Essential genes encoded by the mating-type locus of the human fungal pathogen Cryptococcus neoformans.

Fungal sexual reproduction is controlled by the mating-type (MAT) locus. In contrast to a majority of species in the phylum Basidiomycota that have tetrapolar mating-type systems, the opportunistic human pathogen Cryptococcus neoformans employs a bipolar mating-type system, with two mating types (a and α) determined by a single MAT locus that is unusually large (~120 kb) and contains more than 20 genes. While several MAT genes are associated with mating and sexual development, others control conserved cellular processes (e.g., cargo transport and protein synthesis), of which five (MYO2, PRT1,

Sporothrix schenckii (sensu strict S. globosa) mating type 1-2 (MAT1-2) gene.

Sporotix schenckii is a pathogenic fungus that causes human and animal sporotrichosis, and based on morphology of the sessile conidia and molecular analysis, it was recently recognized as a species complex comprising at least the following six sibling species: S. albicans, S. brasiliensis, S. globosa, S. luriei, S. mexicana and S. schenckii. However, apart from S. schenckii sensu strict, only S. brasiliensis, S. globosa and S. luriei are associated with human and animal infection. S. globosa has been most commonly isolated in Asia, Europe and the USA; therefore, molecular epidemiological study

Phylogenomic insights into the taxonomy, ecology, and mating systems of the lorchel family Discinaceae (Pezizales, Ascomycota).

Lorchels, also known as false morels (Gyromitra sensu lato), are iconic due to their brain-shaped mushrooms and production of gyromitrin, a deadly mycotoxin. Molecular phylogenetic studies have hitherto failed to resolve deep-branching relationships in the lorchel family, Discinaceae, hampering our ability to settle longstanding taxonomic debates and to reconstruct the evolution of toxin production. We generated 75 draft genomes from cultures and ascomata (some collected as early as 1960), conducted phylogenomic analyses using 1542 single-copy orthologs to infer the early evolutionary history

Relief of bHD1 autoinhibition and modular coupling support MAT-A pathway activation in Flammulina filiformis.

Flammulina filiformis is a commercially important edible mushroom whose sexual compatibility is determined by genes at two unlinked mating-type loci, A and B. However, the molecular basis of MAT-A activation in this species remains unclear. Here, we established a pair of compatible monokaryotic strains, F11 and F22, derived from the dikaryotic parent ACCC50407, and used them to investigate the genomic and functional basis of MAT-A activation. Genome analysis showed that the MAT-A locus is bipartite, comprising the Aa and Ab subloci. Five HD genes were identified, including one Aa-encoded gene

Mechanism for Restoration of Fertility in Hybrid Zygosaccharomyces rouxii Generated by Interspecies Hybridization.

The mechanism of whole-genome duplication (WGD) in yeast has been intensively studied because it has a large impact on yeast evolution. WGD has shaped the genomic architecture of modern Saccharomyces cerevisiae; however, the mechanism for restoring fertility after interspecies hybridization, which would be involved in the process of WGD, has not been thoroughly elucidated. In this study, we obtained a draft genome sequence of the salt-tolerant yeast Zygosaccharomyces rouxii NBRC110957 and revealed that it is a hybrid lineage of Z. rouxii (allodiploid) with two subgenomes equivalent to NBRC1876

Distribution of incompatibility factors in natural populations of Hypsizygus marmoreus in Japan.

Hypsizygus marmoreus (bunashimeji in Japanese) is a major cultivated edible mushroom in Japan. Characterizing its mating incompatibility factors in natural populations is essential for improving breeding efficiency. In this study, 71 wild isolates of H. marmoreus collected from different regions in Japan were analyzed to determine the composition and distribution of incompatibility factors. Mating tests using tester monokaryotic strains identified 82 cases of monokaryons that were carrying different alleles at both the A and B mating locus, whereas another 60 factors were shared among isolates

Association between fertility and molecular sub-type of global isolates of Cryptococcus gattii molecular type VGII.

The basidiomycetous yeast Cryptococcus gattii, is a primary pathogen which causes disease in apparently healthy humans and a wide range of animals. Recently, an outbreak of cryptococcosis caused by a previously uncommon genotype of C. gattii, VGII, emerged on Vancouver Island, British Columbia, Canada. Two pathogenic sub-types of VGII (designated VGIIa and VGIIb) were identified among these isolates. All of the isolates proved to be mating type alpha and had exceptionally high sporulation capacity. The common subtype, VGIIa, was more virulent than VGIIb in mice, suggesting a linkage between su

Mating-type-specific and nonspecific PAK kinases play shared and divergent roles in Cryptococcus neoformans.

Cryptococcus neoformans is an opportunistic fungal pathogen with a defined sexual cycle involving fusion of haploid MATalpha and MATa cells. Virulence has been linked to the mating type, and MATalpha cells are more virulent than congenic MATa cells. To study the link between the mating type and virulence, we functionally analyzed three genes encoding homologs of the p21-activated protein kinase family: STE20alpha, STE20a, and PAK1. In contrast to the STE20 genes that were previously shown to be in the mating-type locus, the PAK1 gene is unlinked to the mating type. The STE20alpha, STE20a, and

Chromosome-scale genome assembly and characterization of Saccharomycopsis schoenii, a necrotrophic predatory yeast.

The necrotrophic predatory yeast Saccharomycopsis schoenii preys on diverse fungal species including multidrug-resistant Candida auris, making it a promising candidate for fungal biocontrol. Despite growing interest in its predation mechanisms, a high-quality reference genome is lacking. We constructed a chromosome-scale genome assembly of S. schoenii using a hybrid approach combining Pacific Biosciences HiFi long-read sequencing with Hi-C chromatin conformation capture. The 14.3 Mb genome is resolved into 6 nuclear and 1 mitochondrial chromosome, demonstrating high contiguity and near-complet

Regulatory basis for reproductive flexibility in a meningitis-causing fungal pathogen.

Pathogenic fungi of the genus Cryptococcus can undergo two sexual cycles, involving either bisexual diploidization (after fusion of haploid cells of different mating type) or unisexual diploidization (by autodiploidization of a single cell). Here, we construct a gene-deletion library for 111 transcription factor genes in Cryptococcus deneoformans, and explore the roles of these regulatory networks in the two reproductive modes. We show that transcription factors crucial for bisexual syngamy induce the expression of known mating determinants as well as other conserved genes of unknown function.

Comparative genomics suggests primary homothallism of Pneumocystis species.

UNLABELLED: Pneumocystis species are fungal parasites of mammal lungs showing host specificity. Pneumocystis jirovecii colonizes humans and causes severe pneumonia in immunosuppressed individuals. In the absence of in vitro cultures, the life cycle of these fungi remains poorly known. Sexual reproduction probably occurs, but the system of this process and the mating type (MAT) genes involved are not characterized. In the present study, we used comparative genomics to investigate the issue in P. jirovecii and Pneumocystis carinii, the species infecting rats, as well as in their relative Taphrin

Transcriptional divergence among different mating-type strains and their cross of Morchella sextelata during the asexual stage.

Fungi differ from other eukaryotes in being hermaphroditic, lacking sexual dimorphism and relying on mating-type (MAT) loci for sexual compatibility. Previous studies indicate that mating types of Morchella sextelata exhibit sex-like characteristics, with MAT1-1 tending to act maternally and being primarily responsible for stipe and infertile hymenial tissue formation. Given that conidia have been regarded as spermatia in ascomycetes and play a paternal role, this raises questions about whether MAT1-2 possesses stronger regulatory capacity during conidiation in Morchella. In this study, the ge

Serotype and mating type characterization of Cryptococcus neoformans by multiplex PCR.

Cryptococcus neoformans is an encapsulated yeast, etiological agent of cryptococcosis. The species is commonly associated with pigeon droppings and plant materials. The aim of the present work was to verify the presence of the yeast in pigeon droppings, and to identify the isolates obtained in serotypes and mating types (MAT). Ten samples of pigeon droppings were collected in the rural area of the city of Alfenas, Brazil. Samples were inoculated in agar Niger medium for fungal isolation and 22 isolates with characteristics of C. neoformans were obtained. The serotypes and MAT were determined b

Sitting on the fence: Individuals, distributions, and reproductive strategies in the bird's nest fungus Nidula candida.

Bird's nest fungi (Nidula candida) growing along ~18-year-old cedar fences in Pacific Spirit Regional Park, Vancouver, British Columbia, Canada, produce abundant, persistent peridia containing basidiospore-bearing, egg-like peridioles. To reconstruct the initial mode of reproduction of the populations, 37 peridia were collected following a geometric sampling design, with 8-10 peridia collected 6-222 cm apart from each of four fence segments. We cultured dikaryotic mycelium from one whole peridiole from each peridium and 105 monokaryons from basidiospores from 8 peridioles. We then tested mycel

Homeodomain protein Sxi1α independently controls cell-cell fusion and gene expression during sexual reproduction in Cryptococcus deneoformans.

Sex-specific homeodomain (HD) proteins are key regulators of cell identity and sexual development in fungi, typically functioning as heterodimers to govern transcription. In the human fungal pathogens Cryptococcus neoformans and Cryptococcus deneoformans, the HD proteins Sxi1α and Sxi2a (Sex-inducer 1α and 2a) have been characterized as interacting components that play critical roles in sexual development during α x a sexual reproduction. α cells are the predominant mating type in natural populations of Cryptococcus, and unisexual (same-sex) mating can also occur in certain genetic backgrounds

Transcriptional Heterogeneity of Cryptococcus gattii VGII Compared with Non-VGII Lineages Underpins Key Pathogenicity Pathways.

Cryptococcus gattii is a pathogenic yeast of humans and other animals which causes disease predominantly in immunocompetent hosts. Infection begins when aerosolized yeast or spores enter the body, triggering an immune response, including engulfment by macrophages. To understand the early transcriptional signals in both the yeast and its mammalian host, we performed a time-course dual-transcriptome sequencing (RNA-seq) experiment for four lineages of C. gattii (lineages VGI to IV) interacting with mouse macrophages at 1, 3, and 6 h postinfection. Comparisons of in vitro to ex vivo gene expressi

Genetic basis for coordination of meiosis and sexual structure maturation in Cryptococcus neoformans.

In the human fungal pathogen Cryptococcus neoformans, sex can benefit its pathogenicity through production of meiospores, which are believed to offer both physical and meiosis-created lineage advantages for its infections. Cryptococcus sporulation occurs following two parallel events, meiosis and differentiation of the basidium, the characteristic sexual structure of the basidiomycetes. However, the circuit integrating these events to ensure subsequent sporulation is unclear. Here, we show the spatiotemporal coordination of meiosis and basidial maturation by visualizing event-specific molecule

Virulence of "white-gray-opaque" tri-stable transformation in clinical Candida albicans in vitro and in vivo.

The study aimed to induce the white-opaque-gray tri-stable transformation in clinical C. albicans and to explore their potential pathogenicity. Sixty-four clinical strains were used to induce the white, opaque and gray cells of C. albicans. Secreted aspartyl proteinases (Sap) activity of the three phenotypes was then measured, and a vulvovaginal candidiasis (VVC) animal model was constructed. Of the 64 clinical strains, only 3 strains successfully underwent white-gray-opaque tri-stable transformation, and the three strains all belonged to MTL homozygous strains. A novel Cryphonectria species (Cryphonectriaceae, Diaporthales) discovered on Eucalyptus, including insights into the mating biology of the genus.

The Cryphonectriaceae (Diaporthales, Ascomycota) accommodates numerous important fungal pathogens that cause branch and stem canker diseases on woody plants worldwide. These include, for example, Cryphonectria (Cry.) parasitica, the causal agent of chestnut blight, which caused the near eradication of the American chestnut (Castanea dentata) in North America. The profound ecological and economic impact of Cry. parasitica has prompted considerable research interest in the diversity, biology, and pathogenicity of other species in the Cryphonectriaceae globally. An extremely poor nutritional condition enables efficient white cell mating in Candida albicans.

Pathogenic fungi employ diverse strategies to undergo sexual reproduction. It was previously believed that the major human fungal pathogen Candida albicans must switch from the "sterile" white state to the mating-competent opaque state to mate. We recently reported that glucose depletion could induce C. albicans white cell mating at a moderate frequency. In this study, we demonstrate that white cells can undergo efficient mating when incubated on agar medium (water and agar, without additional nutritional components). Cell survival assays revealed that a subpopulation of white cells underwent
