Free radical scavengers in anaesthesiology and critical care.

Free radicals are highly reactive and unstable compounds. These highly reactive molecules cause oxidative damage to cellular components such as DNA, proteins and lipids. They play central role in the mechanism of cell injury and cell death. Free radical scavengers either prevent these reactive species from being formed, or remove them before they can damage vital components of the cell. Oxidative stress defines an imbalance in production of oxidizing chemical species and their effective removal by protective antioxidants and scavenger enzymes. Evidence of massive oxidative stress is well estab

Reduced free radical scavengers and chronic renal failure.

Endogenous oxygen radical scavengers such as glutathione peroxidase (GSH-Px), catalase (CAT), glutathione and vitamin E are powerful regulatory systems against free radical toxicity. These oxidative injuries are increased in patients with chronic renal failure leading to various abnormalities including anemia. In this study, activities of GSH-Px, CAT, glutathione and vitamin E were measured in the erythrocytes of 54 chronic renal failure patients compared with 32 healthy controls. GSH-Px activities were lower significantly from controls (20.5 +/- 6.79 vs 28.3 +/- 9.0 u/gHb, p < 0.001). Erythro

[Possible clinical application of SOD and free radical scavengers].

Ischemia-reperfusion injury in various organs has been discussed in connection with reactive oxygen species (ROS). Xanthine oxidase (XOD) has been believed to be the source of O2- to produce this injury of dogs and rats but XOD is not detected in hearts of men, pigs or rabbits. This suggests the importance of O2- produced by leukocyte NAD(P)H oxidase. We demonstrated in 1976 that 3 injections of Cu, Zn-SOD (superoxide dismutase) (i.v.) suppressed rat carrageenan paw edema. McCord succeeded with a single injection in the same model using polyethyleneglycol (PEG-SOD) of long retention time in th

Free radical scavengers prevent intestinal ischemia-reperfusion-mediated cardiac dysfunction.

Our previous studies show that intestinal ischemia impairs cardiac function. This present study examined the contribution of oxygen-derived free radicals to cardiac dysfunction after intestinal ischemia-reperfusion in a rat model of superior mesenteric artery (SMA) occlusion (atraumatic clip for 20 min) and ligation of collateral arcades from the right colic and jejunal arteries. Controls were sham operated (Group 1, n = 10); in Group 2, 20 rats with SMA occlusion were sacrificed 2-5 hr after reperfusion without treatment. Superoxide dismutase (SOD) and catalase, scavengers of oxygen-derived f

Oxygen-derived free-radical scavengers prolong survival in colonic cancer.

The influence of scavengers of oxygen-derived free radicals on survival in colonic cancer was studied. Following curative surgery for carcinoma of the sigmoid colon at Dukes' stage C, 198 patients making an uneventful recovery from surgery were randomized to the control group or to receive allopurinol (50 mg orally 4 times a day) or dimethyl sulphoxide (DMSO, 500 mg orally 4 times a day). In 144 fully evaluable patients who were studied for 5 years, allopurinol and DMSO incurred a significant (p less than 0.01) survival advantage over the whole period of study. The similarity in efficacy betwe

Biphasic effects of free radical scavengers against bleomycin-induced pulmonary fibrosis.

Oxidative stress plays a critical role in the development of pulmonary fibrosis. However, the effects of treatment with anti-oxidant agents against pulmonary fibrosis have not yet been thoroughly investigated. In this study, the effect of MCI-186, a novel free radical scavenger, on bleomycin-induced pulmonary fibrosis was investigated. Bleomycin (0.05units/mouse) was administered intratracheally into C57Bl/6 mice. MCI-186 was given to bleomycin-treated mice intraperitoneally from (i) day -3 to day 7, or from (ii) day 10 to day 28 after bleomycin administration in successive days. At 28 days af

Melatonin as a free radical scavenger in the ovarian follicle.

This review summarizes new findings related to beneficial effects of melatonin (N-acetyl-5-methoxytryptamine) on reproductive physiology. Recently many researchers have begun to study the local role of melatonin as an antioxidant. We focused on intra-follicular role of melatonin in the ovary. Melatonin, secreted by the pineal gland, is taken up into the follicular fluid from the blood. Reactive oxygen species (ROS) are produced within the follicles, during the ovulatory process. Melatonin reduces oxidative stress as an antioxidant, and contribute to oocyte maturation, embryo development and lu

Radioprotective effect of free radical scavenging enzymes.

Ionizing radiation is an important treatment modality in the management of head and neck squamous cell carcinoma. The superoxide (O2-) and hydroxyl (OH.) radicals produced from oxygen and the radio-hydrolysis of water are responsible for most of the DNA and lipid membrane injury caused by radiotherapy. Superoxide dismutase (SOD) and catalase (CAT) are intracellular enzymes that scavenge the superoxide and hydroxyl radicals respectively. The effect of intravenous SOD and CAT on acute and delayed radiation injury was investigated in a rat model. Catalase was shown to reduce the severity of radia

Free radicals and scavenging enzymes in chronic tonsillitis.

OBJECTIVE: This study aimed to define the relationship between chronic tonsillitis and levels of malondialdehyde and superoxide dismutase in free radical and antioxidant forms. It is suggested that free oxygen radicals may play a role in chronic tonsillitis. Materials and methods One hundred twenty-four patients were enrolled in the study. Tonsillectomy was performed via the usual dissection-snare method. Venous blood was taken preoperatively and at 2 weeks postoperatively. Blood samples and tonsil specimens were evaluated for malondialdehyde and superoxide dismutase analysis. RESULTS: The le

Application of oxygen free radical scavengers to diminish the occurrence of myringosclerosis.

The present study was designed to establish whether or not an increased production of oxygen-derived free radicals is involved in the causation of myringosclerosis. Sclerotic lesions in the tympanic membrane were experimentally elicited by keeping rats with perforated tympanic membranes in an atmosphere containing roughly 40% oxygen. The animals were treated daily with a solution containing either copper zinc-superoxide dismutase plus catalase, deferoxamine, or copper sulfate plus iron chloride, applied to the traumatized area. After 1 week the extension of myringosclerotic plaques was determi

Protection against stress-induced acute gastric mucosal injury by free radical scavengers.

This study investigated whether the free radical scavengers allopurinol (50 mg p.o. q.i.d.) and dimethyl sulphoxide (DMSO, 500 mg p.o. q.i.d.) influence the incidence of stress-induced acute gastric mucosal injury in patients with pelvic fractures and hypovolaemic shock. In 177 fully evaluable patients (control n = 58, allopurinol n = 62, DMSO n = 57), endoscopically proven stress-induced injury evolved in a significantly (p less than 0.01) larger number of controls relative to either group on active therapy. During the first 3 days after hospitalization, 13 controls (22%) developed the injury

[Antioxidant and/or free radical scavenger vitamins in tropical medicine].

Antioxidant and/or free radical scavenger vitamins (A, E) as beta carotene are unequally distributed among intertropical peoples from Africa. In Ivory coast for example the values observed are clearly enhanced in the regions where Palm oil is usually eaten than in savanna regions. Primary liver cancer (PLC) is more frequently observed in savanna regions. Furthermore it has been recently suggested that retinoic acid which is derived from vitamin A and beta carotene could interact with the genes which are involved in the primary liver carcinogenesis. In PLC patients as in subjects suffering from

Hepatoprotective and free radical scavenging effects of Nelumbo nucifera.

Ethanol extracts from Nelumbo nucifera (ENN) seeds were studied for possible antioxidative and hepatoprotective effects. Antioxidative effects were measured spectrophotometrically by reduction of 2,2'-Diphenyl-1-picrylhydrazyl (DPPH) radicals. Hepatoprotective effects were tested using carbon tetrachloride (CCl4) and aflatoxin B1 (AFB1)-induced hepatocyte toxicity models. ENN showed potent free radical scavenging effects with a median inhibition concentration of 6.49 microg/ml. Treatment of hepatocytes with ENN inhibited both the production of serum enzymes and cytotoxicity by CCl4. The genoto
