Glucagon-Like Peptide-1 Receptor Agonist-Associated Hepatotoxicity.

PURPOSE: Glucagon-like peptide-1 receptor agonists (GLP-1RAs) have transformed the management of obesity, type 2 diabetes and metabolic dysfunction-associated steatohepatitis. Despite their numerous clinical benefits they are not without risk. Although the product labels contain multiple organ-specific warnings and precautions, the liver is not identified as a target organ of safety concern. Drug-induced liver injury (DILI) is a leading safety-related cause of pipeline attrition, boxed warnings and postmarketing drug withdrawal. This review explores GLP-1RA associated hepatotoxicity. METHODS:

Association of glucagon-like peptide-1 receptor-targeted imaging probe with in vivo glucagon-like peptide-1 receptor agonist glucose-lowering effects.

AIMS/INTRODUCTION: Glucagon-like peptide-1 receptor agonists (GLP-1RA) are used for treatment of type 2 diabetes mellitus worldwide. However, some patients do not respond well to the therapy, and caution must be taken for certain patients, including those with reduced insulin secretory capacity. Thus, it is clinically important to predict the efficacy of GLP-1RA therapy. GLP-1R-targeted imaging has recently emerged to visualize and quantify β-cells. We investigated whether GLP-1R-targeted imaging can predict the efficacy of GLP-1RA treatment. MATERIALS AND METHODS: We developed 111 Indium-lab

Glucagon-like peptide-1 receptor agonists across anti-inflammation.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) can ignite the downstream signal transduction in various tissues/organs due to the receptor expression across cell types, thus playing multifaceted biological roles in and beyond anti-diabetes. Besides metabolic benefits, anti-inflammatory pathology is emerging as an attractive mode to inspire expanding therapeutic utilities of GLP-1RAs, through the synergistic cellular/molecular mechanisms of modulating meta-inflammation or immuno-metabolism. Thus this article presents a trending profile of representative inflammation-mediated diseases whic

Glucagon-like peptide 1 receptor agonists in type 1 diabetes mellitus.

PURPOSE: The role of glucagon-like peptide 1 receptor agonists (GLP-1 RAs) in the treatment of type 1 diabetes mellitus (T1DM), including efficacy and safety evidence, is reviewed. SUMMARY: Currently approved treatment options for glycemic control in T1DM include insulin, which combats insulin deficiency but does not effectively target disease progression or alpha cell dysfunction; and pramlintide, whose use requires multiple daily doses and involves a high likelihood of gastrointestinal side effects. GLP-1 RAs have a unique mechanism of action in T1DM, addressing alpha cell dysfunction and t

Trends in glucagon-like peptide 1 receptor agonist prescribing patterns.

OBJECTIVE: Obesity affects more than 40% of US adults, increasing risks for cardiovascular disease and type 2 diabetes. Glucagon-like peptide 1 receptor agonists (GLP-1 RAs), initially indicated for diabetes, show promise in weight loss but face coverage issues, high costs, and premature prescribing from physicians. Research is needed to assess prescribing patterns, especially in patients without diabetes. STUDY DESIGN: We conducted a retrospective, population-based, observational study using the Merative MarketScan Commercial Database and the Merative MarketScan Medicare Supplemental Databas

Glucagon-like peptide-1 receptor agonists and muscle mass effects.

Glucagon-like peptide-1 receptor agonists (GLP-1 RAs) have transformed the management of obesity, demonstrating significant efficacy in inducing weight loss and improving metabolic parameters. However, emerging clinical and paraclinical evidence suggests that these agents may also contribute to an unintended reduction in skeletal muscle mass, potentially exacerbating or precipitating sarcopenic obesity, particularly in older or frail individuals with limited muscular reserves. This review critically examines current data on the effects of GLP-1 RAs on body composition, explores the underlying

Glucagon-Like Peptide-1 Receptor Agonists for Arthritis and Osteoarthritis.

Glucagon-like peptide-1 receptor agonists (GLP-1RAs) play a pivotal role in managing type 2 diabetes mellitus (T2DM), obesity, and other conditions, with proven benefits including, but not limited to, weight loss, glycemic control, and cardiovascular protection. However, their implications for arthritis are still debatable. GLP-1RAs may reduce systemic inflammation, alleviate joint stress through overall weight loss, and exhibit chondroprotective effects. Nevertheless, concerns such as augmented gout flares, joint pain, and indefinite long-term outcomes temper their promise. This editorial hig

Glucagon-Like Peptide-1 Receptor Agonist Treatment for Pediatric Obesity.

Obesity is a complex and retractable disease for which effective and durable treatments are elusive. Successful treatment of severe obesity with lifestyle modification therapy alone is highly unlikely, particularly for adolescents. Pharmacotherapy, if appropriately prescribed, can be an effective tool to use in conjunction with lifestyle modification therapy to achieve better weight loss outcomes. Only a few obesity medications have been evaluated in children and adolescents with results suggesting modest efficacy. However, a new pipeline of obesity drugs has been recently approved for use amo
