Polyphasic taxonomic description of Frankia abietis sp. nov., Frankia limosa sp. nov., Frankia palustris sp. nov. and Frankia luti sp. nov. isolated from Alnus spp.

Frankia strain AiPa1T was isolated from root nodules of Alnus incana induced from soil under Picea abies in Finland. Strains AgKG'84/4T, AgB32T and AgB1.8T were isolated from Alnus glutinosa nodules from Germany. AiPa1T, AgB32T and AgKG'84/4T are infective on A. glutinosa and fix nitrogen in symbiotic nodules and pure culture, and strain AgB1.8T is infective on A. glutinosa but does not fix nitrogen and does not have the nif genes. Colonies of the four strains are white, and they form hyphae and sporangia. N2-fixing vesicles are formed by AiPa1T, AgB32T and AgKG'84/4T in NH4+-free medium but n

Isolation of frankia strains from alder actinorhizal root nodules.

A simple procedure, based on the rapid filtration and washing of Frankia vesicle clusters, was devised for the isolation of Frankia strains from alder actinorhizal root nodules. Of 46 Alnus incana subsp. rugosa nodules prepared, 42 yielded isolates. A simple medium containing mineral salts, Casamino Acids, and sodium pyruvate proved to be the most effective for isolation. In general, colonies appeared 6 to 20 days after inoculation. On the basis of hyphal morphology, two distinct types of Frankia strains were characterized. Randomly selected isolates were tested for infectivity, and all formed

Molecular cloning and complementation analysis of nifV gene from Frankia EuIK1 strain.

The nifV gene from the Frankia EuIK1 strain, a symbiont of Elaeagnus umbellata, was cloned and a complementation test using the Klebsiella pneumoniae nifV mutant was performed to verify its function. The nifV ORF consists of 1245 bp, which encodes 414 amino acids. However, the putative promoter and Shine-Dalgarno sequences were not found in the 5' region of the ORF. The Frankia EuIK1 nifV ORF showed about a 70% nucleotide identity and 80% amino acid similarity with that of Frankia sp. FaC1. In the upstream region of the nifV, a putative ORF that showed a 51% nucleotide identity with the afcD g

Frankobactin Metallophores Produced by Nitrogen-Fixing Frankia Actinobacteria Function in Toxic Metal Sequestration.

A series of new metallophores, referred to as frankobactins, were extracted from cultures of the symbiotic and nitrogen-fixing actinobacterium Frankia sp. CH37. Structure elucidation revealed a 2-hydroxyphenyl-substituted oxazoline core and a chain composed of five proteinogenic and nonproteinogenic amino acids, suggesting nonribosomal peptide synthesis as the biosynthetic origin. By whole-genome sequencing, bioinformatic analysis, and comparison with other Frankia strains, the genetic locus responsible for the biosynthesis was detected. Spectrophotometric titration of frankobactin with Fe(III

The actinorhizal root-nodule symbiont Frankia sp. strain CpI1 has two glutamine synthetases.

Frankia sp. strain CpI1 has two glutamine synthetases. Glutamine synthetase I (GSI) is present during growth on ammonium or N(2) and is similar to classical prokaryotic glutamine synthetases. Gel-filtration chromatography gave a molecular weight estimate of about 680,000 for the GSI holoenzyme, and denaturing polyacrylamide gel electrophoresis yielded a subunit molecular weight of about 59,000, indicating that GSI is most likely a dodecamer. GSI is regulated by adenylylation, as shown by the presence of two spots on two-dimensional polyacrylamide gel electrophoresis and by its behavior during

Preliminary evidence on photoreactivation of Frankia spores with visible light after exposure to UV-C radiation.

Spores of four Frankia strains, the nitrogen-fixing actinomycete, were exposed to short wavelength UV-C radiation of 254 nm at 1 lux cm(2) (0.24 mw cm2 of energy) for 10 min. The used strains were HFP020203, UGL020604, UGL020602q and ORS021001. Exposure to UV was followed by reactivation with visible white light at 327.4 lux cm(2) for the same period of time. Spore germination percentage, spore protein content, and cell growth were damaged by this treatment. The lower and higher percentages of reduction in spore germination were 32 and 63% and, for the same strains, the recovery by white light

Influence of iron and chelator on siderophore production in Frankia strains nodulating Hippophae salicifolia D. Don.

Effect of iron and chelator on the growth and siderophore production in the ten newly Frankia strains isolated from the root nodules of Hippophae salicifolia D. Don and the two reference strains were studied. Growth of the strains was greatly affected when grown in the iron and EDTA deprived conditions. All the strains were capable of producing both the hydroxamate and catecholate type siderophore that was detected using the Csaky and Arnow assays. Production of siderophore was enhanced in the EDTA replenish condition in contrast to the iron supplemented medium suggesting that EDTA reduces the

How Does Salinity Shape Bacterial and Fungal Microbiomes of Alnus glutinosa Roots?

Black alder (Alnus glutinosa Gaertn.) belongs to dual mycorrhizal trees, forming ectomycorrhizal (EM) and arbuscular (AM) root structures, as well as represents actinorrhizal plants that associate with nitrogen-fixing actinomycete Frankia sp. We hypothesized that the unique ternary structure of symbionts can influence community structure of other plant-associated microorganisms (bacterial and fungal endophytes), particularly under seasonally changing salinity in A. glutinosa roots. In our study we analyzed black alder root bacterial and fungal microbiome present at two forest test sites (salin

Is salt stress tolerance in Casuarina glauca Sieb. ex Spreng. associated with its nitrogen-fixing root-nodule symbiosis? An analysis at the photosynthetic level.

Casuarina glauca is an actinorhizal tree which establishes root-nodule symbiosis with N2-fixing Frankia bacteria. This plant is commonly found in saline zones and is widely used to remediate marginal soils and prevent desertification. The nature of its ability to survive in extreme environments and the extent of Frankia contribution to stress tolerance remain unknown. Thus, we evaluated the ability of C. glauca to cope with salt stress and the influence of the symbiosis on this trait. To this end, we analysed the impact of salt on plant growth, mineral contents, water relations, photosynthetic

High-Resolution X-Ray Structures of Two Functionally Distinct Members of the Cyclic Amide Hydrolase Family of Toblerone Fold Enzymes.

The Toblerone fold was discovered recently when the first structure of the cyclic amide hydrolase, AtzD (a cyanuric acid hydrolase), was elucidated. We surveyed the cyclic amide hydrolase family, finding a strong correlation between phylogenetic distribution and specificity for either cyanuric acid or barbituric acid. One of six classes (IV) could not be tested due to a lack of expression of the proteins from it, and another class (V) had neither cyanuric acid nor barbituric acid hydrolase activity. High-resolution X-ray structures were obtained for a class VI barbituric acid hydrolase (1.7 Å)

High-throughput screening of microbial adaptation to environmental stress.

We developed a microwell plate, high-throughput, screening method aimed at quantitating the tolerance of a panel of Gram-positive and Gram-negative bacteria to metals (Frankia sp., Escherichia coli, Cupriavidus metallidurans, Rhizobium leguminosarum, and Streptomyces scabies). Microbial viability was quantified using MTS; a tetrazolium salt converted to a water-soluble formazan through microbial reduction. In this paper, we present the stepwise development of the method, highlighting the main elements underlying its reliability, and compare results obtained with literature. We conclude the met

Conserved biosynthetic pathways for phosalacine, bialaphos and newly discovered phosphonic acid natural products.

Natural products containing phosphonic or phosphinic acid functionalities often display potent biological activities with applications in medicine and agriculture. The herbicide phosphinothricin-tripeptide (PTT) was the first phosphinate natural product discovered, yet despite numerous studies, questions remain surrounding key transformations required for its biosynthesis. In particular, the enzymology required to convert phosphonoformate to carboxyphosphonoenolpyruvate and the mechanisms underlying phosphorus methylation remain poorly understood. In addition, the model for non-ribosomal pepti

Diversity of cultivated and uncultivated actinobacterial endophytes in the stems and roots of rice.

A dual approach consisting of cultivation and molecular retrieval of actinobacterial 16S rRNA genes was used to characterize the diversity of actinobacterial community inhabiting interior of rice stems and roots. Streptomyces is the most frequently isolated genus from rice stems and roots. Forty-five clones chosen randomly among 250 clones in the 16S rRNA gene clone library from roots were affiliated with nine genera of actinobacteria and uncultured actinobacteria (Mycobacterium, Streptomyces, Micromonospora, Actinoplanes, Frankia, Dactylosporangium, Amycolatopsis, Corynebacterium, Rhodococcus

Brazilian Cerrado soil Actinobacteria ecology.

A total of 2152 Actinobacteria strains were isolated from native Cerrado (Brazilian Savannah) soils located in Passos, Luminárias, and Arcos municipalities (Minas Gerais State, Brazil). The soils were characterised for chemical and microbiological analysis. The microbial analysis led to the identification of nine genera (Streptomyces, Arthrobacter, Rhodococcus, Amycolatopsis, Microbacterium, Frankia, Leifsonia, Nakamurella, and Kitasatospora) and 92 distinct species in both seasons studied (rainy and dry). The rainy season produced a high microbial population of all the aforementioned genera.

Dithionite promoted microbial dechlorination of hexachlorobenzene while goethite further accelerated abiotic degradation by sulfidation in paddy soil.

It is of great scientific and practical importance to explore the mechanisms of accelerated degradation of Hexachlorobenzene (HCB) in soil. Both iron oxide and dithionite may promote the reductive dechlorination of HCB, but their effects on the microbial community and the biotic and abiotic mechanisms behind it remain unclear. This study investigated the effects of goethite, dithionite, and their interaction on microbial community composition and structure, and their potential contribution to HCB dechlorination in a paddy soil to reveal the underlying mechanism. The results showed that goethit

Zearalenone exposure mediated hepatotoxicity via mitochondrial apoptotic and autophagy pathways: Associated with gut microbiome and metabolites.

Zearalenone (ZEN), a mycotoxin is frequently detected in different food products and has been widely studied for its toxicity. However, the underlying mechanisms of hepatotoxic effects, relationship between gut microbiome and liver metabolite mediated hepatotoxicity mechanisms induced by ZEN are still not clear. Here, we reported that the different microscopic changes like swelling of hepatocyte, disorganization of hepatocytes and extensive vacuolar degeneration were observed, and the mitochondrial functions decreased in exposed mice. Results exhibited up-regulation in expression of signals of

Combined effects of super absorbent polymers and microbial fertilizers Co-application on apple production in the Loess Plateau.

Super absorbent polymers (SAP) and microbial fertilizers (MF) are pivotal in ameliorating soil physicochemical properties, regulating plant growth, and enhancing fruit quality. However, the combined effects of their co-application on apple plants remain unclear. To address this, a two-year field experiment was conducted on the Loess Plateau to evaluate different co-application ratios of SAP and MF. Results showed that the combined application improved soil properties, with higher contents of available nitrogen and organic matter, better moisture retention and lower soluble salt content. These

Comparative genomics clarifies phylogenetic relationships and genome evolution in Hippophae from the Qinghai-Tibet plateau.

The uplift of the Qinghai-Tibet Plateau (QTP) and associated climatic oscillations have shaped plant evolution, yet how adaptive strategies diversify along elevational, moisture, and latitudinal gradients within a single clade remains poorly understood. Hippophae (Elaeagnaceae) is distributed along these gradients, with all members sharing Frankia-mediated nitrogen-fixing symbiosis and dioecy. Here, we assembled chromosome-level genomes of H. neurocarpa, H. rhamnoides subsp. yunnanensis, and H. rhamnoides subsp. turkestanica, integrated them with four published assemblies, covering seven taxa
