Dihydrotestosterone, a non-aromatizable androgen has been hypothesized to arrest follicular growth. The concentration of androgens in the prepubertal, adult and pregnant mammals leads one to presume that high levels of estrogens cause follicular growth and ovulation while increase in DHT results in follicular atresia. However, DHT probably has no definitive role to play during lactation and menopause. In polycystic ovarian syndrome, inhibition of normal follicular growth may be due to increased DHT secretion.

Programmed cell death, constitutes a common fundamental incident that occurs during oogenesis in a variety of different animals. It plays a significant role in the maturation process of the female gamete and also in the removal of abnormal and superfluous cells at certain checkpoints of development. In the present study, we demonstrate the existence of follicular atresia during mid-oogenesis in the olive fruit fly Dacus oleae (Tephritidae). The number of atretic follicles increases following the age of the fly, suggesting for the presence of an age-susceptible process. The atretic follicles co

Bovine follicles having a higher concentration of progesterone than estradiol in the follicular fluid can be considered as atretic. Since we observed previously that there was an inverse relationship between the follicular fluid estradiol to progesterone (E/P) ratio and the prorenin level, we have proposed that a high prorenin level may be associated with follicular atresia. The aim of the present study was to corroborate this hypothesis by including additional indices to distinguish unambiguously between atretic and nonatretic follicles and to compare the prorenin levels in these two groups o

MicroRNAs (miRNAs) are short, noncoding RNAs that posttranscriptionally regulate gene expression. In the past decade, studies on miRNAs in ovaries have revealed the key roles of miRNAs in ovarian development and function. In this review, we first introduce the development of follicular atresia research and then summarize genome-wide studies on the ovarian miRNA profiles of different mammalian species. Differentially expressed miRNA profiles during atresia and other biological processes are herein compared. In addition, current knowledge on confirmed functional miRNAs during the follicular atre

Follicular atresia is a critical physiological process that ensures the selection of high-quality oocytes by eliminating non-viable follicles. In women, 99.9% of follicles undergo atresia naturally, the premature or dysregulated atresia follicle can lead to pathological conditions such as polycystic ovary syndrome (PCOS) and premature ovarian failure (POF). Recent studies highlight the roles of apoptosis, autophagy, and ferroptosis in regulating follicular atresia. This review integrates molecular mechanisms of programmed cell death with follicular dynamics, emphasizing how aberrant atresia di

Infertility in hyperprolactinemic females is attributed to the dysregulation of GnRH release, subsequently affecting gonadotropin levels, and ultimately leading to anovulation. However, in addition to the hypothalamus, prolactin receptor (PRLR) is expressed in ovaries as well, suggesting potential local effects of PRL in cases of hyperprolactinemia. We have developed an experimental model of sulpiride (SPD)-induced hyperprolactinemia using a wild rodent, the plains vizcacha, and studied the implications of pharmacological PRL levels on folliculogenesis and steroid production. Ovaries of SPD fe

Most of follicles undergo a degenerative process called follicular atresia. This process directly affects the egg production of laying hens and is regulated by external and internal factors. External factors primarily include nutrition and environmental factors. In follicular atresia, internal factors are predominantly regulated at 3 levels; organic, cellular and molecular levels. At the organic level, the hypothalamic-pituitary-ovary (HPO) axis plays an essential role in controlling follicular development. At the cellular level, gonadotropins and cytokines, as well as estrogens, bind to their

Eighteen normal ovulatory cynomolgus (Macaca fascicularis) monkeys were given an extended fixed-dose regimen of human menopausal gonadotropin (hMG), which was followed by human chorionic gonadotropin (hCG) administration. Ovarian hyperstimulation developed in responsive subjects to various degrees. Based on their serum estradiol (E2) response, 16 were arbitrarily categorized as high responders (E2 greater than 1000 pg/ml), 3 were medium responders (E2 500 to 1000 pg/ml), and 2 were low responders (E2 150 to 500 pg/ml). Interestingly, one female did not respond to exogenous gonadotropin therapy

The B6.Y(TIR) mouse fails to develop normal testes despite transcription of Sry, the primary testis-determining gene on the Y chromosome. Consequently, B6.Y(TIR) fetuses with bilateral ovaries develop into apparently normal but infertile females. This infertility can be mainly attributed to oocyte incompatibility for postfertilization development. In addition, abnormality in preovulatory follicles and rapid loss of oocytes have been observed in XY ovaries. This study examined the effect of gonadotropins on follicular development and atresia in B6.Y(TIR) prepubertal females. The results show th

We have demonstrated that 17 beta-estradiol (E2) administered to monkeys for 24 or 48 h from day 6 of the menstrual cycle results consistently in degeneration of the preovulatory follicle. The present experiments were conducted to determine the strength-duration characteristics of this effect and to evaluate the occurrence of partial atresia together with subsequent effects on luteal function. Square wave increments in circulating amounts of estrogen were achieved by varying the number and size of Silastic capsules containing E2. Treatments included 0.5, 1, 2, 4, or 10 capsules placed sc for 6

Atresia is a process in ovarian follicles that is regulated by hormone-induced apoptosis. During atresia, granulosa cell (GC) apoptosis is a key mechanism orchestrated through diverse signaling pathways. Cocaine- and amphetamine-regulated transcript (CART) signaling within ovarian GCs has been demonstrated to play a key role in the regulation of follicular atresia in cattle, pigs, and sheep. The present work aimed to investigate the potential local regulatory role of CART in GC apoptosis-induced follicular atresia in buffalo, focusing on the modulation of the AKT/GSK3β/β-catenin signaling path

The mammalian ovary is an extremely dynamic organ in which a large majority of follicles are effectively eliminated throughout their reproductive life. Due to the numerous efforts of researchers, mechanisms regulating follicular growth and atresia in mammalian ovaries have been clarified, not only their systemic regulation by hormones (gonadotropins) but also their intraovarian regulation by gonadal steroids, growth factors, cytokines and intracellular proteins. Granulosa cells in particular have been demonstrated to play a major role in deciding the fate of follicles, serving molecules that a

Polycystic ovary syndrome (PCOS) is a common endocrine disorder affecting women of reproductive age, characterized by a spectrum of reproductive, endocrine, and metabolic disturbances. The etiology of PCOS encompasses a complex interplay of genetic, metabolic, inflammatory, and oxidative factors, though the precise pathological mechanisms remain inadequately understood. Despite considerable variability in the clinical characteristics and biochemical profiles among individuals with PCOS, abnormalities in follicular development are a hallmark of the condition. Granulosa cells, integral to follic

Epidemiological investigations and animal studies demonstrate a significantly positive relationship between polycyclic aromatic hydrocarbons (PAHs) exposure and reproductive disorders. However, few researches are focused on the reproductive toxicity of low-molecular-weight PAHs (number of benzene ring ≤ 3) which occupy a large part of PAHs. Phenanthrene (Phe), a typical low-molecular-weight PAH, is one of the most abundant PAHs detected in foods. In the present study, oral treatment with Phe at a human exposure related level during gestation (60 μg/kg body weight every three days, six times in

BACKGROUND: Aflatoxin B1 (AFB1) is one of the most potent naturally occurring mycotoxins and has been implicated in reproductive toxicity. The present study is aimed at evaluating the time-dependent effects of experimental AFB1 exposure on ovarian follicular dynamics, reproductive hormone profiles, and intrinsic apoptosis-related markers in female rats.
METHODOLOGY: Twenty-four mature female Wistar rats were assigned to control and AFB1-exposed groups (N = 6 per time point). AFB1 was administered intraperitoneally at 20 mg/kg body weight for 7, 14, and 21 days. Ovarian tissue and serum sample

STUDY QUESTION: Are key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH involved in follicular atresia in the human adult ovary?
SUMMARY ANSWER: In this study, we used spatial transcriptomics to investigate the progression of follicular atresia, focusing on genes of interest associated with steroidogenesis and key signalling pathways WNT, TGFβ/BMP, NOTCH, and HH.
WHAT IS KNOWN ALREADY: While extensive research has focused on the mechanisms driving follicular growth, much less is known about the process of follicular atresia, despite its relevance for ovarian aging and reproductive longevity

It is well known that during mammalian ovarian follicular development, the majority of follicles undergo atresia at various stages of their development. However, the mechanisms controlling this selection process remain unknown. In this study, we investigated apoptosis in granulosa cells during goat follicular atresia by terminal deoxynucleotidyl transferase-mediated dUTP nick end labeling (TUNEL). The changes in the levels of steroids, insulin-like growth factors (IGFs) and IGF receptors were studied by radioimmunoassay (RIA) and semi-quantitative reverse transcription-PCR. We found that the p

A new model for the investigation of atresia in rhesus monkeys is presented. This model is based upon the reliable induction of an atresia-like process in the dominant preovulatory follicle (DF) by estradiol-17 beta administered subcutaneously via Silastic capsules for 24 h. Data obtained from follicular contents aspirated from treated animals demonstrated alterations in the putative markers of atresia similar to those described in other models of atresia. Although follicle size and appearance and volume of follicular fluid (FF) were unaltered in treated animals, FF was much more viscous than

PROBLEM: Endometriosis is a chronic inflammatory disease that leads to pelvic pain and infertility. Recent studies have indicated that immunological, endocrine, biochemical, and genetic irregularities, along with suboptimal quality of oocytes, embryos, and the endometrial environment, significantly impact infertility associated with endometriosis. Ectopic endometrial cells in endometriosis have the capacity to avoid apoptosis. Therefore, apoptosis has supposed to play a fundamental role in the pathogenesis of endometriosis. This study aimed to investigate the association between the messenger
