Ultrastructure of mycobacterial surfaces by freeze-substitution.

The global status of tuberculosis has recently received much public attention, particularly in some developed countries that are now reporting an increase in cases after several years of decline. A number of factors have contributed to the resurgence in tuberculosis and other mycobacterial infections, including homelessness, increased urban overcrowding among the poor, increased drug abuse and the AIDS epidemic. In addition, the intrinsic nature of mycobacterial impermeability to some antibiotics, and their ability to survive in host environments has been attributed to the unique chemistry and

A STUDY OF EXTRACELLULAR SPACE IN CENTRAL NERVOUS TISSUE BY FREEZE-SUBSTITUTION.

It was attempted to preserve the water distribution in central nervous tissue by rapid freezing followed by substitution fixation at low temperature. The vermis of the cerebellum of white mice was frozen by bringing it into contact with a polished silver mirror maintained at a temperature of about -207 degrees C. The tissue was subjected to substitution fixation in acetone containing 2 per cent OsO(4) at -85 degrees C for 2 days, and then prepared for electron microscopy by embedding in Maraglas, sectioning, and staining with lead citrate or uranyl acetate and lead. Cerebellum frozen within 30

The ultrastructure of the connective tissue matrix of skin and cartilage after high-pressure freezing and freeze-substitution.

Studies designed to investigate the ultrastructure of the connective tissue matrix have historically relied on chemical fixatives to stabilize tissue microarchitecture. However, conventional fixatives are not completely effective in retaining many matrix constituents, including proteoglycans. Fixative recipes have been modified to include agents that retain proteoglycans, but they precipitate the glycosaminoglycan moiety into electron-dense granules and therefore do not preserve native microstructure. To avoid the structural artifacts introduced by aqueous fixatives, we prepared cartilage and

Endochondral ossification: Insights into the cartilage mineralization processes achieved by an anhydrous freeze substitution protocol.

Growth plate cartilage (GP) serves as a dynamic site of active mineralization and offers a unique opportunity to investigate the cell-regulated matrix mineralization process. Transmission electron microscopy (TEM) provides a means for the direct observation of these mechanisms, offering the necessary resolution and chemical analysis capabilities. However, as mineral crystallinity is prone to artifacts using aqueous fixation protocols, sample preparation techniques are critical to preserve the mineralized tissue in its native form. We optimized cryofixation by high-pressure freezing followed by

Sample preparation of some plant tissues for transmission electron microscopy using high pressure freezing and freeze substitution.

For sample preparation of biological block sectioning, high pressure freezing and freeze substitution (HPF-FS) is a better choice than chemical fixation. Freezing fixation can retain the original structures and preserve the soluble materials, and avoid the disadvantage of chemical fixation. Using high-pressure freezing, plant tissues are more difficult to freeze successfully than animal tissues and microbes because they have cell walls and varying degrees of vacuolation. For plant tissues, including the roots of Arabidopsis thaliana, 1-hexadecene is an ideal cryoprotectant/filler. The ability

Electron microscopic analysis of the mouse and human alveolar epithelial glycocalyx using high-pressure freezing and freeze substitution.

Alveolar epithelial cells (AECs) are covered apically by the alveolar lining layer (ALL). The alveolar epithelial glycocalyx (AEGCX), which is part of the ALL, consists of a mixture of glycoproteins, proteoglycans, and glycosaminoglycans. In comparison with the endothelial glycocalyx (GCX), much less is known about the composition and structure of the AEGCX, although it is presumed to be involved in many cellular processes. The challenges of in vivo visualization of the AEGCX, coupled with the distorting effects of aldehyde fixation, make it difficult to accurately determine its structural org

Immunocytochemical localization of amylase in gerbil salivary gland acinar cells processed by rapid freezing and freeze-substitution fixation.

We examined the immunocytochemical localization of amylase in cryofixed serous acinar cells of gerbil major salivary glands by indirect immunostaining, using anti-gerbil parotid amylase antibody and protein A-gold complex. Fresh tissue blocks were quickly frozen by the metal-contact method, using liquid helium, and were freeze-substituted with either osmium-acetone solution or glutaraldehyde-containing acetone. They were then embedded in an epoxy resin mixture which was polymerized at 60 degrees C. Some tissue blocks substituted with aldehyde-acetone solution were embedded in Lowicryl K4M, pol

Optimisation of freeze substitution protocols for the examination of malaria parasite structure by volumetric electron microscopy.

Malaria is one of the deadliest infectious diseases in the world, annually responsible for over 400,000 deaths. It is caused by parasites of the genus Plasmodium, which undergo remarkable structural changes during their development within different cells across various hosts. An important approach to understand the structural basis of biochemical and physiological processes during Plasmodium infection has been the quantitative measurement of dimensional parameters obtained by different microscopy techniques. In this regard, sample preparation, particularly electron microscopy protocols that re

Closer to the native state: Sandwich freezing and freeze-substitution of microorganisms, cultured cells, and animal and plant tissues for electron microscopy.

Since the invention of the electron microscope, morphologists have developed numerous techniques to visualise the native ultrastructure of biological cells and tissues. Although conventional chemical fixation followed by room temperature dehydration remains widely used, it often fails to preserve native structures, leading to distortion and leakage of cellular components. In contrast, cryofixation combined with freeze substitution offers a superior alternative, producing micrographs that more accurately reflect native ultrastructure. We have developed a 'sandwich freezing method' for rapid cry

Review article fine structure of the exocrine cells of rat sublingual gland revealed by rapid freezing and freeze substitution method.

The ultrastructure of mucous cells of rat sublingual gland processed by rapid freezing, followed by freeze substitution, was compared with that obtained by the standard chemical fixation technique. The rapid freezing method gave a very good preservation of membrane structure with round and discrete mucous droplets (granules) not showing any sign of coalescence. The cisterns of the Golgi apparatus and the trans Golgi network also were well preserved. Upon secretory stimulation by pilocarpine, mucous droplets were discharged by the usual mechanism of exocytosis. From all these findings it emerge

Dynamic ultrastructure of mouse pulmonary alveoli revealed by an in vivo cryotechnique in combination with freeze-substitution.

A morphological approach to cell dynamics is usually difficult, since routine preparative techniques for electron microscopy always induce artifacts due to cessation of the blood supply into organs. An in vivo cryotechnique followed by the freeze-substitution method probably reduces such problems. It was applied for examining the pulmonary alveoli of BALB/c mice in vivo. The following ultrastructural features were revealed. (1) A surfactant layer provided a continuous covering to the alveolar epithelium. (2) Pleural epithelial cells, alveolar cells and endothelial cells contained many small ve

Ultrastructural study of glomerular capillary loops at different perfusion pressures as revealed by quick-freezing, freeze-substitution and conventional fixation methods.

Wistar rat kidneys were perfused with some fixatives at different pressures of 100, 150, 200 or 250 cm H2O via the aorta and were studied by the quick-freezing and freeze-substitution (QF-FS) or deep-etching (QF-DE) methods, in addition to the conventional immersion or perfusion fixation method. Different parts of glomerular capillary walls were selected for morphometric analyses. It was demonstrated that the widths of glomerular slit diaphragms and the spaces between foot processes were more widely dilated at higher perfusion pressures (200 and 250 cm H2O) than those seen at both normal perfu

Structural components in the synaptic cleft captured by freeze-substitution and deep etching of directly frozen cerebellar cortex.

Structural components in the synaptic cleft were examined in cerebellar excitatory synapses by conventional electron microscopy and by rapid freezing followed by freeze-substitution or deep etching. Two transverse components and one parallel element were identified in the clefts of rapidly frozen and freeze-substituted synapses: (i) bridging fibrils, 4-6 nm in diameter, that span the cleft; (ii) columnar pegs, 4-6 nm wide and 8-15 nm high, projecting from the postsynaptic surface; and (iii) intervening fine fibrils running parallel to the apposed synaptic membranes. These were more clearly vis

The fine structure of sublingual gland acinar cells of the Mongolian gerbil, Meriones unguiculatus, processed by rapid freezing followed by freeze-substitution fixation.

We compare the ultrastructure of the gerbil sublingual gland as seen after cryofixation followed by substitution with osmium tetroxide, with the more familiar appearance of material processed by glutaraldehyde-osmium chemical fixation. After primary cryofixation of fresh salivary glands, the nuclei of the mucous cells are found to be spherical in shape and, rather than being displaced toward the cell base, occupy a nearly central position in the cytoplasm, even in the storage phase of the secretory cycle. The mucous secretory granules are seen as membrane-limited inclusions, only rarely partia

Demonstration of intra- and extracellular localization of bullous pemphigoid antigen using cryofixation and freeze substitution for postembedding immunoelectron microscopy.

Using low-temperature postembedding techniques for immunoelectron microscopy, we succeeded in demonstrating the precise localization of bullous pemphigoid antigen (BP-Ag) in normal human skin. Small pieces (less than 1 mm3) of normal adult skin were rapidly frozen in liquid propane at -190 degrees C and subjected to freeze substitution with 100% methanol at -80 degrees C. Specimens were embedded in Lowicryl K11M at -60 degrees C which was polymerized under ultraviolet radiation at -60 degrees C. Ultrathin sections were incubated with BP sera followed by rabbit anti-human IgG and colloidal-gold

Freeze-Thaw-Induced Hybrid Porous PVA/PEG Hydrogels with Dynamic Load-Dissipation Capability for Cartilage Substitutes.

Osteoarthritis is the most prevalent age-related joint disease, yet the limited regenerative capacity of articular cartilage severely constrains spontaneous repair. Here, we present a freeze-thaw polyvinyl alcohol (PVA)/polyethylene glycol (PEG) hydrogel platform featuring a hybrid open-closed macroporous architecture that enables cartilage-mimetic load dissipation for artificial cartilage applications. The hybrid porous structure provides synergistic advantages, where closed pores enhance load-bearing stiffness while open pores facilitate energy dissipation. By systematically tuning polymer c

Evaluation of freezing point depression osmolality for classifying random urine specimens defined as substituted under HHS/DOT criteria.

This study evaluates the analytical performance characteristics of freezing point depression osmolality in urine and osmolality as a suitable analytical indicator for determining the concentration of urine specimens submitted for workplace drug testing. Specifically, this study attempted to determine the utility of urine osmolality to serve as an indicator of substitution as defined by HHS/SAMHSA criteria. Urine osmolality was validated by determining the accuracy, precision, analytical sensitivity, reportable range, and reference interval for the method. Osmolality was measured in workplace u
