Molecular markers used in forensic genetics.

Forensic genetics is a field that has become subject to increasing interest in recent years. Both the technology and the markers used for forensic purposes have changed since the 1980s. The minisatellite sequences used in the famous Pitchfork case introduced genetics to the forensic sciences. Minisatellite sequences have now been replaced by more sensitive microsatellite markers, which have become the basis for the creation of genetic profile databases. Modern molecular methods also exploit single nucleotide polymorphisms, which are often the only way to identify degraded DNA samples. The same

Past, Present and Future Perspectives of Forensic Genetics.

Forensic genetics has experienced remarkable advancements over the past decades, evolving from the analysis of a limited number of DNA segments to comprehensive genome-wide investigations. This progression has significantly improved the ability to establish genetic profiles under diverse conditions and scenarios. Beyond individual identification, forensic genetics now enables the inference of physical traits (e.g., eye, hair, and skin color, as well as body composition), biogeographic ancestry, lifestyle habits such as alcohol and tobacco use, and even the transfer of genital microbiomes post-

DNA Commission of the International Society for Forensic Genetics (ISFG): recommendations regarding the role of forensic genetics for disaster victim identification (DVI).

The ISFG membership consists of scientists and medical professionals specialized in using genetic testing for kinship analysis and the individualization of biological material. This expertise makes the forensic geneticist a resource of advice to international and national organizations dealing with human identifications and causes many DNA laboratories to get involved in DVI tasks. The present recommendations are meant to educate more forensic geneticists about their potential involvement in mass fatality preparedness and possible DVI efforts, as well as to provide practical guidance for each

Cleaning protocols in forensic genetic laboratories.

It is pivotal to avoid cross-sample contamination in forensic genetic laboratories and optimal cleaning protocols for the removal of DNA are essential. A survey was performed, and ten forensic genetic laboratories shared their cleaning protocols in pre-PCR and post-PCR laboratories. The cleaning frequencies on different surface areas were somewhat similar, whereas none of the laboratories used the same cleaning reagents. Therefore, the efficiencies of the cleaning protocol utilised were tested and compared. The results showed that freshly made household bleach and Virkon® removed all amplifiab

Can Hemorrhagic Stroke Genetics Help Forensic Diagnosis in Pediatric Age (<5 Years Old)?

When stroke occurs in pediatric age, it might be mistakenly interpreted as non-accidental head injury (NAHI). In these situations, a multidisciplinary approach is fundamental, including a thorough personal and familial history, along with accurate physical examination and additional investigations. Especially when the clinical picture is uncertain, it is important to remember that certain genetic conditions can cause bleeding inside the brain, which may resemble NAHI. Pediatric strokes occurring around the time of birth can also be an initial sign of undiagnosed genetic disorders. Hence, it is

Preparing for Shotgun Sequencing in Forensic Genetics-A Workflow for Analysis of Monozygotic Twins.

BACKGROUND/OBJECTIVES: Identifying the true perpetrator among monozygotic (MZ) twins has long posed a challenge in forensic genetics and for police investigations because MZ twins are likely identical in their short tandem repeat (STR) profile. In this study, we propose a workflow to address this issue, anchored in a case study involving MZ twins. METHODS: The workflow includes shotgun sequencing of reference samples from one twin and trace samples, followed by a targeted amplicon-based approach (AmpliSeq™ Custom Panel) to validate the identified variants. RESULTS: Biological traces from the

The genetic aetiology of male infertility: clinical and forensic implications.

The escalating prevalence of infertility on a global scale represents a critical public concern, with over 50% cases attributed to male-related factors. Y chromosome microdeletion, within the azoospermia factor region, has emerged as a significant genetic contributor to male infertility, with its occurrence varying from 2% to 24% based on the geographic location and genetic background. The prevalence of such microdeletions in infertile individuals varies broadly, with AZFc being the most frequently reported deletion. The gr/gr partial deletion demonstrates a significant ethnic correlation, hig

Forensic DNA databases: genetic testing as a societal choice.

In this brief report, the authors argue that while a lot of concerns about forensic DNA databases have been raised using arguments from biomedical ethics, these databases are used in a complete different context from other biomedical tools. Because they are used in the struggle against crime, the decision to create or store a genetic profile cannot be left to the individual. Instead, this decision is made by officials of a society. These decisions have to be based on a policy that is the concretisation of some of society's most fundamental ideas about its own nature and function. Individuals w

[The use of clinical molecular and genetic tests in forensic medical opinions].

Modern molecular biology and clinical genetics have the ability to diagnose many diseases, the presence or absence of which is important in various areas of forensic medical opinion. The aim of the study is to present examples of possible non-standard application of clinical molecular-genetic tests in forensic medicine. From among all the opinions prepared so far at the Department of Forensic Medicine in Katowice, three were selected two of which concerned post-autopsy conclusions relating to the cause of death, and in one case - opinion based on the files of a case related to the suspicion of

Unearthing forensic genetics: preliminary results on the ability to generate DNA profiles from buried human biological samples.

Burying objects with biological material is a common procedure in many crimes, intending to hide and/or accelerate the process of degradation of the possible biological information they may contain. In this sense, the central question of this preliminary study concerns investigating the impact of a burial process on the degradation of genetic material, specifically on blood and saliva samples, compared to the same type of samples deposited on the surface. Saliva and blood samples were methodically positioned on a white cotton T-shirt, with half of the samples being buried and the other half pl

The genetics of skin, hair, and eye color variation and its relevance to forensic pigmentation predictive tests.

This review examines the potential application of single nucleotide polymorphism (SNP)-based predictive tests for skin, hair, and eye color to forensic analysis in support of police investigations lacking DNA database matches or eyewitness testimony. Brief descriptions of the biology of melanogenesis and the main genes involved are presented in order to understand the basis of common pigmentation variation in humans. We outline the most recently developed forensically sensitive multiplex tests that can be applied to investigative analyses. The review also describes the biology of the SNPs with

Forensic pathological and genetic landmarks in sudden cardiac death in the young: An update.

An episode of sudden death in a young individual is a dramatic event for family members but also a challenge for cardiologists, pediatricians, forensic pathologists and researchers. In the young population, most of sudden deaths are of cardiac origin, in particular due to hereditary cardiac disorders. The autopsy protocol includes a proper macroscopic heart examination and a comprehensive histological analysis. The identification of pathognomonic histopathologic findings may help to unravel the cause of death, but microscopic features are often non-specific and highly ambiguous. Negative autop

[Genetic markers in forensic medical examination of acute and chronic opioid intoxication].

The objective of the present study was to analyse cases of intoxication with opioid narcotics at different blood morphine levels, from low and very low (below 0.5 mg/l) to regular toxic ones (1-4 mg/I). It was shown that molecular genetic studies at postmortem morphine concentrations of up to 0.5 mg/l as a rule identify mutant alleles (CYP2D6*3*4; CYP2C19*2*3). Mutant alleles CYP2D6* and CYP2C19* are most frequently detected at postmortem blood morphine levels ranging from 1 to 4 mg/I. These findings make it possible to regard death cases at low morphine concentrations as due to genetic suscep

Single Nucleotide Polymorphisms in Distant Kinship Inference and Forensic Genetic Genealogy.

Forensic genetics is moving from locus-based DNA profiling toward genome-wide inference enabled by high-density single-nucleotide polymorphism (SNP) data. While short tandem repeats remain central to routine human identification, SNP-based technologies and massively parallel sequencing have expanded the analysis of distant kinship through detection of identity-by-descent (IBD) segments and shared autosomal DNA. This narrative review synthesizes the biological basis of SNP-based distant kinship inference, the statistical and computational frameworks used to model genomic relatedness, and the op

Benchmarking KinSNP®: A study on genetic relationship prediction for forensic applications.

Accurate kinship inference is a cornerstone of forensic DNA analysis, particularly in cases involving unidentified human remains in the absence of direct reference samples and when antemortem samples lack reliable chain of custody. This study evaluates the performance of KinSNP®, a forensic software tool developed by Othram, for predicting genetic relationships using whole genome single nucleotide polymorphism (SNP) data. Using a well-characterized family cohort of 12 individuals with known relationships extending to the 6th degree, we benchmarked KinSNP® against GEDmatch PRO™ and the Shared c

Forensic DNA Phenotyping: Genes and Genetic Variants for Eye Color Prediction.

In recent decades, the use of genetic polymorphisms related to specific phenotypes, such as eye color, has greatly contributed to the development of the research field called forensic DNA phenotyping (FDP), enabling the investigators of crime cases to reduce the number of suspects, making their work faster and more precise. Eye color is a polygenic phenotype, and many genetic variants have been highlighted, with the major contributor being the HERC2-OCA2 locus, where many single nucleotide variations (SNPs) were identified. Interestingly, the HERC2-OCA2 locus, containing the intronic SNP rs129

Genetic landscape of forensic DNA phenotyping markers among Mediterranean populations.

Forensic DNA Phenotyping can reveal the appearance of an unknown individual by predicting the External Visible Characteristics (EVC) from DNA obtained at the crime scene. Our aim is to characterize the genetic landscape of Human identification markers responsible for EVC among Mediterranean populations compared to other worldwide groups. We conducted an exhaustive search for genes involved in EVC variation. Then, variants located on these genes were extracted from public genotypic data of Mediterranean, American, African and East Asiatic populations. The genetic landscape of these Human identi

Assessing the usefulness of Raman spectroscopy and lipid analysis of decomposed human bones in forensic genetics and molecular taphonomy.

Bones are among the structures most likely to be recovered after death. However, the low quantity of preserved DNA and complex processing from sample to DNA profile make forensic DNA analysis of bones a challenging task. Raman spectroscopy and gas chromatography-mass spectrometry (GC/MS), have the potential to be useful as screening tools for DNA analysis and in decomposition studies. The objective of this research was to assess the usefulness of such molecular investigations. Femur samples collected from 50 decomposing human bodies were subjected to Raman spectroscopy and GC/MS.
