A model plant, Nicotiana benthamiana, was examined as a host for persistent fungal viruses capable of crossing organismal kingdoms. Protoplasts of N. benthamiana were transfected with a mixture of virions of a betapartitivirus, Rosellinia necatrix partitivirus 18 (RnPV18), and an alphapartitivirus, RnPV19, and were then subjected to plantlet regeneration. Primary RT-PCR-based screening showed that nearly 100% of the resulting calli tested positive for RnPV18, whereas approximately 90% were positive for RnPV19. However, secondary screening performed at a later stage of tissue culture revealed t

This study aimed to characterize the gut virome in children with allergic rhinitis (AR) and explore its interactions with immune markers and allergens. Metagenomic sequencing was performed on fecal samples from 16 AR and 17 healthy control (HC) children. Viral genes (VGs) were identified and taxonomically annotated using BLASTP against the NCBI NR database. Virome diversity, differential abundance, and correlations with IgE were analyzed using LEfSe, random forest, and Spearman correlation. While alpha diversity did not differ, beta diversity revealed subtle compositional trends. Taranisvirus

BACKGROUND: Patients with chronic obstructive pulmonary disease (COPD) develop acute exacerbations (AE), with varying natural history. The exacerbation is triggered by infection, leading to increased morbidity and mortality. The study on infectious aetiology of AECOPD is largely restricted to only viral or only bacterial aetiology. There are no studies from India that have investigated multiple viral, bacterial, and fungal associations from the same group of patients. This prospective study was conducted over 2 years to estimate the incidence and profile of viral infections in AECOPD patients,

The family Potyviridae represents the largest and most economically important group of plant-infecting RNA viruses. Despite extensive study of crop-associated members, the full diversity, host range, and evolutionary history of potyvirids remain poorly understood. Here, we conducted a large-scale mining of publicly available RNA-seq datasets to systematically search for novel potyvirid sequences. This approach enabled the identification and assembly of 47 previously undescribed members of the family, distributed across 8 recognized genera and, importantly, putative new groups. Beyond expanding

Hypovirulence-associated mycoviruses have the potential as biocontrol agents for plant fungal disease management, and exploration of the interactions between these mycoviruses and phytopathogenic fungi can provide opportunities to elucidate the underlying mechanisms of hypovirulence and antiviruses. We previously found that Sclerotinia sclerotiorum endornavirus 3 (SsEV3), belonging to the genus Betaendornavirus within the family Endornaviridae, confers hypovirulence on the phytopathogenic fungus Sclerotinia sclerotiorum, but the underlying mechanisms remains unclear. In this study, we found th

Mycoviruses are widespread in fungi and can influence fungal physiology, pathogenicity, and ecological interactions. In the plant pathogenic fungus Sclerotinia sclerotiorum, increasing attention has been directed toward understanding the diversity and functional roles of its associated viruses. This review synthesizes current knowledge on mycovirus detection methods and summarizes the diversity of viruses reported in S. sclerotiorum. Early detection strategies based on double-stranded RNA profiling, virion purification, and RT-PCR laid the foundation for mycovirus discovery but were limited in

Giant viruses of the phylum Nucleocytoviricota have emerged as particularly notable due to their increasingly recognized impacts on eukaryotic genome evolution. Their origins are hypothesized to predate or coincide with the diversification of eukaryotes, and they have been detected in hosts that span the eukaryotic tree of life. But surprisingly, such viruses have not been definitively found in Kingdom Fungi, though earlier genomic and metagenomic work suggests putative associations. Here we report both "viral fossils" and active infection by giant viruses in fungi, particularly in the zoospor

The complex interaction between viruses and fungi has profound implications, especially given the significant impact of these microorganisms on human health. While well-known examples such as HIV, influenza, and SARS-CoV-2 are recognized as risk factors for invasive fungal diseases, the relationship between viruses and fungi remains largely underexplored outside of these cases. Fungi and viruses can engage in symbiotic or synergistic interactions. Remarkably, some viruses, known as mycoviruses, can directly infect fungi, may influencing their phenotype and potentially their virulence. In addit

Interspecies transmission of viruses is a well-known phenomenon in animals and plants whether via contacts or vectors. In fungi, interspecies transmission between distantly related fungi is often suspected but rarely experimentally documented and may have practical implications. A newly described double-strand RNA (dsRNA) virus found asymptomatic in the phytopathogenic fungus Leptosphaeria biglobosa of cruciferous crops was successfully transmitted to an evolutionarily distant, broad-host range pathogen Botrytis cinerea. Leptosphaeria biglobosa botybirnavirus 1 (LbBV1) was characterized in L.

The Polymycoviridae family is an expanding group of mycoviruses, generally characterized by atypical capsids and segmented double-stranded (ds) RNA genomes. Polymycoviruses have been associated with various changes in their hosts' traits, such as shifts in spore production, changes in pigmentation, and alterations in growth and virulence, either enhancing or reducing them. Here, we report the complete genome sequence of a polymycovirus identified in an entomopathogenic, fungicolous, and keratinophilic, filamentous ascomycete species Lecanicillium aphanocladii, isolated from a patient with foot

Although viruses are primarily characterized as pathogenic agents, certain viruses confer advantages to their hosts. The extent to which a virus can improve host biological performance, however, remains a fascinating topic in virology. An endornavirus, Rhizoctonia solani endornavirus IM (RsEV-IM), was identified as prevalent in Rhizoctonia solani isolates obtained from potato plants. Comparative analysis with a virus-free isogenic strain demonstrated that RsEV-IM infection enhances mycelial growth, sclerotium formation, stress tolerance, and fungal virulence across multiple plant species. Mycoviruses are viruses that infect fungi and are widespread across all major fungal taxa, exhibiting great biological diversity. Since their discovery in the 1960s, researchers have observed a myriad of fungal phenotypes altered due to mycoviral infection. In this review, we examine the nuanced world of mycoviruses in the context of the medically and agriculturally important fungal genus, Aspergillus. The advent of RNA sequencing has revealed a previous underestimate of viral prevalence in fungi, in particular linear single-stranded RNA viruses, and here we outline the diverse viral families

Liver transplantation : official publication of the American Association for the Study of Liver Diseases and the International Liver Transplantation Society2006eng
Viral and fungal infections in liver transplant recipients are important to recognize and treat early because of their association with substantial morbidity and mortality. Some viruses, such as cytomegalovirus and human herpesvirus 6, have immunomodulatory properties and can facilitate other infections, including fungal infections. Cytomegalovirus has long been recognized as an important virus in transplantation, but in the past decade other viruses have also received attention in the medical literature because of their association with particular clinical syndromes. Although human herpesviru

Mycoviruses, or fungal viruses, are widespread throughout the fungal kingdom. This study aimed to identify environmental factors associated with mycoviruses, explore their transmission mechanisms, including vector-mediated transmission, and assess their potential practical applications. A systematic, retrospective, and cross-sectional bibliographic review was conducted. These viruses are closely linked to their fungal hosts, thus developing a symbiotic relationship. Among environmental variables, temperature appears to play a more significant role in limiting fungal virulence than other factor

Ambiviruses are fungal-infecting circular RNA viruses that uniquely combine viroid-like and viral features, yet the function of their conserved ORF-B protein and their effects on hosts remain unknown, hindered by the lack of a reverse genetics system. Here, we constructed the first infectious cDNA clone of an ambivirus, Fusarium graminearum ambivirus 1 (FgAV1), using a head-to-tail dimer placed downstream of a fungal promoter. FgAV1 was horizontally transmitted via hyphal anastomosis to virus-free Fusarium graminearum strains. Notably, a reverse-oriented dimer construct was also infectious and

Rhizopus microsporus is a species in the order Mucorales that is known to cause mucormycosis, but it is poorly understood as a host of viruses. Here, we examined 25 clinical strains of R. microsporus for viral infection with a conventional double-stranded RNA (dsRNA) assay using agarose gel electrophoresis (AGE) and the recently established fragmented and primer-ligated dsRNA sequencing (FLDS) protocol. By AGE, five virus-infected strains were detected. Then, full-length genomic sequences of 12 novel RNA viruses were revealed by FLDS, which were related to the families Mitoviridae, Narnavirida

Cotton verticillium wilt caused by Verticillium dahliae severely restricts global cotton production. Mycovirus-induced hypovirulence provides a promising strategy for sustainable disease management. In this study, V. dahliae isolates from Xinjiang were screened by virome sequencing and RT-PCR, and strain 121-11C-1 co-infected with Verticillium dahliae partitivirus 1 (VdPV1) and Verticillium dahliae magoulivirus 1 (VdMoV1) was identified. Virus-free isogenic strains were obtained via single-conidium purification, verifying that both viruses can be vertically transmitted through fungal conidia.

Cross-kingdom virus transmission between plants and fungi remains rare in nature. Here, a novel circular ssDNA virus, Diaporthe pseudophoenicicola DNA virus 1 (DpDV1), was identified from the phytopathogenic fungus Diaporthe pseudophoenicicola. DpDV1 attenuates the pathogenicity of both its original and experimental host fungi, demonstrating its potential as a biocontrol agent against fungal phytopathogens. Remarkably, DpDV1 displays bidirectional plant-fungal transmission capability without inducing observable phenotypic changes in plant hosts, overcoming the limitations imposed by vegetative

Fungi are known to be frequently infected by mycoviruses, which could play an important yet underexplored role in the fungal holobiont. Mycoviruses can enhance fungal traits, such as salinity tolerance and resistance to fungicides, and may also benefit plant hosts in tripartite interactions. While research on arbuscular mycorrhizal fungi (AMF) has focused on their ecological and agricultural value, few studies have explored their virome, possibly due to the difficulty of culturing AMF in the lab. This study presents an updated bioinformatic approach for virome characterization, emphasizing RNA

Nowadays, the focus of mycovirology research has expanded from plant pathogenic fungi and mycovirus mediated hypovirulence to include insect and human pathogenic fungi together with a range of mycovirus mediated phenotypes, such as hypervirulence, control of endophytic traits, regulation of metabolite production and drug resistance. In fungus-mycovirus-environmental interactions, the environment and both abiotic and biotic factors play crucial roles in whether and how mycovirus mediated phenotypes are manifest. Mycovirus infections result in alterations in the host transcriptome profile, via p
