Identification and Quantification of Gluconic, Metasaccharinic, Glyceric, Formic, and Acetic Acids as Novel Glucose Degradation Products in Peritoneal Dialysis Fluids.

Peritoneal dialysis (PD) fluids often contain glucose as the osmotic agent. Heat sterilization ensures their microbiological safety, but the heat impact degrades glucose. The altered composition and reduced biocompatibility of the fluids limit the long-term use of PD. A key indicator of glucose degradation is a decrease in pH poststerilization suggesting that carboxylic glucose degradation products (carboxyl GDPs) were formed. The present study employed a precursor ion mass spectrometry screening using 3-nitrophenylhydrazine derivatization to investigate the presumed carboxylic products. Based

Glyceric Prodrug of Ursodeoxycholic Acid (UDCA): Novozym 435-Catalyzed Synthesis of UDCA-Monoglyceride.

Bile acids (BAs) are a family of steroids synthesized from cholesterol in the liver. Among bile acids, ursodeoxycholic acid (UDCA) is the drug of choice for treating primary biliary cirrhosis and dissolving cholesterol gallstones. The clinical effectiveness of UDCA includes its choleretic activity, the capability to inhibit hydrophobic bile acid absorption by the intestine under cholestatic conditions, reducing cholangiocyte injury, stimulation of impaired biliary output, and inhibition of hepatocyte apoptosis. Despite its clinical effectiveness, UDCA is poorly soluble in the gastro-duodeno-je

A method for determination of the absolute configuration of chiral glycerol residues in natural products using TEMPO oxidation and characterization of the glyceric acids formed.

A method has been developed for determination of the absolute configuration of glycerol residues in natural products. It is required that the glycerol moiety contain a primary nonsubstituted hydroxymethyl group or that such a group can be obtained by modification without racemization. The method employs TEMPO oxidation of the primary hydroxyl group, hydrolysis, butanolysis with chiral 2-butanol, and acetylation. The acetylated (+)-2-butyl esters of the glyceric acid formed by oxidation are analyzed by gas-liquid chromatography. The esterification can also be performed with other chiral alcohol

Chebulic acid prevents hepatic fibrosis induced by advanced glycation end-products in LX-2 cell by modulating Nrf2 translocation via ERK pathway.

Advanced glycation end-products (AGEs) are formed during normal aging, and at an accelerated rate in metabolic syndrome patients. Nonalcoholic steatohepatitis (NASH) can be caused by the AGEs in plasma, while glyceraldehyde-derived AGEs (glycer-AGEs) are significantly higher in the serum of NASH patients. In this study, we investigated the molecular mechanisms of chebulic acid, isolated from Terminalia chebula Retz., in the inhibition of glycer-AGEs induced production of reactive oxygen species (ROS) and collagen accumulation using the LX-2 cell line. Chebulic acid significantly inhibited the

D-glyceric aciduria.

Inherited metabolic diseases are a heterogeneous group of diseases caused by a punctual defect in cell metabolism, resulting in the accumulation of toxic intermediate metabolites or in the lack of important biomolecules for adequate cell functioning. D-glyceric aciduria is an inherited disease caused by a deficiency of glycerate 2-kinase activity, whose pathophysiological mechanisms remain unknown. The main clinical and neurological symptoms seen in affected patients include progressive encephalopathy, hypotonia, psychomotor and mental retardation, microcephaly, seizures, speech delay, metabol

A transport system for phosphoenolpyruvate, 2-phosphoglycerate, and 3-phosphoglycerate in Salmonella typhimurium.

Salmonella typhimurium strain LT-2 was found to utilize phosphoenolpyruvate, 2-phosphoglycerate, and 3-phosphoglycerate as sole sources of carbon and energy for growth, but Escherichia coli strains did not. The following evidence suggests that this growth difference was due to the presence in Salmonella cells of an inducible phosphoglycerate permease distinct from previously studied transport systems: (a) The ability of cells to take up 3-phospho[14-C]glycerate was induced by growth in the presence of phosphoenolpyruvate, 2-phosphoglycerate, or 3-phosphoglycerate, but not glycerate, alpha-glyc

Molecular cloning and functional expression of enolase from Trichinella spiralis.

The cDNA library was constructed from muscle larvae of Trichinella spiralis, and immunoscreened with sera from mice infected with T. spiralis. A cDNA clone, designated as TsENO, encoded 2-phospho-D-glycerate hydro-lyase (enolase) that catalyzed a reversible conversion of 2-phospho-D-glyceric acid (2PGA) to phosphoenolpyruvate (PEP) in the glycolytic pathway. The recombinant TsENO protein was produced in an Escherichia coli expression system. The recombinant full-length TsENO protein had no activity in the conversion of 2PGA to PEP, but gained enolase activity after cutting off the signal pepti

d-Glycerate kinase deficiency in a neuropediatric patient.

d-Glyceric aciduria (DGA) due to d-glycerate kinase deficiency (DGKD) is a rare autosomal-recessive inborn error of metabolism that is usually linked to the metabolism of fructose and serine. We describe a Moroccan patient with DGKD whose metabolic defect has been characterized by metabolite studies, sequencing of genomic DNA and by studies on the RNA level. Since birth the index patient presented with severe muscular hypotonia, joint hypermobility and tremor. Enantioselective analysis showed elevated d-glyceric acid in the urine of the patient, but not in that of his parents. DNA analysis rev

Structural characteristics of endogenous sugar acids and relations to feeding modulation.

Structural specificity among short-chain organic acids for effects on feeding behavior, blood glucose and insulin was investigated by infusion of 1 exogenous and 6 endogenous derivatives into the rat third cerebral ventricle. Glyceric acid (GEA) (1.0 mumol), 3,4-dihydroxybutanoic acid gamma-lactone (3,4-DB) and 3,4,5-trihydroxypentanoic acid gamma-lactone (3,4,5-TP) (2.50 mumol) decreased food intake for, at most, 24 h. These acids depressed the size of the first meal after infusion, but did not affect latency to the first meal, eating speed, drinking or ambulation. Infusion of 2,4-dihydroxybu

Precise Ga engineering of surface reaction energetics in Pd2Sn for enhanced alcohol electrooxidation.

Alcohol electrooxidation is an important anodic reaction in alkaline electrochemical energy-conversion systems, yet its performance is often limited by sluggish reaction kinetics and catalyst deactivation associated with strongly adsorbed intermediates. Herein, Ga was introduced into the Pd2Sn intermetallic to regulate its surface reaction energetics. A series of composition-controlled ternary Pd2(Sn1-xGax) intermetallic nanoparticles were synthesized through a ligand-regulated colloidal strategy. Systematic compositional optimization identified Pd2(Sn0.87Ga0.13) as the optimal catalyst, which

Severe child form of primary hyperoxaluria type 2 - a case report revealing consequence of GRHPR deficiency on metabolism.

BACKGROUND: Primary hyperoxaluria type 2 is a rare monogenic disorder inherited in an autosomal recessive pattern. It results from the absence of the enzyme glyoxylate reductase/hydroxypyruvate reductase (GRHPR). As a consequence of deficient enzyme activity, excessive amounts of oxalate and L-glycerate are excreted in the urine, and are a source for the formation of calcium oxalate stones that result in recurrent nephrolithiasis and less frequently nephrocalcinosis. CASE PRESENTATION: We report a case of a 10-month-old patient diagnosed with urolithiasis. Screening of inborn errors of metabo

Time-resolved targeted metabolomics shows an abrupt switch from Calvin-Benson-Bassham cycle to tricarboxylic acid cycle when the light is turned off.

In leaves, major CO₂ fluxes alternate between fixation by the Calvin-Benson-Bassham (CBB) cycle during light and release by the tricarboxylic acid (TCA) cycle in darkness. The speed at which leaf metabolism transitions between these pathways likely influences plant tolerance to fluctuating light conditions. To investigate these rapid metabolic shifts, we exposed leaves to ¹³CO₂ for 20 min to establish a quasi-steady state before abruptly turning off the light while maintaining ¹³CO₂ feeding. Within 10 s of dark transition, 3-phosphoglycerate levels rose significantly while most other CBB cycle

A recombinant bisphosphoglycerate mutase variant with acid phosphatase homology degrades 2,3-diphosphoglycerate.

To date no definite and undisputed treatment has been found for sickle cell anemia, which is characterized by polymerization of a deoxygenated hemoglobin mutant (HbS) giving rise to deformed erythrocytes and vasoocclusive complications. Since the erythrocyte glycerate 2,3-bisphosphate (2,3-DPG) has been shown to facilitate this polymerization, one therapeutic approach would be to decrease the intraerythrocytic level of 2,3-DPG by increasing the phosphatase activity of the bisphosphoglycerate mutase (BPGM; 3-phospho-D-glycerate 1,2-phosphomutase, EC 5.4.2.4). For this purpose, we have investiga

Biotransformations of glafenine in the rat and in man.

The biotransformations of a therapeutic dose of the non-narcotic analgesic, glafenine, have been studied in the rat and in man. In the rat, the ester bond is extensively hydrolysed to give glafenic acid which is the major metabolite excreted in bile and in urine. Two minor pathways have been identified one leading by hydroxylation of the benzene ring of glafenine or glafenic acid in para of the amino-substituent to the corresponding phenols, the other, by oxidation of the quinoline nitrogen of glafenic acid, to its N-oxide. In vivo this N-oxide is partly reduced into the parent compound. Hydro

Molecular responses of watermelon seedlings to MgO nanoparticles: transcriptome and metabolome insights.

MgO nanoparticles (MgO NPs) exhibit considerable potential in regulating plant development. This study firstly demonstrates that MgO NPs at 100-200 mg/L enhance the growth of watermelon (Citrullus lanatus) seedlings, resulting in 18.7-32.8 % and 23.8-46.5 % increases in the over ground fresh and dry weights, respectively. Significant changes in the accumulation of multiple metabolites were identified after MgO NPs treatment, such as carboxylic acids and derivatives, benzene and substituted derivatives, fatty acyls, and organooxygen compounds. MgO NPs enhance watermelon growth primarily by upre

Glycolytic Metabolite 3-Phosphoglycerate Induced by Inflammation Inhibits Chondrocyte Survival.

The reduction of chondrocytes is an important pathological manifestation in the cartilage degeneration, and the abnormal metabolism of chondrocytes triggered by inflammation is the key reason for the inhibition of chondrocyte survival. The enhancement of glycolysis is an important feature of chondrocyte metabolism in inflammatory environments, but the effects of metabolic enzymes and metabolites on chondrocyte survival in this process are still unclear. In this study, we used transcriptomics to analyze the expression of glycolytic metabolic enzymes in condylar chondrocytes under inflammatory e

Sarcoidosis resolvers and progressors demonstrate distinct systemic metabolomic profiles.

Sarcoidosis is an idiopathic syndrome with striking disparities in clinical outcome. We have previously identified immune distinctions between sarcoidosis clinical cohorts that either resolve or progress. A significantly higher percentage of Programmed Death 1 (PD-1)+Th17 cells was present in sarcoidosis and IPF subjects experiencing disease progression. Metabolites, or bioactive compounds, have been shown to interact with immune cells, such as Th17 cells by regulating cellular processes such as proliferation, signaling and differentiation.
