Germinal Center Reaction.

Germinal centers (GCs) are transient microstructures formed within the follicles of secondary lymphoid tissues in response to certain types of immunization and foreign pathogens. A mature GC comprises two functionally distinct compartments, a dark zone (DZ) and a light zone (LZ). DZ B cells undergo rapid clonal expansion during which their antibody genes are modified by activation-induced cytidine deaminase (AID)-mediated immunoglobulin variable region (IgV) gene hypermutation to generate a repertoire of antibody mutants with varying affinities to the immunizing antigen. With the help of other

The biology of the germinal center.

The immune system requires the production of high affinity antibodies of different subclasses to accomplish its many effector functions. Specific steps in B-cell ontogeny that occur within germinal centers of secondary lymphoid organs create much of the diversity in the immune system. This process also provides the raw material for the genesis of B-cell lymphomas as misdirection of the molecular machinery that regulate these steps can cause chromosomal translocations, prevent apoptosis and promote proliferation of abnormal clones. Many recent avenues of investigation have elucidated that the g

Comparison of germinal center markers CD10, BCL6 and human germinal center-associated lymphoma (HGAL) in follicular lymphomas.

BACKGROUND: Recently, human germinal center-associated lymphoma (HGAL) gene protein has been proposed as an adjunctive follicular marker to CD10 and BCL6. METHODS: Our aim was to evaluate immunoreactivity for HGAL in 82 cases of follicular lymphomas (FLs)--67 nodal, 5 cutaneous and 10 transformed--which were all analysed histologically, by immunohistochemistry and PCR. RESULTS: Immunostaining for HGAL was more frequently positive (97.6%) than that for BCL6 (92.7%) and CD10 (90.2%) in FLs; the cases negative for bcl6 and/or for CD10 were all positive for HGAL, whereas the two cases negative f

Monocytes asphyxiate germinal centers.

Some bacteria and parasites, such as Salmonella, actively disrupt germinal centers and elicit only low affinity antibodies, but the mechanisms by which microbes alter these responses is poorly understood. In this issue of Immunity, Biram et al. (2022) uncover a mechanism by which Salmonella recruits Sca-1+ monocytes to germinal centers and impairs metabolic adaptation.

The life and death of the germinal center.

The formation, development and dissolution of germinal centers is a major part of immune system function. It is important to differentiate neoplastic processes from follicular hyperplasia and regressive follicular changes. Better understanding of germinal center development and dissolution also provides diagnostic clues to the underlying pathologic process. It is also important in identifying the immune basis of different pathologic entities as well as in immunotherapy decision making and follow up. In this study, we characterize the immunoarchitecture of lymphoid follicles with a focus on ger

Synaptic Interactions in Germinal Centers.

The germinal center (GC) is a complex, highly dynamic microanatomical niche that allows the generation of high-affinity antibody-producing plasma cells and memory B cells. These cells constitute the basis of long-lived highly protective antibody responses. For affinity maturation to occur, B cells undergo multiple rounds of proliferation and mutation of the genes that encode the immunoglobulin V region followed by selection by specialized T cells called follicular helper T (TFH) cells. In order to achieve this result, the GC requires spatially and temporally coordinated interactions between th

[The structure of the germinal centers of the thymus].

A morphological investigation of the thymus was performed in 82 patients dead from rheumatism. The germinal centers of the thymus localized in the cortical layer and medullary substance were found in 42% of observations. These centers were not homogeneous in their structure. Some of them represent germinal centers of lymphoid nodules, others have epithelioreticulocytes. The latters having a substantial difference from the centers of the lymphoid nodules should be referred to as thymic islands. Necrosis of their cells appear in these islands, its spreading being responsible for transformation o

CD28 is required for germinal center formation.

Previous studies have demonstrated that the T cell costimulatory molecule, CD28, is important in the development of humoral immunity. CD28-deficient mice exhibit defects in isotype switching and are more susceptible to pathogens that depend on an effective Ab response. To determine the basis of these defects, we have examined B cell responses of CD28-deficient mice at the microenvironmental level. Early in a normal T-dependent immune response, small numbers of B cells undergo activation in the T cell-rich zone of secondary lymphoid tissues and then migrate to B cell areas. These migrant B cell

Germinal Center Selection and Plasma Cell Differentiation.

Germinal centers (GCs) are specialized microenvironments in which B cells undergo affinity maturation through iterative cycles of somatic hypermutation, proliferation and selection, generating antibodies with improved antigen-binding properties. Although GC B cells do not themselves secrete antibody, they differentiate into memory B cells and plasma cells that provide long-term humoral immunity. GC selection has traditionally been viewed as a process that preferentially expands B cells expressing the highest-affinity B cell receptors. However, recent studies have revealed that physiological GC

Centrocytoid plasma cells of the germinal center.

Germinal centers within the lymph node follicles are T-cell-dependent, antigen-driven B-cell proliferations that develop from the rapid clonal expansion of a few founder cells. The end results of this B-cell expansion are memory B cells or plasma cells. Two morphologic forms of plasma cell can be recognized in the germinal center: classic plasma cells, characterized morphologically by peripherally clumped arrangement of nuclear chromatin, and cells with a nuclear morphology more resembling that of the centrocytes, which the authors have termed "centrocytoid plasma cells." In this study the aut

Tracking CD40 signaling during germinal center development.

Substantial evidence indicates that signaling through the CD40 receptor (CD40) is required for germinal center (GC) and memory B-cell formation. However, it is not fully understood at which stages of B-cell development the CD40 pathway is activated in vivo. To address this question, we induced CD40 signaling in human transformed GC B cells in vitro and identified a CD40 gene expression signature by DNA microarray analysis. This signature was then investigated in the gene expression profiles of normal B cells and found in pre- and post-GC B cells (naive and memory) but, surprisingly, not in GC

Expression of the human germinal center-associated lymphoma (HGAL) protein, a new marker of germinal center B-cell derivation.

We identified the human germinal center-associated lymphoma (HGAL) in gene-expression profiling studies of diffuse large B-cell lymphoma (DLBCL). The expression of HGAL correlated with survival in patients with DLBCL. The HGAL gene is the human homolog of M17, a mouse gene expressed specifically in normal germinal center (GC) B cells. We generated a monoclonal antibody against the HGAL protein and show that HGAL is expressed in the cytoplasm of GC lymphocytes and in lymphomas of GC derivation. Among 727 lymphomas tested by immunohistochemistry on tissue microarrays, HGAL staining was found in

Diffuse large B-cell lymphoma with composite germinal center and non-germinal center types: A report of two cases.

We report two cases of diffuse large B-cell lymphoma (DLBCL) with composite germinal center B-cell (GCB) and non-GCB types. Case 1 was a 72-year-old woman with inguinal lymph node swelling. Two morphologically different lesions were concurrently observed in needle biopsy specimens. One lesion was DLBCL with centroblastic morphology and a GCB phenotype (CD10+, BCL6+, and MUM1-), according to the Hans algorithm. The other lesion was DLBCL with anaplastic morphology and a non-GCB phenotype (CD10-, BCL6+, and MUM1+). Considering cellular atypia, the GCB-type DLBCL likely progressed to non-GCB-type

HGAL protein expression persists in disorders of germinal center dissolution: potential role of HGAL in the germinal center microenvironment.

Human Germinal Center-associated Lymphoma (HGAL) is a germinal center (GC) B-cell marker associated with a favorable outcome in diffuse large B-cell and classic Hodgkin lymphomas (CHL). To test its potential role in GC function, 75 cases involving GC disruption including 23 progressive transformation of germinal centers (PTGC), 25 follicle lysis and 27 Castleman disease (CD) were studied. HGAL protein expression uniformly correlated with GC B-cells in all except a subset of hyaline-vascular CD that showed severe regression of GCs. HGAL staining highlighted dismantled GCs in PTGC, in contrast t

Highlights of 2025: advances in germinal centers.

The germinal center (GC) is the engine room of the humoral immune response, driving the evolution of B cells into memory and long-lived plasma cells that can protect against (re)infection. Understanding GC biology remains a key research focus for promoting lifelong health. In this annual update, we highlight several influential studies on GC biology published in 2025 (Fig. 1). Space constraints prevent us from discussing in depth all published work.

The sweet truth about germinal centers.

Glycan-specific antibodies are classically associated with limited durability. In a recent Immunity article, Fryer et al. showed that repeated Streptococcus pyogenes exposure elicits glycan-specific germinal center responses in tonsils despite reduced T cell help, revealing key limitations of anti-glycan immunity and opportunities to improve vaccine strategies against bacteria.

Follicular dendritic cells and germinal centers.

Follicular dendritic cells (FDCs) are stromal cells unique to primary and secondary lymphoid follicles. Recirculating resting B cells migrate through the FDC networks, whereas antigen-activated B cells undergo clonal expansion within the FDC networks in a T cell-dependent fashion, thereby generating germinal centers. Here, B cells undergo somatic mutation, positive and negative selection, isotype switching and differentiation into high-affinity plasma cells and memory B cells. Since the discovery of FDCs by electron microscopy as long-term antigen-retaining cells 30 years ago isolation of FDCs

Linking signaling and selection in the germinal center.

Germinal centers (GC) are sites of rapid B-cell proliferation in response to certain types of immunization. They arise in about 1 week and can persist for several months. In GCs, B cells differentiate in a unique way and begin to undergo somatic mutation of the Ig V regions at a high rate. GC B cells (GCBC) thus undergo clonal diversification that can affect the affinity of the newly mutant B-cell receptor (BCR) for its driving antigen. Through processes that are still poorly understood, GCBC with higher affinity are selectively expanded while those with mutations that inactivate the BCR are l

A microRNA involved in the germinal center reaction.

In this issue of Blood, de Yébenes et al identify miR-217 as a promoter of the germinal center reaction, partly by suppressing the DNA damage response. Overactivity of miR-217 is hence potentially dangerous, and, indeed, its uncontrolled expression causes lymphomas.

TLR7 influences germinal center selection in murine SLE.

TLR7 enhances germinal center maturation and migration of B cells to the dark zone where proliferation and somatic hypermutation occur. Our goal was to determine how Tlr7 dose influences selection of the autoreactive B cell repertoire in NZW/BXSB. Yaa mice bearing the site-directed heavy chain transgene 3H9 that encodes for the TLR7 regulated anti-CL response. To create a physiologic setting in which autoreactive B cells compete for survival with non-autoreactive B cells, we generated bone marrow chimeras in which disease onset occurred with similar kinetics and the transferred 3H9+ female non
