Hydrophilic Polymers Enable Gastrointestinal Transit Monitoring by Fluorine-19 Magnetic Resonance Imaging.

Gastrointestinal (GI) transit shapes nutrient absorption and oral drug delivery. However, GI anatomy, function, and motility disorders are primarily examined by radiographic imaging, which exposes patients to ionizing radiation and yields only coarse proxies of how a meal moves through the gut. By contrast, fluorine-19 (19F) MRI offers background-free, quantitative tracking, but most oral 19F agents are hydrophobic and phase-separate from chyme, generating layering artifacts, which spoil motility and anatomy readouts. Here, we developed nonresorbable, hydrophilic fluorinated polymer tracers mi

First in vivo fluorine-19 magnetic resonance imaging of the multiple sclerosis drug siponimod.

Theranostic imaging methods could greatly enhance our understanding of the distribution of CNS-acting drugs in individual patients. Fluorine-19 magnetic resonance imaging (19F MRI) offers the opportunity to localize and quantify fluorinated drugs non-invasively, without modifications and without the application of ionizing or other harmful radiation. Here we investigated siponimod, a sphingosine 1-phosphate (S1P) receptor antagonist indicated for secondary progressive multiple sclerosis (SPMS), to determine the feasibility of in vivo 19F MR imaging of a disease modifying drug. Methods: The 19F

Evaluation of fluorine-19 magnetic resonance imaging of the lungs using octafluorocyclobutane in a rat model.

PURPOSE: To test octafluorocyclobutane (OFCB) as an inhalation contrast agent for fluorine-19 MRI of the lung, and to compare the image quality of OFCB scans with perfluoropropane (PFP) scans THEORY AND METHODS: After normalizing for the number of signal averages, a theoretical comparison between the OFCB signal-to-noise ratio (SNR) and PFP SNR predicted the average SNR advantage of 90% using OFCB during gradient echo imaging. The OFCB relaxometry was conducted using single-voxel spectroscopy and spin-echo imaging. A comparison of OFCB and PFP SNRs was performed in vitro and in vivo. Five heal

Flexible and Zwitterionic Fluorinated Hydrogel Scaffold with High Fluorine Content for Noninvasive 19 F Magnetic Resonance Imaging under Ultrahigh Scanning Resolution.

The imaging of hydrogel scaffolds by 19 F magnetic resonance imaging (MRI) represents an attractive tool for straightforward and noninvasive monitoring of their morphology and in vivo fate. However, their further applications are significantly limited by a dilemma of insufficient signal resolution with low 19 F content, and/or hydrophobic aggregation of fluorine moieties-induced signal attenuation with high 19 F content. Herein, a novel label-free fluorinated hydrogel (PFCB) is fabricated with high fluorine content to realize noninvasive monitoring through 19 F MRI under ultrahigh scanning res

In vivo and ex vivo 19-fluorine magnetic resonance imaging and spectroscopy of beta-cells and pancreatic islets using GLUT-2 specific contrast agents.

The assessment of the β-cell mass in experimental models of diabetes and ultimately in patients is a hallmark to understand the relationship between reduced β-cell mass/function and the onset of diabetes. It has been shown before that the GLUT-2 transporter is highly expressed in both β-cells and hepatocytes and that D-mannoheptulose (DMH) has high uptake specificity for the GLUT-2 transporter. As 19-fluorine MRI has emerged as a new alternative method for MRI cell tracking because it provides potential non-invasive localization and quantification of labeled cells, the purpose of this project

In Vivo Quantification of Inflammation in Experimental Autoimmune Encephalomyelitis Rats Using Fluorine-19 Magnetic Resonance Imaging Reveals Immune Cell Recruitment outside the Nervous System.

Progress in identifying new therapies for multiple sclerosis (MS) can be accelerated by using imaging biomarkers of disease progression or abatement in model systems. In this study, we evaluate the ability to noninvasively image and quantitate disease pathology using emerging "hot-spot" 19F MRI methods in an experimental autoimmune encephalomyelitis (EAE) rat, a model of MS. Rats with clinical symptoms of EAE were compared to control rats without EAE, as well as to EAE rats that received daily prophylactic treatments with cyclophosphamide. One-dimensional chemical shift imaging of fluorinated neuroleptics in rat brain in vivo by 19F NMR rotating frame zeugmatography.

One-dimensional chemical shift imaging reflecting local tissue contents of the fluorinated neuroleptics fluphenazine and trifluoperazine in rat brain was obtained noninvasively in vivo using one-dimensional 19-fluorine (19F) nuclear magnetic resonance (NMR) rotating frame zeugmatography. Activities of these fluorinated neuroleptics showed a differential distribution in the brain, maximal in the deep structures presumably within the basal ganglia. An additional resonance of an unidentified fluorinated metabolite was localized to the subcutaneous tissue. The present data indicate the feasibility

Fluorodopa is a Promising Fluorine-19 MRI Probe for Evaluating Striatal Dopaminergic Function in a Rat Model of Parkinson's Disease.

Parkinson's disease (PD) is a progressive neurodegenerative disorder characterized by the loss of dopaminergic neurons in the substantia nigra projecting to the striatum. It has been estimated that approximately 80% of the striatal dopamine and 50% of nigral dopaminergic neurons are lost before the onset of typical motor symptoms, indicating that early diagnosis of PD using noninvasive imaging is feasible. Fluorine-19 (19 F) magnetic resonance imaging (MRI) represents a highly sensitive, easily available, low-background, and cost-effective approach to evaluate dopaminergic function using non-r

Efficient Synthesis of Polymeric Fluorinated Nanoparticles with High Fluorine Content via Aqueous Photo-Polymerization-Induced Self-Assembly for 19F MRI Application.

Polymeric fluorinated nanoparticles (PFNPs) are useful materials in many applications, especially in the field of 19F magnetic resonance imaging (MRI). Despite the development of numerous PFNPs with diverse chemical compositions and structures, those with high fluorine content and capable of highly sensitive 19F MRI remain scarce. Here we report an elegantly designed aqueous photo-polymerization-induced self-assembly (photo-PISA) system for the synthesis of PFNPs with high fluorine content for effective 19F MRI applications. This innovative photo-PISA system is enabled by two analogous fluorin

19F magnetic resonance imaging-informed fate models of PFAS in porous media.

Per- and polyfluoroalkyl substances (PFAS) are persistent contaminants. Predicting their fate in natural or engineered porous media, using accurate models, is essential for effective remediation and contamination management strategies. The mechanisms of transport and retention included in such models, and the associated parameters, are mostly inferred from PFAS concentration vs. time breakthrough curves (BTCs) measured during transport experiments. Still, the interpretation of BTCs may not be unique as they result from a succession of mechanisms taking place inside the porous media. We address

Imaging of Thromboinflammation by Multispectral 19F MRI.

The close interplay between thrombotic and immunologic processes plays an important physiological role in the immune defence after tissue injury and has the aim to reduce damage and to prevent the spread of invading pathogens. However, the uncontrolled or exaggerated activation of these processes can lead to pathological thromboinflammation. Thromboinflammation has been shown to worsen the outcome of cardiovascular, autoinflammatory, or even infectious diseases. Imaging of thromboinflammation is difficult because many clinically relevant imaging techniques can only visualize either inflammator

Visualization and Quantification of Base-Level SO2 in Live Cells without Intracellular Background Interference Using Sensitive 19F-NMR.

Sulfur dioxide (SO2), as an endogenous gasotransmitter, is involved in a variety of physiological processes in living systems, however, distinction and quantification of base-level SO2 in live cells pose great challenges for the current analytical techniques. We report an efficient way to visualize and quantify base-level SO2 in different live mammalian cells by 19F-NMR. This method relies on a high-performance 19F-probe (P1-CF3) that reacts with the reactive sulfur species (RSS) in the generation of distinct chemical shift profiles, which are well-resolved in the NMR spectrum. The quantitativ

Fluorinated charge-reversible phthalocyanine nanoemulsions for glioma-targeted dual-modal imaging and self-oxygenated phototherapy.

Gliomas are highly aggressive brain tumors with poor prognosis, limited by inadequate targeting, tumor hypoxia, and the restrictive blood-brain barrier (BBB). Here, we present a fluorinated phthalocyanine manganese-based nanoemulsion for dual-modal imaging and effective phototherapy of gliomas. The amphiphilic phthalocyanine, engineered through side-chain modification and Mn2+ chelation, exhibits "turn-on" near-infrared fluorescence (NIRF), sensitive fluorine-19 magnetic resonance imaging (19F MRI), and high phototherapy efficacy. Fluorinated 1,7-DO2A-Based Iron(II) Complexes as Sensitive 19F MRI Molecular Probes for Visualizing Renal Dysfunction in Living Mice.

Kidney diseases have become an important global health concern due to their high incidence, inefficient diagnosis, and poor prognosis. Devising direct methods, especially imaging means, to assess renal function is the key for better understanding the mechanisms of various kidney diseases and subsequent development of effective treatment. Herein, we developed a fluorinated ferrous chelate-based sensitive probe, 1,7-DO2A-Fe(II)-F18 (Probe 1), for 19F magnetic resonance imaging (MRI). This highly fluorinated probe (containing 18 chemically equivalent 19F atoms with a fluorine content at 35 wt %)

Parallelized 61-Sample Inhibition Screening Accelerated by 19F-MRI Compressed Sensing.

19F NMR is a well-established technique for screening ligands in early drug discovery. However, conventional NMR measurements are inherently sequential, which limits experimental throughput. We recently demonstrated that 19F MR imaging (MRI) enables spatial parallelization of such measurements. In this study, we leverage the reporter assay format, where the transverse relaxation ( T 2 ) of a fluorinated reporter ligand is monitored in the presence of potential competitors. Using T 2 -weighted MR images, up to 61 miniaturized samples can be resolved simultaneously. The screening assay operate

Impact of fluorine-containing nanoparticle PEGylation on inflammation imaging by 19F MRI.

Fluorine-containing nanoparticles (FNPs) are widely used for inflammation imaging by fluorine-19 magnetic resonance imaging (19F MRI) due to their biocompatibility and suitability to track immune cells via phagocytic uptake. For targeting approaches beyond passive incorporation, surface PEGylation of FNPs is required to reduce cellular uptake, but is known to prolong blood half-life of the particles. This study investigates the efficacy of FNP PEGylation for inflammation imaging in vivo. FNPs and PEGylated FNPs (PEGFNPs) of different size were synthesized and characterized for particle propert

Fluorinated Core-Shell Metal-Organic Framework-Based Dual-Activated 19F MRI Nanoprobes for In Vivo Visualization of the Tumor Microenvironment.

In vivo simultaneous visualization of the dynamic changes in biomarkers within the tumor microenvironment (TME) is crucial for elucidating the mechanisms underlying tumor initiation and progression and providing pivotal insights for tumor management. Herein, we develop an acidosis- and inorganic phosphate-dual-responsive fluorinated core-shell nanoprobe (Zr@Zn-MOF) from corresponding metal ions and fluorinated ligands for visualizing the TME in vivo via multiplexed 19F magnetic resonance imaging (MRI). In normal tissues, the insignificant dual-channel 19F MRI signals of Zr@Zn-MOF could be ascr

A 19F MRI/NIR-FL Nanoprobe for "Turn-On" Imaging of β-Secretase (BACE1) Activity in Alzheimer's Disease.

Alzheimer's disease (AD) is the most common progressive neurodegenerative disorder with very few therapeutic methods; thus, its early detection is urgently important. β-secretase (BACE1) is a well-recognized biomarker of early-stage AD, but its in vivo imaging-based diagnostic method is rarely reported. Herein, we report a BACE1-disassemblable nanoprobe, 19F-Cy5.5-NP for "Turn-On" 19F magnetic resonance/near-infrared fluorescence (19F MR/NIR-FL) imaging of the enzyme activity in AD in vivo. The "Turn-On" 19F MRI signals are employed for imaging BACE1 activity with high specificity, while the "

Intracellular Binding of Novel Fluorinated Compounds to Carbonic Anhydrase Isoforms Explored by In-Cell 19F NMR.

One of the most important challenges of drug development is identifying whether a candidate molecule can efficiently reach and engage intracellular targets. Cell-based assays are usually employed to address this issue; nevertheless, many compounds that appear promising in vitro fail in cellular or in vivo contexts due to low intracellular activity or off-target effects. In this work, we employed ligand-based in-cell 19F NMR to characterize ligand-target interactions in the cellular environment. We investigated the binding of newly synthesized fluorinated sulfonamide derivatives to various cyto
