Identification of ZmGLN5, a candidate gene for resistance to Gibberella ear rot in maize.

BACKGROUND: Gibberella ear rot (GER), caused by the fungal pathogen Fusarium graminearum (F. graminearum), severely affects maize yield and grain quality worldwide. Although numerous genetic loci associated with maize resistance to GER have been identified, the functional genes underlying resistance to GER remain largely uncharacterized. METHODS AND RESULTS: In this study, we performed comparative transcriptome analysis using two maize lines with contrasting resistance to GER. Differently expressed genes were mainly involved in defense-related pathway, including phenylpropanoid biosynthesis,

[Induction and genetic analysis of laboratory mutants of Gibberella zeae resistance to carbendazim].

The mutants of wild-type Gibberella zeae resistance to carbendazim were generated in laboratory by ultra-violet (UV) irradiating and fungicide taming. Two levels of resistance (low, LR; high, HR) were identified among these mutants. LR mutants could grow at the critical concentration of 1.4 +/- microg/mL of carbendazim while completely inhibited above 10 microg/ mL concentration, but this phenotype of isolates was not detected in the field. HR mutants could grow at 100 microg/mL and showed negative cross-resistance with N-phenylcarbamate fungicides, such as N-( 3,5-dichlorophenyl) carbamate (M

Insights into the genetic and biochemical basis of Gibberella ear rot resistance in maize.

Fusarium graminearum colonizes the maize ear, causing Gibberella ear rot (GER) and producing harmful mycotoxins, including deoxynivalenol (DON) and zearalenone (ZEA). The disease can be managed in part by breeding and planting resistant maize cultivars. Resistance to GER is a quantitative and complex trait. Evaluation of diverse germplasm to identify regions and candidate genes associated with resistance may be useful for crop improvement efforts. Screening for GER is time-consuming and costly. Thus, identifying other traits that may serve as a proxy for GER resistance may accelerate resistanc

Integrated analysis of transcriptomics and defense-related phytohormones to discover hub genes conferring maize Gibberella ear rot caused by Fusarium Graminearum.

BACKGROUND: Gibberella ear rot (GER) is one of the most devastating diseases in maize growing areas, which directly reduces grain yield and quality. However, the underlying defense response of maize to pathogens infection is largely unknown. RESULTS: To gain a comprehensive understanding of the defense response in GER resistance, two contrasting inbred lines 'Nov-82' and 'H10' were used to explore transcriptomic profiles and defense-related phytohormonal alterations during Fusarium graminearum infection. Transcriptomic analysis revealed 4,417 and 4,313 differentially expressed genes (DEGs) fr

Stereoselective 15α-hydroxylation of androsta-1,4-diene-3,17-dione by Gibberella sp. for efficient production of 15α-OH-ADD and 15α-OH-AD.

Androsta-1,4-diene-3,17-dione (ADD) and its hydroxylated derivatives are important steroidal drug intermediates, playing a crucial role in the pharmaceutical industry. This study evaluated the whole-cell biotransformation of ADD by Gibberella sp. CICC 2498 and Absidia sp. CICC 41050. While Absidia sp. showed low efficiency, Gibberella sp. selectively produced two major metabolites, identified as 15α-OH-ADD and 15α-OH-AD via HPLC, MS, and NMR. Through orthogonal design and single-factor experiments, the transformation rate of ADD to 15α-OH-ADD was enhanced from 30.35% to 61.74% under optimal co

Fungal biotransformation of chlorogenic and caffeic acids by Fusarium graminearum: New insights in the contribution of phenolic acids to resistance to deoxynivalenol accumulation in cereals.

Fusarium Head Blight and Gibberella Ear Rot, mainly caused by the fungi Fusarium graminearum and Fusarium culmorum, are two of the most devastating diseases of small-grain cereals and maize. In addition to yield loss, these diseases frequently result in contamination of kernels with toxic type B trichothecenes. The potential involvement of chlorogenic acid in cereal resistance to Fusarium Head Blight and Gibberella Ear Rot and to trichothecene accumulation was the focus of this study. The effects of chlorogenic acid and one of its hydrolyzed products, caffeic acid, on fungal growth and type B

Characterization and complete genome analysis of the surfactin-producing, plant-protecting bacterium Bacillus velezensis 9D-6.

BACKGROUND: Bacillus velezensis is an endospore-forming, free-living soil bacterium with potential as a biopesticide against a broad spectrum of microbial pathogens of plants. Its potential for commercial development is enhanced by rapid replication and resistance to adverse environmental conditions, typical of Bacillus species. However, the use of beneficial microbes against phytopathogens has not gained dominance due to limitations that may be overcome with new biopesticidal strains and/or new biological knowledge. RESULTS: Here, we isolated B. velezensis strain 9D-6 and showed that it inhi

NMR solution structure of a cyanovirin homolog from wheat head blight fungus.

Members of the cyanovirin-N homolog (CVNH) lectin family are found in bacteria, fungi and plants. As part of our ongoing work on CVNH structure-function studies, we determined the high-resolution NMR solution structure of the homolog from the wheat head blight disease causing ascomycetous fungus Gibberella zeae (or Fusarium graminearum), hereafter called GzCVNH. Like cyanovirin-N (CV-N), GzCVNH comprises two tandem sequence repeats and the protein sequence exhibits 30% identity with CV-N. The overall structure is similar to those of other members of the CVNH family, with the conserved pseudo-s

Effect of Hydroxyl or Methoxy Group in B-Ring of Flavonols on Structural Change and Antiseptic Ability of Soy Protein Isolate.

Soy protein isolate (SPI) enhances the functionality of flavonols, with their interaction and bioactivity critically dependent on B-ring substitution. Multispectral analysis and molecular docking were used to investigate the interaction between SPI and flavonols (Kaempferide (Kaem), Galangin (Gal), Kaempferol (Kae), Myricetin (Myr)) and their resulting antiseptic/antifungal performance. The results indicated that hydroxylation on the B-ring of flavonols significantly enhanced their binding affinity to SPI through static quenching, primarily driven by hydrogen bonds and van der Waals forces, wh
