Glucagon-like peptide-1, a matter of taste?

Understanding of gustatory coding helps to predict, and perhaps even modulate the ingestive decision circuitry, especially when eating behaviour becomes dysfunctional. Preclinical research demonstrated that glucagon like peptide 1 (GLP-1) is locally synthesized in taste bud cells in the tongue and that GLP-1 receptor exists on the gustatory nerves in close proximity to GLP-1 containing taste bud cells. In humans, the tongue has not yet been addressed as clinically relevant target for GLP-1 based therapies. The primary aim of the current review was to elaborate on the role of GLP- 1 in mammalia

Glucagon-Like Peptide-1 Receptor Agonist-Associated Hepatotoxicity.

PURPOSE: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have transformed the management of obesity, type 2 diabetes and metabolic dysfunction-associated steatohepatitis. Despite their numerous clinical benefits they are not without risk. Although the product labels contain multiple organ-specific warnings and precautions, the liver is not identified as a target organ of safety concern. Drug-induced liver injury (DILI) is a leading safety-related cause of pipeline attrition, boxed warnings and postmarketing drug withdrawal. This review explores GLP-1RA associated hepatotoxicity. METHODS:

Glucagon-like peptide 1 and cardiac cell survival.

During myocardial infarction (MI), a variety of mechanisms contribute to activation of cell death processes in cardiomyocytes, which determines the final MI size, subsequent mortality, and post-MI remodeling. The deleterious mechanisms activated during the ischemia and reperfusion phases in MI include oxygen deprival, decreased availability of nutrients and survival factors, accumulation of waste products, generation of oxygen free radicals, calcium overload, neutrophil infiltration in the ischemic area, depletion of energy stores, and opening of the mitochondrial permeability transition pore,

Glucagon-like peptide-1 secretion in bulimia nervosa.

Blood concentrations of the satiety-inducing glucagon-like peptide-1 (GLP-1) were compared in 20 bulimic patients and 20 healthy controls to examine whether secretory impairment of the peptide could be involved in bulimia nervosa (BN). Basal GLP-1 concentrations were measured by means of an enzyme-linked immunosorbent assay (ELISA) in blood samples taken four times over a 12-h period (08.00h, 12.00h, 16.00h, 20.00h) and seven times over a 3-h period following administration of a test meal. Eating-related and non-eating related patients' psychopathological aspects were measured by the use of a

Glucagon-like peptide-2 inhibits antral emptying in man, but is not as potent as glucagon-like peptide-1.

BACKGROUND: GLP-1 (glucagon-like peptide-1) and GLP-2 (glucagon-like peptide-2) are released in equimolar amounts in response to meal ingestion. GLP-1 inhibits gastric emptying and reduces postprandial gastric and exocrine pancreatic secretion and may play a physiological regulatory role in controlling appetite and energy intake in humans. The role of GLP-2 is more uncertain. Based on the results of animal studies, it has been suggested that GLP-2 may induce intestinal epithelial growth and inhibit gastric motility. The aim of this study was to determine to what extent GLP-2 alone or together

Glucagon-like peptide 1(GLP-1) in biology and pathology.

Post-translational proteolytic processing of the preproglucagon gene in the gut results in the formation of glucagon-like peptide 1 (GLP-1). Owing to its glucose-dependent insulinotropic effect, this hormone was postulated to primarily act as an incretin, i.e. to augment insulin secretion after oral glucose or meal ingestion. In addition, GLP-1 decelerates gastric emptying and suppresses glucagon secretion. Under physiological conditions, GLP-1 acts as a part of the 'ileal brake', meaning that is slows the transition of nutrients into the distal gut. Animal studies suggest a role for GLP-1 in

A Review of Glucagon-like Peptide-1 in Dermatology.

OBJECTIVE: Glucagon-like peptide-1 (GLP-1) is a hormone produced in response to meal intake by endocrine intestinal cells. GLP-1 binds to its receptors which are expressed on various cells throughout the body. GLP-1 receptors (GLP-1R) have become a target for the treatment of diabetes mellitus and weight loss, and GLP-1 receptor agonist (GLP-1RA) use has become more common among patients. In addition to the anti-hyperglycemic effects, recent studies have exhibited anti-inflammatory effects of GLP-1RAs. Current research surrounding GLP-1Rs and GLP-1R agonism in dermatology is limited. This revi

Association of glucagon-like peptide-1 receptor-targeted imaging probe with in vivo glucagon-like peptide-1 receptor agonist glucose-lowering effects.

AIMS/INTRODUCTION: Glucagon-like peptide-1 receptor agonists (GLP-1RA) are used for treatment of type 2 diabetes mellitus worldwide. However, some patients do not respond well to the therapy, and caution must be taken for certain patients, including those with reduced insulin secretory capacity. Thus, it is clinically important to predict the efficacy of GLP-1RA therapy. GLP-1R-targeted imaging has recently emerged to visualize and quantify β-cells. We investigated whether GLP-1R-targeted imaging can predict the efficacy of GLP-1RA treatment. MATERIALS AND METHODS: We developed 111 Indium-lab

Glucagon-Like Peptide-1 Receptor Agonists and Chronic Cough.

IMPORTANCE: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have become substantially more popular as a medication to treat obesity and type 2 diabetes (T2D). Despite their known association with gastroesophageal reflux disease and vagal nerve stimulation, the association between GLP-1RAs and chronic cough has not been previously studied. OBJECTIVE: To assess the clinical association between GLP-1RAs and chronic cough. DESIGN, SETTING, AND PARTICIPANTS: This large, multicenter cohort study used clinical records from a US-based collection of electronic medical record data from April 28,

Glucagon-like peptide-1 analogs: Miracle drugs are blooming?

Glucagon-like peptide-1 (GLP-1), secreted by L cells in the small intestine, assumes a central role in managing type 2 diabetes mellitus (T2DM) and obesity. Its influence on insulin secretion and gastric emptying positions it as a therapeutic linchpin. However, the limited applicability of native GLP-1 stems from its short half-life, primarily due to glomerular filtration and the inactivating effect of dipeptidyl peptidase-IV (DPP-IV). To address this, various structural modification strategies have been developed to extend GLP-1's half-life. Despite the commendable efficacy displayed by curre

Glucagon-Like Peptide-1: New Regulator in Lipid Metabolism.

Glucagon-like peptide-1 (GLP-1) is a 30-amino acid peptide hormone that is mainly expressed in the intestine and hypothalamus. In recent years, basic and clinical studies have shown that GLP-1 is closely related to lipid metabolism, and it can participate in lipid metabolism by inhibiting fat synthesis, promoting fat differentiation, enhancing cholesterol metabolism, and promoting adipose browning. GLP-1 plays a key role in the occurrence and development of metabolic diseases such as obesity, nonalcoholic fatty liver disease, and atherosclerosis by regulating lipid metabolism. It is expected t

Creatine for glucagon-like peptide-1-associated muscle loss.

Glucagon-like peptide-1 (GLP-1)-based therapies have transformed obesity treatment, yet reductions in lean tissue may accompany pharmacological weight loss. We hypothesize that rapid weight loss and associated reductions in energy and protein intake, mechanical loading, and potentially physical activity may create conditions of increased skeletal-muscle bioenergetic vulnerability. Creatine supplementation represents a biologically plausible adjunct that could support muscle energetics and function in this setting. However, this proposal is hypothesis-generating, and no randomized clinical tria

Glucagon-like peptide-1 receptor agonists across anti-inflammation.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) can ignite the downstream signal transduction in various tissues/organs due to the receptor expression across cell types, thus playing multifaceted biological roles in and beyond anti-diabetes. Besides metabolic benefits, anti-inflammatory pathology is emerging as an attractive mode to inspire expanding therapeutic utilities of GLP-1RAs, through the synergistic cellular/molecular mechanisms of modulating meta-inflammation or immuno-metabolism. Thus this article presents a trending profile of representative inflammation-mediated diseases whic
