The multicomponent residue depletion of Gelsemium elegans in pig tissues, urine, and plasma.

INTRODUCTION: Gelsemium elegans (G. elegans) as a traditional medicinal plant used in livestock production. The use of G. elegans in veterinary clinics may pose safety risks to human health. OBJECTIVES: The aim of this study was to investigate tissue residue depletion in pigs fed G. elegans powder. METHODS: A precise quantitation method and a simultaneous semi-quantitation method for multiple components independently of standards in pig tissues were developed for the first time. The two methods were validated in terms of specificity, LODs, LOQs, linearity, accuracy, precision, and matrix eff

Multiple animal intoxications associated with Carolina jessamine (Gelsemium sempervirens) ingestions.

Neurological signs characterized by marked progressive weakness and convulsions culminating in death were observed in 3 goats over a 24-h period. Affected animals were in a group of 5 goats confined toa fenced paddock: a domestic goose within the paddock was also found dead. Present in the same paddock, but unaffected, were 2 other goats and an adult cow. Five days prior to the animals' deaths, the owner had trimmed the surrounding brush and had thrown the cuttings into the enclosure. Post mortem examination of 2 of the dead goats and the goose revealed reduced muscle mass and fat stores, sero

Gelsenicine from Gelsemium elegans attenuates neuropathic and inflammatory pain in mice.

Gelsemium elegans BENTH and its crude extract are widely used to treat pain in China despite its apparent toxicity. The analgesic effects of gelsenicine, an active component of G. elegans, however, have not been reported. The current study examined potential analgesic effects of subcutaneously injected gelsenicine using acetic acid-induced writhing, formalin-induced nociceptive behavior, and thermal hyperalgesia caused by chronic constriction injury (CCI) in mice. Gelsenicine produced dose-dependent analgesic effects in both inflammatory and neuropathic pain models. The ED(50), for either the

Six New Dimeric Monoterpenoid Indole Alkaloids With Antiviral Activity From Gelsemium elegans.

Six new dimeric monoterpenoid indole alkaloids, geleganidines D-I (1-6), together with six known analogues (7-12), were isolated from the roots of Gelsemium elegans. Compounds 1 and 2 represent the first dimeric Gelsemium alkaloids composed of a humantenine-type unit and a gelsedine-type unit, connected through a carbonyl-derived urea bridge. Compounds 3-6 were new dimeric monoterpenoid indole alkaloids featuring distinct dimerization patterns, including a carbonyl-derived urea bridge (3), azo linkages (4 and 5), and a symmetric C-C-linked dimer (6). Their structures and absolute configuration

Preparative separation of alkaloids from Gelsemium elegans Benth. using pH-zone-refining counter-current chromatography.

Alkaloids in Gelsemium elegans possess a variety of therapeutic properties, including tumor suppression, analgesic and anti-inflammatory effects. In China, G. elegans has been used for centuries to treat a variety of medical conditions, including chronic pain and skin ulcer. Methods currently used to separate the active components of G. elegans are time-consuming and have low recovery. In the present study, we used pH-zone-refining counter-current chromatography to separate major alkaloids from a crude extract of G. elegans. The two-phase solvent system was methyl tert-butyl ether (MtBE)/aceto

Excretion, Metabolism, and Tissue Distribution of Gelsemium elegans (Gardn. & Champ.) Benth in Pigs.

Gelsemium elegans (Gardn. & Champ.) Benth is a toxic flowering plant in the family Loganiaceae used to treat skin diseases, neuralgia and acute pain. The high toxicity of G. elegans restricts its development and clinical applications, but in veterinary applications, G. elegans has been fed to pigs as a feed additive without poisoning. However, until now, the in vivo processes of the multiple components of G. elegans have not been studied. This study investigates the excretion, metabolism and tissue distribution of the multiple components of G. elegans after feeding it to pigs in medicated feed

Gelsemium elegans poisoning-induced acute respiratory failure treated with respiratory support and hemoperfusion: A case report.

We report a case of severe Gelsemium elegans poisoning in a 64-year-old male following ingestion of approximately 40 mL of a Gelsemium decoction. Shortly after presentation with blurred vision, the patient developed sudden respiratory arrest during gastric lavage. Immediate endotracheal intubation and mechanical ventilation were performed, followed by hemoperfusion and supportive care in the intensive care unit. The patient regained spontaneous respiration, was weaned off ventilation, and achieved full recovery without residual complications. This case highlights the critical role of prompt re

Fatal gelsemium alkaloid poisoning versus cardiovascular cause in a sudden death: a forensic case report.

Gelsemium alkaloids, such as gelsemine and koumine, are highly toxic compounds that can induce rapid respiratory and cardiovascular failure. Their clinical manifestations-dizziness, ptosis, and sudden collapse-closely resemble those of acute myocardial infarction, presenting diagnostic challenges in forensic practice, especially in distinguishing between poisoning and cardiovascular events. We describe a sudden death in a middle-aged male following a restaurant meal. The decedent developed acute dizziness and eyelid drooping shortly after ingestion and died within hours despite resuscitation.

Chronic Exposure to Gelsemium Preparations Alters Mitochondrial Bioenergetics, Neurite Outgrowth, and Akt/mTOR Signaling in Human Neuronal Cells.

Stress-related psychiatric disorders are frequently associated with impaired mitochondrial function, altered neuronal energy metabolism, and reduced neuroplasticity. Intracellular pathways such as PI3K/Akt and mTOR play central roles in regulating mitochondrial bioenergetics and neuronal structural adaptation. Gelsemium is traditionally used in integrative and homeopathic practice; however, the cellular effects of prolonged exposure to high serial dilutions remain insufficiently characterized. This study aimed to examine the effects of chronic exposure to Gelsemium preparations on mitochondria

Effects of koumine, an alkaloid of Gelsemium elegans Benth., on inflammatory and neuropathic pain models and possible mechanism with allopregnanolone.

Crude alkaloidal extraction from Gelsemium elegans Benth. produces analgesic property. However, its clinical utility has been obstructed by its narrow therapeutic index. Here, we investigated the potential of koumine, a monomer of Gelsemium alkaloids, to reduce both inflammatory and neuropathic pain. Interestingly, allopregnanolone, a neurosteroid, appeared to mediate the reduction of neuropathic pain. The potential anti-inflammatory pain effects of koumine were evaluated by acetic acid-, formalin- and complete Freund's adjuvant (CFA) -induced nociceptive behaviors in mice. Chronic constrictio

Gelsemium elegans extract powder alleviates high stocking density-related adverse effects in broilers via immune-intestinal microbiota synergistic mechanisms.

This study aimed to investigate the effects of different doses of Gelsemium elegans extract powder (GEE) on broilers under high stocking density (HSD). A total of 78 one-day-old male yellow-feathered broilers were randomly assigned to 5 treatment groups: normal stocking density (NSD), HSD, HSD + 0.06% GEE, HSD + 0.12% GEE, and HSD + 0.6% GEE. The results showed that HSD impaired broiler growth and physiological functions. Dietary supplementation with 0.06% GEE (200 mg/kg diet Gelsemium elegans extract) significantly improved growth performance (marked increases in body weight, average daily fe
