Zebrafish ventral gastrula fate map reveals neural crest progenitor domain.

BACKGROUND: Fate maps relate progenitor cell positions to later fates and locations of their progeny, revealing early embryonic organization. Previous zebrafish fate maps identified the origin of germ layers and derivative cell fates, but ventral gastrula progenitor domains were not fully resolved. In particular, the neural crest, a multipotent cell type central to vertebrate development and evolution, was not characterized in the zebrafish fate map. RESULTS: We generated a region-based fate map to resolve the origin of the neural crest and overall organization of the ventral gastrula. This f

[Situs inversus of the Newt (Triturus alpestris) after UV-Irradiation of Gastrulae].

Gastrulae (Harrison 9-13), and for comparison neurulae (Harrison 15-17) ofTriturus alpestris were irradiated with mixed ultraviolet light of various doses and at various body regions. Bilateral irradiation leads to a markedly higher mortality than does unilateral irradiation. Gastrulae possess a better regulation capability than neurulae. The frequency of alterations of the situs (irregularities, partial and total inversions), lethality and rate of malformations are dose dependent. Irradiation of the right side ofearly gastrulae (Harrison 9-11a) leads to a higher frequency of inversions than d

Core pathway proteins and the molecular basis of planar polarity in the zebrafish gastrula.

The planar polarization of cells and subcellular structures is critical for embryonic development. Coordination of this polarity can provide cells a sense of direction in relation to the anterior-posterior and dorsal-ventral body axes. Fly epithelia use a core pathway comprised of transmembrane (Van Gogh/Strabismus, Frizzled, and Flamingo/Starry night) and cytoplasmic (Prickle or Spiny-legs, Dishevelled, and Diego) proteins to communicate directional information between cells and thereby promote the uniform orientation of structures such as hairs. In the zebrafish gastrula, planar polarity und

Ethanol downregulates gastrula gene expression and cell movement, causing symptoms of foetal alcohol spectrum disorders.

Foetal alcohol spectrum disorders (FASDs) occur in embryos when they are exposed to maternally supplied alcohol. To study the mechanisms of FASDs, the zebrafish embryo can serve as an excellent model as ethanol-exposed zebrafish embryos exhibit common symptoms of human FASDs including microcephaly, incomplete neural plate closure, eye defects, craniofacial disorders and many other defects. Here, we investigated the embryo development at gastrula stage where three germ layers develop with specific gene expressions and undergo dynamic cell movement including extension, convergence and epiboly, e

[Provocation of situs inversus by means of material defects in the lateral ectoderm of the gastrula ofTriturus alpestris].

Roundish pieces were removed from the lateral polar region of young gastrulae ofTriturus alpestris. These consisted mainly of undetermined ectoderm and partly of ventral mesoderm. The majority developed into normal embryos or embryos with minor abnormalities. After injury of the left side (101 cases) the gut situs of the larvae was normal in 85.2% median in 6.9% and inverted in 7.9%. When injured on the right side (106 cases), the larval gut situs was regular in 79,2%, median in 8.5% and inverted in 12.3%.These results support the statements of v.KRAFT (1968b) who also found a higher rate of si

Dorsal convergence of gastrula cells requires Vangl2 and an adhesion protein-dependent change in protrusive activity.

Lateral zebrafish hypoblast cells initiate dorsal convergence near mid-gastrulation and exhibit non-polarized morphologies, limited cell-cell contact and indirect migration trajectories. By late gastrulation, mesodermal cells become packed as they engage in planar cell polarity (PCP)-dependent movement. Here, we aimed to understand this transition in cell behavior by examining the relationship between protrusion dynamics and establishment of PCP and directed migration. We found that wild-type cells undergo a reduction in bleb protrusions near late gastrulation accompanied by a VANGL planar cel

BMP antagonism protects Nodal signaling in the gastrula to promote the tissue interactions underlying mammalian forebrain and craniofacial patterning.

Holoprosencephaly (HPE) is the most common forebrain and craniofacial malformation syndrome in humans. The genetics of HPE suggest that it often stems from a synergistic interaction of mutations in independent loci. In mice, several combinations of mutations in Nodal signaling pathway components can give rise to HPE, but it is not clear whether modest deficits of Nodal signaling along with lesions in other pathways might also cause such defects. We find that HPE results from simultaneous reduction of Nodal signaling and an organizer BMP (bone morphogenetic protein) antagonist, either Chordin o

Brachyury drives formation of a distinct vascular branchpoint critical for fetal-placental arterial union in the mouse gastrula.

How the fetal-placental arterial connection is made and positioned relative to the embryonic body axis, thereby ensuring efficient and directed blood flow to and from the mother during gestation, is not known. Here we use a combination of genetics, timed pharmacological inhibition in living mouse embryos, and three-dimensional modeling to link two novel architectural features that, at present, have no status in embryological atlases. The allantoic core domain (ACD) is the extraembryonic extension of the primitive streak into the allantois, or pre-umbilical tissue; the vessel of confluence (VOC

Basic fibroblast growth factor induces differentiation of neural tube and neural crest lineages of cultured ectoderm cells from Xenopus gastrula.

The vertebrate nervous system is initially induced from a section of dorsal ectoderm by signal(s) from the underlying dorsal mesoderm during gastrulation. In an effort to identify the neural inducing factor(s) emanating from the dorsal mesoderm, we have examined the inductive action of various growth factors by applying them to ectoderm cells from Xenopus gastrulae (8- to 12.5-hour age; embryonic stage 9+ to 11 1/2) in a microculture system. Vitamin A deficiency in the late gastrula stage rat embryo results in a one to two vertebral anteriorization that extends throughout the axial skeleton.

Vitamin A and its metabolites are known to be involved in patterning the vertebrate embryo. Study of the effect of vitamin A on axial skeletal patterning has been hindered by the fact that deficient embryos do not survive past midgestation. In this study, pregnant vitamin A-deficient rats were maintained on a purified diet containing limiting amounts of all-trans retinoic acid (12 microg atRA/g diet) and given a daily oral bolus dose of retinol starting at embryonic day 0.5, 8.25, 8.5, 8.75, 9.25, 9.5, 9.75, or 10.5. Embryos were recovered at E21.5 for analysis of the skeleton and at earlier t

Does the respiratory rate in sea urchin embryos increase during early development without proliferation of mitochondria?

During early development of the sea urchin, the respiratory rate, enhanced upon fertilization, is maintained up to hatching (pre-hatching period) and then gradually increases to a maximum at the gastrula stage (post-gastrula period). Except for a short duration after fertilization, respiration in embryos is strongly inhibited by CN- and antimycin A. During the whole span of early development, the amounts of proteins, cytochromes and the specific activities of cytochrome c oxidase and reduced nicotinamide adenine dinucleotide (NADH) cytochrome c reductase in mitochondria are practically the sam

Stat3-Efemp2a modulates the fibrillar matrix for cohesive movement of prechordal plate progenitors.

Recently, emerging evidence has shown that Stat3 controls tumor cell migration and invasion. However, the molecular mechanisms by which Stat3 controls the cell movement remain largely unknown. Embryonic gastrula progenitors display coordinated and orientated migration, called collective cell migration. Collective cell migration is the simultaneous movement of multiple cells and is universally involved in physiological and pathological programs. Stat3 activity is required for the migration of gastrula progenitors, but it does not affect cell specification, thus suggesting that gastrula movement

Unveiling Gene Expression Dynamics during Early Embryogenesis in Cynoglossus semilaevis: A Transcriptomic Perspective.

Commencing with sperm-egg fusion, the early stages of metazoan development include the cleavage and formation of blastula and gastrula. These early embryonic events play a crucial role in ontogeny and are accompanied by a dramatic remodeling of the gene network, particularly encompassing the maternal-to-zygotic transition. Nonetheless, the gene expression dynamics governing early embryogenesis remain unclear in most metazoan lineages. We conducted transcriptomic profiling on two types of gametes (oocytes and sperms) and early embryos (ranging from the four-cell to the gastrula stage) of an eco

CLDN10-driven lineage decision in an amnion and primordial germ cell progenitor at the amnion-epiblast boundary in primates.

BACKGROUND: A growing body of evidence from primate embryos as well as in vitro systems supports the notion that amnion and primordial germ cell (PGC) lineage progressing cells share a common precursor. RESULTS: To gain comprehensive transcriptomic insights into this critical but poorly understood precursor and its progeny, we examine the evolving transcriptome of a developing human pluripotent stem cell-derived model of amnion and PGC formation at the single cell level. This analysis reveals several continuous amniotic fate progressing states with state-specific markers. Additionally, a prog

[The situs viscerum in the newt (Triturus alpestris) after material defects in the ectoderm of the blastula].

In the blastula stage ofTriturus alpestris roundish pieces were removed from the animal half of the embryo, which consisted of undetermined ectoderm; some of them could perhaps contain some mesoderm on their vegetal edge. Since the median plane could not be ascertained in this stage, a chance distribution around the animal-vegetal axis has to be assumed. Most of the embryos developed to externally normal larvae. The gut situs of 193 larvae was regular in 97.9% of the cases, median in 0.5% and inverted in 1.5%. This corresponds to the inversion rate of non operated animals. Therefore mechanic def

Inhibition of ecto-5'-nucleotidase and adenosine deaminase is able to reverse long-term behavioural effects of early ethanol exposure in zebrafish (Danio rerio).

The behavioural impacts of prenatal exposure to ethanol include a lower IQ, learning problems, anxiety and conduct disorders. Several components of the neurochemical network could contribute to the long-lasting effects of ethanol embryonic exposure. Adenosine is an important neuromodulator, that has been indicated to be affected by acute and chronic exposure to ethanol. Here, embryos of zebrafish exposed to 1% ethanol during the developmental stages of gastrula/segmentation or pharyngula exhibited anxiolytic effect, increased aggressiveness, and decreased social interaction. Zebrafish as a Model for Fetal Alcohol Spectrum Disorders.

In this review, we will discuss zebrafish as a model for studying mechanisms of human fetal alcohol spectrum disorders (FASDs). We will overview the studies on FASDs so far and will discuss with specific focus on the mechanisms by which alcohol alters cell migration during the early embryogenesis including blastula, gastrula, and organogenesis stages which later cause morphological defects in the brain and other tissues. FASDs are caused by an elevated alcohol level in the pregnant mother's body. The symptoms of FASDs include microcephaly, holoprosencephaly, craniofacial abnormalities, and car

Isolation and developmental expression of the amphibian homolog of the fibroblast growth factor receptor 3.

Recent observations suggest that fibroblast growth factors (FGFs) and their receptors are involved in the control of embryogenesis. Several FGF receptor genes have been identified so far and their expression is differentially regulated. As part of a continuing effort to analyse the differential expression of FGF receptors and their potential role during amphibian development, we have isolated a Pleurodeles homolog of FGF receptor 3 (FGFR-3), which we designated PFR-3 because of its highest homology to human FGFR-3 (75% overall identity). PFR-3 is a maternally derived mRNA. While a low level of

RNA synthesis in nuclei isolated from early embryos of Xenopus laevis.

1. Rates of RNA synthesis in isolated Xenopus embryo nuclei decrease from blastula through gastrula and neurula stages to hatching tadpoles. 2. In blastula and gastrula nuclei, net synthesis of RNA continues for over 30 min, both in the presence of KCl at 0.4 M and in its absence. In nuclei from later stages, net synthesis continues for only about 10 min in the absence of KCl. 3. At low ionic strength, RNA synthesis in all nuclei is greater with optimum Mg-2+ (6 mM) than with optimum Mn-2+ (1 mM). At high ionic strength the reverse is true. 4. Experimental evidence for autonomous action of the periodic albinism (ap) gene within developing retinal pigment cells and melanophores of Xenopus laevis.

Genes which affect pigment elaboration may do so by autonomous action within the developing pigment cells or by way of tissue interactions leading to pigment cell differentiation. The site of action of the periodic albinism (ap) gene was investigated by substituting presumptive neural ectoderm of gastrulae of one genotype with uncommitted ectoderm of different genotype. Retinal pigment cells and melanophores arising from such grafts were found to differentiate according to their own genotype in spite of having spent their entire developmental history in tissues of different genotype. This find
