Glycopeptide antibiotic resistance genes in glycopeptide-producing organisms.

The mechanism of high-level resistance to vancomycin in enterococci consists of the synthesis of peptidoglycan terminating in D-alanyl-D-lactate instead of the usual D-alanyl-D-alanine. This alternate cell wall biosynthesis pathway is ensured by the collective actions of three enzymes: VanH, VanA, and VanX. The origin of this resistance mechanism is unknown. We have cloned three genes encoding homologs of VanH, VanA, and VanX from two organisms which produce glycopeptide antibiotics: the A47934 producer Streptomyces toyocaensis NRRL 15009 and the vancomycin producer Amycolatopsis orientalis C3

Wolfberry glycopeptides alleviate levodopa-induced dyskinesia.

Levodopa-induced dyskinesia is a prevalent and almost inevitable complication of long-term levodopa therapy in Parkinson's disease, significantly impairing patients' quality of life. Despite its clinical significance, levodopa-induced dyskinesia remains insufficiently understood, and effective therapeutic strategies are still limited. Wolfberry glycopeptides, bioactive components extracted from Lycium barbarum L. have recently been widely reported to possess neuroimmune activities. In this study, levodopa-induced dyskinesia was induced through chronic levodopa administration in multiple Parkin

Glycopeptide resistance in enterococci.

Glycopeptide resistance in enterococci results from the production of peptidoglycan precursors with low affinity for these antibiotics. The mobility of the resistance genes by transposition and conjugation and the ability of the resistance proteins to interfere with synthesis of normal precursors in different hosts indicate that dissemination into other bacterial species should be anticipated.

Resistance of enterococci to aminoglycosides and glycopeptides.

High-level resistance to aminoglycosides in enterococci often is mediated by aminoglycoside-modifying enzymes, and the corresponding genes generally are located on self-transferable plasmids. These enzymes are similar to those in staphylococci but differ from the modifying enzymes of gram-negative bacteria. Three classes of enzymes are distinguished, depending upon the reaction catalyzed. All but amikacin and netilmicin confer high-level resistance to the antibiotics that are modified in vitro. However, the synergistic activity of these last two antibiotics in combination with beta-lactam agen

Antiendotoxin activity of antimicrobial peptides and glycopeptides.

An animal study was performed to investigate the efficacy of two glycopeptides and two cationic peptides in the prevention of lethality in a septic shock rat model. Adult Wistar rats were given an intraperitoneal injection of 2x10(10) CFU of Escherichia coli ATCC 25922, with the exception of an uninfected control group (C0). Animals were randomized to receive, immediately after bacterial challenge, intraperitoneally isotonic sodium chloride solution (control group C1), 3 mg/Kg teicoplanin (group 1), 7 mg/Kg vancomycin (group 2), 1 mg/Kg colistin (group 3), 1 mg/Kg buforin II (group 4), or 60 m

Glycopeptides in the treatment of staphylococcal infections.

Gram-positive bacteria are rapidly becoming the most important pathogens in nosocomial infections. In recent years, attention and concern have been focused on the gram-positive bacteria, Staphylococcus aureus, Staphylococcus epidermidis and Enterococcus faecalis. These microorganisms are well equipped to exert their pathogenic effects and to display virulence. Treatment of severe infections caused by gram-positive bacteria remains difficult because of the increase in infections caused by methicillin-resistant staphylococci, and this has renewed interest in the glycopeptide antibiotics, vancomy

Glycopeptides in pus of acute pleurisy patients.

Glycopeptides were isolated from tryptic digests of pus from acute pleurisy patients. The hexose, hexosamine, and sialic acid contents rose over the first 2-4 days after admission to the hospital, continued at high levels for 5-8 hospital days, and fell to low levels after 9 hospital days. The course of duration after admission to the hospital was divided into three stages: 1-4 days after admission to the hospital, 5-8 hospital days, and 9-21 hospital days. Materials corresponding to these three stages were then collected for fractionation by DEAE-cellulose column chromatography. Fractionation

Glycopeptide Hypersensitivity and Adverse Reactions.

Glycopeptides, such as vancomycin and teicoplanin, are primarily used in the treatment of methicillin-resistant Staphylococcus aureus (MRSA) infections, such as cellulitis, endocarditis, meningitis, pneumonia, and septicemia, and are some of the most commonly prescribed parenteral antimicrobials. Parenteral glycopeptides are first-line therapy for severe MRSA infections; however, oral vancomycin is used as a first-line treatment of Clostridioides difficile infections. Also, we currently have the longer-acting lipoglycopeptides, such as dalbavancin, oritavancin, and telavancin to our armamentar

Safety and efficacy of glycopeptide antibiotics.

It would be difficult to envision the practice of infectious diseases over the past 20 years without the availability of the glycopeptide antibiotics. The two agents currently in clinical use, vancomycin and teicoplanin, have proven remarkably versatile in many common applications. Several attributes of these agents account for this favourable profile: (i) their broad spectrum of activity against Gram-positive bacteria, including strains resistant to many other antimicrobials; (ii) their favourable pharmacokinetic properties that allow the once- or twice-daily dosing regimens that have made ou

Teicoplanin: an investigational glycopeptide antibiotic.

The chemistry, mechanism of action, antimicrobial spectrum, pharmacokinetics, adverse effects, and clinical uses of teicoplanin are reviewed. Teicoplanin, a novel glycopeptide that is similar to vancomycin, was isolated in the mid-1970s. A fermentation product of Actinoplanes teicomyceticus, teicoplanin is a structurally complex compound made up of six fatty-acid components attached to a common aglycone. Teicoplanin's mechanism of action, like that of vancomycin, is inhibition of cell-wall biosynthesis. In vitro activity is comparable to that of vancomycin and includes staphylococci, streptoco

Experimental study on the efficacy of combinations of glycopeptides and beta-lactams against Staphylococcus aureus with reduced susceptibility to glycopeptides.

OBJECTIVES: The combination of glycopeptides and beta-lactams has been proposed as an alternative therapy against infections due to Staphylococcus aureus with reduced susceptibility to glycopeptides, though its role is still controversial. Our aim was to evaluate the efficacy (decrease in bacterial concentration after 24 h therapy) of these combinations both in vitro and in vivo. METHODS: Four strains of S. aureus with different glycopeptide susceptibility (MICs of vancomycin from 1 to 8 mg/L) were used. In vitro experiments were performed by means of time-kill curves while we used the mouse

Glycopeptide vs. non-glycopeptide antibiotics for prophylaxis of surgical site infections: a systematic review.

BACKGROUND: Patients receive prophylactic antibiotics against surgical site infections (SSIs) before or during many procedures. Glycopeptide antibiotics are effective against most strains of methicillin-resistant Staphylococcus aureus (MRSA), but their wider use risks increasing resistance. Our objective was to review the evidence for clinical effectiveness that might help to determine whether there is a threshold of MRSA prevalence at which switching from non-glycopeptide to glycopeptide antibiotic prophylaxis might be justified. METHODS: We performed a systematic review of randomized trials

A systematic review and economic model of switching from non-glycopeptide to glycopeptide antibiotic prophylaxis for surgery.

OBJECTIVES: To determine whether there is a level of methicillin-resistant Staphylococcus aureus (MRSA) prevalence at which a switch from non-glycopeptide to glycopeptide antibiotics for routine prophylaxis is indicated in surgical environments with a high risk of MRSA infection. DATA SOURCES: Major electronic databases were searched up to September 2005. REVIEW METHODS: The effectiveness review included controlled clinical trials comparing a glycopeptide with an alternative antibiotic regimen that reported effectiveness and/or adverse events. Controlled observational studies were also inclu

Glycopeptides as a Tool for Early Detection of Cancer.

Protein glycosylation, as one of the most common and complex posttranslational modifications, plays an important role in many biological processes. Along with the intensive progress in MS techniques and development of glycan search tools and databases, glycoproteomics has become a popular subject of studies. The possibility of simultaneous identification of amino acid sequence, glycosylation sites, and glycan composition enabled the monitoring of changes in glycosylation patterns in various pathological states. In this issue, Saraswat et al. describe MS-based investigations of glycopeptide cha

Chemoenzymatic synthesis of sulfated O-glycopeptides from human CD34.

O-Glycosylation is a key modulator of CD34 biofunction. To investigate the distinct roles of different glycoform modifications, we report an efficient chemoenzymatic strategy to rapidly assemble a library of CD34 O-glycopeptides bearing sulfated and sialylated glycan variants, enabling the elucidation of how specific glycan modifications govern molecular recognition events.

Gallium-68 Labeled Cyclic Glycopeptides for Liver Function Imaging.

Asialoglycoprotein receptor (ASGPR) is specifically expressed on hepatocytes, making it an attractive target for imaging functional liver reserve. Clinically used ASGPR-targeting radiopharmaceuticals, such as [99mTc]Tc-GSA, are widely employed for this purpose. Here, we report a series of cyclic glycopeptides and evaluate their performance in hepatic imaging in healthy mice. Using a solid-phase peptide synthesis approach, we readily accessed low-molecular-weight cyclic glycopeptides. Cellular uptake studies in hepatocytes identified IPM-G1005 as a lead compound with pronounced uptake. Radiolab

Pursuit of next-generation glycopeptides: a journey with vancomycin.

Vancomycin, a blockbuster antibiotic of the glycopeptide class, has been a life-saving therapeutic against multidrug-resistant Gram-positive infections. The emergence of glycopeptide resistance has however enunciated the need to develop credible alternatives with potent activity against vancomycin-resistant bacteria. Medicinal chemistry has responded to this challenge through various strategies, one of them being the development of semisynthetic analogues. Many groups, including ours, have been contributing towards the development of semisynthetic vancomycin analogues to tackle vancomycin-resi
