[Mice SI/Col: a study model in the research of genes involved in situs inversus].

The SI/Col mouse is carrier of the i.v. mutation which expresses itself in an autosomal recessive mode. Fifty per cent of mice born of homozygotic i.v./i.v. parents are normal and 50% present an abnormality of lateralisation of the thoracic and abdominal organs with cardiac malformations involving the venous, atrial, ventricular and arterial segments. Identifying the i.v. mutation may improve our understanding of the genesis of the heterotaxic syndrome in man. With the help of return crossing with wild mice, the authors studied segregation of 10 makers of the murine, chromosome 12 of the situs

Novel mutations in the human sucrase-isomaltase gene (SI) that cause congenital carbohydrate malabsorption.

Disaccharide intolerance I or congenital sucrase-isomaltase deficiency (CSID) is a disorder leading to maldigestion of disaccharides, which is autosomal recessively inherited. Here we analyzed the sucrase-isomaltase (SI) gene from 11 patients of Hungarian origin with congenital sucrase-isomaltase deficiency. Variants in the SI gene had previously been described in CSID patients, which cause amino acid exchanges that affect the transport, the processing, or the function of the SI protein. None of our patients had known mutations for CSID. Our analyses revealed 43 SI variants in total, 15 within

Chromosomal translocation t(1;22) and sis oncogene variant with gene amplification in a case of atypical malignant lymphoma.

sis Oncogene amplification and PstI restriction enzyme variant were found in a patient of atypical malignant lymphoma with chromosomal translocation t(1;22). A diagnosis of atypical malignant lymphoma was made because dual rearrangements of both beta chain of T-cell receptor gene and heavy and kappa chains of immunoglobulin genes were observed by DNA analysis extracted from the lymph node. Amplification of sis was estimated to be about fivefold by dot blot. BamHI and EcoRI digested DNA hybridized with sis probe from blood and lymph nodes revealed identical bands. Lymph node DNA digested with P

Exploring congenital sucrase-isomaltase deficiency in autism spectrum disorder patients with irritable bowel syndrome symptoms: A prospective SI gene sequencing study.

Congenital sucrase-isomaltase deficiency (CSID) is an inherited metabolic disorder causing chronic gastrointestinal symptoms and malnutrition when untreated. Most CSID patients are likely to remain under- or misdiagnosed. This study aimed to investigate prevalence of CSID among patients with autism spectrum disorder (ASD) presenting with irritable bowel syndrome (IBS) symptoms via prospective SI gene sequencing. A prospective cross-sectional study was conducted on 98 ASD patients exhibiting gastrointestinal symptoms consistent with IBS. Participants were assessed according to Rome IV criteria

Influences of Co-Exposure to Fatty Acids and SiO2 Nanoparticles on Gene Expression in Zebrafish Intestines and Livers and Si Accumulation and Lipid Staining in Caco-2 and HepG2 Cells.

The interaction between fatty acids and nanoparticles (NPs) is increasingly recognized. This study investigated how stearic acid (SA)/linolenic acid (LNA) and SiO2 NPs co-exposure affects mRNA abundance of efflux, autophagy, fatty acid metabolism, and synthesis genes in adult zebrafish, and Si concentrations and lipid accumulation in Caco-2 and HepG2 cells. In vivo, SA or LNA inhibited SiO2 NPs from inducing abcg1 expression, and SiO2 NPs and SA/LNA co-exposure decreased agap1 expression in intestines. In livers, SiO2 NPs and SA co-exposure increased abcg1 expression. SiO2 NPs increased autoph

Highly efficient, label free, ultrafast plasmonic SERS biosensor (silver nanoarrays/Si) to detect GJB2 gene expressed deafness mutations in real time validated with PCR studies.

Rapid diagnostics of any gene mutations related to organ loss is highly demanded now-a days to consume time as well to reduce cost. Currently, Surface enhanced Raman spectroscopy (SERS) is evolved to be a rapid investigating tool to screen gene mutations down to single molecule sensing with regard to the design and development of substrates used for sensing. The current research focuses on particular towards direct detection of deafness mutations associated with single and dual sites related to GJB2 gene. SERS Sensor construction is achieved with plasmonic silver nanoarrays on Si (SNA/Si) subs

A semi-automated genetic screen using genome-wide association analysis identifies modulators of stress-induced sleep in Caenorhabditis elegans.

Animals sleep more when they are sick. In C. elegans, stress-induced sleep (SIS) follows cellular injury such as exposure to ultraviolet (UV) light. The genetic regulators of SIS remain incompletely defined. Using robotic automation, high-throughput imaging, and genome-wide association analysis, we performed a semi-automated screen of 940 whole-genome sequenced Million Mutation Project (MMP) strains. We quantified behavioral activity and quiescence before and after ultraviolet (UV) radiation. We applied the Sequence Kernel Association Test (SKAT) to this behavioral data to prioritize a set of

Activation of a diverse set of genes during the tobacco resistance response to TMV is independent of salicylic acid; induction of a subset is also ethylene independent.

Through differential screening of a cDNA library, we cloned six groups of genes that are expressed relatively early in the inoculated leaves of tobacco resisting infection by tobacco mosaic virus (TMV). Induction of all these genes was subsequently detected in the uninoculated leaves; thus, their expression is associated with the development of both local and systemic acquired resistance. Exogenously applied salicylic acid (SA) was observed to induce these genes transiently. However, analyses with transgenic NahG plants, which are unable to accumulate SA, demonstrated that expression of these

Pig-derived ECM-SIS provides a novel matrix gel for tumor modeling.

The absence of effective extracellular matrix to mimic the natural tumor microenvironment remains a significant obstacle in cancer research. Matrigel, abundant in various biological matrix components, is limited in its application due to its high cost. This has prompted researchers to explore alternative matrix substitutes. Here, we have investigated the effects of the extracellular matrix derived from pig small intestinal submucosa (ECM-SIS) in xenograft tumor modeling. Our results showed that the pig-derived ECM-SIS effectively promotes the establishment of xenograft tumor models, with a tum

Transcriptomic analysis of chronic social instability stress during adolescence reveals major shifts in the extracellular matrix.

Adolescence represents a sensitive window of neural plasticity, when social experiences exert enduring influences on emotional and cognitive development. Adverse social conditions, such as social instability stress (SIS), disrupt peer relationships and lead to long-term changes in social and affective behaviour. Here, we used RNA sequencing to characterize the immediate (P46) and enduring (P70) molecular consequences of adolescent SIS in male and female rats in two stress-sensitive regions: the dorsal hippocampus (dHPC) and basolateral amygdala (BLA). SIS produced region- and sex-specific tran

Stress-Induced Sleep After Exposure to Ultraviolet Light Is Promoted by p53 in Caenorhabditis elegans.

Stress-induced sleep (SIS) in Caenorhabditis elegans is important for restoration of cellular homeostasis and is a useful model to study the function and regulation of sleep. SIS is triggered when epidermal growth factor (EGF) activates the ALA neuron, which then releases neuropeptides to promote sleep. To further understand this behavior, we established a new model of SIS using irradiation by ultraviolet C (UVC) light. While UVC irradiation requires ALA signaling and leads to a sleep state similar to that induced by heat and other stressors, it does not induce the proteostatic stress seen wit

Phenotypic and genetic overlap between autistic traits at the extremes of the general population.

OBJECTIVE: To investigate children selected from a community sample for showing extreme autistic-like traits and to assess the degree to which these individual traits--social impairments (SIs), communication impairments (CIs), and restricted repetitive behaviors and interests (RRBIs)--are caused by genes and environments, whether all of them are caused by the same genes and environments, and how often they occur together (as required by an autism diagnosis). METHOD: The most extreme-scoring 5% were selected from 3,419 8-year-old pairs in the Twins Early Development Study assessed on the Child

Angiogenesis-facilitating and inflammation-modulating SIS-based patches coupled with functional peptides for abdominal wall repair.

Abdominal wall defects caused by trauma, tumors, infections, abdominal surgery, and congenital factors can lead to functional impairments. The use of patches remains the most effective treatment approach. However, current repair materials still have limitations in regulating inflammation and promoting vascularization. Here, a small intestinal submucosa (SIS) extracellular patch was developed via conjugation with functional peptides PR1P and LL37 (i.e., SIS-PR1P-LL37), to achieve angiogenesis and inflammation modulation for abdominal wall repair. In vitro experiments confirmed its superior pro-

A female-specific lethal lesion in an X-linked positive regulator of the Drosophila sex determination gene, Sex-lethal.

Characterization of a partial-loss-of-function, female-specific lethal mutation has identified an X-linked genetic element (1-34.3; 10B4) that functions as a positive regulator of Sxl, a central gene controlling sex determination in Drosophila melanogaster. The name, sisterless-a, was chosen both to suggest functional similarities that exist between this gene and another positive regulator of Sxl, the maternally acting gene daughterless (da), and also to highlight an important difference; namely, that in contrast to da, it is the zygotic rather than maternal functioning of sis-a that is involv

The NALCN channel complex is necessary for stabilizing sleep in Caenorhabditis elegans.

Sleep is a fundamental physiological state that is widely conserved across diverse animal species. Caenorhabditis elegans exhibits developmentally timed sleep (DTS) and stress-induced sleep (SIS), which meet the behavioral definition of sleep and are regulated by molecular mechanisms shared with other animals. Here, we studied the sleep-regulating functions of UNC-80, which forms a complex with NCA-1 and NCA-2 (hereafter collectively referred to as NCA), and UNC-79. We found that the unc-80 loss-of-function (lf) mutants experienced lower quiescence and a more fragmented and longer DTS than the

Decellularized Matrix Scaffold Functionalized with GRGDS Peptide and Nanohydroxyapatite for Alveolar Bone Defect Repair.

Periodontitis is a common inflammatory disease of the oral cavity that often leads to alveolar bone loss and eventually causes tooth detachment, seriously damaging the physical and mental health of patients. This study developed an osteogenic scaffold (G-nHA/SIS) derived from porcine small-intestinal submucosa (SIS), functionalized with Gly-Arg-Gly-Asp-Ser (GRGDS) peptide and nanohydroxyapatite (nHA) to promote alveolar bone regeneration. The scaffold has a three-dimensional (3D) porous structure, similar to that of the natural extracellular matrix (ECM), providing physical support for cell in

Effects of fractionated x-irradiation on the Ly-6--Ril-1--Pol-5 region.

Our laboratory has focused on defining, localizing, and understanding the mode of action of genes involved in fractionated x-irradiation (FXI) leukemia in susceptible and restraint mouse strains. We have described the genetic and molecular evidence suggesting the existence of multiple independent loci involved in FXI-induced leukemogenesis. These studies indicated that one of these, Ril-1, a locus on the distal portion of chromosome 15, is the major locus influencing susceptibility to the disease. Our data unequivocally place Ril-1 in the gene complex Ly-6--Ril-1--Sis--H-30--Pol-5. Ril-1 appea

Ellagic acid through attenuation of neuro-inflammatory response exerted antidepressant-like effects in socially isolated mice.

Recent studies have been demonstrated that neuroinflammation plays a crucial role in the pathophysiology of depression. Therefore, anti-inflammatory medications could be regarded as a potentially effective treatments for depression. Ellagic acid (EA) is a natural polyphenol with antioxidant and anti-inflammatory properties. This study aimed to evaluate the antidepressant-like effect of EA in a mouse model of social isolation stress (SIS), considering its potential anti-neuroinflammatory properties. In this study, 48 male mice were divided into six groups (n = 8), including saline-treated contr

Modulation of c-myc, c-myb, c-fos, c-sis and c-fms proto-oncogene expression and of CSF-1 transcripts and protein by phorbol diester in human malignant histiocytosis DEL cell line with 5q 35 break point.

Following exposure to phorbol ester (TPA), DEL cell line, a human malignant histiocytosis (MH) cell line, is able to differentiate along a macrophage phenotype and thus it provides a suitable model for analyzing the sequential and differential gene expression associated with monocyte/macrophage differentiation. C-myc, c-myb, c-fos, c-sis and c-fms expression were determined by Northern analysis at various times following TPA treatment. The results showed that TPA down-modulated the constitutive expression of c-myc, c-myb, and c-fms, mRNA to low but still detectable levels. Conversely, TPA-indu

Architecture of the superintegron in Vibrio cholerae: identification of core and unique genes.

BACKGROUND: Vibrio cholerae, the etiologic agent of cholera, is indigenous to aquatic environments. The V. cholerae genome consists of two chromosomes; the smallest of these harbors a large gene capture and excision system called the superintegron (SI), of ~120 kbp. The flexible nature of the SI that results from gene cassette capture, deletion and rearrangement is thought to make it a hotspot of V. cholerae diversity, but beyond the basic structure it is not clear if there is a core genome in the SI and if so how it is structured. The aim of this study was to explore the core genome structure
