Biomineralization of Cu2S Nanoparticles by Geobacter sulfurreducens.

Biomineralization of Cu has been shown to control contaminant dynamics and transport in soils. However, very little is known about the role that subsurface microorganisms may play in the biogeochemical cycling of Cu. In this study, we investigate the bioreduction of Cu(II) by the subsurface metal-reducing bacterium Geobacter sulfurreducens Rapid removal of Cu from solution was observed in cell suspensions of G. sulfurreducens when Cu(II) was supplied, while transmission electron microscopy (TEM) analyses showed the formation of electron-dense nanoparticles associated with the cell surface. Lack of Specificity in Geobacter Periplasmic Electron Transfer.

Reduction of extracellular acceptors requires electron transfer across the periplasm. In Geobacter sulfurreducens, three separate cytoplasmic membrane cytochromes are utilized depending on redox potential, and at least five cytochrome conduits span the outer membrane. Because G. sulfurreducens produces 5 structurally similar triheme periplasmic cytochromes (PpcABCDE) that differ in expression level, midpoint potential, and heme biochemistry, many hypotheses propose distinct periplasmic carriers could be used for specific redox potentials, terminal acceptors, or growth conditions. Anaerobic Mercury Methylation and Demethylation by Geobacter bemidjiensis Bem.

Microbial methylation and demethylation are two competing processes controlling the net production and bioaccumulation of neurotoxic methylmercury (MeHg) in natural ecosystems. Although mercury (Hg) methylation by anaerobic microorganisms and demethylation by aerobic Hg-resistant bacteria have both been extensively studied, little attention has been given to MeHg degradation by anaerobic bacteria, particularly the iron-reducing bacterium Geobacter bemidjiensis Bem. Here we report, for the first time, that the strain G. bemidjiensis Bem can mediate a suite of Hg transformations, including Hg(II

Microbial Reduction of Antimony(V)-Bearing Ferrihydrite by Geobacter sulfurreducens.

The reduction of Sb(V)-bearing ferrihydrite by Geobacter sulfurreducens was studied to determine the fate of the metalloid in Fe-rich systems undergoing redox transformations. Sb(V) added at a range of concentrations adsorbed readily to ferrihydrite, and the loadings had a pronounced impact on the rate and extent of Fe(III) reduction and the products formed. Magnetite dominated at low (0.5 and 1 mol%) Sb(V) concentrations, with crystallite sizes decreasing at higher Sb loadings: 37-, 25-, and 17-nm particles for no-Sb, 0.5% Sb, and 1% Sb samples, respectively. In contrast, goethite was the dom

Direct Observation of Electrically Conductive Pili Emanating from Geobacter sulfurreducens.

Geobacter sulfurreducens is a model microbe for elucidating the mechanisms for extracellular electron transfer in several biogeochemical cycles, bioelectrochemical applications, and microbial metal corrosion. Multiple lines of evidence previously suggested that electrically conductive pili (e-pili) are an essential conduit for long-range extracellular electron transport in G. sulfurreducens. However, it has recently been reported that G. sulfurreducens does not express e-pili and that filaments comprised of multi-heme c-type cytochromes are responsible for long-range electron transport. Mercury reduction and cell-surface adsorption by Geobacter sulfurreducens PCA.

Both reduction and surface adsorption of mercuric mercury [Hg(II)] are found to occur simultaneously on G. sulfurreducens PCA cells under dark, anaerobic conditions. Reduction of Hg(II) to elemental Hg(0) initially follows a pseudo-first order kinetics with a half-life of <2 h in the presence of 50 nM Hg(II) and 10(11) cells L(-1) in a phosphate buffer (pH 7.4). Multiple gene deletions of the outer membrane cytochromes in this organism resulted in a decrease in reduction rate from ∼0.3 to 0.05 h(-1), and reduction was nearly absent with heat-killed cells or in the cell filtrate. Adsorption of

[Research on mercury methylation by Geobacter sulfurreducens and its influencing factors].

Mercury methylation by Geobacter sulfurreducens and the effects of environmental factors were studied under laboratory conditions. The results showed that G. sulfurreducens could grow well in the presence of low concentrations of mercuric chloride, but its growth was inhibited to a certain extent, mainly expressed in the prolonged lag phase. G. sulfurreducens could transform inorganic mercury into methylmercury, and this process was affected by many environmental factors. The efficiency of mercury methylation reached 38% under anaerobic conditions with 1 mg x L(-1) HgCl2 and 0.9% salinity at 3

Quinolone antibiotics stimulate bacterial mercury methylation by Geobacter metallireducens GS-15.

Bacterial mercury (Hg) methylation is critical for bioremediating Hg pollution, but the impact of emerging antibiotics on this process has rarely been reported. This study innovatively investigated the interactions between Hg-methylating bacteria of Geobacter metallireducens GS-15 and two quinolone antibiotics: lomefloxacin (LOM) and ciprofloxacin (CIP) at 5 μg/L. Short-term LOM exposure increased methylmercury (MeHg) yield by 36 % compared to antibiotic-free conditions, caused by hormesis to alter bioactivities of single GS-15 cells. Long-term CIP exposure led to more antibiotic resistance an

FliW regulates biofilm formation in Geobacter sulfurreducens through interaction with CsrA.

CsrA is a post-transcriptional regulator that controls a wide range of bacterial phenotypes, including carbon metabolism, motility, quorum sensing, virulence, and biofilm formation. In Geobacter sulfurreducens, CsrA modulates both biofilm development and extracellular electron transfer in microbial fuel cells. In this study, we further investigated the regulatory mechanism of CsrA and its role in the formation of electroconductive biofilms in G. sulfurreducens. Bioelectrochemical analyses revealed that a ΔcsrA strain produces biofilms with enhanced electroconductivity compared with the wild-ty

Facet Dependence of Biosynthesis of Vivianite from Iron Oxides by Geobacter sulfurreducens.

Vivianite plays an important role in alleviating the phosphorus crisis and phosphorus pollution. The dissimilatory iron reduction has been found to trigger the biosynthesis of vivianite in soil environments, but the mechanism behind this remains largely unexplored. Herein, by regulating the crystal surfaces of iron oxides, we explored the influence of different crystal surface structures on the synthesis of vivianite driven by microbial dissimilatory iron reduction. The results showed that different crystal faces significantly affect the reduction and dissolution of iron oxides by microorganis

Inhibitory effects of Skeletonema costatum on mercury methylation by Geobacter sulfurreducens PCA.

Algae and mercury (Hg) are ubiquitous in marine environments. In this study, we investigated the effects of a typical marine algae of diatom Skeletonema costatum on Hg methylation by an iron-reducing bacterium of Geobacter sulfurreducens (G. sulfurreducens) PCA. In the absence of Skeletonema costatum, the bacterial MeHg production rate maximized at 104.06 ± 11.7 ng L-1 h-1 with a high Hg level, while the highest methylation efficiency was achieved at a low Hg concentration. The existence of Skeletonema costatum greatly inhibited the capability of G. sulfurreducens PCA to methylate Hg. With the

Geobacter-based syntrophy drives biogenic element cycling in anoxic soils: a review.

Microbial syntrophy is a central organizing principle in anoxic soils, where the degradation of organic matter and the cycling of nitrogen, sulfur, and iron are constrained by electron availability and thermodynamics. Among soil anaerobes, Geobacter species occupy a distinctive ecological anoxic niche in which they form syntrophy relationship with partner cells to exchange electrons for anaerobic carbon mineralization, methane production, nitrate reduction, sulfur and Fe(III) reduction. Here, we review how Geobacter-based syntrophy contributes to C/N/S/Fe cycling in anoxic soils. We first summ

Mercury methylation by Geobacter metallireducens GS-15 in the presence of Skeletonema costatum.

In this study, bacterial mercury (Hg) methylation was investigated under the influence of red-tide algae of Skeletonema costatum (S. costatum). The distribution and speciation of total mercury (THg) and methylmercury (MeHg) were profiled by employing Geobacter metallireducens (G. metallireducens GS-15) as the methylating bacteria. G. metallireducens GS-15 showed different capabilities in methylating different inorganic forms of Hg(II) (HgCl2) and Hg(II)-Algae (HgCl2 captured by S. costatum) to MeHg. In the absence of S. costatum, a maximum methylation efficiency of 4.31 ± 0.47% was achieved wi

Genomic determinants of organohalide-respiration in Geobacter lovleyi, an unusual member of the Geobacteraceae.

BACKGROUND: Geobacter lovleyi is a unique member of the Geobacteraceae because strains of this species share the ability to couple tetrachloroethene (PCE) reductive dechlorination to cis-1,2-dichloroethene (cis-DCE) with energy conservation and growth (i.e., organohalide respiration). Strain SZ also reduces U(VI) to U(IV) and contributes to uranium immobilization, making G. lovleyi relevant for bioremediation at sites impacted with chlorinated ethenes and radionuclides. G. lovleyi is the only fully sequenced representative of this distinct Geobacter clade, and comparative genome analyses ident

Extracellular electron transfer proteins contribute to reduction of ferric minerals by Geobacter biofilms.

To investigate how the thickness and extracellular electron transfer (EET) capabilities of microbial biofilms influence the reduction of ferric iron [Fe(III)]-containing minerals, we utilized four strains of Geobacter sulfurreducens with varying biofilm thicknesses and EET capabilities. These strains were engineered by modulating intracellular levels of dinucleotide second messengers. We systematically investigated the capacity of biofilms formed by four strains to reduce different Fe(III)-containing minerals including ferrihydrite, goethite, and lepidocrocite. By growing the G. sulfurreducens

Characterization of CbcS from Geobacter sulfurreducens' Cbc4 complex: a putative novel respiratory pathway.

Electroactive bacteria mediate electron exchange with external compounds through a process known as extracellular electron transfer (EET). A key step in EET is the transfer of electrons from the menaquinone pool to inner membrane-associated quinol-cytochrome c oxidoreductase complexes, which subsequently relay electrons to periplasmic redox partners. Gene-knockout and proteomic analyses have identified several critical components involved in EET in Geobacter sulfurreducens, including six inner membrane oxidoreductase gene clusters. Of these, three - CbcL, ImcH, and CbcBA - have been linked to

Phenol degradation in the strictly anaerobic iron-reducing bacterium Geobacter metallireducens GS-15.

Information on anaerobic phenol metabolism by physiological groups of bacteria other than nitrate reducers is scarce. We investigated phenol degradation in the strictly anaerobic iron-reducing deltaproteobacterium Geobacter metallireducens GS-15 using metabolite, transcriptome, proteome, and enzyme analyses. The results showed that the initial steps of phenol degradation are accomplished by phenylphosphate synthase (encoded by pps genes) and phenylphosphate carboxylase (encoded by ppc genes) as known from Thauera aromatica, but they also revealed some distinct differences. The pps-ppc gene clu

Geobacter sulfurreducens' Unique Metabolism Results in Cells with a High Iron and Lipid Content.

Geobacter sulfurreducens is a ubiquitous iron-reducing bacterium in soils, and in engineered systems, it can respire an electrode to produce measurable electric current. Its unique metabolism, heavily dependent on an extensive network of cytochromes, requires a unique cell composition. In this work, we used metallomics, cell fraction and elemental analyses, and transcriptomics to study and analyze the cell composition of G. sulfurreducens. Elemental composition studies (C, H, O, N, and ash content) showed high C:O and H:O ratios of approximately 1.7:1 and 0.25:1, indicative of more reduced cel

Graphite electrode as a sole electron donor for reductive dechlorination of tetrachlorethene by Geobacter lovleyi.

The possibility that graphite electrodes can serve as the direct electron donor for microbially catalyzed reductive dechlorination was investigated with Geobacter lovleyi. In an initial evaluation of whether G. lovleyi could interact electronically with graphite electrodes, cells were provided with acetate as the electron donor and an electrode as the sole electron acceptor. Current was produced at levels that were ca. 10-fold lower than those previously reported for Geobacter sulfurreducens under similar conditions, and G. lovleyi anode biofilms were correspondingly thinner.
