Tomato auxin biosynthesis/signaling is reprogrammed by the geminivirus to enhance its pathogenicity.

Tomato leaf curl New Delhi virus-derived AC4 protein interacts with host proteins involved in auxin biosynthesis and reprograms auxin biosynthesis/signaling to help in viral replication and manifestation of the disease-associated symptoms. Perturbations of phytohormone-mediated gene regulatory network cause growth and developmental defects. Furthermore, plant viral infections cause characteristic disease symptoms similar to hormone-deficient mutants. Tomato leaf curl New Delhi Virus (ToLCNDV)-encoded AC4 is a small protein that attenuates the host transcriptional gene silencing, and aggravated

Characterization of the C4 proteins encoded by okra-infecting geminiviruses in India.

BACKGROUND: Okra (bhendi) is a widely cultivated food crop in warm regions of the world, with India contributing approximately 60% of global production. However, okra cultivation in India is severely constrained by viral diseases, among which infections caused by the geminiviruses bhendi yellow vein mosaic virus (BYVMV) and okra enation leaf curl virus (OELCuV), in association with their satellites, represent major limitations to crop productivity. In recent years, the geminivirus-encoded C4 protein has emerged as a key pathogenicity determinant in this viral family, with functions that includ

Genome characterization of an Argentinean isolate of alfalfa leaf curl virus.

We investigated the molecular characteristics of an Argentinean isolate of alfalfa leaf curl virus (ALCV-Arg), a virus of the genus Capulavirus in the family Geminiviridae that was isolated from alfalfa plants showing dwarfism. The genome was found to be 2,750 nucleotides in length. In pairwise comparisons, this ALCV isolate shared 83.2% to 92.6% sequence identity with European ALCV isolates. Sequence comparisons and phylogenetic analysis showed that this isolate combines features of strains A and B of ALCV. Recombination analysis showed that ALCV-Arg is a recombinant isolate that was generate

Surveying for potential grapevine red blotch virus vectors in an Oregon vineyard.

Grapevine red blotch virus (GRBV) reduces grapevine vigor, delays grape ripening, reduces sugar content and quality of wines in North America and other continents. To date, the three-cornered alfalfa hopper, Spissistilus festinus, has been clearly identified as a vector in the United States. Since this hopper has not been observed in some vineyards where secondary spread occurs, additional vectors of GRBV are likely. To search for vectors, we conducted a two-year survey collecting live hemipterans from infected vines and surrounding grass, trees, and shrubs. Screening a wide variety of hemipte

Temporal dynamics of Grapevine red blotch virus accumulation in grapevine leaves is influenced by fruit maturity stages.

Grapevine red blotch disease (GRBD) poses a serious threat to viticulture in the United States. Grapevine red blotch virus (GRBV), a member species of Grablovirus vitis (family Geminiviridae), the causal agent of GRBD, disrupts grapevine physiology and metabolism, thereby interfering with the natural processes of fruit ripening. To date, there is limited knowledge about how grape maturity stages influence the timing of changes in viral load and disease progression. This study elucidates the effect of fruit maturity stages on GRBV accumulation in Vitis vinifera L. cv. Merlot grapevines in a Cen

Development of a new Collateral Cleavage-independent CRISPR/Cas12a based easy detection system for plant viruses.

Plant virus spread through various means, from mechanically to the insect vectors and act as obligate parasite, therefore, are extremely challenging to eradicate. Geminiviruses are an important class of viruses which have reported extensively in last two decades on several new hosts. They infect wide range of annual crops and perineal shrubs, therefore, essentially required to detect them on field and dispose to check their vector transmission to healthy crops. In this study, we have chosen two important begomovirus viz. Mungbean yellow mosaic India virus which infect wide range of leguminous

The divergent V2 protein encoded by parsley yellow leaf curl virus is a suppressor of transcriptional and post-transcriptional gene silencing and a potential symptom determinant.

The Geminiviridae family encompasses over 500 species of economically important plant viruses. A new geminivirus species named parsley yellow leaf curl virus (PYLCV) was first identified in 2020 in Southeastern Iran. The PYLCV genome, comprising 2,779 nucleotides, exhibits a pairwise sequence identity of less than 66% with other geminiviruses and encodes a divergent V2 protein that lacks sequence similarity to known proteins in GenBank. This study explores the functions of this V2 through various analyses. An assessment of the V2 amino acid composition indicates a higher prevalence of acidic r

Alfalfa leaf curl virus is transmitted by Aphis craccivora in a highly specific circulative manner.

Two members of the genus Capulavirus (Geminiviridae) are transmitted by aphids including Alfalfa leaf curl virus (ALCV) transmitted by Aphis craccivora. The capulavirus Euphorbia caput-medusae latent virus was shown here to be transmitted also by A. craccivora, using the population EuphorbiaSA. ALCV was transmissible by several A. craccivora populations including Robinia, but not the EuphorbiaSA population, reflecting a high transmission specificity. Typical of the circulative-persistent mode of transmission, ALCV persists through insect molts. ALCV accumulation and localization were analyzed

Spatiotemporal spread of grapevine red blotch-associated virus in a California vineyard.

Grapevine red blotch-associated virus (GRBaV), the causative agent of red blotch disease, is a member of the genus Grablovirus, in the family Geminiviridae and the first known geminivirus of Vitis spp. Limited information is available on the epidemiology of red blotch disease. A 2-hectare Vitis vinifera cv. 'Cabernet franc' vineyard in Napa County, California, USA was selected for monitoring GRBaV spread over a three-year period (2014-2016) based on an initially low disease incidence and an aggregation of symptomatic vines at the edge of the vineyard proximal to a wooded riparian area. The inc

Leaf Bacteriome in Sugar Beet Shows Differential Response against Beet curly top virus during Resistant and Susceptible Interactions.

Beet curly top virus (BCTV) significantly reduces sugar beet yield in semi-arid production areas. Genetic resistance to BCTV is limited; therefore, identification of additional resistance-associated factors is highly desired. Using 16S rRNA sequencing and BCTV resistant (R) genotypes (KDH13, KDH4-9) along with a susceptible (S) genotype (KDH19-17), we investigated leaf bacteriome changes during BCTV post inoculation (pi). At day 6 (~6-week-old plants), Cyanobacteria were predominant (~90%); whereas, at week 4 (~10-week-old plants) Firmicutes (11-66%), Bacteroidetes (17-26%), and Verrucomicrobi

Immunolocalization of Beet Curly Top Virus (BCTV) and GroEL Chaperon Protein of Endosymbionts in Beet Leafhopper (Circulifer tenellus) Vector Tissue.

Beet curly top virus (BCTV, curtovirus, geminiviridae) causes one of the most economically significant viral diseases in crops in the Western United States and is transmitted only by the beet leafhopper (Circulifer tenellus) in a non-propagative circulative manner. A better understanding of how this virus overcomes insect vector cellular barriers is essential to understanding virus-vector interactions. The distribution of BCTV in its beet leafhopper vector was investigated using immunofluorescence confocal laser scanning microscope analysis (iCLSM) on the whole-mount-dissected organs of leafho

Evidence for transovarial transmission of tomato yellow leaf curl virus by its vector, the whitefly Bemisia tabaci.

The whitefly Bemisia tabaci is the only vector of the tomato yellow leaf curl geminivirus (TYLCV). The insect transmits the virus in a persistent-circulative manner. TYLCV DNA was detected by polymerase chain reaction and by Southern blot hybridization in progeny (eggs, first and second instars, adults) of single viruliferous whiteflies that developed on eggplant or on cotton (two TYLCV nonhost plants). Furthermore, TYLCV DNA was present in the progeny of insects that had acquired the virus through the egg. The adult progeny of the viruliferous insects and their own progeny were able to infect

A Rapid and Sensitive Loop-Mediated Isothermal Amplification Assay for the Detection of Mulberry Mosaic Dwarf-Associated Virus.

Mulberry mosaic dwarf-associated virus (MMDaV), a member of the genus Mulcrilevirus in the family Geminiviridae, poses a serious threat to mulberry cultivation and growth in China. Early and accurate diagnosis is a prerequisite for the effective prevention and control of MMDaV-related disease. In this study, a loop-mediated isothermal amplification (LAMP)-based detection method was established for the specific identification of MMDaV. Three pairs of specific primers were designed targeting the nucleotide sequences of the MMDaV V2 gene. The optimized LAMP reaction was performed at a constant te

Defender or accomplice? Dual roles of plant vesicle trafficking in restricting and enabling geminiviral systemic infection.

The vesicle trafficking system enables multidirectional cargo fluxes between endomembrane compartments. However, vesicle trafficking plays dual roles during pathogen infections. In plants, it mediates autophagic immune responses but can also be hijacked by pathogens to facilitate successful infections. We demonstrate that vesicle trafficking machinery acts as a double-edged sword during infection by the geminivirus tomato yellow leaf curl Sardinia virus (TYLCSaV) in Nicotiana benthamiana. Virus-induced gene silencing of eight genes encoding key vesicle trafficking regulators revealed contrasti

Analysis of bidirectional promoter activity and structure of the large intergenic region (LIR) in mulberry crinkle leaf virus.

The mulberry crinkle leaf virus (MCLV) is a representative member of the genus Mulcrilevirus of the family Geminiviridae. Here, the bidirectional promoter activity of the long intergenic region (LIR) of MCLV and the roles of a specific element consisting of five GAAAAA repeats [(GAAAAA)5] in LIR were investigated using Agrobacterium-mediated transient expression and transfection of Nicotiana benthamiana protoplasts. Transient expression results demonstrated that, similar to other geminiviruses, MCLV’s LIR also exhibits bidirectional promoter activity. The promoter activity of the C-sense is si
