyme acetylcholinesterase (AChE). Over, all this review refers to 128 flavonoids, which are classified in chemical structure, and summarizes 64 references.

Flavonoid antioxidants.

In order to ascertain the role of dietary flavonoids as antioxidants in vivo it is necessary to understand the chemical nature of the absorbed forms in the circulation in vivo and how the multiplicity of research findings in vitro reflect the bioactivity of flavonoids in vivo. Only when we gain adequate information on the circulating forms can we begin to understand the targeting to the tissues, whether flavonoids cross the blood-brain barrier, for example, and in what forms. Flavonoids are powerful antioxidants in vitro, but their overall function in vivo has yet to be clarified, whether anti

Functionalization of Flavonoids with Lipases.

Flavonoids, valued for their health benefits, face challenges due to low solubility. Enzymatic acylation offers promise in enhancing properties, with the use of lipase B sourced from Candida antarctica (N435). This study has the aim to improve naringin solubility via esterification using as acyl donors short- and medium-chain fatty acids (vinyl acetate, propionate, and laurate) catalyzed by N435. Lipases play a crucial role as catalysts, facilitating the acylation process. Our study encompasses executing acylation and characterizing products. This process significantly enhances aqueous and org

Flavonoids and polycystic ovary syndrome.

Polycystic ovary syndrome (PCOS) is a heterogeneous endocrine disorder affecting women, characterized by irregular menstrual cycles, elevated androgen levels, and ovulatory dysfunction. Current clinical treatments, such as combined oral contraceptive pills (COCP), metformin, and clomiphene, can alleviate symptoms but are often associated with adverse effects including gastrointestinal discomfort, the risk of ovarian hyperstimulation syndrome (OHSS), and potential deterioration of cardiometabolic indices. Thus, there is an urgent need for safer and well-tolerated alternative therapies. Flavonoi

[Flavonoids: antithrombotic agents or nutrients?].

Flavonoids are a vast group of natural substances, but their pharmacological properties have not all been explored. The term flavonoid is used at large to designate a series of more than 4,000 molecules, which in fact can have very heterogenous molecular structures. We have shown that some flavonoids are good inhibitors of cyclic nucleotide phosphodiesterase (PDE). The most active PDE inhibitors among the flavonoids were also good inhibitors of the aggregation of human platelets in vitro. This suggests that flavonoids could serve as a template for the development of new anti-platelet drugs. Ho

NMR Chemical Shifts of Common Flavonoids.

We present 1H and 13C nuclear magnetic resonance (NMR) chemical shifts for the most common flavonoids, a class of natural products widely present in the plant kingdom. Our data are acquired in three common NMR solvents: methanol d4, dimethyl sulfoxide d6, and pyridine d5 and may be used as a reference for isolated compounds, for detecting flavonoids within mixtures like plant extracts as shown in an example, as reference data for natural product databases, for refined training of prediction software, and for machine learning in resonance assignment and NMR-based structure elucidation. Biological valorization of lignin to flavonoids.

Lignin is the most affluent natural aromatic biopolymer on the earth, which is the promising renewable source for valuable products to promote the sustainability of biorefinery. Flavonoids are a class of plant polyphenolic secondary metabolites containing the benzene ring structure with various biological activities, which are largely applied in health food, pharmaceutical, and medical fields. Due to the aromatic similarity, microbial conversion of lignin derived aromatics to flavonoids could facilitate flavonoid biosynthesis and promote the lignin valorization. This review thereby prospects a

Glycosylated flavonoids: fruit's concealed antifungal arsenal.

Fruit defense against pathogens relies on induced and preformed mechanisms. The present contribution evaluated performed resistance of red and green mango fruit against the fungal pathogen Colletotrichum gloeosporioides and identified the main active antifungal components. High-performance liquid chromatography analysis of nonhydrolyzed mango peel extracts identified major anthocyanin peaks of glycosylated cyanidin and methylcyanidin, and flavonol peaks of glycosylated quercetin and kaempferol, which were more abundant on the 'red side' of red mango fruit. Organic extracts of red vs green mang

Flavonoids interfere with NLRP3 inflammasome activation.

NOD-like receptor (NLR) family, pyrin domain-containing 3 (NLRP3) inflammasome is a component of innate immunity, and is responsible for producing mature IL-1β and -18. Several flavonoids were found to affect inflammasome pathway, but the mechanism of action is still obscure. To elucidate the effects on NLRP3 inflammasome pathway and to determine the structure-activity relationships, NLRP3 inflammasome in differentiated THP-1 cells was activated via treatment with monosodium urate (MSU) crystals. Levels of mature IL-1β, NLRP3 inflammasome components and apoptosis-associated speck-like protein

Flavonoids in Kidney Health and Disease.

This review summarizes the latest advances in knowledge on the effects of flavonoids on renal function in health and disease. Flavonoids have antihypertensive, antidiabetic, and antiinflammatory effects, among other therapeutic activities. Many of them also exert renoprotective actions that may be of interest in diseases such as glomerulonephritis, diabetic nephropathy, and chemically-induced kidney insufficiency. They affect several renal factors that promote diuresis and natriuresis, which may contribute to their well-known antihypertensive effect. Flavonoids prevent or attenuate the renal i

Protection against peroxynitrite reactions by flavonoids.

Peroxynitrite is believed to contribute to pathogenesis of various diseases. Search for drugs and nutraceuticals able to limit peroxynitrite reactions is thus of interest. This study aimed to compare the capacity of 16 flavonoids and several other compounds present in food to prevent peroxynitrite reactions in three different test systems, based on fluorescein bleaching, tyrosine nitration and serum albumin thiol oxidation. The sequence of protective capacity of the antioxidants was different in various systems. Correlation analysis revealed that the hydroxyl group at the R'4 position of the f

Flavonoids: recent advances as anticancer drugs.

Flavonoids belong to polyphenolic secondary metabolites with broad-spectrum pharmacological activities and extensive biological effects, and the most prominent activity is their potential role as anticancer agents. In recent years, flavonoids and their synthetic analogues have been intensely investigated in the treatment of ovarian, breast, cervical, pancreatic, and prostate cancer. To some extent, chemical structures of flavonoids will influence their anticancer activities. Thus, some new analogues based on the structural skeleton of these flavonoids have been synthesized and investigated. Th

An Arabidopsis Protein-Flavonoid Interactome Identifies Peroxiredoxin A as a Candidate for Flavonoid Action in Chloroplasts.

The ability of phytochemicals to act as small molecule effectors of protein function is a largely overlooked dimension of plant biochemistry. This is particularly true for the ubiquitous flavonoids where, despite abundant examples of functional interactions with human proteins, biological activities in plants are primarily attributed to ROS scavenging. We used affinity capture to explore the protein interactome of the flavonoid glycoside, rutin, in Arabidopsis seedlings. Unexpectedly, the 397 high-confidence candidates included numerous proteins associated with chloroplasts, where flavonoids a

Flavonoids as potential anti-asthmatic agents.

Asthma affects approximately 260 million people globally and causes an estimated 450,000 deaths annually, posing a serious threat to human health and life quality. Current mainstay pharmacotherapies are effective for symptom control but are associated with well-documented systemic side effects and do not offer a cure. Flavonoids have garnered increasing scientific interest due to their pleiotropic biological activities; however, there is still a lack of summary on the mechanism of flavonoids in key pathological processes of asthma. This review summarizes the evidence on how flavonoids modulate

Citrus Flavonoids for Cystic Fibrosis Treatment.

Studies conducted since the late 1990s show substantial activity of selected citrus flavonoids as cystic fibrosis transmembrane conductance regulator (CFTR) membrane protein activators. Aimed at reinvigorating fundamental and applied research on citrus flavonoids for the treatment of mucoviscidosis, the thesis of this work is that aptly formulated citrus flavonoids hold significant potential for developing a multitarget treatment of cystic fibrosis combining in a single treatment antimicrobial, anti-inflammatory, CFTR-stimulating, and immunomodulatory properties, as required by a genetic disea
