Enhanced biodegradation of sticky deposits through cutinase-lipase synergy in designer cellulosomes.

Sticky deposits hinder the recycling of waste paper by causing equipment clogging and poor paper quality. Enzymatic degradation is constrained by poor enzyme stability and limited degradation capacity. Herein, Humicola insolens cutinase and Archeoglobus fulgidus carboxylesterase, were selected to construct a cellulosome-based complex (namely HACOM) to enhance the binding and degradation of sticky deposits and the stability of enzymes. Computational analysis and protein adsorption experiments revealed that the carbohydrate-binding module could interact with the substrate poly (vinyl acetate) (P

Mechanisms and optimization Strategies for surfactant-mediated enhancement of enzymatic hydrolysis of Poly(ethylene terephthalate).

Polyethylene terephthalate (PET) pollution has become a worldwide environmental challenge, and enzymatic hydrolysis represents a green and sustainable strategy to address this contamination. To enhance enzymatic PET hydrolysis, this study systematically compared the effects of five common surfactants-sodium dodecyl sulfate (SDS), cetyltrimethylammonium bromide (CTAB), di-rhamnolipid (DRL), lauryl betaine (LAB), and polyethylene glycol (PEG300)-on Humicola insolens cutinase (HiC)-catalyzed PET hydrolysis with two different crystallinities. Integrated experimental and computational approaches el

Rational engineering of cutinase for enhanced biodegradation of polyvinyl acetate and polyacrylates to control stickies in pulp and paper processing.

In the pulp and paper industry, residual sticky substances accumulate during white water circulation, forming deposits that cause paper defects and increase costs. Cutinase is a promising biocatalyst for mitigating stickies, but its industrial application is limited by inherent catalytic constraints. To address this, a strategy integrating structural alignment, virtual screening, and computer-aided design was employed to remodel the substrate-binding pocket of cutinase from Humicola insolens (HiC). Through iterative recombination, variants C11 and E12 were developed, exhibiting enhanced degrad

Identification of rare Candida species and other yeasts by polymerase chain reaction and slot blot hybridization.

Invasive candidiasis has become a major cause of morbidity and mortality in immunocompromised hosts. Here we describe a fast and reliable DNA extraction and PCR amplification method in combination with a slot blot hybridization assay. A genus-specific probe was designed that allowed to detect DNA from a broad range of Candida species and 3 other yeasts. In addition, species-specific oligonucleotides for emerging Candida and other yeast species allowed to identify DNA extracted from Candida lusitaniae, Candida humicola, Candida kefyr, Candida inconspicua, Candida solani, Malassezia furfur and T

Food Habit Associated Mycobiota Composition and Their Impact on Human Health.

Mycobiota is not only associated with healthy homeostasis in the human gut but also helps to adapt to the environment. Food habits, alcohol consumption, intake of probiotics, and contaminated food with a mycotoxin, often lead to the alteration in the mycobiota composition. Impaired immunity of the host may affect fungal symbiosis leading to mycosis. The human gut adapts to the commensalism fungi belonging to the phylum Ascomycota and Basidiomycota. Diet habits such as plant-or animal-based, phytoestrogens enriched plant products, fat-rich diets also influence the colonization of certain fungal

Diversity and taxonomy of endophytic Chaetomiaceae (Sordariales) associated with roots of wild Orchidaceae in Brazil.

Fungi in the family Chaetomiaceae are ubiquitous, colonizing diverse substrates and frequently occurring as endophytes in various plant species. Despite their prevalence, knowledge of their association with the Orchidaceae remains limited. In this study, we investigated the diversity of endophytic fungi associated with the roots of wild orchids. Based on comprehensive morphological and multigene phylogenetic analysis (ITS, LSU, tub2, and rpb2), 51 isolates were identified as belonging to six Chaetomiaceae genera: Arcopilus, Chaetomium, Collariella, Dichotomopilus, Humicola, and Pseudohumicola.

[Soil microorganisms and physicochemical properties in marshlands along the Yangtze River basin at different types of land use and their effects on distribution of Oncomelania hupensis snails].

OBJECTIVE: To investigating the microbial communities and physicochemical properties of soil and distribution of Oncomelania hupensis snails in marshlands along the Yangtze River basin at different types of land use, and to examine the effects of soil microorganisms and physicochemical properties on snail distribution, so as to provide insights into snail control and schistosomiasis prevention in marshland along the Yangtze River basin. METHODS: Marshlands with four types of land use were selected along the Yangtze River basin on April 2021, including poplar forest-crops integrated planting,

Chemopreventive Potential of Caryophyllane Sesquiterpenes: An Overview of Preliminary Evidence.

Chemoprevention is referred to as a strategy to inhibit, suppress, or reverse tumor development and progression in healthy people along with high-risk subjects and oncologic patients through using pharmacological or natural substances. Numerous phytochemicals have been widely described in the literature to possess chemopreventive properties, although their clinical usefulness remains to be defined. Among them, caryophyllane sesquiterpenes are natural compounds widely occurring in nature kingdoms, especially in plants, fungi, and marine environments. Several structures, characterized by a commo
