Alterations in gene expression of complement components in chronic rhinosinusitis.

BACKGROUND: The complement cascade forms part of the initial innate response to pathogens in the airway. Complement activation is important in the maintenance of host homeostasis, but excessive and uncontrolled activation may lead to inflammation and disease. The role of the complement pathway in the innate response in chronic rhinosinusitis (CRS) is poorly characterized Methods: Sinus mucosa biopsy specimens from the anterior ethmoid or uncinate process of patients with allergic fungal rhinosinusitis (AFRS), CRS without NPs (CRS-NPs), and controls were harvested and gene and protein expressio

Gene editing of clock components in Solanum lycopersicum: Effects on gene expression, development, and productivity.

The circadian clock plays a crucial role in regulating key biological processes, including growth and development. While studies in the model plant Arabidopsis thaliana have significantly advanced our understanding of circadian function, recent research has also focused on crop species for improved yield and quality. In this study, we examined the rhythmic behavior and regulatory function of circadian clock components in tomato (Solanum lycopersicum). Time course analyses of gene expression over the circadian cycle revealed robust rhythmic oscillations in tomato leaves under free-running condi

Integrating gene delivery and gene-editing technologies by adenoviral vector transfer of optimized CRISPR-Cas9 components.

Enhancing the intracellular delivery and performance of RNA-guided CRISPR-Cas9 nucleases (RGNs) remains in demand. Here, we show that nuclear translocation of commonly used Streptococcus pyogenes Cas9 (SpCas9) proteins is suboptimal. Hence, we generated eCas9.4NLS by endowing the high-specificity eSpCas9(1.1) nuclease (eCas9.2NLS) with additional nuclear localization signals (NLSs). We demonstrate that eCas9.4NLS coupled to prototypic or optimized guide RNAs achieves efficient targeted DNA cleavage and probe the performance of SpCas9 proteins with different NLS compositions at target sequences

Intron retention as a component of regulated gene expression programs.

Intron retention has long been an exemplar of regulated splicing with case studies of individual events serving as models that provided key mechanistic insights into the process of splicing control. In organisms such as plants and budding yeast, intron retention is well understood as a major mechanism of gene expression regulation. In contrast, in mammalian systems, the extent and functional significance of intron retention have, until recently, remained greatly underappreciated. Technical challenges to the global detection and quantitation of transcripts with retained introns have often led t

Expression of a sperm flagellum component encoded by the Als2cr12 gene.

Genes exclusively expressed in male germ cells encode proteins that play important roles in spermatogenesis and fertilization. In this study, we investigated the expression of a novel spermatogenic cell-specific gene known as amyotrophic lateral sclerosis 2 chromosome region candidate 12 (Als2cr12). Our in silico and in vitro analyses revealed that the mouse Als2cr12 gene produces two transcript isoforms by alternative splicing and that one of the isoforms is unique to spermatogenic cells. Using an antibody against the ALS2CR12 protein, we found that a protein from the germ cell-specific Als2c

Investigation of genes coding for inflammatory components in Parkinson's disease.

Several findings obtained recently indicate that inflammation may contribute to the pathogenesis in Parkinson's disease (PD). Genetic variants of genes coding for components involved in immune reactions in the brain might therefore influence the risk of developing PD or the age of disease onset. Five single nucleotide polymorphisms (SNPs) in the genes coding for interferon-gamma (IFN-gamma; T874A in intron 1), interferon-gamma receptor 2 (IFN-gamma R2; Gln64Arg), interleukin-10 (IL-10; G1082A in the promoter region), platelet-activating factor acetylhydrolase (PAF-AH; Val379Ala), and intercell

Artificial intelligence-curated repository of gene-encoded natural diverse components from herbal medicines.

Natural components, evolved to help organisms adapt and defend against threats, are also vital sources for drug discovery due to their diverse and potent bioactivities. In the present work, we proposed the Gene-encoded Natural Diverse Components Repository (GNDC, https://cbcb.cdutcm.edu.cn/gndc/), a primary and most extensive database dedicated to cataloging diverse natural components. GNDC currently catalogs over 234 million natural components that are organized into four specialized sub-databases: HerbalMDB for 2.32 million secondary metabolites, HerbalPDB for 229 million small peptides, Her

Gene Variants in Components of the microRNA Processing Pathway in Chronic Myeloid Leukemia.

Current therapy in chronic myeloid leukemia (CML) has improved patient life expectancy close to that of healthy individuals. However, molecular alterations other than BCR::ABL1 fusion gene in CML are barely known. MicroRNAs are important regulators of gene expression, and variants in some of the components of microRNA biosynthesis pathways have been associated with genetic susceptibility to different types of cancer. Thus, the aim of this study was to evaluate the association of variants located in genes involved in the biogenesis of microRNAs with susceptibility to CML. Fifteen variants in ei

Gene polymorphisms associated with corneal curvature, astigmatism and its vector components in children.

BACKGROUND: While parental astigmatism is a known risk factor for childhood astigmatism, the molecular genetic basis remains elusive. Previous genetic studies, largely confined to adult corneal and refractive astigmatism (CA and RA), did not address internal astigmatism (IA) and astigmatism vector components. We aimed to determine whether genes previously identified to have associations with corneal curvature (CR), CA, and RA in adults similarly occur for CR, CA, RA, IA, and astigmatism vector components (J0 and J45) in children. METHODS: Fourteen polymorphisms in nine loci were genotyped in

Dissecting components of the Campylobacter jejuni fetMP-fetABCDEF gene cluster under iron limitation.

Campylobacter jejuni is a bacterium that is prevalent in the ceca of farmed poultry such as chickens. Consumption of ill-prepared poultry is thus the most common route by which C. jejuni infects the human gut to cause a typically self-limiting but severe gastrointestinal illness that can be fatal to very young, old, or immunocompromised people. The lack of a vaccine and an increasing resistance to current antibiotics highlight a need to better understand the mechanisms that make C. jejuni a successful human pathogen. This study focused on the functional components of one such mechanism-a molec

Global analysis of the human pathophenotypic similarity gene network merges disease module components.

The molecular complexity of genetic diseases requires novel approaches to break it down into coherent biological modules. For this purpose, many disease network models have been created and analyzed. We highlight two of them, "the human diseases networks" (HDN) and "the orphan disease networks" (ODN). However, in these models, each single node represents one disease or an ambiguous group of diseases. In these cases, the notion of diseases as unique entities reduces the usefulness of network-based methods. We hypothesize that using the clinical features (pathophenotypes) to define pathophenotyp

Differential gene expression profiles of the mitochondrial respiratory components in illuminated Arabidopsis leaves.

Plant mitochondria have multiple energy-dissipating components in the respiratory chain. It is known that these components are induced under several stress conditions. Here we examined whether the gene expression pattern and its regulatory mechanism under high light (HL) conditions are different among the respiratory components in Arabidopsis leaves. Alternative oxidase (AOX) gene expression (AOX1a and AOX1c) and amount of protein were elevated after exposure to HL. In addition to AOX, the expression of other respiratory genes, including NDA1, NDB2, NDC1, UCP1, UCP5, COX6b and CI76, was also i

Rat arylalkylamine N-acetyltransferase gene: upstream and intronic components of a bipartite promoter.

The daily rhythm in the activity of arylalkylamine N-acetyltransferase (AA-NAT) controls the rhythm in melatonin synthesis in the pineal gland. In the rat, transcriptional regulatory mechanisms play a major role in determining the observed pattern of AA-NAT gene expression. Remarkably, high levels of AA-NAT transcripts can only be detected in the night pineal; significant levels can also be found in the retina. To characterize the regulatory events that impinge upon the activity of the AA-NAT gene we embarked on the systematic analysis of the AA-NAT promoter. To this end we transfected several

Genes representing the stress-dependent component in arterial hypertension development.

Hypertension is among the major risk factors of many cardiovascular diseases. Chronic psychoemotional stress is one of its key causes. Studies of molecular mechanisms of human hypertension development are conducted in animals, including artificial rat strains that model various forms of the disease. The RatDEGdb database, used in our work, includes 144 hypothalamic genes that represent the common response to single short-term restraint stress in hypertensive ISIAH and normotensive WAG rats. These rat genes were annotated with changes in the expression of the human orthologs using data on 17,45

Downregulation of Genes for Skeletal Muscle Extracellular Matrix Components by Cisplatin.

The extracellular matrix (ECM) in skeletal muscle is involved in a variety of physiological functions beyond the mechanical support of muscle tissue, nerves, and blood vessels; however, the role of the ECM in skeletal muscle remains unclear. There is little information regarding changes in the expression of factors comprising the ECM during cisplatin-induced muscle atrophy. In the present study, we examined the changes in gene expressions for skeletal muscle extracellular matrix components in skeletal muscle during cisplatin-induced muscle atrophy. Intraperitoneal administration of cisplatin c

Identification of novel components of the retinal determination gene network in Drosophila cell lines.

The retinal determination genetic network controls the development of the visual system in all seeing animals through the molecular regulation of cells to adopt an eye tissue fate. The compound eye of the fruit fly, Drosophila melanogaster, is an excellent model system to study the complex mechanisms within the network that regulate specification of cellular identity during embryogenesis. In Drosophila, the two Pax6 paralogues, eyeless and twin of eyeless, sit at the very pinnacle of the network and their expression early in development activates critical downstream components of the retinal d

Gene expression profiling in Entamoeba histolytica identifies key components in iron uptake and metabolism.

Entamoeba histolytica is an ameboid parasite that causes colonic dysentery and liver abscesses in humans. The parasite encounters dramatic changes in iron concentration during its invasion of the host, with relatively low levels in the intestinal lumen and then relatively high levels in the blood and liver. The liver notably contains sources of iron; therefore, the parasite's ability to use these sources might be relevant to its survival in the liver and thus the pathogenesis of liver abscesses. The objective of the present study was to identify factors involved in iron uptake, use and storage

Recurrence Score® gene group components and outcomes in the RxPONDER trial (SWOG S1007).

BACKGROUND: The RxPONDER trial has guided adjuvant chemotherapy use in node-positive HR+/HER2- breast cancer; however, prior analyses showed poorer outcomes among non-Hispanic Black (NHB) women compared with non-Hispanic White (NHW) women despite similar 21-gene Recurrence Score® (RS) results. This suggests contributors to disparities may not be captured by the composite RS. We evaluated RS gene group components-proliferation, estrogen receptor (ER), HER2 (GRB7), and invasion-by ethnicity, body mass index, and menopausal status, and assessed associations with outcomes. METHODS: RxPONDER enrol
