A novel recombinant serotype 4 fowl adenovirus efficiently protects both serotype 4 and 11 fowl adenovirus.

Recently, the diseases caused by the infection of multiple serotypes of fowl adenoviruses (FAdVs) have spread worldwide and resulted in huge economic losses to the poultry industry. However, the commercial multivalent or bivalent vaccines against different serotypes of FAdV are not available. In this study, a novel recombinant FAdV-4, designated FAdV-4-F11, expressing the Fiber of FAdV-11, was generated using the template virus FA4-EGFP through the CRISPR-Cas9 and Cre-LoxP system. The recombinant virus FAdV-4-F11 could efficiently replicate in LMH cells with a high yield of 109.6 TCID50/mL. Mo

Fowl Adenovirus-Based Vaccine Platform.

Nonpathogenic fowl adenoviruses (FAdVs) are amenable for engineering multivalent vaccine platforms due to large stretches of nonessential DNA sequences in their genomes. We describe the generation of FAdV-9-based vaccine platforms by targeted homologous recombination in an infectious clone (pPacFAdV-9 or wild type FAdmid) containing the entire viral genome in a cosmid vector. The viral DNA is subsequently released from the cosmid by restriction enzyme digestion followed by transfection in a chicken hepatoma cell line (CH-SAH). Virus is harvested, propagated, and verified for foreign gene expre

Necrotizing Salpingitis by Fowl Adenovirus in a Backyard Hen.

A 3-yr-old Ameraucana hen was received for postmortem examination following a 1-day history of lethargy and death. Gross lesions observed during necropsy were limited to pulmonary congestion and a small clump of egg yolk material in the oviductal lumen. On histopathology, there was a necrotizing salpingitis of the infundibular and isthmus mucosa with amphophilic, intranuclear inclusion bodies in superficial epithelial cells. Transmission electron microscopy identified the intranuclear inclusions as aggregates of adenovirus virions. Fowl adenovirus (FAdV) type A was identified with PCR and sequ

Detection and Typing of a Fowl Adenovirus Type 1 Agent of Pancreatitis in Guinea Fowl.

Adenoviral pancreatitis has been amply described for decades in guinea fowl. Although its pathologic picture has been characterized fairly well, its etiology still remains only partially clarified. Based on several outbreaks diagnosed on commercial guinea flocks raised in France since 2017, we performed direct whole-genome sequencing from pancreatic lesional tissue by using the Oxford Nanopore Technologies (ONT) sequencing method. We generated 4781 viral reads and assembled a whole genome of 43,509 bp, clustering within fowl adenovirus type 1 (FAdV-1). A phylogenetic analysis based on a partia

Emerging of a novel natural recombinant fowl adenovirus in China.

Outbreaks of hydropericardium syndrome and inclusion body hepatitis caused by fowl adenovirus (FAdV) have occurred in China since June 2015, resulting in significant economic loss to poultry industry. In this study, a novel FAdV, designated as AH720, with recombination among serotype FAdV-8a and FAdV-8b was isolated and characterized in China. Full genome analysis revealed that the AH720 has the genome backbone from FAdV-8b and the fibre gene from FAdV-8a. In an infection study, although AH720 was not lethal to chickens, AH720 did cause characteristic lesions of inclusion body hepatitis in the

Airborne Transmission of a Serotype 4 Fowl Adenovirus in Chickens.

Serotype 4 fowl adenovirus (FAdV-4) is the main pathogen for hydropericardium syndrome (HPS) in chickens. It has caused major economic losses in the global poultry industry. Currently, FAdV-4's transmission routes in chickens remain unclear. Here we investigate the airborne transmission routes of FAdV-4 in chickens. A total of 45 ten-day-old chickens were equally divided into three groups (infected group/isolator A, airborne group/isolator B, and control group/isolator C). Of note, isolators A and B were connected by a leak-free pipe. The results showed that the virus could form a viral aeroso

Cytokine patterns associated with a serotype 8 fowl adenovirus infection.

This study examined cytokine gene expression patterns associated with fowl adenovirus (FAdV) infection. The selected cytokine mRNA was quantified by quantitative real-time reverse transcription-PCR in spleen, liver, and cecal tonsil during the course of infection of chickens with a serotype 8 FAdV (FAdV-8). Compared to uninfected chickens, infected birds had higher mRNA expression of interleukin (IL)-18 and IL-10 in spleen and liver, respectively. Interferon gamma (IFN-γ) mRNA expressed in spleen and liver of infected chickens was significantly upregulated, while the expression of IL-8 mRNA in

Biological features of fowl adenovirus serotype-4.

Fowl adenovirus serotype 4 (FAdV-4) is highly pathogenic to broilers aged 3 to 5 weeks and has caused considerable economic loss in the poultry industry worldwide. FAdV-4 is the causative agent of hydropericardium-hepatitis syndrome (HHS) or hydropericardium syndrome (HPS). The virus targets mainly the liver, and HPS symptoms are observed in infected chickens. This disease was first reported in Pakistan but has now spread worldwide, and over time, various deletions in the FAdV genome and mutations in its major structural proteins have been detected. This review provides detailed information ab

Immunosuppressive potential of fowl adenovirus serotype 4.

Fowl adenovirus serotype 4 (FAdV-4) is the causative agent of hydropericardium syndrome. To clarify the effects of FAdV-4 on immune organs in birds, we conducted a detailed examination of dynamic morphology and damage mechanisms in chickens randomly divided into 4 groups (FAdV-4, vaccination, FAdV-4 plus vaccination, and control). FAdV-4 caused the depletion of lymphocytes and subsequent growth impairment in the thymus and bursa. Chickens infected with FAdV-4 and subjected to vaccination experienced greater inhibition of antibody responses to inactivated vaccines against Newcastle disease and

Rapid generation of fowl adenovirus 9 vectors.

Fowl adenoviruses (FAdV) have the largest genomes of any fully sequenced adenovirus genome, and are widely considered as excellent platforms for vaccine development and gene therapy. As such, there is a strong need for stream-lined protocols/strategies for the generation of recombinant adenovirus genomes. Current genome engineering strategies rely upon plasmid based homologous recombination in Escherichia coli BJ5183. This process is time-consuming, involves multiple cloning steps, and low efficiency recombination. This report describes a novel system for the more rapid generation of recombina

A novel recombinant serotype 4 fowl adenovirus expressing fiber-2 protein of duck adenovirus 3.

Recently, the highly pathogenic serotype 4 fowl adenovirus (FAdV-4) and duck adenovirus 3 (DAdV-3) were outbroken and widespread, causing substantial economic losses to the duck industry. Therefore, there is an urgent need to generate a recombinant genetic engineering vaccine candidate against both FAdV-4 and DAdV-3. In this study, a novel recombinant FAdV-4 expressing the Fiber-2 protein of DAdV-3, designated as rFAdV-4-Fiber-2/DAdV-3, was generated based on CRISPR/Cas9 and Cre-LoxP systems. Indirect immunofluorescence assay (IFA) and western blot (WB) showed that the Fiber-2 protein of DAdV-

Genomic Epidemiology and Evolution of Fowl Adenovirus 1.

Fowl adenovirus 1 (FAdV-1) is the main cause of gizzard erosion in chickens. Whole genome sequencing and sequence analyses of 32 FAdV-1 strains from a global collection provided evidence that multiple recombination events have occurred along the entire genome. In gene-wise phylogenies, only the adenoviral pol gene formed a tree topology that corresponded to whole genome-based phylogeny. Virus genetic features that were clearly connected to gizzard erosion were not identified in our analyses. However, some genome variants tended to be more frequently identified from birds with gizzard erosion a

Immunopathogenesis and physical maps of fowl adenovirus serotype 9.

Restriction endonuclease maps of the genome of fowl adenovirus (FAV) serotype 9 have been constructed for the restriction endonucleases NdeI, NotI and XbaI. The total size of the FAV-9 genome was estimated to be 44.5 kb pairs, consistent with previous reports that FAV genomes are approximately 10 kb larger than human adenovirus (HAV). The pathogenicity of this virus in day-old chickens was intermediate between the pathogenicity of the non-pathogenic and the highly pathogenic FAVs.

LAMP real-time turbidity detection for fowl adenovirus.

BACKGROUND: Fowl adenovirus (FAdV) is an infectious agent that can cause jaundice, severe anaemia, dyspnoea and reproductive disorders in fowls. Since 2015, FAdV disease has been rapidly spreading among broiler chickens in China, where it has caused significant economic losses. In this study, a loop-mediated isothermal amplification (LAMP) real-time turbidity method with strong specificity to FAdV was established. RESULTS: The established assay was specific to FAdV-4, and the lower limit of detection was 75 copies/μL of extracted DNA. The positive detection rate for the suspected tissue sampl

Characterization and functional studies of fowl adenovirus 9 dUTPase.

Deoxyuridine 5'-triphosphate pyrophosphatase (dUTPase), a ubiquitous enzyme that catalyzes the hydrolysis of dUTP to dUMP and found in many viruses, has yet to be identified in fowl adenovirus 9 (FAdV-9). By a multiple alignment of dUTPase amino acid sequences, FAdV-9 ORF1 contained the five conserved motifs that define the protein family, and encoded a functional dUTPase. Moreover, transcription and protein expression patterns were characterized, indicating that dUTPase was transcribed from 2h post-infection (h.p.i.) and translated from 6h.p.i., and both continued to the late phase of virus i

Generation and characterization of a fowl adenovirus 9 dual-site expression vector.

Fowl adenoviruses (FAdVs) are widely considered as excellent platforms for vaccine development and gene therapy. We improved on our right-end partial TR-2 deleted or a left-end 2.3 kb deleted vectors by developing a single, dual-site delivery vector. We demonstrated that, in addition to ORF11, the right end ORF17 is also dispensable. To further improve the capacity and flexibility of the FAdV-9 based vector system, we generated an infectious recombinant FAdV-9 dual-site expression clone lacking 1.9 kb of the left end and replaced with mCherry under the control of a native promoter, and 3.6 kb

Characterization of a natural recombinant fowl adenovirus of serotypes 8a and 8b.

Fowl adenoviruses (FAdV) infections have been reported in numerous countries worldwide. The hepatitis-hydropericardium syndrome (HHS) caused by FAdV-4 infection has inflicted enormous economic losses on the poultry industry in China, but it has been effectively controlled with the advancement of vaccine development. Currently, the main disease affecting poultry industry is chicken inclusion body hepatitis (IBH). FAdV strain XH2109, isolated from the liver of an IBH-affected broiler, was identified as a recombinant strain between FAdV-8a and -8b through whole-genome sequencing and recombination

Gallus gallus coxsackievirus and adenovirus receptor facilitates the binding of fowl adenovirus serotype 1 in chickens.

Coxsackievirus and adenovirus receptor (UXADR) is an integral membrane protein that serves as a receptor for coxsackie B viruses and adenovirus types 2 and 5. Previous studies demonstrated that Fowl adenovirus (FAV) can also utilize Homo sapiens CXADR to infect cells. FAV is a double-stranded DNA virus of the family Adenoviridae. FAV causes inclusion body hepatitis and hydropericardium syndrome in chickens. In addition, FAV serotypes 1 and 8 have recently been shown to cause gizzard erosion in chickens. These chicken diseases and growth insufficiency caused by FAV infection result in great eco
